    Validity and measurement invariance of the Unified Multidimensional Calling Scale (UMCS): A three-wave survey study

    The accumulation of scientific knowledge on calling is limited by the absence of a common theoretical and measurement framework. Many different models of calling have been proposed, and we do not know how much research results that refer to a specific model are generalizable to different theoretical accounts of calling. In this article, we investigate whether two leading models of calling tackle the same construct. The two models were merged into a comprehensive framework that measures calling across seven facets: Passion, Purposefulness, Sacrifice, Pervasiveness, Prosocial Orientation, Transcendent Summons, and Identity. We then developed the Unified Multidimensional Calling Scale (UMCS) drawing from previous published items. Across two surveys involving college students (N = 5886) and adult employees (N = 205) the UMCS was proved to be valid and reliable. We also observed that the UMCS is invariant across time and calling domains. Finally, we found that facets of calling have very different relationships with outcomes and concurrent measures, suggesting that results obtained with a smaller set of facets are not generalizable to the higher-order construct of calling or to a different model that does not share the same facets. \ua9 2018 Vianello et al.     A Universal Kriging predictor for spatially dependent functional data of a Hilbert Space

    We address the problem of predicting spatially dependent functional data belonging to a Hilbert space, with a Functional Data Analysis approach. Having defined new global measures of spatial variability for functional random processes, we derive a Universal Kriging predictor for functional data. Consistently with the new established theoretical results, we develop a two-step procedure for predicting georeferenced functional data: first model selection and estimation of the spatial mean (drift), then Universal Kriging prediction on the basis of the identified model.     Aberrant ribonucleotide incorporation and multiple deletions in mitochondrial DNA of the murine MPV17 disease model

    All DNA polymerases misincorporate ribonucleotides despite their preference for deoxyribonucleotides, and analysis of cultured cells indicates that mammalian mitochondrial DNA (mtDNA) tolerates such replication errors. However, it is not clear to what extent misincorporation occurs in tissues, or whether this plays a role in human disease. Here, we show that mtDNA of solid tissues contains many more embedded ribonucleotides than that of cultured cells, consistent with the high ratio of ribonucleotide to deoxynucleotide triphosphates in tissues, and that riboadenosines account for three-quarters of them. The pattern of embedded ribonucleotides changes in a mouse model of Mpv17 deficiency, which displays a marked increase in rGMPs in mtDNA. However, while the mitochondrial dGTP is low in the Mpv17−/− liver, the brain shows no change in the overall dGTP pool, leading us to suggest that Mpv17 determines the local concentration or quality of dGTP. Embedded rGMPs are expected to distort the mtDNA and impede its replication, and elevated rGMP incorporation is associated with early-onset mtDNA depletion in liver and late-onset multiple deletions in brain of Mpv17−/− mice. These findings suggest aberrant ribonucleotide incorporation is a primary mtDNA abnormality that can result in pathology
