    Quantification of eDNA to map the distribution of cold-water coral reefs

    An effective management of vulnerable marine ecosystems is dependent on thorough knowledge of their location. Multibeam bathymetric mapping and targeted remotely operated vehicle (ROV) surveys are currently used to map areas impacted by industrial activities when vulnerable species are expected. However, multibeam bathymetric mapping is not always a possibility and surveying large areas using ROVs is expensive. Here, we developed a species-specific eDNA assay targeting a 178 bp fragment in the control region of the mitochondrial DNA of the cold-water coral (CWC) Lophelia pertusa. The aim was to test if concentrations of L. pertusa eDNA in seawater, determined using droplet digital PCR (ddPCR) technology, could be used to assess the broad scale distribution of CWCs in a region, to supplement multibeam mapping and direct targeted ROV surveys. Our assay successfully amplified L. pertusa DNA from seawater. In laboratory we documented an exponential decay rate of the targeted DNA fragment and a linear correlation between coral biomass and eDNA concentrations in flow through microcosms. The ability of the method to detect CWC reefs in situ was tested in the fjords south of Bergen, Norway, where such reefs are common. We tested five sites with, and five sites without, known reefs. Lophelia pertusa eDNA was detected in all 10 sites. However, concentrations were elevated by 5 to 10 times in water sampled off the two large reefs growing on vertical surfaces. Water sampled 10 m above CWC reefs growing on the flat seabed did not produce an equally clear eDNA signal, nor did single CWC colonies growing on vertical surfaces. Treating the eDNA as a passive particle with no active vertical or horizontal movement, we successfully modeled the dispersal of eDNA from the known CWC reefs in the region and achieved a good fit with measured eDNA concentrations. In all, our study demonstrated a great potential for eDNA measurements as a cost-efficient tool for a rapid screening of the broad scale distribution of CWC reefs growing on vertical surfaces (so called wall reefs) that cannot be imaged using traditional ship mounted downward looking multibeam echo-sounders and difficult to detect using ROVs alone.publishedVersio

    Family Planning Decisions, Perceptions and Gender Dynamics among Couples in Mwanza, Tanzania: A Qualitative Study.

    Contraceptive use is low in developing countries which are still largely driven by male dominated culture and patriarchal values. This study explored family planning (FP) decisions, perceptions and gender dynamics among couples in Mwanza region of Tanzania. Twelve focus group discussions and six in-depth interviews were used to collect information from married or cohabiting males and females aged 18-49. The participants were purposively selected. Qualitative methods were used to explore family planning decisions, perceptions and gender dynamics among couples. A guide with questions related to family planning perceptions, decisions and gender dynamics was used. The discussions and interviews were tape-recorded, transcribed verbatim and analyzed manually and subjected to content analysis. Four themes emerged during the study. First, "risks and costs" which refer to the side effects of FP methods and the treatment of side -effects as well as the costs inherit in being labeled as an unfaithful spouse. Second, "male involvement" as men showed little interest in participating in family planning issues. However, the same men were mentioned as key decision-makers even on the number of children a couple should have and the child spacing of these children. Third, "gender relations and communication" as participants indicated that few women participated in decision-making on family planning and the number of children to have. Fourth, "urban-rural differences", life in rural favoring having more children than urban areas therefore, the value of children depended on the place of residence. Family Planning programs should adapt the promotion of communication as well as joint decision-making on FP among couples as a strategy aimed at enhancing FP use

    Caribbean Corals in Crisis: Record Thermal Stress, Bleaching, and Mortality in 2005

    BACKGROUND The rising temperature of the world's oceans has become a major threat to coral reefs globally as the severity and frequency of mass coral bleaching and mortality events increase. In 2005, high ocean temperatures in the tropical Atlantic and Caribbean resulted in the most severe bleaching event ever recorded in the basin. METHODOLOGY/PRINCIPAL FINDINGS Satellite-based tools provided warnings for coral reef managers and scientists, guiding both the timing and location of researchers' field observations as anomalously warm conditions developed and spread across the greater Caribbean region from June to October 2005. Field surveys of bleaching and mortality exceeded prior efforts in detail and extent, and provided a new standard for documenting the effects of bleaching and for testing nowcast and forecast products. Collaborators from 22 countries undertook the most comprehensive documentation of basin-scale bleaching to date and found that over 80% of corals bleached and over 40% died at many sites. The most severe bleaching coincided with waters nearest a western Atlantic warm pool that was centered off the northern end of the Lesser Antilles. CONCLUSIONS/SIGNIFICANCE Thermal stress during the 2005 event exceeded any observed from the Caribbean in the prior 20 years, and regionally-averaged temperatures were the warmest in over 150 years. Comparison of satellite data against field surveys demonstrated a significant predictive relationship between accumulated heat stress (measured using NOAA Coral Reef Watch's Degree Heating Weeks) and bleaching intensity. This severe, widespread bleaching and mortality will undoubtedly have long-term consequences for reef ecosystems and suggests a troubled future for tropical marine ecosystems under a warming climate.This work was partially supported by salaries from the NOAA Coral Reef Conservation Program to the NOAA Coral Reef Conservation Program authors.     Marine litter harms marine life, human wellbeing and the provision of clean seafood. In the Barents Sea, a high portion of this source of pollution derives from fisheries. However, there remains a knowledge gap between fishers' attitudes towards litter and their practices when it comes to marine litter onboard their vessels and in harbors. By applying the conceptual lenses of social institutions, this study aims to explore the institutionalization of new practices among fishers to handle marine waste, as well as possible measures to reduce it. Empirical data derives from 21 qualitative interviews with fishers from Northern Norway. The data indicate that there has been a shift in fishers' attitude, partly because of media focus on this environmental issue and awareness campaigns from fishers’ organizations. However, increased sensitivity to the problem has not completely translated into effective practices to address it. The institutionalization of desirable values and practices is hindered by the lack of infrastructure, institutional support and appropriate practical arrangements in harbors. We conclude that increased awareness has limited impact (and can potentially be invalidated), if not accompanied by a system designed to support environmentally adequate choices. Progress on addressing the problem of marine litter depends on the institutionalization of environmental practices, and these are contingent upon an integrated approach between operations at sea and on land.publishedVersio

    Exploring Mechanisms of Change in a Dyadic Relationship Intervention for Siblings in Foster Care

    There is increasing recognition of the potential for sibling relationships to promote the well‐being of youth in out‐of‐home care. Efficacious interventions now exist to strengthen the sibling relationships for youth in out‐of‐home care, yet the processes through which these interventions work to enhance sibling relationships remains largely speculative. The current study therefore aimed to identify the critical components of an efficacious dyadic relationship enhancement intervention for siblings in foster care through a secondary analysis of fidelity of implementation and trial outcome data. Data for 168 youth from the treatment condition of the Supporting Siblings in Foster Care study were analysed. Fidelity of implementation was assessed across seven intervention domains: Sibs 4 Life, Positive/Sib Thinking, Activity Planning, Problem Solving, Cooperation, Managing Feelings and Adult Allies. Trial outcome data were drawn from efficacy test results and included a multi‐agent construct of sibling relationship quality. Descriptive statistics detailed intervention implementation, and hierarchical linear models examined associations between intervention coverage, comprehension, and engagement, and 18‐month improvements to the sibling relationship. Results indicate high fidelity to implementation, and Positive/Sib Thinking was the primary intervention domain associated with sibling relationship improvements. Considerations for future mechanisms‐based intervention research with siblings in out‐of‐home care are provided

    Generation of a Biobank From Two Adult Thoroughbred Stallions for the Functional Annotation of Animal Genomes Initiative

    Following the successful creation of a biobank from two adult Thoroughbred mares, this study aimed to recapitulate sample collection in two adult Thoroughbred stallions as part of the Functional Annotation of the Animal Genome (FAANG) initiative. Both stallions underwent thorough physical, lameness, neurologic, and ophthalmic (including electroretinography) examinations prior to humane euthanasia. Epididymal sperm was recovered from both stallions immediately postmortem and cryopreserved. Aseptically collected full thickness skin biopsies were used to isolate, culture and cryopreserve dermal fibroblasts. Serum, plasma, cerebrospinal fluid, urine, and gastrointestinal content from various locations were collected and cryopreserved. Under guidance of a board-certified veterinary anatomic pathologist, 102 representative tissue samples were collected from both horses. Whole tissue samples were flash-frozen and prioritized tissues had nuclei isolated and cryopreserved. Spatially contemporaneous samples of each tissue were submitted for histologic examination. Antemortem and gross pathologic examination revealed mild abnormalities in both stallions. One stallion (ECA_UCD_AH3) had unilateral thoracic limb lameness and bilateral chorioretinal scars. The second stallion (ECA_UCD_AH4) had subtle symmetrical pelvic limb ataxia, symmetrical prostatomegally, and moderate gastrointestinal nematodiasis. DNA from each was whole-genome sequenced and genotyped using the GGP Equine 70K SNP array. The genomic resources and banked biological samples from these animals augments the existing resource available to the equine genomics community. Importantly we may now improve the resolution of tissue-specific gene regulation as affected by sex, as well as add sex-specific tissues and gametes

    Vacation (after-) effects on employee health and well-being, and the role of vacation activities, experiences and sleep

However, the recovery-potential of relatively long vacations and the underlying processes have been disregarded. Therefore, our study focused on vacations longer than 14 days and on the psychological processes associated with such a long respite from work. In the present study, we investigated (1) how health and well-being (H&W) develop during and after a long summer vacation, (2) whether changes in H&W during and after vacation relate to vacation activities and experiences and (3) whether changes in H&W during and after vacation relate to sleep. Fifty-four employees reported their H&W before, three or four times during and five times after vacation. Vacations lasted 23 days on average. Information on vacation experiences, work-related activities and sleep was collected during vacation. Vacation activities were assessed immediately after vacation. H&W increased quickly during vacation, peaked on the eighth vacation day and had rapidly returned to baseline level within the first week of work resumption. Vacation duration and most vacation activities were only weakly associated with H&W changes during and after vacation. Engagement in passive activities, savoring, pleasure derived from activities, relaxation, control and sleep showed strong relations with improved H&W during and to a lesser degree after vacation. In conclusion, H&W improved during long summer vacations, but this positive effect was short-lived. Vacation experiences, especially pleasure, relaxation, savoring and control, seem to be especially important for the strength and persistence of vacation (after-) effects
