ANALISIS KONFLIK ANTARA KONTRAKTOR DENGAN PEKERJA DALAM PROYEK KONSTRUKSI DI DILI, TIMOR - LESTE
Pada proyek konstruksi, konflik merupakan hal yang tidak dapat dihindari
akibat sifat dinamis dari lingkungan proyeknya. Konflik tersebut harus
diselesaikan dengan cara yang tepat untuk meminimalkan pengaruh buruknya
terhadap usaha-usaha pencapaian sasaran proyek. Oleh karenanya, identifikasi
terhadap konflik yang berpotensi terjadi menjadi sangat penting untuk dilakukan
guna pengelolaan konflik secara efektif.
Penelitian ini dilakukan dengan membuat kuesioner yang berisi tentang
parameter penyebab konflik, solusi dari konflik, dan akibat terjadinya konflik
terhadap organisasi. Kuesioner ini diberikan kepada kontraktor dan pekerja dalam
proyek konstruksi yang sedang berjalan di Dili, Timor-Leste. Data kemudian
diolah dan dianalisis dengan nilai mean, standar deviasi dan uji-t.
Hasil analisis menunjukkan bahwa faktor yang perlu diperhatikan pada
penyebab konflik menurut kontraktor dan pekerja adalah sama yaitu kondisi alat
kerja yang sering rusak dengan nilai mean 3,13 dan 3,35 dengan hasil uji T sig 2-
tailed hitung 0,000 < 0,05 sehingga tolak H๐ dan terdapat perbedaan dari
penyebab konflik. Sedangkan faktor yang harus diperhatikan dalam solusi dari
konflik menurut kontraktor dan pekerja adalah sama yaitu pembagian kerja
disesuaikan dengan keahlian tiap pekerja dengan nilai mean 4,70 dan 4,12 dengan
hasil Uji T signifikansi 2-tailed hitung 0,010 < 0,05 sehingga tolak H๐ dan
terdapat perbedaan dari solusi konflik. Dan akibat terjadinya konflik terhadap
organisasi menurut kontraktor adalah bersemangat menyelesaikan setiap
pekerjaan yang diberikan dan menurut pekerja adalah dapat menghasilkan ide-ide
yang lebih baik. Pada analisis uji-t, nilai signifikan t yang didapatkan hasilnya
0,311 > 0,05 sehingga terima H๐ dan persepsi responden tidak ada perbedaan
dalam kategori akibat terjadinya konflik terhadap organisasi
Cultural intelligence: Measuring the CQ level in a Portuguese university
The objective of this thesis is to discover the cultural intelligence (CQ) level of ISCTE professors and their cross-cultural effectiveness through the comparison and validation of two constructs ICK & ECF two-factor model and the four-factor Cultural Intelligence Scale (CQS) using a self-report survey, involving Metacognitive, Cognitive, Motivational and Behavioral aspects. The respondents are professors in the University Institute of Lisbon – ISCTE-IUL, in Portugal. That is, professors with foreign experience and contact frequency with other cultures.
The understanding of cultural intelligence (CQ), which according to Ang & Van Dyne (2008), and Earley & Ang (2003) is the capability to function effectively in intercultural contexts, is the core of this work. Therefore, one may find the theoretical part written chronologically with selected references by the researcher to build the conceptual framework. To support the empirical research a quantitative methodological analysis using SPPS was performed. Farther, the results are analyzed to identify more characteristics of the sample.
Finally, findings shows that the original CQ model was the best one to analyze the data and the alternative model could not been validated. Also, surprisingly the time spent abroad and cultural intelligence do not have a significant relationship. This study delivered the cultural intelligence level and effectiveness of the university professor from ISCTE-IUL.O objetivo desta dissertação é conhecer o nível de inteligência cultural (CQ) dos professores do ISCTE e a sua eficácia intercultural por meio da comparação e validação de dois constructos, o modelo ICK & ECF de dois fatores e da Escala de Inteligência Cultural (CQS) de quatro fatores por meio de uma pesquisa de autorrelato, envolvendo aspetos Metacognitivos, Cognitivos, Motivacionais e Comportamentais. Os respondentes são docentes do Instituto Universitário de Lisboa - ISCTE-IUL, em Portugal, ou seja, docentes com experiência estrangeira e frequência de contacto com outras culturas.
A compreensão da inteligência cultural (CQ), que segundo Ang & Van Dyne (2008), e Earley & Ang (2003) é a capacidade de funcionar eficazmente em contextos interculturais, é o cerne deste trabalho. Portanto, pode-se encontrar a parte teórica escrita cronologicamente com referências selecionadas pelo pesquisador para a construção do quadro conceitual. Para dar suporte a pesquisa empírica foi realizada uma análise metodológica quantitativa utilizando o SPPS. Em seguida, os resultados foram analisados entre si para identificar mais características da amostra.
Finalmente, os resultados mostram que o constructo original foi o melhor para analisar os dados e o modelo alternativo não pôde ser validado. Além disso, surpreendentemente, o tempo passado no exterior e a inteligência cultural não têm uma relação significativa. MensSana: Design of a mental well-being self-report interface for shop floor workers
A ascensão da Indústria 4.0 trouxe consigo novas tecnologias e oportunidades que estão a mudar a natureza do trabalho, especialmente em ambientes de chão de fábrica. No entanto, essas mudanças também trouxeram novos desafios para os trabalhadores, incluindo desafios na sua saúde mental. Estes trabalhadores, em particular, enfrentam no seu trabalho estressores físicos e mentais que podem afetar seu bem-estar geral, apesar dos esforços da Indústria 4.0. O conceito de Operador 4.0 na Indústria 4.0 introduz muitos operadores, como o Operador Saudável, que enfatiza a centralidade no ser humano e visa melhorar a eficiência e o bem-estar do trabalhador por meio de tecnologias avançadas e análise de dados.
Esta tese propõe o desenvolvimento de uma ferramenta protótipo, co-criada e validada no contexto da Indústria 4.0 para medir métricas do trabalhador e do local de trabalho, criando uma imagem holística do trabalhador, sua competência e bem-estar, alinhado ao conceito de um trabalhador "mais saudável" de Romero et al. Essas informações são devolvidas ao trabalhador e apresentadas de maneira legível e compreensível para identificar tendências e informar decisões futuras relacionadas ao trabalho e bem-estar.The rise of Industry 4.0 has brought about new technologies and opportunities that are changing the nature of work, particularly in factory floor settings. However, these changes have also brought about new challenges for workers, including mental health issues. Shop floor workers, in particular, face physical and mental stressors in their work that can impact their overall well-being, despite Industry 4.0 efforts. The Operator 4.0 concept in Industry 4.0 introduces a lot of operators like the Healthy Operator that emphasises human-centricity and aims to improve worker efficiency and well-being through advanced technologies and data analytics.
This thesis proposes the development of a prototype tool co-created and validated in the context of Industry 4.0 to measure metrics from the worker and the workplace, creating a holistic picture of the worker, their competence and well-being in line with Romero's et al. concept of a "healthier" worker. This information is returned to the worker and presented in a readable and understandable manner to identify trends and inform future decisions concerning their work and well-being
Classification of skin tumours through the analysis of unconstrained images
Skin cancer is the most frequent malignant neoplasm for Caucasian individuals. According to the Skin Cancer Foundation, the incidence of melanoma, the most malignant of skin tumours, and resultant mortality, have increased exponentially during the past 30 years, and continues to grow. [1]. Although often intractable in advanced stages, skin cancer in general and melanoma in particular, if detected in an early stage, can achieve cure ratios of over 95% [1,55].
Early screening of the lesions is, therefore, crucial, if a cure is to be achieved.
Most skin lesions classification systems rely on a human expert supported dermatoscopy, which is an enhanced and zoomed photograph of the lesion zone. Nevertheless and although contrary claims exist, as far as is known by the author, classification results are currently rather inaccurate and need to be verified through a laboratory analysis of a piece of the lesion’s tissue.
The aim of this research was to design and implement a system that was able to automatically classify skin spots as inoffensive or dangerous, with a small margin of error; if possible, with higher accuracy than the results achieved normally by a human expert and certainly better than any existing automatic system.
The system described in this thesis meets these criteria. It is able to capture an unconstrained image of the affected skin area and extract a set of relevant features that may lead to, and be representative of, the four main classification characteristics of skin lesions: Asymmetry; Border; Colour; and Diameter.
These relevant features are then evaluated either through a Bayesian statistical process - both a simple k-Nearest Neighbour as well as a Fuzzy k-Nearest Neighbour classifier - a Support Vector Machine and an Artificial Neural Network in order to classify the skin spot as either being a Melanoma or not.
The characteristics selected and used through all this work are, to the author’s knowledge, combined in an innovative manner. Rather than simply selecting absolute values from the images characteristics, those numbers were combined into ratios, providing a much greater independence from environment conditions during the process of image capture.
Along this work, image gathering became one of the most challenging activities. In fact several of the initially potential sources failed and so, the author had to use all the pictures he could find, namely on the Internet. This limited the test set to 136 images, only. Nevertheless, the process results were excellent.
The algorithms developed were implemented into a fully working system which was extensively tested. It gives a correct classification of between 76% and 92% – depending on the percentage of pictures used to train the system. In particular, the system gave no false negatives. This is crucial, since a system which gave false negatives may deter a patient from seeking further treatment with a disastrous outcome. These results are achieved by detecting precise edges for every lesion image, extracting features considered relevant according to the giving different weights to the various extracted features and submitting these values to six classification algorithms – k-Nearest Neighbour, Fuzzy k-Nearest Neighbour, Naïve Bayes, Tree Augmented Naïve Bayes, Support Vector Machine and Multilayer Perceptron - in order to determine the most reliable combined process. Training was carried out in a supervised way – all the lesions were previously classified by an expert on the field before being subject to the scrutiny of the system.
The author is convinced that the work presented on this PhD thesis is a valid contribution to the field of skin cancer diagnostics. Albeit its scope is limited – one lesion per image – the results achieved by this arrangement of segmentation, feature extraction and classification algorithms showed this is the right path to achieving a reliable early screening system. If and when, to all these data, values for age, gender and evolution might be used as classification features, the results will, no doubt, become even more accurate, allowing for an improvement in the survival rates of skin cancer patients

Os padrões repetitivos como actividade de investigação matemática, na sala de 4 anos do pré-escolar
Dissertação de Mestrado em Estudos da Criança - Especialização em Ensino e Aprendizagem da MatemáticaA incidência deste estudo está focada na introdução de actividades de
investigação matemática num contexto de padrões repetitivos junto de crianças com
quatro anos.
Começa-se por caracterizar o que são actividades de investigação no Pré–
Escolar, e quais os exemplos justificativos no desenvolvimento do trabalho com padrões. Segue-se a especificação de como se desenvolve o processo de actividade de
investigação, focando a atenção na curiosidade, no sentido crítico e no relacionamento das crianças durante as actividades realizadas, conteúdos e competências, envolvimento nas actividades e interacções, e qual o papel da educadora neste contexto. Finalizando, procura-se dar um contributo para ajudar outros profissionais a aumentarem os seus conhecimentos na área de padrões, e a conhecerem a forma como as crianças enfrentam e exploram as tarefas e as potencialidades desta experiência ao nível da aprendizagem
da matemática.
Não se limitando à aplicação de conhecimentos ou procedimentos pré-definidos, as actividades de investigação estimulam uma abordagem globalizante, relacionando diferentes temas, e permitindo um tipo de trabalho de carácter transversal.
No início da intervenção entendeu-se propor às crianças um conjunto de tarefas
consistentes com o seu nível de conhecimento, de compreensão ou competência. Assim numa primeira fase as crianças desenvolveram actividades “desafiadoras” a partir dos
seus interesses, de cunho progressivamente mais aberto, referentes a contextos variados
tais como: rotinas e formação de conjuntos utilizando material manipulativo, reconhecimento das propriedades e formulação de hipóteses sobre a figura geométrica “o quadrado”, numa perspectiva de envolvimento futuro com actividades de
investigação.
Numa segunda fase temporal, pretendeu-se implementar uma actividade de investigação matemática num contexto de padrões repetitivos, já que é um conceito que
é pouco explorado com crianças de 4 anos.
O estudo segue uma abordagem de investigação qualitativa baseada no estudo de
caso, configurando trajectórias hipotéticas de aprendizagem.
O investigador assumiu um papel de observador participante, dirigindo e testemunhando presencialmente as sessões de trabalho das crianças. Foi privilegiada a recolha de dados de ordem qualitativa, durante oito meses, incluindo-se nestes os dados recolhidos durante os três meses em que decorreram as investigações com padrões.
A análise dos dados recaiu sobre os processos desenvolvidos durante as
actividades investigativas, mais do que sobre os resultados ou produtos obtidos. A
finalidade foi a de perceber as questões de relevo, os processos utilizados pelas crianças, as conjecturas, generalizações e argumentações, tendo por base o processo de interacção e de comunicação, na tentativa de perceber as tomadas de decisões e as aprendizagens conseguidas.
Neste contexto, pudemos certificar-nos de que as crianças de quatro anos conseguem envolver-se em actividades de investigação matemática.The present study focuses on the introduction of mathematical investigation
activities in a context of repetitive patterns among four year old children.
First, we have characterized preschool investigation activities, and given
justifying examples for the development of the work with patterns.
Following, we have specified how to develop the investigation activity process,
focusing our attention on the curiosity, critical sense, and relationship of the children
during the activities, as well as on the contents, competences, involvement in the
activities, interactions, and the role of the educator concerning this matter.
Finally, we tried to make a contribution to help other professionals both to
increase their knowledge in the area of patterns, as well as to realise the way that
children face and explore the tasks and the potentialities of this experiment considering
mathematics learning.
Investigation activities are not limited to the application of knowledge or
pre-defined procedures, also stimulating a global approach, relating different subjects,
and allowing a transversal kind of work.
At the beginning of the intervention, we have decided to propose to the children
some tasks consistent with their level of knowledge, understanding or competence.
Thus, the children have first developed challenging activities, according to their own
interests, which became more and more open, referring to varied contexts, such as
routines, formation of sets using manipulative material, recognition of properties and
formulation of hypothesis on the geometrical figure “the square”, into a perspective
of future involvement in investigation activities.
Then, we attempted to implement a mathematical investigation activity in a
context of repetitive patterns, since it is a concept which is usually not explored with
four year olds.
Our study follows a qualitative investigation approach, based on a case-study,
configuring hypothetical learning trajectories.
The investigator took the role of participant observer, both directing and
witnessing the children’s work sessions. It was given priority to the collecting of
qualitative data, for a period of eight months, including the data which have been
collected during the three months in which the investigations with patterns took place.
The analysis of the data focused more on the investigation process than on its
results or obtained products. The main objective of this study was to understand
important issues, the procedures used by the children, their conjectures, generalisations
and argumentations, emphasising the interaction and communication process, in an
attempt to understand their decision making and learning achievements.
In this context, we could conclude that that four year old children can, actually,
involve themselves into mathematical investigation activities
Development and characterization of sensors fabricated from polymer based magnetoelectric nanocomposites
Tese de Doutoramento em Engenharia Electrónica e de ComputadoresSensors are increasingly used in many applications areas, integrated in structures,
industrial machinery, or in the environment, contributing to improve the society level of
well-being. It is expected that sensorization will play on of the most relevant roles in the
fourth industrial revolution, and allow, together with mechanization and informatization,
a full automation. Particularly, magnetic sensors allow measurements, without physical
contact, of parameters such as direction, presence, rotation, angle, or current, in addition
to magnetic field. In this way, for most applications, such sensors offer a safe, noninvasive
and non-destructive measurement, as well as provide a reliable and almost
maintenance-free technology.
Industry demands for smaller, cheaper and low-powered magnetic sensors,
motivating the exploration of new materials and different technologies, such as polymerbased
magnetoelectric (ME) composites. These composites are flexible, versatile,
lightweight, low cost, easy to model in complicated shapes, and typically involve a lowtemperature
fabrication process, being in this way, a solution for innovative magnetic
sensor device applications. Therefore, the main objective of this thesis is the development
of polymer-based ME sensors to be incorporated into technological devices.
Thus, the ME effect is increasingly being considered an attractive alternative for
magnetic field and current sensing, being able to sense static and dynamic magnetic fields.
In order to obtain a wide-range ME response, a nanocomposite of Tb0.3Dy0.7Fe1.92
(Terfenol-D)/CoFe2O4/poly(vinylidene fluoride-trifluoroethylene) (P(VDF-TrFE)) was
produced and their morphological, piezoelectric, magnetic and magnetoelectric properties
investigated. The obtained composites reveals a high piezoelectric response (≈-18 pC∙N-
1) that is independent of the weight ratio between the fillers. In turn, the magnetic
properties of the composites are influenced by the composite composition. It was found
that the magnetization saturation values decrease with increasing CoFe2O4 content (from
18.5 to 13.3 emu∙g-1) while the magnetization and coercive field values increase (from
3.7 to 5.5 emu∙g-1 and from 355.7 to 1225.2 Oe, respectively) with increasing CoFe2O4
content. Additionally, the films show a wide-range dual-peak ME response at room
temperature with the ME coefficient increasing with increasing weight content of
Terfenol-D, from 18.6 mV∙cm-1∙Oe-1 to 42.3 mV∙cm-1∙Oe-1.
The anisotropic ME effect on a Fe61.6Co16.4Si10.8B11.2 (FCSB)/poly(vinylidene
fluoride) (PVDF)/FCSB laminate composite has been used for the development of a magnetic field sensor able to detect both magnitude and direction of ac and dc magnetic
fields. The accuracy (99% for both ac and dc sensors), linearity (92% for the dc sensor
and 99% for the ac sensor), sensitivity (15 and 1400 mV∙Oe-1 for the dc and ac fields,
respectively), and reproducibility (99% for both sensors) indicate the suitability of the
sensor for applications.
A dc magnetic field sensor based on a PVDF/Metglas composite and the
corresponding readout electronic circuits for processing the output ME voltage were
developed. The ME sensing composite presents an electromechanical resonance
frequency close to 25.4 kHz, a linear response (r2=0.997) in the 0–2 Oe dc magnetic field
range, and a maximum output voltage of 112 mV (ME voltage coefficient α33 of ≈30
V∙cm-1∙Oe-1). By incorporating a charge amplifier, an ac–rms converter and a
microcontroller with an on chip analog-to-digital converter (ADC), the ME voltage
response is not distorted, the linearity is maintained, and the ME output voltage increases
to 3.3 V (α33effective=1000 V∙cm-1∙Oe-1). The sensing device, including the readout
electronics, has a maximum drift of 0.12 Oe with an average total drift of 0.04 Oe, a
sensitivity of 1.5 V∙Oe-1 (15 kV∙T-1), and a 70 nT resolution. Such properties allied to the
accurate measurement of the dc magnetic field in the 0–2 Oe range makes this polymerbased
device very attractive for applications, such as Earth magnetic field sensing, digital
compasses, navigation, and magnetic field anomaly detectors.
A dc current sensor device based on a ME PVDF/Metglas composite, a solenoid, and
the corresponding electronic instrumentation were developed. The ME sample exhibits a
maximum α33 of 34.48V∙cm-1∙Oe-1, a linear response (r2=0.998) and a sensitivity of 6.7
mV∙A-1. With the incorporation of a charge amplifier, a precision ac/dc converter and a
microcontroller, the linearity is maintained (r2=0.997), the ME output voltage increases
to a maximum of 2320 mV and the sensitivity is increased to 476.5 mV∙A-1. Such features
indicate that the fabricated ME sensing device is suitable to be used in non-contact electric
current measurement, motor operational status checking, and condition monitoring of
rechargeable batteries, among others.
In this way, polymer-based ME composites proved to be suitable for magnetic field
and current sensor applications.Os sensores estão a ser cada vez mais utilizados em diversas áreas, integrados em
estruturas, máquinas industriais ou projetos ambientais, contribuindo para melhorar o
nível de bem-estar e eficiência da nossa sociedade. Espera-se que a “sensorização”
contribua decisivamente para a quarta revolução industrial, e que permita, em conjunto
com a mecanização e a informatização, uma completa automação. Em particular, os
sensores magnéticos permitem medir parâmetros como a direção, presença, rotação,
ângulo ou corrente, para além do campo magnético, tudo isto sem qualquer contacto
físico. Assim, para a maioria das aplicações, estes sensores oferecem uma medição
segura, não invasiva e não destrutiva, para além de garantirem uma tecnologia confiável
e de escassa manutenção.
A indústria procura e exige sensores magnéticos mais pequenos, mais baratos e de
baixo consumo, daí a motivação para explorar novos materiais e diferentes tecnologias,
tais como os compósitos magnetoelétricos (ME) baseados em polímeros. Estes
compósitos são flexíveis, versáteis, leves, de baixo custo, fáceis de se modelar em formas
complexas e tipicamente envolvem um processo de fabricação a baixa temperatura,
constituindo uma solução fiável e de qualidade para os sensores magnéticos. É da
constatação deste potencial que surge este estudo e o objetivo desta tese: o
desenvolvimento de sensores ME de base polimérica.
O efeito ME é cada vez mais considerado como uma alternativa credível para a
medição de campo magnético e da intensidade da corrente elétrica, podendo detetar
campos magnéticos estáticos e dinâmicos.
De modo a obter uma gama mais alargada de resposta ME, produziram-se
nanocompósitos de Tb0.3Dy0.7Fe1.92 (Terfenol-D)/CoFe2O4/poli(fluoreto de vinilideno
trifluor-etileno) (P(VDF-TrFE) e as suas propriedades morfológicas, piezoelétricas,
magnéticas e magnetoelétricas foram investigadas. Os compósitos obtidos revelam uma
elevada resposta piezoelétrica (≈-18 pC∙N-1) que é independente da percentagem de cada
material magnetoestrictivo. Por sua vez, as propriedades magnéticas são influenciadas
pela composição dos compósitos. Verificou-se que a magnetização de saturação diminuí
com o aumento da percentagem de CoFe2O4 (de 18.5 para 13.3 emu∙g-1) enquanto que a
magnetização e o campo coercivo aumentam (de 3.7 para 5.5 emu∙g-1 e de 355.7 para
1225.2 Oe, respetivamente) com o aumento da percentagem em massa de CoFe2O4. O efeito ME anisotrópico num compósito Fe61.6Co16.4Si10.8B11.2 (FCSB)/
poli(fluoreto de vinilideno) (PVDF)/FCSB laminado foi utilizado para desenvolver um
sensor de campo magnético capaz de detetar tanto a magnitude como a direção de campos
magnéticos ac e dc. A exatidão (99% para ambos os sensores ac e dc), linearidade (92%
para o sensor dc e 99% para o ac), sensibilidade (15 e 1400 mV∙Oe-1 para o sensor dc e
ac, respetivamente) e reprodutibilidade (99% para ambos os sensores) indicam a aptidão
destes sensores para aplicações avançadas.
Desenvolveu-se ainda um sensor de campo magnético dc baseado num compósito
ME de PVDF/Metglas, bem como a correspondente eletrónica de leitura para processar a
tensão de saída ME. O compósito ME apresenta uma ressonância eletromecânica de
aproximadamente 25.4 kHz, uma resposta linear (r2=0.997) para uma gama de campos
magnéticos dc entre 0–2 Oe e uma tensão de saída máxima de 112 mV (coeficiente ME
α33≈30 V∙cm-1∙Oe-1). Ao incorporar um amplificador de carga, um conversor ac–rms e
um microcontrolador com um conversor analógico-digital (ADC), a tensão ME não é
distorcida, a linearidade manteve-se e a tensão ME aumentou para 3.3 V (α33efectivo=1000
V∙cm-1∙Oe-1). O sensor, incluindo a eletrónica de leitura, obteve um desvio máximo de
0.12 Oe com um desvio total médio de 0.04 Oe, uma sensibilidade de 1.5 V∙Oe-1 (15
kV∙T-1) e 70 nT de resolução. Tais propriedades aliadas à medida exata do campo
magnético dc entre 0–2 Oe tornam este dispositivo indicado para aplicações como
sensores de campo magnético terrestre, compassos digitais, navegação e detetores de
anomalia no campo magnético.
Foi ainda possível desenvolver e otimizar um sensor de corrente baseado num
compósito ME de PVDF/Metglas, num solenoide e na correspondente eletrónica de
instrumentação. A amostra ME exibe um α33 máximo de 34.48V∙cm-1∙Oe-1, uma resposta
linear (r2=0.998) e uma sensibilidade de 6.7 mV∙A-1. Com a incorporação de um
amplificador de carga, um conversor ac/dc de precisão e um microcontrolador, a
linearidade manteve-se, a tensão ME aumentou para um máximo de 2320 mV e a
sensibilidade subiu para 476.5 mV∙A-1. Estas propriedades tornam este sensor ME
apropriado para a medição de corrente elétrica sem contato, para a verificação do estado
de funcionamento de motores e para monitorização da condição de baterias recarregáveis,
entre outros.
Concluindo-se deste modo que os compósitos de ME com base em polímeros
provaram ser adequados para aplicações na medição de campos magnéticos e intensidade
de corrente elétrica
Cognitive flexibility and its relation with forecasting : the Ukrainian crise as a case study
Does cognitive flexibility enhance forecasting abilities? Cognitive flexibility “is the human
ability to adapt the cognitive processing strategies to face new and unexpected conditions in the
environment” (Cañas, 2006, p. 296), and has already been proven to have a positive impact on
decision-making (Cañas et al., 2003; Laureiro-Martínez et al., 2009; Laureiro-Martínez &
Brusoni, 2018). I focused my research on forecasting abilities, using the war in Ukraine as a
study case. Participants had to first test their flexibility with the Cognitive Flexibility scale
(Martin & Rubin, 1995) and then answer forecasting questions about the war. The sample
consisted of 95 valid answers. The results of my analysis showed no correlation between
cognitive flexibility and forecasting abilities. Nevertheless, I highlight several limitations of my
work, which explain why these results should not be considered a definitive answer to this
study’s question and why researchers should further explore this topic. Previous researchers
have shown the advantages brought by cognitive flexibility, and I emphasize the importance of
continuing to study this matter, in order to be able to teach, foster and enhance cognitive
flexibility to reach a competitive advantage – if indeed these advantages are eventually proved.A flexibilidade cognitiva aumenta as capacidades de previsão? A flexibilidade cognitiva "é a
capacidade humana de adaptar as estratégias de processamento cognitivo para enfrentar
condições novas e inesperadas no ambiente" (Cañas, 2006, p. 296) e já foi provado que tem um
impacto positivo na tomada de decisões (Cañas et al., 2003; Laureiro-Martínez et al., 2009;
Laureiro-Martínez & Brusoni, 2018). Concentrei a minha investigação na capacidade de
previsão, utilizando a guerra na Ucrânia como caso de estudo. Os participantes tiveram primeiro
de testar a sua flexibilidade através de uma escala de flexibilidade cognitiva (Martin & Rubin,
1995) e depois responderam a perguntas de previsão sobre a guerra. A amostra consistiu em 95
respostas válidas. Os resultados da minha análise não mostraram qualquer correlação entre a
flexibilidade cognitiva e as capacidades de previsão. No entanto, destaco várias limitações do
meu trabalho que explicam porque é que estes resultados não devem ser utilizados como uma
resposta definitiva à pergunta deste estudo e porque é que os investigadores devem continuar a
explorar este tópico. A investigação anterior mostra as vantagens trazidas pela flexibilidade
cognitiva, e sublinho a importância de continuar a estudar este assunto de modo a poder ensinar,
fomentar e melhorar a flexibilidade cognitiva para alcançar uma vantagem competitiva - se de
facto estas vantagens acabem por ser provadas
Understanding the disease and supporting clinical decisions
Barrett’s esophagus (BE) is a recognized premalignant condition of the distal
esophagus that constitutes the major risk factor for the development of esophageal
adenocarcinoma (EA). Despite the known low rates of BE progression to EA, the
incidence of both has increased profoundly over the last decades and esophageal
malignancy remains to be a deadly cancer with high morbidity and mortality unless
diagnosed at early stages. Current BE clinical management has revealed unsuccessful in
reverting this worrisome epidemiological picture and a major hurdle has been the
incapacity to discriminate among BE patients those who have a higher risk of
malignant progression. In fact, to this date, besides dysplasia none of the existing
clinical and histologic criteria could anticipate malignant progression. It is therefore
imperative to find reliable molecular biomarkers to guide medical practice and improve
the standard of care for BE patients.
The global aim of this thesis was to better understand the pathways underlying BE
malignant progression and thereby identify reliable biomarkers relevant for the
diagnosis, prognosis and management of BE patients thus contributing to an improved
understanding of BE biology and an optimized support for clinical decisions.
To accomplish these challenging goals an unbiased and a hypothesis-driven strategies
were followed. Through the unbiased approach, a meta-analysis of transcriptome
datasets and subsequent experimental validation in a cohort BE patients in follow-up
was used to define a gene set associated with BE cancer development and, therefore,
identify early biomarkers predictive of BE malignant progression. In silico analysis
singled out two genes, CYR61 and TAZ as candidate predictive markers for BE
malignant progression and experimental validation using quantitative PCR and
immunohistochemistry revealed that both genes are upregulated and overexpressed in
non-dysplastic BE index biopsies from progressors years before cancer development
when compared with index biopsies from BE patients that did not progressed. We also
found that EMT and stemness-related genes were also significantly over represented in
BE associated with progression. Together, these results support that CYR61 and TAZ
are promising early biomarkers to stratify BE patients according to their cancer risk
and suggest a novel mechanist route for BE neoplastic progression.
Using an hypothesis-driven approach, we explored when and how centrosome
abnormalities arise along BE malignant pathway, from the early premalignant condition stage to metastatic disease, by establishing an accurate method to identify
and score centrosomes, at the single-cell level, in patient samples and cell lines. We
found that centrosome amplification arises as early as the premalignant condition of
patients that progress to malignancy and significantly expands at dysplasia stage, which
is dependent of p53 loss of function, being then present along cancer progression,
namely in EA and metastasis. So, these finding suggest that centrosome amplification
could contribute to BE initiation and malignant progression. Considering that
centrosome amplification is specific of patients that progress to cancer, this could be
further explored to be translated into useful tools to be used in the clinical setting and
potentially improve its diagnosis, prognosis and treatment. Moreover, given
widespread occurrence of both p53 mutations and centrosome abnormalities in human
tumors, our findings are likely to be extended to other cancers.
Collectively, both research avenues suggest the existence of different cellular and
molecular abnormalities dictating different pathological propensity for malignant
progression in BE, right from the beginning, and this could be further explored to trace
a cancer risk profile for every patient and guide medical decisions and improve patient
care.O esófago de Barrett (EB) é uma reconhecida condição pré-maligna que surge no
esófago distal associada ao refluxo gastroesofágico crónico e constitui o maior factor de
risco para o desenvolvimento do adenocarcinoma esofágico (AE). Apesar da taxa de
progressão maligna do EB ser baixa, a incidência de ambos tem aumentado
drasticamente nas últimas décadas. Como agravante, esta neoplasia em estadios
avançados está associada a taxas elevadas de morbilidade e mortalidade a menos que
diagnosticada e tratada em estadios iniciais, pelo que todos os doentes com EB são
integrados em programas de vigilância com vista à detecção precoce de progressão
neoplásica. Contudo, esta prática clínica tem-se mostrado ineficaz para reverter este
cenário epidemiológico e um dos factores limitantes relaciona-se com o facto da
displasia continuar a ser o único marcador de risco de progressão maligna e não haver
marcadores moleculares e clínicos que possam predizer e estratificar o potencial
maligno do EB. É assim urgente encontrar biomarcadores sensíveis e específicos
capazes de guiar a prática médica, melhorar a relação custo-benefício dos programas de
vigilância, mas principalmente a qualidade de vida dos doentes com EB.
O objectivo principal desta tese foi descobrir novas vias moleculares subjacentes à
progressão maligna do EB de forma a identificar biomarcadores fidedignos passíveis de
serem aplicados à clínica apoiando o diagnóstico, prognóstico e manejo destes doentes,
e assim contribuir para aprofundar o conhecimento relativo ao processo de
cancerigénese desta doença e potencialmente melhorar a abordagem clínica aos
doentes com EB.
Para tal foram seguidas duas linhas de investigação diferentes. De modo a encontrar
biomarcadores com potencial preditivo para progressão maligna em doentes com EB
foi realizada uma meta-análise de dados de transcriptomas previamente publicados,
seguida de uma validação experimental utilizando amostras de EB de doentes
acompanhados no programa de vigilância do Instituto Português de Oncologia
Francisco Gentil. Esta estratégia permitiu identificar dois promissores biomarcadores,
o CYR61 e o TAZ, capazes de estratificar o risco de progressão maligna do EB, não só
pela sua expressão diferencial ser a mais significativa na análise bioinformática mas
porque a validação experimental revelou que estes dois genes se encontravam sobreexpressos
logo na primeira biopsia (vários anos antes do desenvolvimento de cancro)
onde foi feito o diagnóstico de EB dos doentes que progrediram comparativamente com a expressão detetada no EB dos não progressores. Adicionalmente, foram ainda
identificados outros genes diferencialmente expressos em EB associados a progressão
maligna, cujas funções estão associadas a fenótipos de células estaminais e fenómenos
de transição epitélio-mesenquimal (TEM) em cancro. Assim, foi também descoberto
um potencial novo processo molecular associado ao desenvolvimento de cancro em EB.
Paralelamente, e considerando que alterações numéricas dos centrossomas podem
estar presentes ao longo da progressão maligna em EB e assim contribuir para o seu
processo de cancerigénese, decidimos explorar quando e como surgem as alterações
numéricas dos centrossomas ao longo da progressão do EB, desde a condição prémaligna
até às metástases ganglionares, tanto em amostras de doentes como em linhas
celulares, usando um método de dupla marcação por imunofluorescência para
identificar e quantificar fidedignamente o número de centrossomas por célula. Esta
análise revelou a existência de células com centrossomas supranumerários logo na fase
de metaplasia dos doentes que progrediram para cancro, e que a sua incidência
aumenta significativamente na fase de displasia, a qual é dependente da perda de
função do gene supressor tumoral p53, estando também depois presentes ao longo das
restantes fases de progressão. Estes resultados sugerem assim que a desregulação dos
centrossomas pode contribuir para a iniciação e progressão neoplásica do EB. No
futuro será importante aprofundar o contributo dos centrossomas na cancerigénese do
EB e tentar perceber qual o seu impacto no diagnóstico, prognóstico e tratamento
destes doentes. Uma vez que tanto as alterações numéricas dos centrossomas como a
perda de função do p53 são achados prevalentes em cancro, os resultados deste estudo
poderão ser relevantes para outros modelos tumorais.
Em conjunto, os resultados obtidos sugerem que a propensão maligna não é igual em
todos os EB. Logo muito cedo no processo, aqueles cujo risco de virem a desenvolver
cancro é maior sofrem alterações moleculares e celulares passíveis de serem detetadas e
utilizadas como biomarcadores preditivos estratificando o risco de progressão maligna,
e desta forma orientar as decisões clínicas, adequar tempos de vigilância e melhorar a
abordagem terapêutica
