Computational model of 18650 lithium-ion battery with coupled strain rate and SOC dependencies
Highly nonlinear structures and constituent materials and hazardous experiment situations have resulted in a pressing need for a numerical mechanical model for lithium-ion battery (LIB). However, such a model is still not well established. In this paper, an anisotropic homogeneous model describing the jellyroll and the battery shell is established and validated through compression, indentation, and bending tests at quasi-static loadings. In this model, state-of-charge (SOC) dependency of the LIB is further included through an analogy with the strain-rate effect. Moreover, with consideration of the inertia and strain rate effects, the anisotropic homogeneous model is extended into the dynamic regime and proven capable of predicting the dynamic response of the LIB using the drop-weight test. The established model may help to predict extreme cases with high SOCs and crashing speeds with an over 135% improved accuracy compared to traditional models. The established coupled strain rate and SOC dependencies of the numerical mechanical model for the LIB aims to provide a solid step toward unraveling and quantifying the complicated problems for research on LIB mechanical integrity. Antisense-induced suppression of taxoid 14β- hydroxylase gene expression in transgenic Taxus × media cells
The enzyme taxoid 14β-hydroxylase (14OH) directs a side-route of taxol pathway to 14β-hydroxy taxoids. Suppression of this side-route could increase the production of taxol. To suppress taxoid 14β- hydroxylase gene (14OH) expression in the Taxus × media TM3 cell line, antisense RNA inhibition approach was used in this study. Following the construction of an antisense RNA expression vector of 14OH from Taxus chinensis, the antisense 14OH cDNA (as14OH) was introduced into TM3 cells by Agrobacterium tumefaciens-mediated transformation. Southern blot analysis of hygromycin phosphotransferase gene (HYG) revealed that this selection gene was integrated successfully into the genome of Taxus × media cells. Reverse transcription-polymerase chain reaction (RT-PCR) analysis showed that the 14OH mRNA level in transgenic cells dropped dramatically, suggesting that the expression of endogenous14OH gene was significantly suppressed by the exogenous as14OH gene. Correspondingly, the total yield of three major C-14 oxygenated taxoids (yunnanxane, taxuyunnanine C, sinenxan C) was markedly reduced in the silenced cell lines when compared with those of the nontransgenic controls. These results indicated that the antisense RNA strategy is a useful tool in suppressing the expression of genes in Taxus and this method could be used to silence other important genes that divert Taxol pathway to side-route metabolites.Key words: Taxus × media, taxoid 14β-hydroxylase, antisense, gene suppression
Coexisting conical bipolar and equatorial outflows from a high-mass protostar
The BN/KL region in the Orion molecular cloud is an archetype in the study of
the formation of stars much more massive than the Sun. This region contains
luminous young stars and protostars, but it is difficult to study because of
overlying dust and gas. Our basic expectations are shaped to some extent by the
present theoretical picture of star formation, the cornerstone of which is that
protostars acrete gas from rotating equatorial disks, and shed angular momentum
by ejecting gas in bipolar outflows. The main source of the outflow in the
BN/KL region may be an object known as radio source I, which is commonly
believed to be surrounded by a rotating disk of molecular material. Here we
report high-resolution observations of silicon monoxide (SiO) and water maser
emission from the gas surrounding source I; we show that within 60 AU (about
the size of the Solar System), the region is dominated by a conical bipolar
outflow, rather than the expected disk. A slower outflow, close to the
equatorial plane of the protostellar system, extends to radii of 1,000 AU.Comment: 10 pages, 2 figures. Accepted by Nature. To appear December 199
Experimental Characterization Of Electrical Current Leakage In Poly(Dimethylsiloxane) Microfluidic Devices
Poly(dimethylsiloxane) (PDMS) is usually considered as a dielectric material and the PDMS microchannel wall can be treated as an electrically insulated boundary in an applied electric field. However, in certain layouts of microfluidic networks, electrical leakage through the PDMS microfluidic channel walls may not be negligible, which must be carefully considered in the microfluidic circuit design. In this paper, we report on the experimental characterization of the electrical leakage current through PDMS microfluidic channel walls of different configurations. Our numerical and experimental studies indicate that for tens of microns thick PDMS channel walls, electrical leakage through the PDMS wall could significantly alter the electrical field in the main channel. We further show that we can use the electrical leakage through the PDMS microfluidic channel wall to control the electrolyte flow inside the microfluidic channel and manipulate the particle motion inside the microfluidic channel. More specifically, we can trap individual particles at different locations inside the microfluidic channel by balancing the electroosmotic flow and the electrophoretic migration of the particle
Mice lacking NF-κB1 exhibit marked DNA damage responses and more severe gastric pathology in response to intraperitoneal tamoxifen administration
Tamoxifen (TAM) has recently been shown to cause acute gastric atrophy and metaplasia in mice. We have previously demonstrated that the outcome of Helicobacter felis infection, which induces similar gastric lesions in mice, is altered by deletion of specific NF-κB subunits. Nfkb1-/- mice developed more severe gastric atrophy than wild-type (WT) mice 6 weeks after H. felis infection. In contrast, Nfkb2-/- mice were protected from this pathology. We therefore hypothesized that gastric lesions induced by TAM may be similarly regulated by signaling via NF-κB subunits. Groups of five female C57BL/6 (WT), Nfkb1-/-, Nfkb2-/- and c-Rel-/- mice were administered 150 mg/kg TAM by IP injection. Seventy-two hours later, gastric corpus tissues were taken for quantitative histological assessment. In addition, groups of six female WT and Nfkb1-/- mice were exposed to 12 Gy γ-irradiation. Gastric epithelial apoptosis was quantified 6 and 48 h after irradiation. TAM induced gastric epithelial lesions in all strains of mice, but this was more severe in Nfkb1-/- mice than in WT mice. Nfkb1-/- mice exhibited more severe parietal cell loss than WT mice, had increased gastric epithelial expression of Ki67 and had an exaggerated gastric epithelial DNA damage response as quantified by γH2AX. To investigate whether the difference in gastric epithelial DNA damage response of Nfkb1-/- mice was unique to TAM-induced DNA damage or a generic consequence of DNA damage, we also assessed gastric epithelial apoptosis following γ-irradiation. Six hours after γ-irradiation, gastric epithelial apoptosis was increased in the gastric corpus and antrum of Nfkb1-/- mice. NF-κB1-mediated signaling regulates the development of gastric mucosal pathology following TAM administration. This is associated with an exaggerated gastric epithelial DNA damage response. This aberrant response appears to reflect a more generic sensitization of the gastric mucosa of Nfkb1-/- mice to DNA damage
Validation of the Spanish version of the 9-item Shared Decision-Making Questionnaire
OBJECTIVE: To translate and assess the psychometric properties of the 9-item Shared Decision-Making Questionnaire (SDM-Q-9) for measuring patients' perceptions of how clinicians' performance fits the SDM process. DESIGN: Cross-sectional study. SETTING AND PARTICIPANTS: Data were collected in primary care health centres. Patients suffering from chronic diseases and facing a medical decision were included in the study. MEASUREMENTS: The original German SDM-Q-9 was translated to Spanish using the process of cross-cultural adaptation of self-reported measures as the methodological model for Spanish translation. Reliability (internal consistency) and construct validity [exploratory (EFA) and confirmatory factor analysis (CFA)] were assessed. RESULTS: The final Spanish version of the SDM-Q-9 was tested in a primary care sample of 540 patients. The SDM-Q-9 presented adequate reliability and acceptable validity. Internal consistency yielded a Cronbach's alpha of 0.885 for the whole scale. EFA showed a two-factorial solution, and for the CFA, the best solution was obtained with a one-dimensional factor with the item 1 excluded, which produced the best indexes of fit. DISCUSSION AND CONCLUSIONS: The Spanish version of the SDM-Q-9 showed adequate reliability and acceptable validity parameters among primary care patients. The SDM-Q-9 is suitable for use in Spain and other Spanish-speaking countries with similarly organized health-care systems. The use of the SDM-Q-9 may contribute to the evaluation of SDM process from the patient's perspective
Vectorial dissipative solitons in vertical-cavity surface-emitting Lasers with delays
We show that the nonlinear polarization dynamics of a vertical-cavity
surface-emitting laser placed into an external cavity leads to the formation of
temporal vectorial dissipative solitons. These solitons arise as cycles in the
polarization orientation, leaving the total intensity constant. When the cavity
round-trip is much longer than their duration, several independent solitons as
well as bound states (molecules) may be hosted in the cavity. All these
solutions coexist together and with the background solution, i.e. the solution
with zero soliton. The theoretical proof of localization is given by the
analysis of the Floquet exponents. Finally, we reduce the dynamics to a single
delayed equation for the polarization orientation allowing interpreting the
