Algılanan Örgütsel Destek ve Şirkete Bağlılık: Örgüt Temelli Öz-Saygının Rolü
148 beyaz yakalı çalışanın katıldığı bu çalışma, algılanan örgütsel destek ile örgütsel bağlılık arasında ilişkiyi ve ayrıca bu değişkenler arasında örgüt temelli özsaygının ara değişken olarak rolünü incelemektedir. Sonuçlar algılanan örgütsel destek ve duygusal bağlılık ve normative bağlılık arasında anlamlı pozitif bir ilişkinin olduğunu ancak devam bağlılığı ile negatif bir ilişkinin olduğunu ortaya koymuştur. Örgüt temelli özsaygının ise algılanan örgütsel destek ile duygusal bağlılık arasında kısmi, algılanan örgütsel destek ve devam bağlılığı arasında ise tam olarak ara değişken rolü gördüğü saptanmıştır. Araştırma sonuçları örgüt temelli özsaygının normatif bağlılık açısından herhangi bir ara değişken rolü olmadığını göstermiştir.Present study examines the relationship between perceived organizational support (POS) and organizational commitment and the mediating role of organization based self-esteem between these variables. 148 white collared employees participated to the study. Results indicated a significant relationship between POS and affective commitment and normative commitment, but a negative relationship between POS and continuance commitment. Results reveal that organization based self-esteem (OBSE) has a partial mediating role between perceived organizational support and affective commitment and full mediating role between perceived organizational support and continuance commitment. Findings also indicate that OBSE has no mediating role between perceived organizational support and normative commitment
Recent Advances in Health Biotechnology During Pandemic
The outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which
emerged in 2019, cut the epoch that will make profound fluctuates in the history of the world
in social, economic, and scientific fields. Urgent needs in public health have brought with
them innovative approaches, including diagnosis, prevention, and treatment. To exceed the
coronavirus disease 2019 (COVID-19) pandemic, various scientific authorities in the world
have procreated advances in real time polymerase chain reaction (RT-PCR) based diagnostic
tests, rapid diagnostic kits, the development of vaccines for immunization, and the purposing
pharmaceuticals for treatment. Diagnosis, treatment, and immunization approaches put for-
ward by scientific communities are cross-fed from the accrued knowledge of multidisciplinary
sciences in health biotechnology. So much so that the pandemic, urgently prioritized in the
world, is not only viral infections but also has been the pulsion in the development of novel
approaches in many fields such as diagnosis, treatment, translational medicine, virology, mi-
crobiology, immunology, functional nano- and bio-materials, bioinformatics, molecular biol-
ogy, genetics, tissue engineering, biomedical devices, and artificial intelligence technologies.
In this review, the effects of the COVID-19 pandemic on the development of various scientific
areas of health biotechnology are discussed
Mikro ölçekli güç üretimi için mems tabanli mikrolitre hacimli mikrobiyal yakit pili
Fuel cells can be a part of the solution to energy problem in the world. They can
supply power in both macro and micro scales. Especially, MEMS based microscale
microbial fuel cells (μMFC) may hold the answer to manufacture easy, cheap, fast,
and mobile power sources and sensors. μMFCs are electrochemical devices
converting chemical energy into electrical energy utilizing microorganisms as
biocatalyst, instead of precious metal catalysts used in conventional fuel cells. They
can be integrated to power, for example, lab-on-a-chip systems, or they can be used
as stand-alone biosensors for sensing applications.
This study focused on the development of a compact microbial fuel cell with
microliter volume fabricated using silicon MEMS technology. The aim was to have
high power density and low start-up time to be integrated as a power source for small
devices.
Several µMFC systems were operated under different conditions throughout the
study. Effects of external load, anolyte type, operating conditions, and chemical
modification of gold anode surfaces were compared in terms of start-up time and
power densities using Shewanella oneidensis MR-1. Performances were evaluated
using polarization curves, Electrochemical Impedance Spectroscopy, and Scanning
Electron Microcopy. The results showed that µMFCs modified with cysteamine selfassembled monolayers resulted in more than a 50% reduction in start-up times due
to better bacterial attachment on the anode surface. The volumetric power density
(330 µW/cm3
) was found to be similar in cysteamine-modified and bare gold µMFCs
and was comparable to results reported in similar studies in the literature.Yakıt pilleri, dünyadaki enerjine probleminin çözümünün bir parçası olabilirler. Hem makro hem de mikro düzeyde güç üretebilirler. Özellikle, MEMS tabanlı mikro ölçekli mikrobiyal yakıt pilleri (μMYP), kolay, ucuz, hızlı ve taşınabilir güç kaynaklarının ve sensörlerin üretimi için bir cevap olabilir. μMYP’ler, konvansiyonel yakıt pillerinde kullanılan metal katalizörlerin aksine mikroorganizmaların katalizör olarak kullanıldığı, kimyasal enerjiyi elektriksel enerjiye dönüştüren elektrokimyasal cihazlardır. Örneğin, çip-üstü-laboratuvar sistemlerine güç sağlamak üzere entegre edilebilecekleri gibi kendi başlarına bir biyosensör olarak kullanılabilirler. Bu çalışma, silisyum MEMS teknolojisi ile üretilmiş, mikrolitre hacimli, kompakt bir mikrobiyal yakıt pilinin geliştirilmesi üzerine odaklanmıştır. Küçük cihazlar için güç kaynağı olarak entegre edilmek üzere yüksek güç yoğunluğuna ve düşük başlatma süresine sahip olacak şekilde tasarlanması amaçlanmıştır. Çalışma boyunca birçok μMYP farklı koşullarda çalıştırılmıştır. Dış yükün, anolit tipinin, çalışma koşullarının ve altın anot yüzeylerinin kimyasal modifikasyonunun başlatma süresi ve güç yoğunluğu üzerine etkileri Shewanella oneidensis MR-1 kullanılarak karşılaştırılmıştır. Performans parametreleri polarizasyon eğrileri, Elektrokimyasal Empedans Spektroskopisi ve Taramalı Elektron mikroskopu kullanılarak karşılaştırılmıştır. Sonuçlar, kendiliğinden oluşan sisteamin tek katmanıyla işlem görmüş μMYP’lerinin anot yüzeyinde daha iyi bakteriyel tutunma sağladığı için başlatma süresini %50’den fazla kısalttığını göstermiştir. Sisteaminle işlenmiş ve işlenmemiş altın yüzeyler kullanılan μMYP’lerde, hacimsel güç yoğunluğu (330 µW/cm3 ) benzer olarak bulunmuştur ve bu değerler literatürdeki benzer çalışma sonuçlarıyla aynı ölçektedir.Ph.D. - Doctoral Progra
Validity and reliability of the Turkish version of the Child Perceptions Questionnaire 8-10
WOS: 000583583400001PubMed: 33051908Background A reliable measurement tool is needed to gain more knowledge about Turkish children's oral health status that affects the quality of life. Aim To develop a translated version of CPQ(8-10) into Turkish language and evaluate its reliability, validity, and reproducibility. Design the intra-class correlation coefficient (ICC) was used for the test-retest reliability in the 50 children. For the internal consistency, 418 children aged 8-10 years were evaluated using Cronbach's alpha Coefficient. Construct validity and discriminant validity were calculated using Spearman's correlation analysis, and exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) was also employed. Results the mean CPQ(8-10) score was 13.3 +/- 9.2. the translated Turkish version of CPQ(8-10) showed high test-retest reliability (ICC = 0.90) and internal consistency (Cronbach's alpha = 0.82). A statistically significant correlation was observed between the scores of the CPQ(8-10) and oral health and overall well-being, which revealed good construct validity (r = 0.50; P < .001 and r = 0.47; P < .001, respectively). Seven factors identified from EFA were statistically verified using CFA. Discriminant validity was supported by significant differences between CPQ and OHIS scores (P < .001). Conclusions the translated version of CPQ(8-10) to the Turkish language is reliable, valid, and reproducible for use in the Turkish culture in this age group
Capture and release of viable CTCs in microfluidic channel
The number of circulating tumor cells (CTCs) in blood is associated with prognosis in several types of cancer. Isolation and characterization of CTCs have important clinical significance in terms of prognosis and early detection of response to treatment. Moreover, downstream characterization of CTCs may help better patient stratification and therapy guidance. However, CTCs are extremely rare and highly sensitive and specific technology is required to isolate viable CTCs from blood cells. In this study, a surface modification strategy is developed for (i) on-chip detection of CTCs by using immunostaining and (ii) viable cell release for downstream analysis. In the first application, selective CTC capture is demonstrated using breast cancer cells (MCF-7) spiked in buffer containing background leukocytes with cell concentration ratio of 1:100. Cells captured by anti-EpCAM coated microchannel are stained with immunofluorescence conjugated anti-pan cytokeratin antibody for epithelial cells and anti-CD45 antibody for hematologic cells, and by DAPI for nuclear staining. Cytokeratin and DAPI stained cells are scored as CTCs whereas CD45+ cells are scored as leukocytes. In the second application, captured cells are released with the combination of enzymatic treatment and high flow rate washing. Development of EpCAM Sensitive Surface Functionalization Strategy for CTC Capture in Microfluidic Channels
Dolaşımdaki tümör hücrelerinin (CTC'ler) izolasyonu ve karakterizasyonu, prognoz ve tedaviye yanıtın erken tespiti açısından önemli klinik öneme sahiptir. Ayrıca, CTC'lerin aşağı akış karakterizasyonu, daha iyi hasta sınıflandırmasına ve tedavi kılavuzluğuna yardımcı olabilir. Bununla birlikte, CTC'ler son derece nadirdir (~ 10 CTC / 1010 periferik kan hücresi) ve oldukça hassastır ve izolasyonları için özel teknoloji gereklidir. Hızla gelişen mikroakışkan teknolojiler, nadir hücre izolasyonunda hızlı, düşük maliyetli ve otomatik numune işleme dahil olmak üzere çeşitli avantajlar ve biyolojik hücrelere benzer fiziksel boyutları nedeniyle daha yüksek hassasiyet ve özgüllük sunar. Bu teknolojilerin çoğu, immünoafinite tabanlı CTC yakalama kullanır, epitel hücre yüzeyi biyobelirtecine karşı anti-EpCAM antikoru, CTC'ye özgü hücre yakalama için yaygın olarak kullanılmaktadır. Bu tür uygulamalarda, uygun bir antikor immobilizasyonu, yüksek verimli hücre yakalama için çok önemli bir rol oynar. Bu çalışmada, verimli, seçici ve EpCAM'a duyarlı CTC yakalama için bir oksit yüzey modifikasyon stratejisi geliştirilmiştir. Hücre yakalama etkinliği, bir CTC modeli olarak MCF-7 göğüs kanseri hücre dizisi kullanılarak belirlenmiş ve ortalama hücre yakalama etkinliği, 8 bağımsız deneyden% 89 olarak bulunmuştur. Seçici CTC yakalama, hücre konsantrasyonu oranı 1: 104 olan arka plan lökositleri içeren tamponda spike edilmiş göğüs kanseri hücreleri (MCF-7) kullanılarak gösterildi ve sırasıyla% 90 ve% 87'lik yakalama verimliliği ve özgüllüğü elde edildi.
