    Pyridoxine analysis by high performance liquid chromatography and validation in fortified milk powder

    Vitamin B6 analysis by reverse-phase high performance liquid chromatography and its validation in milk powder prompted this study. The calibration curves for pyridoxal, pyridoxamine and pyridoxine were evaluated and method precision and accuracy assessed. It was found that the working range was adequate for all the analytes. The analytical procedure was verified by testing the enzymatic efficiency, precision and accuracy. In milk powder samples pyridoxine was the only vitamer found in quantities above the considered limit of quantification and the accuracy showed Z-scores lower than 2. Uncertainty estimation based on method validation results was 0.066 mg/100g, for a vitamin B6 content of 0.61 mg/100 g. It was concluded that the method is adequate for vitamin B6 quantification in powder milk

    Assessment of PCR in the detection of Leishmania spp in experimentally infected individual phlebotomine sandflies (Diptera: Psychodidae: Phlebotominae)

    DNA amplification by the polymerase chain reaction (PCR) was applied in the investigation of the presence of Leishmania (Kinetoplastida: Trypanosomatidae) parasites in single phlebotomine sandflies. Three phlebotomine/parasite pairs were used: Lutzomyia longipalpis/Leishmania chagasi, Lutzomyia migonei/Leishmania amazonensis and Lutzomyia migonei/Leishmania braziliensis, all of them incriminated in the transmission of visceral or cutaneous leishmaniasis. DNA extraction was performed with whole insects, with no need of previous digestive tract dissection or pooling specimens. The presence of either mouse blood in the digestive tract of the sandflies or the digestive tract itself did not interfere in the PCR. Infection by as few as 10 Leishmania sp. per individual were sufficient for DNA amplification with genus-specific primers. Using primers for L. braziliensis and L. mexicana complexes, respectively, it was possible to discriminate between L. braziliensis and L. amazonensis in experimentally infected vectors (L. migonei).Neste trabalho avaliamos o uso da reação em cadeia da polimerase (PCR) na investigação da presença de parasitas Leishmania (Kinetoplastida:Trypanosomatidae) em flebotomíneos individuais. Para isso, foram utilizados três pares flebotomíneo/parasita: Lutzomyia longipalpis/Leishmania chagasi, Lutzomyia migonei/Leishmania amazonensis e Lutzomyia migonei/Leishmania braziliensis, todos eles incriminados na transmissão de leishmaniose cutânea ou visceral. O DNA total a ser amplificado foi extraído de flebotomíneos inteiros, contendo ou não o parasita, sem dissecção prévia do trato digestivo ou combinação de indivíduos. Conteúdos do trato digestivo de flebotomíneos, em especial sangue de camundongo, não interferiram na reação de amplificação. Dez parasitas Leishmania sp. por flebotomíneo foram suficientes para detecção com iniciadores gênero-específicos. Com a utilização de iniciadores para os complexos L. braziliensis e L. mexicana, respectivamente, foi possível discriminar entre L. braziliensis e L. amazonensis, em flebotomíneos infectados experimentalmente (L. migonei)

    Characterization of anti-crotalic antibodies

    AbstractCrotalus durissus terrificus, C. d. collilineatus, C. d. cascavella and C. d. marajoensis are responsible minor but severe snake bites in Brazil. The venoms of these snakes share the presence of crotoxin, a neurotoxin comprising of two associated components, crotapotin and phospholipase A2 (PLA2). Treatment of the victims with specific antiserum is the unique effective therapeutic measure. The ability of anti-Crotalus antisera produced by the routine using crude venom to immunize horses or purified crotoxin and PLA2 as individual immunogens was compared. Antisera obtained from horses immunized with C. durissus terrificus crude venom were able to recognize and neutralize not only the toxins presents in C. durissus terrificus, but also the ones present in the venoms from C. d. collilineatus, C. d. cascavella and C. d. marajoensis. Antisera from horses immunized with individual crotoxin or PLA2, although in lesser titers, were also able of recognizing the toxins in all four Crotalus species and neutralize the lethality of the C. d. terrificus venom

    NEW SEISMIC SOURCE ZONE MODEL FOR PORTUGAL AND AZORES

    The development of seismogenic source models is one of the first steps in seismic hazard assessment. In seismic hazard terminology, seismic source zones (SSZ) are polygons (or volumes) that delineate areas with homogeneous characteristics of seismicity. The importance of using knowledge on geology, seismicity and tectonics in the definition of source zones has been recognized for a long time [1]. However, the definition of SSZ tends to be subjective and controversial. Using SSZ based on broad geology, by spreading the seismicity clusters throughout the areal extent of a zone, provides a way to account for possible long-term non-stationary seismicity behavior [2,3]. This approach effectively increases seismicity rates in regions with no significant historical or instrumental seismicity, while decreasing seismicity rates in regions that display higher rates of seismicity. In contrast, the use of SSZ based on concentrations of seismicity or spatial smoothing results in stationary behavior [4]. In the FP7 Project SHARE (Seismic Hazard Harmonization in Europe), seismic hazard will be assessed with a logic tree approach that allows for three types of branches for seismicity models: a) smoothed seismicity, b) SSZ, c) SSZ and faults. In this context, a large-scale zonation model for use in the smoothed seismicity branch, and a new consensus SSZ model for Portugal and Azores have been developed. The new models were achieved with the participation of regional experts by combining and adapting existing models and incorporating new regional knowledge of the earthquake potential. The main criteria used for delineating the SSZ include distribution of seismicity, broad geological architecture, crustal characteristics (oceanic versus continental, tectonically active versus stable, etc.), historical catalogue completeness, and the characteristics of active or potentially-active faults. This model will be integrated into an Iberian model of SSZ to be used in the Project SHARE seismic hazard assessment

    COMPILATION OF ACTIVE FAULT DATA IN PORTUGAL FOR USE IN SEISMIC HAZARD ANALYSIS

    To estimate where future earthquakes are likely to occur, it is essential to combine information about past earthquakes with knowledge about the location and seismogenic properties of active faults. For this reason, robust probabilistic seismic hazard analysis (PSHA) integrates seismicity and active fault data. Existing seismic hazard assessments for Portugal rely exclusively on seismicity data and do not incorporate data on active faults. Project SHARE (Seismic Hazard Harmonization in Europe) is an EC-funded initiative (FP7) that aims to evaluate European seismic hazards using an integrated, standardized approach. In the context of SHARE, we are developing a fully-parameterized active fault database for Portugal that incorporates existing compilations, updated according to the most recent publications. The seismogenic source model derived for SHARE will be the first model for Portugal to include fault data and follow an internationally standardized approach. This model can be used to improve both seismic hazard and risk analyses and will be combined with the Spanish database for use in Iberian- and European-scale assessments

    Poultry shelf-life enhancing potential of nanofibers and nanoparticles containing Porphyra dioica extracts

    Aqueous extracts of commercially available red macroalgae Porphyra dioica were integrated as inner coatings of food-grade polypropylene (PP) films through use of electrospinning and electrospraying technologies. Two coating formulations (A = 5 wt% P. dioica extract and 7.5 wt% polyvinyl alcohol (PVA); B = 1 wt% P. dioica extract, 1 wt% PVA, and 17% gelatine) were evaluated as to their capacity to delay spoilage of minced chicken breasts, through monitoring of microbial growth (total mesophile aerobic colony counts), colour stability, lipid oxidation (thiobarbituric acid reactive substances (TBARS)), and sensory analysis over a 4-day refrigerated storage. Scanning electron microscopy (SEM) imaging revealed an increased nanofiber and nanoparticle density on extract-enriched fibers, without compromise to their morphology or the homogeneity of the coatings. Total microbial counts on coating B samples was significantly (p < 0.001) reduced compared to uncoated plastic wraps. The coated samples also exhibited fewer colour degradation, though the coatings did not di er substantially from uncoated plastic wrap. Sensory analysis test subjects successfully distinguished the raw samples based on their treatment and gave a positive approval rating (66.7%) to the extract-enriched coatings when asked about edibility post storage.info:eu-repo/semantics/publishedVersio

    Mineralization of Sialoliths Investigated by Ex Vivo and In Vivo X-ray Computed Tomography

    The fraction of organic matter present affects the fragmentation behavior of sialoliths; thus, pretherapeutic information on the degree of mineralization is relevant for a correct selection of lithotripsy procedures. This work proposes a methodology for in vivo characterization of salivary calculi in the pretherapeutic context. Sialoliths were characterized in detail by X-ray computed microtomography (μCT) in combination with atomic emission spectroscopy, Fourier transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, and transmission electron microscopy. Correlative analysis of the same specimens was performed by in vivo and ex vivo helical computed tomography (HCT) and ex vivo μCT. The mineral matter in the sialoliths consisted essentially of apatite (89 vol%) and whitlockite (11 vol%) with average density of 1.8 g/cm3. In hydrated conditions, the mineral mass prevailed with 53 ± 13 wt%, whereas the organic matter, with a density of 1.2 g/cm3, occupied 65 ± 10% of the sialoliths' volume.     An Updated Small Magellanic Cloud and Magellanic Bridge Catalog of Star Clusters, Associations, and Related Objects

    We present a catalog of star clusters, associations, and related extended objects in the Small Magellanic Cloud (SMC) and the Magellanic Bridge with 2741 entries, a factor 2 more than a previous version from a decade ago. Literature data up until 2018 December are included. The identification of star clusters was carried out with digital atlases in various bands currently available in the Digitized Sky Survey and the Machine Automatique à Mésurer pour l'Astronomie. imaging surveys. In particular, we cross-identified recent cluster samples from the Visible and Infrared Survey Telescope for Astronomy near-infrared YJK s survey of the Magellanic System, Optical Gravitational Lensing Experiment IV, and Survey of the MAgellanic Stellar History surveys, confirming new clusters and pointing out equivalencies. A major contribution of the present catalog consists of the accurate central positions for clusters and small associations, including a new sample of 45 clusters or candidates in the SMC and 19 in the Magellanic Bridge, as well as a compilation of the most reliable age and metallicity values from the literature. A general catalog must also deal with the recent discoveries of 27 faint and ultra-faint star clusters and galaxies projected on the far surroundings of the Clouds, most of them from the Dark Energy Survey. The information on these objects has been complemented with photometric, spectroscopic, and kinematical follow-up data from the literature. The underluminous galaxies around the Magellanic System, still very few as compared to the predictions from Λ Cold Dark Matter simulations, can bring constraints to galaxy formation and hierarchical evolution. Furthermore, we provide diagnostics, when possible, of the nature of the ultra-faint clusters, searching for borders of the Magellanic System extensions into the Milky Way gravitational potential. © 2020.
