    Correlation of test results and influence of a mass balance constraint on risks in conformity assessment of a substance or material

    When components of a substance or material are subject to a mass balance constraint, test results of the components’ contents are intrinsically correlated because of the constraint. This so-called ‘spurious’ correlation is observed in addition to possible metrologically-related correlation of test results, and natural and/or technological correlation of the components’ contents. Such correlations may influence understanding of test results and evaluation of risks of false decisions, due to measurement uncertainty, in conformity assessment of the substance or material. The objective of the present paper is the development of a technique for appropriate evaluation of the risks. A Bayesian multivariate approach to evaluate the conformance probability of materials or objects and relevant risks is discussed for different scenarios of the data modelling, taking into account all observed correlations. A Monte Carlo method, including the mass balance constraint, written in the R programming environment, is provided for the necessary calculations

    Energy Consumption Saving in Embedded Microprocessors Using Hardware Accelerators

    This paper deals with the reduction of power consumption in embedded microprocessors. Computing power and energy efficiency are becoming the main challenges for embedded system applications. This is, in particular, the caseof wearable systems. When the power supply is provided by batteries, an important requirement for these systems is the long service life. This work investigates a method for the reduction of microprocessor energy consumption, based on the use of hardware accelerators. Their use allows to reduce the execution time and to decrease the clock frequency, so reducing the power consumption. In order to provide experimental results, authors analyze a case of study in the field of wearable devices for the processing of ECG signals. The experimental results show that the use of hardware accelerator significantly reduces the power consumption

    Maml1 acts cooperatively with Gli proteins to regulate Sonic hedgheog signaling pathway

    Sonic hedgehog (Shh) signaling is essential for proliferation of cerebellar granule cell progenitors (GCPs) and its misregulation is linked to various disorders, including cerebellar cancer medulloblastoma. The effects of Shh pathway are mediated by the Gli family of transcription factors, which controls the expression of a number of target genes, including Gli1. Here, we identify Mastermind-like 1 (Maml1) as a novel regulator of the Shh signaling since it interacts with Gli proteins, working as a potent transcriptional coactivator. Notably, Maml1 silencing results in a significant reduction of Gli target genes expression, with a negative impact on cell growth of NIH3T3 and Patched1−/− mouse embryonic fibroblasts (MEFs), bearing a constitutively active Shh signaling. Remarkably, Shh pathway activity results severely compromised both in MEFs and GCPs deriving from Maml1−/− mice with an impairment of GCPs proliferation and cerebellum development. Therefore Maml1−/− phenotype mimics aspects of Shh pathway deficiency, suggesting an intrinsic requirement for Maml1 in cerebellum development. The present study shows a new role for Maml1 as a component of Shh signaling, which plays a crucial role in both development and tumorigenesis

    Enhanced Healing of Diabetic Wounds by Topical Administration of Adipose Tissue-Derived Stromal Cells Overexpressing Stromal-Derived Factor-1: Biodistribution and Engraftment Analysis by Bioluminescent Imaging

    Chronic ulcers represent a major health problem in diabetic patients resulting in pain and discomfort. Conventional therapy does not guarantee adequate wound repair. In diabetes, impaired healing is partly due to poor endothelial progenitor cells mobilisation and homing, with altered levels of the chemokine stromal-derived factor-1 (SDF-1) at the wound site. Adipose tissue-associated stromal cells (AT-SCs) can provide an accessible source of progenitor cells secreting proangiogenic factors and differentiating into endothelial-like cells. We demonstrated that topical administration of AT-SCs genetically modified ex vivo to overexpress SDF-1, promotes wound healing into diabetic mice. In particular, by in vivo bioluminescent imaging analysis, we monitored biodistribution and survival after transplantation of luciferase-expressing cells. In conclusion, this study indicates the therapeutic potential of AT-SCs administration in wound healing, through cell differentiation, enhanced cellular recruitment at the wound site, and paracrine effects associated with local growth-factors production

    A new severity score index for phenotypic classification and evaluation of responses to treatment in type I Gaucher disease

    Gaucher disease is the first lysosomal storage disease for which specific therapy became available. Over 4800 patients have been treated with enzyme replacement therapy. Analysis of Gaucher disease registry data has outlined the clinical heterogeneity of the disease and the different responses to treatment from patient to patient, and for different organs. This variability in clinical response justifies the development of a severity score index to assess disease activity, stage and prognosis, and to quantify the effects of treatment.The \u201cGaucher Disease Severity Score Index Type I\u201d(GauSSI-I), is based on the clinical experience of the authors and an extensive literature review, including data from the International Gaucher Registry. In particular for skeletal disease, all the available scoring systems have been reviewed and compared in order to provide a skeletal scoring system that allows use of any of the different methods. Siix specific domains, in which different items were scored according to their impact on morbidity, were characterized. GauSSI-I was evaluated in 53 type I Gaucher patients treated with imiglucerase, and it was compared to the Zimran score, the only severity index score so far available. It is a reliable method for staging the severity of adult type I Gaucher disease, and it is more sensitive than the Zimran score for monitoring the response to treatment
