    Novel Virtual Environment for Alternative Treatment of Children with Cerebral Palsy

    Cerebral palsy is a severe condition usually caused by decreased brain oxygenation during pregnancy, at birth or soon after birth. Conventional treatments for cerebral palsy are often tiresome and expensive, leading patients to quit treatment. In this paper, we describe a virtual environment for patients to engage in a playful therapeutic game for neuropsychomotor rehabilitation, based on the experience of the occupational therapy program of the Nucleus for Integrated Medical Assistance (NAMI) at the University of Fortaleza, Brazil. Integration between patient and virtual environment occurs through the hand motion sensor “Leap Motion,” plus the electroencephalographic sensor “MindWave,” responsible for measuring attention levels during task execution. To evaluate the virtual environment, eight clinical experts on cerebral palsy were subjected to a questionnaire regarding the potential of the experimental virtual environment to promote cognitive and motor rehabilitation, as well as the potential of the treatment to enhance risks and/or negatively influence the patient’s development.     ESTRATÉGIAS DE INCORPORAÇÃO E REESTRUTURAÇÕES ORGANIZACIONAIS NO ENSINO SUPERIOR PRIVADO BRASILEIRO: UMA ESCUTA AO TESTEMUNHO DE DOCENTES

    The text socializes results of a qualitative research on organizational restructuring processes caused by incorporation in Brazilian higher education. Some assumptions of structural contingency theory that consider the relationship between organizational and contingencies characteristics affect the organization. The research aims to comprehend the organizational restructuring process of two private colleges before the incorporation of a large educational group in expansionary stage in Brazil, from the testimony of two professors who experienced this process. Methodologically, it is utilized semi-structured interview to collect the data, which started from a guide designed with the intention of evoking testimonial narratives. The results show that the incorporation process implied in great challenges, mainly related to changes in the curricula, the imposition of new teaching practices, the intensification of work, structural changes, and management changes.El texto socializa resultados de una investigación cualitativa sobre los procesos de reestructuración de organización provocados por la incorporación en la educación superior brasileña. Algunos supuestos de la teoría de la contingencia estructural que tengan en cuenta la relación entre las características de organización y contingencias afectan a la organización. La investigación tiene como objetivo comprender el proceso de reestructuración de la organización de dos colegios privados antes de la incorporación de un grupo educativo grande en la etapa expansiva en Brasil, por el testimonio de dos profesores que experimentaron este proceso. Metodológicamente, se utilizó la entrevista semiestructurada para recoger los datos, que se iniciaron a partir de una guía diseñada con la intención de evocar narrativas testimoniales. Los resultados muestran que el proceso de incorporación implícita en grandes retos, principalmente relacionada con los cambios en los planes de estudio, la imposición de nuevas prácticas de enseñanza, la intensificación del trabajo, los cambios estructurales y cambios de gestión.O texto socializa resultados de uma pesquisa qualitativa sobre processos de reestruturação organizacional decorrentes de incorporações no ensino superior brasileiro. Parte de pressupostos da teoria da contingência estrutural que considera a relação entre características organizacionais e contingências que afetam a organização. O objetivo da pesquisa foi compreender o processo de reestruturação organizacional de duas faculdades privadas ante a incorporação de um grande grupo educacional em fase expansionista no Brasil, desde o testemunho de duas docentes que vivenciaram o processo. Utilizou-se a entrevista semiestruturada para coleta de dados, a qual partia de um roteiro concebido com a intenção de evocar narrativas testemunhais. Os resultados mostram que o processo de incorporação implicou em grandes desafios, ligados principalmente às mudanças nas grades curriculares, à imposição de novas práticas de ensino, à intensificação do trabalho, às mudanças estruturais e às alterações na forma de gestão.DOI: 10.21714/2178-8030gep.v18.355

    The Psychedelic State Induced by Ayahuasca Modulates the Activity and Connectivity of the Default Mode Network

    The experiences induced by psychedelics share a wide variety of subjective features, related to the complex changes in perception and cognition induced by this class of drugs. A remarkable increase in introspection is at the core of these altered states of consciousness. Self-oriented mental activity has been consistently linked to the Default Mode Network (DMN), a set of brain regions more active during rest than during the execution of a goal-directed task. Here we used fMRI technique to inspect the DMN during the psychedelic state induced by Ayahuasca in ten experienced subjects. Ayahuasca is a potion traditionally used by Amazonian Amerindians composed by a mixture of compounds that increase monoaminergic transmission. In particular, we examined whether Ayahuasca changes the activity and connectivity of the DMN and the connection between the DMN and the task-positive network (TPN). Ayahuasca caused a significant decrease in activity through most parts of the DMN, including its most consistent hubs: the Posterior Cingulate Cortex (PCC)/Precuneus and the medial Prefrontal Cortex (mPFC). Functional connectivity within the PCC/Precuneus decreased after Ayahuasca intake. No significant change was observed in the DMN-TPN orthogonality. Altogether, our results support the notion that the altered state of consciousness induced by Ayahuasca, like those induced by psilocybin (another serotonergic psychedelic), meditation and sleep, is linked to the modulation of the activity and the connectivity of the DMN.The Brazilian Federal Agencies: CNPq, CAPES; FINEP; The Sao Paulo State financial agency (FAPESP)

    The partial inhibition of hypothalamic IRX3 exacerbates obesity

    The Iroquois homeobox 3 (Irx3) gene has been identified as a functional long-range target of obesity-associated variants within the fat mass and obesity-associated protein (FTO) gene. It is highly expressed in the hypothalamus, and both whole-body knockout and hypothalamic restricted abrogation of its expression results in a lean phenotype, which is mostly explained by the resulting increased energy expenditure in the brown adipose tissue. Because of its potential implication in the pathogenesis of obesity, we evaluated the hypothalamic cell distribution of Irx3 and the outcomes of inhibiting its expression in a rodent model of diet-induced obesity. Methods: Bioinformatics tools were used to evaluate the correlations between hypothalamic Irx3 and neurotransmitters, markers of thermogenesis and obesity related phenotypes. Droplet-sequencing analysis in >20,000 hypothalamic cells was used to explore the types of hypothalamic cells expressing Irx3. Lentivirus was used to inhibit hypothalamic Irx3 and the resulting phenotype was studied. Findings: IRX3 is expressed predominantly in POMC neurons. Its expression is inhibited during prolonged fasting, as well as when mice are fed a high-fat diet. The partial inhibition of hypothalamic Irx3 using a lentivirus resulted in increased diet-induced body mass gain and adiposity due to increased caloric intake and reduced energy expenditure. Interpretation: Contrary to the results obtained when lean mice are submitted to complete inhibition of Irx3, partial inhibition of hypothalamic Irx3 in obese mice causes an exacerbation of the obese phenotype.     Representation of freely-explored objects in the primary somatosensory cortex and hippocampus

    When a rat freely explores an object in the dark and its whiskers touch different parts of the object with different velocities and angles, tactile information reaches the telencephalon with tantalizing spatio-temporal complexity. To investigate the tactile representation of freely-explored objects in the telencephalon, we performed multielectrode extracellular spike recordings from the primary somatosensory cortex (S1) and hippocampus (HP) of adult rats, as they freely explored novel objects in the dark. Neuronal data were fed to five different binary classifiers: multilayer perceptron, radial basis functions, support vector machines, decision tree and naive Bayes classifier. The classifiers were evaluated using the area under a receiver operating characteristic curve (AUROC). In most cases, it was possible to achieve substantial and significant object classification in both S1 and HP (>0.75 AUROC medians, corrected p<0.05). To assess the distribution of information across neuronal ensembles recorded from S1 and HP we performed a neuron dropping analysis, a bootstrap method that reveals how much information is lost, on average, as neuronal ensembles decrease their size from “n” to 1 neuron. We found that significant object classification is achieved with ensembles as small as 10 neurons in both S1 and HP. The best fit for the neuron dropping curves was akin to a type II functional response curve, which in ecology describes the decelerating intake rate of a consumer as a function of food density. We propose that the representation of freely-explored complex objects by neuronal ensembles in S1 and HP is robust, highly distributed, and shaped by the limited availability of non-redundant information to individual neurons
