    Analysis of Logistics in Support of a Human Lunar Outpost

    Strategic level analysis of the integrated behavior of lunar transportation system and lunar surface system architecture options is performed to inform NASA Constellation Program senior management on the benefit, viability, affordability, and robustness of system design choices. This paper presents an overview of the approach used to perform the campaign (strategic) analysis, with an emphasis on the logistics modeling and the impacts of logistics resupply on campaign behavior. An overview of deterministic and probabilistic analysis approaches is provided, with a discussion of the importance of each approach to understanding the integrated system behavior. The logistics required to support lunar surface habitation are analyzed from both 'macro-logistics' and 'micro-logistics' perspectives, where macro-logistics focuses on the delivery of goods to a destination and micro-logistics focuses on local handling of re-supply goods at a destination. An example campaign is provided to tie the theories of campaign analysis to results generation capabilities

    Using Low-Fix Rate GPS Telemetry to Expand Estimates of Ungulate Reproductive Success

    Background Population parameters such as reproductive success are critical for sustainably managing ungulate populations, however obtaining these data is often difficult, expensive, and invasive. Movement-based methods that leverage Global Positioning System (GPS) relocation data to identify parturition offer an alternative to more invasive techniques such as vaginal implant transmitters, but thus far have only been applied to relocation data with a relatively fine (one fix every  \u3c 8 h) temporal resolution. We employed a machine learning method to classify parturition/calf survival in cow elk in southeastern Kentucky, USA, using 13-h GPS relocation data and three simple movement metrics, training a random forest on cows that successfully reared their calf to a week old. Results We developed a decision rule based upon a predicted probability threshold across individual cow time series, accurately classifying 89.5% (51/57) of cows with a known reproductive status. When used to infer status of cows whose reproductive outcome was unknown, we classified 48.6% (21/38) as successful, compared to 85.1% (40/47) of known-status cows. Conclusions While our approach was limited primarily by fix acquisition success, we demonstrated that coarse collar fix rates did not limit inference if appropriate movement metrics are chosen. Movement-based methods for determining parturition in ungulates may allow wildlife managers to extract more vital rate information from GPS collars even if technology and related data quality are constrained by cost

    Lunar Exploration Architecture Level Key Drivers and Sensitivities

    Strategic level analysis of the integrated behavior of lunar transportation and lunar surface systems architecture options is performed to assess the benefit, viability, affordability, and robustness of system design choices. This analysis employs both deterministic and probabilistic modeling techniques so that the extent of potential future uncertainties associated with each option are properly characterized. The results of these analyses are summarized in a predefined set of high-level Figures of Merit (FOMs) so as to provide senior NASA Constellation Program (CxP) and Exploration Systems Mission Directorate (ESMD) management with pertinent information to better inform strategic level decision making. The strategic level exploration architecture model is designed to perform analysis at as high a level as possible but still capture those details that have major impacts on system performance. The strategic analysis methodology focuses on integrated performance, affordability, and risk analysis, and captures the linkages and feedbacks between these three areas. Each of these results leads into the determination of the high-level FOMs. This strategic level analysis methodology has been previously applied to Space Shuttle and International Space Station assessments and is now being applied to the development of the Constellation Program point-of-departure lunar architecture. This paper provides an overview of the strategic analysis methodology and the lunar exploration architecture analyses to date. In studying these analysis results, the strategic analysis team has identified and characterized key drivers affecting the integrated architecture behavior. These key drivers include inclusion of a cargo lander, mission rate, mission location, fixed-versus- variable costs/return on investment, and the requirement for probabilistic analysis.     Protocol for a prospective study evaluating circulating tumour cells status to predict radical prostatectomy treatment failure in localised prostate cancer patients (C-ProMeta-1)

    BACKGROUND: Treatment decisions in prostate cancer (PCa) rely on disease stratification between localised and metastatic stages, but current imaging staging technologies are not sensitive to micro-metastatic disease. Circulating tumour cells (CTCs) status is a promising tool in this regard. The Parsortix® CTC isolation system employs an epitope-independent approach based on cell size and deformability to increase the capture rate of CTCs. Here, we present a protocol for prospective evaluation of this method to predict post radical prostatectomy (RP) PCa cancer recurrence. METHODS: We plan to recruit 294 patients diagnosed with unfavourable intermediate, to high and very high-risk localised PCa. Exclusion criteria include synchronous cancer diagnosis or prior PCa treatment, including hormone therapy. RP is performed according to the standard of care. Two blood samples (20 ml) are collected before and again 3-months after RP. The clinical team are blinded to CTC results and the laboratory researchers are blinded to clinical information. Treatment failure is defined as a PSA ≥ 0.2 mg/ml, start of salvage treatment or imaging-proven metastatic lesions. The CTC analysis entails enumeration and RNA analysis of gene expression in captured CTCs. The primary outcome is the accuracy of CTC status to predict post-RP treatment failure at 4.5 years. Observed sensitivity, positive and negative predictive values will be reported. Specificity will be presented over time. DISCUSSION: CTC status may reflect the true potential for PCa metastasis and may predict clinical outcomes better than the current PCa progression risk grading systems. Therefore establishing a robust biomarker for predicting treatment failure in localized high-risk PCa would significantly enhance guidance in treatment decision-making, optimizing cure rates while minimizing unnecessary harm from overtreatment.     MATH APPS UTILIZATION: ITS PERCEIVED EFFECTS TO THE ACADEMIC PERFORMANCE OF MATHEMATICS MAJOR STUDENTS

    This study aimed to determine the perceived effects of apps utilization to study habits and academic performance. Further, it also sought to determine the views of the participants and respondents on their engagement in math apps and their relationship with study habits and academic performance. This study made use of explanatory-sequential design. The purposive sampling design was used to determine the respondents in quantitative data while the quota sampling design was also used to determine the participants in qualitative data. The study revealed that the mathematical application that is mostly used by the students is the calculator app as it garnered the highest percentage value of 80.85.%. The participants agreed that math applications serve as an aid for math problems, aid for lack of teaching by the teacher, promote learning of specific lessons and increase, and help in improving their academic performance. Respondents’ perception of their utilization of math apps is due to technical problems. The respondents also agreed that math application also affected their study habits in which they become dependent on it which led to laziness and tiredness. In addition, this study also found that there is a significant relationship between respondents’ utilization of math applications and their study habits. Lastly, there is also a significant relationship between the study habits and academic performance of selected math students of Notre Dame of Midsayap College.  Article visualizations

    Pharmacokinetics and Pharmacodynamics of PARP Inhibitors in Oncology

    Olaparib, niraparib, rucaparib, and talazoparib are poly (ADP-ribose) polymerase (PARP) inhibitors approved for the treatment of ovarian, breast, pancreatic, and/or prostate cancer. Poly (ADP-ribose) polymerase inhibitors are potent inhibitors of the PARP enzymes with comparable half-maximal inhibitory concentrations in the nanomolar range. Olaparib and rucaparib are orally dosed twice a day, extensively metabolized by cytochrome P450 enzymes, and inhibitors of several enzymes and drug transporters with a high risk for drug-drug interactions. Niraparib and talazoparib are orally dosed once a day with a lower risk for niraparib and a minimal risk for talazoparib to cause drug-drug interactions. All four PARP inhibitors show moderate-to-high interindividual variability in plasma exposure. Higher exposure is associated with an increase in toxicity, mostly hematological toxicity. For talazoparib, exposure-efficacy relationships have been described, but for olaparib, niraparib, and rucaparib this relationship remains inconclusive. Further studies are required to investigate exposure-response relationships to improve dosing of PARP inhibitors, in which therapeutic drug monitoring could play an important role. In this review, we give an overview of the pharmacokinetic properties of the four PARP inhibitors, including considerations for patients with renal dysfunction or hepatic impairment, the effect of food, and drug-drug interactions. Furthermore, we focus on the pharmacodynamics and summarize the available exposure-efficacy and exposure-toxicity relationships
