    Morphological peculiarites and functional activity of adipose-derived mesenchimal stem cells during in vitro cultivation conditions

    The studies were conducted on 2-3-months-old males of C57BL/6 mice weighing 20–24 g. Obtaining and cultivating of adipose-derived mesenchimal stem cells (AD MSCs) were carried out in a sterile laminar box with compliance of conditions of asepsis and antiseptics. AD MSCs of the 2, 4, 7 and 12 passages were analyzed. Morphometric analysis was performed using a light microscopy. Morphometric parameters such as cell and nucleus area or nuclear-cytoplasmic ratio (NCR) were calculated using the Axiovision light microscope (Carl Zeiss, Germany) and ImageJ 1.45 software. Trypan blue dye used for investigation of the viability of MSC. The morphological characteristics of mesenchymal stem cells from adipose tissue during the process of cultivation changes: at the first passages of cultivation, the cells are spindle-shaped with two, at least three, long long cytoplasmic processes, located bipolar. Near the nucleus the Golgi complex is clearly visible – a sign of active cells. At later passages cells have a small cytoplasmic processes and the bipolar arrangement of processes changes by stellar arrangement. Golgi complex is also clearly visualized. The indicator of the nuclear-cytoplasmic ratio in MSC from adipose tissue is significantly reduced at 7 passage to 0.2189 ± 0.0122 (P < 0.01), and at 12 passage to 0.1111 ± 0.0086 (P < 0.001) compared to the 2 passage. The coefficient of proliferation of MSC from adipose tissue is significantly reduced at 12th passage. The viability of mesenchymal stem cells from adipose tissue with an increasing of a number of passages significantly reduces and at the 12th passage of cultivation reaches 84,67 ± 1,36* (P < 0.05). The content of apoptotic cells that exhibited sensitivity to serum-free significantly increased at 7 and 12 passages and was respectively 21.33 ± 1.36 (P < 0.05) and 23.67 ± 0.97% (P < 0.05)

    ЗАХВОРЮВАНІСТЬ НА ГОСТРИЙ ВІРУСНИЙ ГЕПАТИТ С У ДНІПРОПЕТРОВСЬКОМУ РЕГІОНІ ТА УКРАЇНІ

    Purpose: to study and analyze the incidence of acute viral hepatitis C in the years 2001–2017 in the Dnipropetrovsk region in comparison with the indicators in Ukraine, and to determine the trends and rates of growth.Materials and Methods. The morbidity analysis was conducted using the epidemiological method. Trends in the incidence calculated in terms of growth and increase.Results. The dynamics of the incidence of acute viral hepatitis C in Ukraine and in the Dnipropetrovsk region as a whole and in separate territories in the period from 2004 to 2017 was wavy, with a more pronounced tendency to lower rates in Ukraine. The average incidence rate in the region was (2.04±0.11) per 100.000 population (95 % CI 1.81–2.27), the average rate of growth was (98.48±4.70) % versus (1.72±0.12) per 100 thousand population in Ukraine (p <0.05), average growth rate – (95.18±3.25) %. The high incidence of acute viral hepatitis C in the cities of the oblast was noted (2.30±0.13) per 100 thousand), which in 1.34 times exceeded its level in the city of Dnipro (p <0.01) and in 1.55 times – the corresponding indicator in rural areas (p <0.001).Conclusions. The incidence of acute viral hepatitis C in the Dnipropetrovsk region and in Ukraine tends to decrease. To implement a global WHO strategy for viral hepatitis, a comprehensive screening program is needed to plan effective prevention and anti-epidemic measures.Мета: вивчити та проаналізувати рівень захворюваності на гострий вірусний гепатит С (ВГС) у багаторічній динаміці за 2001–2017 рp. у Дніпропетровському регіоні, порівняно з показниками в Україні, та визначити тенденції й темпи приросту.Матеріали і методи. Аналіз захворюваності проведено епідеміологічним методом. Тенденції захворюваності розраховували за темпами зростання та приросту.Результати. Динаміка захворюваності на ВГС в Україні та в Дніпропетровському регіоні в цілому і на окремих територіях у період із 2004 до 2017 р. мала хвилеподібний характер з більш вираженою тенденцією до зниження показників в Україні. Середній показник захворюваності в області становив 2,04±0,11 на 100 тис. населення (95 % ДІ 1,81–2,27), середній темп зростання – (98,48±4,70) %. Показник захворюваності в Україні – 1,72±0,12 на 100 тис. населення (p<0,05), середній темп зростання – (95,18±3,25) %. Відзначено високий рівень захворюваності на ВГС у містах області (2,30±0,13 на 100 тис. населення), який в 1,34 разу перевищував її рівень у м. Дніпрі (p<0,01) та в 1,55 разу – відповідний показник у сільських районах (p<0,001).Висновки. Захворюваність на гострий вірусний гепатит С у Дніпропетровській області та в Україні має тенденцію до зниження. Для впровадження глобальної стратегії ВООЗ із вірусних гепатитів необхідно провести широку скринінгову програму при плануванні ефективних профілактичних та протиепідемічних заходів

    Pluripotency reprogramming by competent and incompetent POU factors uncovers temporal dependency for Oct4 and Sox2

    Oct4, along with Sox2 and Klf4 (SK), can induce pluripotency but structurally similar factors like Oct6 cannot. To decode why Oct4 has this unique ability, we compare Oct4-binding, accessibility patterns and transcriptional waves with Oct6 and an Oct4 mutant defective in the dimerization with Sox2 (Oct4(defSox2)). We find that initial silencing of the somatic program proceeds indistinguishably with or without Oct4. Oct6 mitigates the mesenchymal-to-epithelial transition and derails reprogramming. These effects are a consequence of differences in genome-wide binding, as the early binding profile of Oct4(defSox2) resembles Oct4, whilst Oct6 does not bind pluripotency enhancers. Nevertheless, in the Oct6-SK condition many otherwise Oct4-bound locations become accessible but chromatin opening is compromised when Oct4(defSox2) occupies these sites. We find that Sox2 predominantly facilitates chromatin opening, whilst Oct4 serves an accessory role. Formation of Oct4/Sox2 heterodimers is essential for pluripotency establishment; however, reliance on Oct4/Sox2 heterodimers declines during pluripotency maintenance

    A Novel Conductometric Urea Biosensor with Improved Analytical Characteristic Based on Recombinant Urease Adsorbed on Nanoparticle of Silicalite

    Development of a conductometric biosensor for the urea detection has been reported. It was created using a non-typical method of the recombinant urease immobilization via adsorption on nanoporous particles of silicalite. It should be noted that this biosensor has a number of advantages, such as simple and fast performance, the absence of toxic compounds during biosensor preparation, and high reproducibility (RSD = 5.1 %). The linear range of urea determination by using the biosensor was 0.05–15 mM, and a lower limit of urea detection was 20 μM. The bioselective element was found to be stable for 19 days. The characteristics of recombinant urease-based biomembranes, such as dependence of responses on the protein and ion concentrations, were investigated. It is shown that the developed biosensor can be successfully used for the urea analysis during renal dialysis

    There are obtained dependences of phase incursion of retransmitting meter signal, obtained in case of deviations of retransmitter phase characteristics from nominal values. New method of compensation of deviations is supposed for the case of linear law of dependence of phase shift from distance to the controlled object, when it considerably reduces an output error. Replacement of phase shift change is considered taking into account the possibility of processing algorithm modification. It is also shown the necessity of some additions for the input phase shifter circuit

    A modification of the numerical technique for calculating the field and delay of the signal reflected by a flat reflector and based on the Fresnel–Kirchhoff equation of the diffraction scalar field has been proposed. The relationships taking into account the size of antennas and reflector as well as changes of the amplitude and phase characteristics of repeater channels were obtained. It was shown that the signal delay characteristics in the Fresnel zone depended on the size of reflector shifting with the signal delay variations
