    This research aims to analyze the employee\u27s performance of woven sarong industries in Pekalongan. The population is 10 woven sarong industries in Pekalongan with a total respondents size of 105 weaving employees obtained using Purposive Sampling technique. SEM analysis was used to examine the effect of job training andwork motivation as the exogenous variable, competence as the intervening variable, and job performance as the endogenous variable. The results show that job trainning has a positive effect on the competence, competence has a positive effect on the job performance, the competence mediates the relationship between job training and job performance, and work motivation has a positive effect on the job performance

    The Bank has a very important role, namely in processing funds or allocating of public funds to be channeled public funds in the form of credit. Credit control is very important. Therefore, credit must be with prudence and with a good credit control system.This research purpose was to know how to analyze the system and procedure ofworking capital crediting applied by Bank Rakyat Indonesia (Persero), Tbk Kawi Malang to support credit control. This type of research used a descriptive research with qualitative approach. From the research, the researcher knew some weaknesses in the procedures of crediting of working capital and credit control as in the analysis stage credits was the number of tasks. In addition, the credit control was also seen some weaknesses that should had been fixed, such as documents used inadequate, and improved the flow in more easily understood at the time of the credit application to credit disbursement. Advice that cloud be given namely the distribution of the duty for the judgment of assurance or collateral and judgment analysis credit, the form register candidates debitor at the time entreaty credit that contains on the identity of customes to minimize fraud in supplication credit

    The LARGE Principle of Cellular Reprogramming: Lost, Acquired and Retained Gene Expression in Foreskin and Amniotic Fluid-Derived Human iPS Cells

    Human amniotic fluid cells (AFCs) are routinely obtained for prenatal diagnostics procedures. Recently, it has been illustrated that these cells may also serve as a valuable model system to study developmental processes and for application in regenerative therapies. Cellular reprogramming is a means of assigning greater value to primary AFCs by inducing self-renewal and pluripotency and, thus, bypassing senescence. Here, we report the generation and characterization of human amniotic fluid-derived induced pluripotent stem cells (AFiPSCs) and demonstrate their ability to differentiate into the trophoblast lineage after stimulation with BMP2/BMP4. We further carried out comparative transcriptome analyses of primary human AFCs, AFiPSCs, fibroblast-derived iPSCs (FiPSCs) and embryonic stem cells (ESCs). This revealed that the expression of key senescence-associated genes are down-regulated upon the induction of pluripotency in primary AFCs (AFiPSCs). By defining distinct and overlapping gene expression patterns and deriving the LARGE (Lost, Acquired and Retained Gene Expression) Principle of Cellular Reprogramming, we could further highlight that AFiPSCs, FiPSCs and ESCs share a core self-renewal gene regulatory network driven by OCT4, SOX2 and NANOG. Nevertheless, these cell types are marked by distinct gene expression signatures. For example, expression of the transcription factors, SIX6, EGR2, PKNOX2, HOXD4, HOXD10, DLX5 and RAXL1, known to regulate developmental processes, are retained in AFiPSCs and FiPSCs. Surprisingly, expression of the self-renewal-associated gene PRDM14 or the developmental processes-regulating genes WNT3A and GSC are restricted to ESCs. Implications of this, with respect to the stability of the undifferentiated state and long-term differentiation potential of iPSCs, warrant further studies

    Proliferation and survival of human amniotic epithelial cells during their hepatic differentiation

    Stem cells derived from placental tissues are an attractive source of cells for regenerative medicine. Amniotic epithelial cells isolated from human amnion (hAECs) have desirable and competitive characteristics that make them stand out between other stem cells. They have the ability to differentiate toward all three germ layers, they are not tumorigenic and they have immunosuppressive properties. Although liver transplantation is the best way to treat acute and chronic hepatic failure patients, there are several obstacles. Recently, stem cells have been spotlighted as alternative source of hepatocytes because of their potential for hepatogenic differentiation. In this work, we aimed to study the proliferation and survival of the hAECs during their hepatic differentiation. We have also analyzed the changes in pluripotency and hepatic markers. We differentiated amniotic cells applying a specific hepatic differentiation (HD) protocol. We determined by qRT-PCR that hAECs express significant levels of SOX-2, OCT-4 and NANOG during at least 15 days in culture and these pluripotent markers diminish during HD. SSEA-4 expression was reduced during HD, measured by immunofluorescence. Morphological characteristics became more similar to hepatic ones in differentiated cells and representative hepatic markers significantly augmented their expression, measured by qRT-PCR and Western blot. Cells achieved a differentiation efficiency of 75%. We observed that HD induced proliferation and promoted survival of hAECs, during 30 days in culture, evaluated by 3H-thymidine incorporation and MTT assay. HD also promoted changes in hAECs cell cycle. Cyclin D1 expression increased, while p21 and p53 levels were reduced. Immunofluorescence analysis showed that Ki-67 expression was upregulated during HD. Finally, ERK 1/2 phosphorylation, which is intimately linked to proliferation and cell survival, augmented during all HD process and the inhibition of this signaling pathway affected not only proliferation but also differentiation. Our results suggest that HD promotes proliferation and survival of hAECs, providing important evidence about the mechanisms governing their hepatic differentiation.     One of applications for evaluating student learning outcomes is quizizz. In the current era of technological development, students are familiar with the use of smartphones in daily activities, both for school activities and outside of school. Not only students but educators are also required to be able to use smartphones to generate interest in student learning. In this way, educators invite students to use technology as a medium for evaluating learning outcomes, especially learning physics. Researchers conducted research at SMK Taruna Satria Pekanbaru on Jl Delima No. 5 Kec.Tampan, Pekanbaru, Riau. When PLP activities with class X with 20 students which coincided in the odd semester of the academic year 2021/2022. This study aims to determine the responses or perceptions of students after using the quizizz application. The method used quantitative method using a questionnaire for data collection. The data analysis technique uses a questionnaire. The results of the questionnaire were analyzed descriptively using percentages. The results of this study are that students have a response that strongly agrees with the use of the Quizizz application as an evaluation medium. Perception which includes three aspects, namely goals, interests and attitudes. This shows that the use of the Quizizz application is considered to have a good response from students as a medium for evaluating physics learning outcome

    Effects of High-intensity Interval Training Treadmill with Changes in Inclination to Body Fat Mass Percentage of Overweight Men

    A higher body fat percentage reflects an increased risk of comorbidities. High-Intensity Interval Training (HIIT) is a more efficient and effective form of exercise in reducing body fat mass. This study aimed to determine the effects of HIIT treadmill with changes in inclination to body fat mass percentage of overweight men. Twenty-two overweight subjects who met the study criteria were randomly assigned to the intervention group and the control group. The intervention group did HIIT using a treadmill with incline changes for 4 weeks. Measurements of the anthropometrics such as weight, body mass index (BMI), waist and hip ratio (WHR), and body fat mass were performed pre and post the intervention in both groups. There was significant decrease in body fat mass percentage (p=0,000) and WHR (p=0,000) in intervention group and significant difference in body fat mass percentage (p=0,000) in the intervention group compared to the control group (p=0,00). However, there is no significant decrease in BW (p=0.598) and BMI (p=0.592) in the intervention group after exercise.     Effects of High-intensity Interval Training Treadmill with Changes in Inclination on TGF-β2 Serum Level of Overweight Male

    A third of adults in Indonesia were overweight. Overweight and abdominal obesity also have associations with musculoskeletal disorders, limited respiratory function, and decreased physical function and quality of life, so public health interventions are needed to overcome them. High-Intensity Interval Training (HIIT) has more efficient time and has similar effectiveness as endurance exercise. Transforming Growth Factor-β2 (TGF-β2) is secreted from adipose tissue in response to exercise and improves glucose tolerance. Sixteen overweight subjects who met the study criteria were randomly assigned to the intervention and control groups. The intervention group did HIIT using a treadmill with incline changes for 4 weeks. This research measures weight, body mass index (BMI), waist and hip ratio (WHR), body fat mass, and TGF-β2 serum levels pre- and post-intervention in both groups. There was significant decrease in body fat mass percentage (p=0,002) and WHR (p=0,003) but no significant decrease in weight (p=0.51) and BMI (p=0.526) and no significant increase in TGF-β2 serum levels (p=0.208). However, the control got a significant decrease in TGF-β2 serum levels (p=0.029). Although HIIT for 4 weeks did not increase the TGF-β2 serum levels, it is effective to prevent a significant decrease in TGF-β2 serum levels in overweight men
