    Comportamento de bactérias deteriorantes e Salmonella enterica subespécie entérica O:4,5 em carne bovina embalada a vácuo durante refrigeração

    In this study, the kinetic parameters of mesophilic, psychrotrophic and lactic acid bacteria in vacuum-packed beef at 1 °C and 4 °C were estimated from experimental growth curves produced by samples stored during 21 and 60 days, respectively. In a separate experiment, the survival of multidrug resistant (MDR) Salmonella enterica O:4,5 at 1°C was also characterized. The shelf-life of vacuum-packed beef stored at 4 °C was estimated at 16.1 days (95% CI: 14.8 – 17.3 days), whereas at 1 °C it was longer than 21 days because the mesophiles count estimated towards the end of the experiment was 12.5 ln CFU.g-1 (95% CI: 11.8 – 13.3 ln CFU.g-1) which is lower than the shelf-life reference value. At 1 °C, inoculated Salmonella was reduced in 6.61 ln CFU.g-1 (2.87 log CFU.g-1). These results demonstrated the importance of establishing in legislation, especially in Brazil, standard values of deteriorating microorganisms in beef for maintaining product quality.Neste estudo, os parâmetros cinéticos de bactérias mesófilas, psicrotróficas e ácido lácticas foram estimados em carne bovina embalada a vácuo a 1 °C e 4 °C, a partir de curvas experimentais produzidas em amostras estocadas durante 21 e 60 dias, respectivamente. Em um experimento separado, a sobrevivência de Salmonella enterica O:4,5 multirresistente (MDR) a 1°C também foi caracterizada. A vida de prateleira da carne bovina embalada a vácuo, estocada a 4°C, foi estimada em 16.1 dias (95% CI: 14.8 – 17.3 dias), enquanto que a 1 °C o período foi maior que 21 dias, porque a contagem estimada de mesófilos ao final do experimento foi de 12.5 ln UFC.g-1 (95% CI: 11.8 – 13.3 ln UFC.g-1), o qual é mais baixo que o valor referência de shelf-life. A 1 °C, Salmonella inoculada reduziu em 6.61 ln UFC.g-1 (2.87 log UFC.g-1).     HYGIENIC-SANITARY QUALITY OF LETTUCE (Lactuca sativa) AND ARUGULA (Eruca sativa) PRODUCED IN AN ORGANIC FARM SYSTEM IN CUIABÁ, MT, BRAZIL

    Demand for organic food has increased as a result of awareness of the health risks posed by the presence of chemical residues. In this context, this study aimed to verify the safety of lettuce (Lactuca sativa) and arugula (Eruca sativa) produced organically in the metropolitan region of Cuiabá, MT, Brazil, and identify the main microbiological contamination sources of this production system. Fifty-five samples, 10 lettuces and 10 arugula, were collected in a farm and supermarket, in addition to five samples of each agricultural adjuvant used in the production system (irrigation water, vegetable and animal fertilizers). No Salmonella spp. was detected, although thermotolerant coliforms above the maximum acceptable limit established by the Commission on Microbiological Specifications for Foods (ICMSF) were observed in 90% (9/10) and 50% (5/10) of the farm arugula and lettuce samples, 20% (2/10) and 10% (1/10) of the supermarket arugula and lettuce samples, 60% of animal and vegetable fertilizers (6/10) and 40% (2/5) of irrigation water samples. Over half of the vegetable samples analyzed herein were, thus, unfit for consumption, indicating the relatively high influence of system inputs on the hygienic-sanitary quality of the arugula and lettuce produced in the investigated organic farm.Demand for organic food has increased as a result of awareness of the health risks posed by the presence of chemical residues. Nesse contexto, este estudo teve como objetivo verificar a segurança de alface (Lactuca sativa) e rúcula (Eruca sativa) produzidas organicamente na região metropolitana de Cuiabá, MT, Brasil, e identificar as principais fontes de contaminação microbiológica desse sistema de produção. Cinquenta e cinco amostras, sendo 10 de alfaces e 10 de rúculas foram coletadas em uma fazenda e supermercado, além de quinze amostras de adjuvantes agrícola utilizados no sistema de produção. Nenhuma Salmonella spp. foi detectada, embora coliformes termotolerantes acima do limite máximo aceitável estabelecido pela Comissão de Especificações Microbiológicas para Alimentos (ICMSF) tenham sido observados em 90% (9/10) e 50% (5/10) das amostras de rúcula e alface da fazenda, 20 % (2/10) e 10% (1/10) das amostras de rúcula e alface de supermercado, 60% de fertilizantes animais e vegetais (6/10) e 40% (2/5) de amostras de água de irrigação. Mais da metade das amostras de hortaliças aqui analisadas estavam, portanto, impróprias para consumo, indicando a influência relativamente alta dos insumos do sistema na qualidade higiênico-sanitária da rúcula e alface produzidas na fazenda orgânica investigada.     Recomendações técnicas para atualização das boas práticas de fabricação de alimentos (BPF) visando a prevenção da COVID-19 em serviços de alimentação: checklist e revisão de manuais BPF

    The COVID-19 pandemic caused by the SARS-CoV-2 virus brought additional challenges to the Food and Nutrition Service, so in addition to meeting the hygienic-sanitary conditions of food, companies now need to incorporate new manufacturing practices. foods that aim to preserve the health of workers as well as consumers. In this context, this study aimed to carry out a literature review on technical recommendations for good food manufacturing practices related to the prevention of COVID-19 and prepare a checklist to facilitate the identification of failures, assess risk COVID-19 transmission in food services and guide how to adapt the good manufacturing practices manuals. Based on technical notes (NT) 47, 48 and 49 of 2020 that were edited by ANVISA, based on the standards of the World Health Organization (WHO), a questionnaire was created containing 100 questions in the various categories of the food production sector, such as physical structure/building, hand washing conditions, general worker protection measures, material storage area, personal hygiene conditions, food storage area, company personnel, customer service area, preparation area, portioning and distribution area, food consumption area for workers, area for reception and food services received and food delivery services. In this way, we understand that biosafety measures must be adopted from the update of the manuals of good manufacturing practices in food services, and, to support this action, we propose the use of the checklist in the appendix to identify non-conformities related to the prevention of COVID -19.A pandemia da COVID-19 causada pelo vírus SARS-CoV-2 trouxe desafios adicionais ao Serviço de Alimentação e Nutrição (SAN), pois, além de atender as condições higiênicas sanitárias dos alimentos, as empresas agora precisam incorporar novas práticas que visem preservar a saúde das pessoas. Nesse contexto, esse estudo buscou realizar uma revisão da literatura sobre as recomendações técnicas de boas práticas de fabricação de alimentos relacionadas com a prevenção da COVID-19 e elaborar uma lista de verificações (check-list) para facilitar a identificação de falhas, avaliar risco de transmissão da COVID-19 nos serviços de alimentação e orientar as adaptações dos manuais de boas práticas de fabricação. Com base nas notas técnicas vigentes no Brasil e baseadas nas diretrizes da Organização Mundial de Saúde (OMS), foi elaborado um questionário contendo 100 perguntas nas diversas categorias do setor de produção de alimentos. Dessa maneira, entendemos que medidas de biossegurança devem ser adotadas a partir da atualização dos manuais de boas práticas de fabricação nos serviços de alimentação, e, para apoiar essa ação, propomos a utilização do checklist em apêndice para identificar as inconformidades relacionadas a prevenção da COVID-19

    A contaminação de alimentos tem aumentado a cada ano, e atualmente representa um risco potencial para a saúde humana. Staphylococcus spp. foi isolado de alimentos processados e in natura e identificados através dos teste de Gram, catalase, coagulase, DNAase, termonuclease, acetoína (VP) e metabolismo de carboidratos (glicose, maltose e manitol). Foram selecionadas cepas com evidente reação de termonuclease, e realizada análise de enterotoxinas estafilocócicas, por teste imunoenzimático (VIDAS - Staph Enterotoxin - bioMerieux). Reação positiva de enterotoxinas estafilocócicas foram produzidas por S. aureus em camarão, queijo de coalho e macarrão e S. intermedius em peixe cozido e pasta de alho. Observou-se SCP (EstafilococosCoagulase Positiva) em queijo de coalho. Contaminação por Staphylococcus spp.ocorreu durante os estágios de produção ou na estocagem dos alimentos, produzindo toxinas. Conclui-se que estes alimentos representam risco à saúde humana se não forem observadas práticas adequadas de higiene no seu manuseio e no armazenamento.<br>Food contamination has increased in the recent years and actually represents a serious risk for human health. The occurrence of Staphylococcus spp. in nature and processed foods were identified by Gram test, catalase, coagulase, DNAase, acetoin production (AP) and by assays of carbohydrate metabolism (glucose, maltose and mannitol). Strains of Staphylococcus spp were selected by thermonuclease reaction and analysis of staphylococcal enterotoxins, detected by immunoenzymatic assays (VIDAS - Staph Enterotoxin - bio Merieux). Positive reaction for staphylococci enterotoxin was produced by S. aureus in shrimps; farmhouse cheese and macaroni mass; S intermedius in cooked fish and garlic paste. SCP (Staphylococci Coagulase Positive) were observed in farmhouse cheese. Contamination by Staphylococcus sppoccurred during food production and storage. We can conclude that practices of hygiene need to be observed during food production and storage

    Os alimentos são passíveis de contaminação por diferentes agentes etiológicos, podendo levar a doenças manifestadas por ação de microorganismos patogênicos ou suas toxinas. Pesquisou-se a ocorrência de cepas de Staphylococcus, assim como a sua capacidade para produção de enterotoxinas em leite produzido e/ou comercializado no Estado de Pernambuco, Brasil. Foram isoladas e selecionadas 109 cepas de Staphylococcus coagulase positiva e negativa de leite in natura. A identificação das cepas isoladas foi realizada por meio de testes morfológicos e bioquímicos, como: testes de catalase, coagulase, hemólise, DNAse, termonuclease, produção de acetoína (VP) e metabolismo de carboidratos (glicose, maltose e manitol). Das 77 cepas coagulase positivas foram identificadas S. aureus (30), S. hyicus (3), S. intermedius (16), S. aureus identificação presuntiva (13) e Estafilococos Coagulase Positiva (SCP) (15). Das 32 cepas coagulase negativa foram identificadas S. capitis (2), S. carnosus (1), S. chromogenes (6), S. hyicus (1), S. schleiferi (1) e Estafilococos Coagulase Negativa (SCN) (21). Foram selecionadas 43 cepas que apresentaram reações de termonuclease evidentes, para análise de enterotoxinas estafilocócicas, realizada pelo teste imunoenzimático ELISA. Os resultados obtidos evidenciaram dez cepas com reação negativa para enterotoxinas: S. aureus (4), S. carnosus (1), S. chromogenes (2), S. hyicus (2) e S. intermedius (1). Entre as cepas enterotoxina positiva, foram encontrados: S. aureus (17), S. chromogenes (2), S. hyicus (1), S. intermedius (8), S. aureus identificado presuntivamente (2), cepas do grupo SCP (1) e as do SCN (2). As espécies que apresentaram maior número de linhagens enterotoxigênicas foram: S. aureus e S. intermedius. Esses resultados podem ser atribuídos à manipulação inadequada do leite e/ou à recontaminação durante o seu armazenamento e distribuição.<br>Foods are susceptible to contamination for different etiological agents, which can take to manifested diseases by action of pathogenic microorganisms or their toxins. It was researched the occurrence of strains of Staphylococcus and your capacity in producing enterotoxins in milk in natura, that have been produced or commercialized in the State Pernambuco, Brazil. 109 strains of Staphylococus positive and negative coagulase of milk in natura were selected. The identification of the isolated strains was accomplished through morphologic and biochemical tests as: catalase, coagulase, haemolysins, DNAse, thermonuclease, acetoin production (VP) and carbohydrates metabolism (glucose, maltose and mannitol). From the 77 coagulase positive strains 30 were identified as S. aureus, 3 as S. hyicus, 16 as S. intermedius, 13 as S. aureus identification presumptive and 15 as SCP. Among 32 coagulase negative strains 2 were identified as S. capitis, 1 as S. carnosus, 6 as S. chromogenes, 1 as S. hyicus, 1 as S. schleiferi and 21 as SCN. Fourty-three strains that presented very evident thermonuclease reaction, were selected in order to perform for staphylococcal enterotoxins analysis by the immuno enzimatic test (ELFA). 10 strains showed negative reaction for enterotoxins: S. aureus (4), S. carnosus (1), S. chromogenes (2), S. hyicus (2) and S. intermedius (1). Strains that gave positive results, were S. aureus (17), S. chromogenes (2), S. hyicus (1), S. intermedius (8), S. aureus identified presumptively (2) and of the groups SCP (1) and SCN (2). The species that presented larger number of enterotoxigenics strains were S. aureus and S. intermedius.     <i>Salmonella</i> spp. in Aquaculture: An Exploratory Analysis (Integrative Review) of Microbiological Diagnoses between 2000 and 2020

    The present study aimed to characterize, through descriptive statistics, data from scientific articles selected in a systematic integrative review that performed a microbiological diagnosis of Salmonella spp. in aquaculture. Data were obtained from research articles published in the BVS, Scielo, Science Direct, Scopus and Web of Science databases. The selected studies were published between 2000 and 2020 on samples of aquaculture animal production (fish, shrimp, bivalve mollusks, and other crustaceans) and environmental samples of aquaculture activity (farming water, soil, and sediments). After applying the exclusion criteria, 80 articles were selected. Data such as country of origin, categories of fish investigated, methods of microbiological diagnosis of Salmonella spp., sample units analyzed and most reported serovars were mined. A textual analysis of the word cloud and by similarity and descending hierarchical classification with the application of Reinert’s algorithm was performed using R® and Iramuteq® software. The results showed that a higher percentage of the selected articles came from Asian countries (38.75%). Fish was the most sampled category, and the units of analysis of the culture water, muscle and intestine were more positive. The culture isolation method is the most widespread, supported by more accurate techniques such as PCR. The most prevalent Salmonella serovars reported were S. Typhimurium, S. Weltevreden and S. Newport. The textual analysis showed a strong association of the terms “Salmonella”, “fish” and “water”, and the highest hierarchical class grouped 25.4% of the associated text segments, such as “aquaculture”, “food” and “public health”. The information produced characterizes the occurrence of Salmonella spp. in the aquaculture sector, providing an overview of recent years. Future research focusing on strategies for the control and prevention of Salmonella spp. in fish production are necessary and should be encouraged
