    Temporal dynamics of spectral reflectance and vegetation indices during canola crop cycle in southern Brazil

    The objective of this study was to characterize the variability of spectral reflectance and temporal profiles of vegetation indices associated with nitrogen fertilization, crop cycle periods, and weather conditions of the growing season in canola canopies in southern Brazil. An experiment was carried out during the 2013 and 2014 canola growing seasons at EMBRAPA Trigo, Passo Fundo, state of Rio Grande do Sul, Brazil. The experiment was conducted in a randomized block design with four replications. Five doses of nitrogen top dressing were used as treatments: 10, 20, 40, 80, and 160kg ha-1. Measurements were obtained with the spectroradiometer positioned above the canopy, to construct spectral reflectance curves for canola and establish temporal profiles for several vegetation indices (SR, NDVI, EVI, SAVI, and GNDVI). In addition, data on shoot dry matter were obtained and phenological stages were determined. The spectral reflectance curves of canola were reported to change with canopy growth and development. Temporal profiles of vegetation indices showed two maximum peaks, one before flowering and other after flowering. The indices SR, NDVI, EVI, SAVI, and GNDVI were able to characterize changes in the canola canopy over time, as a function of phenological phases, weather conditions, and nitrogen fertilization, throughout the development cycle.     Charge effect on the photoinactivation of Gram-negative and Gram-positive bacteria by cationic meso-substituted porphyrins

    <p>Abstract</p> <p>Background</p> <p>In recent times photodynamic antimicrobial therapy has been used to efficiently destroy Gram (+) and Gram (-) bacteria using cationic porphyrins as photosensitizers. There is an increasing interest in this approach, namely in the search of photosensitizers with adequate structural features for an efficient photoinactivation process. In this study we propose to compare the efficiency of seven cationic porphyrins differing in <it>meso</it>-substituent groups, charge number and charge distribution, on the photodynamic inactivation of a Gram (+) bacterium (<it>Enterococcus faecalis</it>) and of a Gram (-) bacterium (<it>Escherichia coli</it>). The present study complements our previous work on the search for photosensitizers that might be considered good candidates for the photoinactivation of a large spectrum of environmental microorganisms.</p> <p>Results</p> <p>Bacterial suspension (10<sup>7 </sup>CFU mL<sup>-1</sup>) treated with different photosensitizers concentrations (0.5, 1.0 and 5.0 μM) were exposed to white light (40 W m<sup>-2</sup>) for a total light dose of 64.8 J cm<sup>-2</sup>. The most effective photosensitizers against both bacterial strains were the Tri-Py<sup>+</sup>-Me-PF and Tri-Py<sup>+</sup>-Me-CO<sub>2</sub>Me at 5.0 μM with a light fluence of 64.8 J cm<sup>-2</sup>, leading to > 7.0 log (> 99,999%) of photoinactivation. The tetracationic porphyrin also proved to be a good photosensitizer against both bacterial strains. Both di-cationic and the monocationic porphyrins were the least effective ones.</p> <p>Conclusion</p> <p>The number of positive charges, the charge distribution in the porphyrins' structure and the <it>meso</it>-substituent groups seem to have different effects on the photoinactivation of both bacteria. As the Tri-Py<sup>+</sup>-Me-PF porphyrin provides the highest log reduction using lower light doses, this photosensitizer can efficiently photoinactivate a large spectrum of environmental bacteria. The complete inactivation of both bacterial strains with low light fluence (40 W m<sup>-2</sup>) means that the photodynamic approach can be applied to wastewater treatment under natural light conditions which makes this technology cheap and feasible in terms of the light source.</p

    Performance, feed utilization, and hepatic metabolic response of weaned juvenile Atlantic bluefin tuna (Thunnus thynnus L.): effects of dietary lipid level and source

    The development of formulated diets and feeds is essential to increase production of farmed tuna species. There is limited knowledge of this topic, mainly on Pacific Bluefin tuna (Thunnus orientalis) in Japan, whereas no major attempts have been made with Atlantic Bluefin tuna (Thunnus thynnus; ABT). In the present study, two trials were performed using inert formulated diets as on-growing feeds for weaned ABT juvenile in order to establish adequate dietary levels of both lipid and omega-3 long-chain polyunsaturated fatty acids (LC-PUFA). In a first trial, ABT (initial weight = 2.9±0.9g) were fed for 10 days with either a commercial (Magokoro®, MGK) or two experimental feeds with two different lipid levels (15 or 20%) using krill oil (KO) as the single lipid source in order to estimate the suitable lipid content. Fish fed MGK displayed the highest growth, followed by 15KO, with no differences in fish survival. Thus, a lipid content of 15% was considered better than 20% for ABT juveniles. In the second trial, fish (initial weight = 3.3 ± 0.6g) were fed either MGK, 15KO or a feed containing 15% lipid with a combination (1:1, v/v) KO and rapeseed oil (RO) (15KORO). Fish fed 15KO and 15KORO showed the highest growth in terms of weight and fork length (including weight gain and SGR). Increasing dietary lipid level or adding RO to the feeds did not increase liver lipid content. The liver fatty acid profile largely reflected dietary intake confirming very limited LC-PUFA biosynthetic activity for this teleost species. In this respect, liver of fish fed 15KO and 20KO displayed the highest contents of docosahexaenoic acid (DHA). The hepatic expression of genes of lipid and fatty acid metabolism, transcription factors, and antioxidant enzymes was investigated with many of the genes showing regulation by both dietary lipid and LC-PUFA contents. The present study showed promising results that suggested ABT juveniles can be on grown on inert dry feeds that supported good fish growth and the accumulation of the health-promoting fatty acid DHA. Further studies are required in order to fully elucidate lipid and fatty acid requirements of this iconic species regarding dietary sources and production costs.En prensa1,52

    Acyl ghrelin improves cognition, synaptic plasticity deficits and neuroinflammation following amyloid beta (Aβ1-40) administration in mice

    Ghrelin is a metabolic hormone that has neuroprotective actions in a number of neurological conditions, including Parkinson's disease (PD), stroke and traumatic brain injury. Acyl ghrelin treatment in vivo and in vitro also shows protective capacity in Alzheimer's disease (AD). In the present study, we used ghrelin knockout (KO) and their wild-type littermates to test whether or not endogenous ghrelin is protective in a mouse model of AD, in which human amyloid β peptide 1-40 (Aβ1-40 ) was injected into the lateral ventricles i.c.v. Recognition memory, using the novel object recognition task, was significantly impaired in ghrelin KO mice and after i.c.v. Aβ1-40 treatment. These deficits could be prevented by acyl ghrelin injections for 7 days. Spatial orientation, as assessed by the Y-maze task, was also significantly impaired in ghrelin KO mice and after i.c.v. Aβ1-40 treatment. These deficits could be prevented by acyl ghrelin injections for 7 days. Ghrelin KO mice had deficits in olfactory discrimination; however, neither i.c.v. Aβ1-40 treatment, nor acyl ghrelin injections affected olfactory discrimination. We used stereology to show that ghrelin KO and Aβ1-40 increased the total number of glial fibrillary acidic protein expressing astrocytes and ionised calcium-binding adapter expressing microglial in the rostral hippocampus. Finally, Aβ1-40 blocked long-term potentiation induced by high-frequency stimulation and this effect could be acutely blocked with co-administration of acyl ghrelin. Collectively, our studies demonstrate that ghrelin deletion affects memory performance and also that acyl ghrelin treatment may delay the onset of early events of AD. This supports the idea that acyl ghrelin treatment may be therapeutically beneficial with respect to restricting disease progression in AD
