Hymenaea courbaril L. (Fabaceae) is a tree species popularly known as 'jatobá'. Its wood is largely sold and its fruits are used as a food and medicinal resources. This species has been subjected to logging and habitat destruction. The knowledge about genetic polymorphism is important for the conservation of species affected by anthropic action. In this study, we analyzed the genetic diversity of natural populations of Hymenaea courbaril in the Mato Grosso Amazon. Seven microsatellite primers were used to genotyping of 43 individuals distributed in five populations in the north region of Mato Grosso State. All the seven primers were polymorphic, with 72 alleles detected, averaging 10.29 per locus. Expected and observed heterozygosities averaged 0.85 and 0.29 per locus, respectively. The fixation index was positive for all populations, suggesting excess homozygotes, probably due to inbreeding. Molecular analysis of variance revealed that 85.33% of genetic diversity is within the populations. Based on the UPGMA dendrogram, the individuals were divided into six groups. In the clustering provided by Structure software, four groups were formed which did not correspond to the geographic distribution of the individuals, indicating that the populations are not geographically structured. All sampled populations present elevated levels of genetic diversity, and so they can be used for conservation projects. Genetic variation is higher within the analyzed populations; therefore, several individuals must be conserved per population

Endotoxin exposure during sensitization to Blomia tropicalis allergens shifts TH2 immunity towards a TH17-mediated airway neutrophilic inflammation: role of TLR4 and TLR2

Experimental evidence and epidemiological studies indicate that exposure to endotoxin lipopolysaccharide (eLPS) or other TLR agonists prevent asthma. We have previously shown in the OVA-model of asthma that eLPS administration during alum-based allergen sensitization blocked the development of lung TH2 immune responses via MyD88 pathway and IL-12/IFN-γ axis. In the present work we determined the effect of eLPS exposure during sensitization to a natural airborne allergen extract derived from the house dust mite Blomia tropicalis (Bt). Mice were subcutaneously sensitized with Bt allergens co-adsorbed onto alum with or without eLPS and challenged twice intranasally with Bt. Cellular and molecular parameters of allergic lung inflammation were evaluated 24 h after the last Bt challenge. Exposure to eLPS but not to ultrapure LPS (upLPS) preparation during sensitization to Bt allergens decreased the influx of eosinophils and increased the influx of neutrophils to the airways. Inhibition of airway eosinophilia was not observed in IFN-γdeficient mice while airway neutrophilia was not observed in IL-17RA-deficient mice as well in mice lacking MyD88, CD14, TLR4 and, surprisingly, TLR2 molecules. Notably, exposure to a synthetic TLR2 agonist (PamCSK4) also induced airway neutrophilia that was dependent on TLR2 and TLR4 molecules. In the OVA model, exposure to eLPS or PamCSK4 suppressed OVA-induced airway inflammation. Our results suggest that B. tropicalis allergens engage TLR4 that potentiates TLR2 signaling. This dual TLR activation during sensitization results in airway neutrophilic inflammation associated with increased frequency of lung TH17 cells. Our work highlight the complex interplay between bacterial products, house dust mite allergens and TLR signaling in the induction of different phenotypes of airway inflammation.State of Sao Paulo Foundation for Research Support (FAPESP - 07/03031-3)State of Sao Paulo Foundation for Research Support (FAPESP - 11/17880-8)CAPES, Brazilian Council of Scientific and Technologic Developmen

A Relativistic Study of the Electronic and Magnetic Properties of Cerocene and Thorocene and Its Anions

In this study, we evaluated the importance of the relativistic effects on the optical and magnetic properties of cerocene and thorocene and its corresponding anions. The optimized molecular structures show <i>D</i>8<i>h</i> symmetry for all systems, in good agreement with the experimental data. Atomic charges were analyzed using different approaches (Mulliken, AIM, multipole, and NBO), and the results suggest that the net charge on the thorium is greater than on the cerium atom; however, none of the methodologies were able to predict the expected net charge Ce(III) and Th(IV) atoms. However, by an energy decomposition analysis, a significant electrostatic, ionic, interaction, ∼58% and ∼61%, was found between the metal and the COT2– rings, respectively. The calculated electronic excitations are underestimated in comparison with the experimental data, while the calculated EPR <i>g</i>-tensors are in agreement with previous theoretical and experimental data. Besides, the NICS analysis shows an increased ring electron delocalization due to the lanthanide and actinide metals

Dual Emissions of Salicylidene-5-chloroaminepyridine Due to Excited State Intramolecular Proton Transfer: Dynamic Photophysical and Theoretical Studies

    No full text
In this study, the photophysical properties of <i>N</i>-salycilidene-5-chloroaminopyridine were evaluated. This molecule presented dual emission from the keto* and enol* species, depending on the solvent used (DMSO or methanol in the absence and presence of trifluoroacetic acid). This molecule was synthesized through a dehydration reaction of salicylaldehyde under sonication and characterized using FTIR, cyclic voltammetry, mass spectrometry, and single-crystal X-ray diffraction, yielding the molecular structure. Theoretical calculations were performed under the DFT framework and its time-dependent counterpart. Our results suggest the occurrence of excited state proton transfer, ESIPT, in the aprotic solvent, leading to a keto*–enol* balance, which is confirmed by the emission bands observed at 425 and 545 nm, respectively. However, in a protic environment (methanol or DMSO with trifluoroacetic acid), only the enol form is emissive due to the hydrogen bonding between the iminic group and the solvent. Molecules with double emissions are potentially useful for white light emitting diodes

The aim of this study was to evaluate the effects of the protein-calorie malnutrition in BALB/c isogenic mice infected with Lacazia loboi, employing nutritional and histopathological parameters. Four groups were composed: G1: inoculated with restricted diet, G2: not inoculated with restricted diet, G3: inoculated with regular diet, G4: not inoculated with regular diet. Once malnutrition had been imposed, the animals were inoculated intradermally in the footpad and after four months, were sacrificed for the excision of the footpad, liver and spleen. The infection did not exert great influence on the body weight of the mice. The weight of the liver and spleen showed reduction in the undernourished groups when compared to the nourished groups. The macroscopic lesions, viability index and total number of fungi found in the footpads of the infected mice were increased in G3 when compared to G1. Regarding the histopathological analysis of the footpad, a global cellularity increase in the composition of the granuloma was observed in G3 when compared to G1, with large numbers of macrophages and multinucleated giant cells, discrete numbers of lymphocytes were present in G3 and an increase was observed in G1. The results suggest that there is considerable interaction between Jorge Lobo's disease and nutrition
