    Identification of New Genes Related to Virulence of Xanthomonas axonopodis Pv. Citri during Citrus Host Interactions

    A mutant library of the bacterium Xanthomonas citri subsp. citri strain 306 pathotype A (Xac), the causative agent of most aggressive Asiatic type A citrus canker, was screened regarding altered canker symptoms after inoculations into Citrus sinensis and Citrus limonia host leaves. Twenty-six mutants have shown phenotypic virulence changes and have respectively knocked out gene identified by sequencing. In vivo growth curves were obtained for nine mutants to quantify how the mutations could affect pathogen's adaptability to growth inside and attack host plant infected tissue. Among identified genes in mutated strains, we could find those that until now had not been reported as being involved in Xac adaptation and/or virulence, such as predicted to encode for xylose repressor-like protein (XAC Delta xylR), Fe-S oxidoredutase (XAC Delta aslB), helicase IV (XAC Delta helD), ubiquinol cytochrome c oxidoreductase iron-sulfur subunit (XAC Delta petA), chromosome partitioning protein (XAC Delta parB) and cell division protein FtsB (XAC Delta ftsB), in addition to genes predicted to encode for hypothetical proteins.     A streamlined approach by a combination of bioindication and geostatistical methods for assessing air contaminants and their effects on human health in industrialized areas: A case study in Southern Brazil

Industrialization in developing countries associated with urban growth results in a number of economic benefits, especially in small or medium-sized cities, but leads to a number of environmental and public health consequences. This problem is further aggravated when adequate infrastructure is lacking to monitor the environmental impacts left by industries and refineries. In this study, a new protocol was designed combining biomonitoring and geostatistics to evaluate the possible effects of shale industry emissions on human health and wellbeing. Futhermore, the traditional and expensive air quality method based on PM2.5 measuring was also used to validate the low-cost geostatistical approach. Chemical analysis was performed using Energy Dispersive X-ray Fluorescence Spectrometer (EDXRF) to measure inorganic elements in tree bark and shale retorted samples in São Mateus do Sul city, Southern Brazil. Fe, S, and Si were considered potential pollutants in the study area. Distribution maps of element concentrations were generated from the dataset and used to estimate the spatial behavior of Fe, S, and Si and the range from their hot spot(s), highlighting the regions sorrounding the shale refinery. This evidence was also demonstrated in the measurements of PM2.5 concentrations, which are in agreement with the information obtained from the biomonitoring and geostatistical model. Factor and descriptive analyses performed on the concentrations of tree bark contaminants suggest that Fe, S, and Si might be used as indicators of industrial emissions. The number of cases of respiratory diseases obtained from local basic health unit were used to assess a possible correlation between shale refinery emissions and cases of repiratory disease. These data are public and may be accessed on the website of the the Brazilian Ministry of Health. Significant associations were found between the health data and refinery activities. The combination of the spatial characterization of air pollution and clinical health data revealed that adverse effects were significant for individuals over 38 years of age. These results also suggest that a protocol designed to monitor urban air quality may be an effective and low-cost strategy in environmentally contaminated cities, especially in low- and middle-income countries.Link_to_subscribed_fulltex

    New genes of Xanthomonas citri subsp. citri involved in pathogenesis and adaptation revealed by a transposon-based mutant library

    <p>Abstract</p> <p>Background</p> <p>Citrus canker is a disease caused by the phytopathogens <it>Xanthomonas citri </it>subsp. <it>citri</it>, <it>Xanthomonas fuscans </it>subsp. <it>aurantifolli </it>and <it>Xanthomonas alfalfae </it>subsp. <it>citrumelonis</it>. The first of the three species, which causes citrus bacterial canker type A, is the most widely spread and severe, attacking all citrus species. In Brazil, this species is the most important, being found in practically all areas where citrus canker has been detected. Like most phytobacterioses, there is no efficient way to control citrus canker. Considering the importance of the disease worldwide, investigation is needed to accurately detect which genes are related to the pathogen-host adaptation process and which are associated with pathogenesis.</p> <p>Results</p> <p>Through transposon insertion mutagenesis, 10,000 mutants of <it>Xanthomonas citri </it>subsp. <it>citri </it>strain 306 (Xcc) were obtained, and 3,300 were inoculated in Rangpur lime (<it>Citrus limonia</it>) leaves. Their ability to cause citrus canker was analyzed every 3 days until 21 days after inoculation; a set of 44 mutants showed altered virulence, with 8 presenting a complete loss of causing citrus canker symptoms. Sequencing of the insertion site in all 44 mutants revealed that 35 different ORFs were hit, since some ORFs were hit in more than one mutant, with mutants for the same ORF presenting the same phenotype. An analysis of these ORFs showed that some encoded genes were previously known as related to pathogenicity in phytobacteria and, more interestingly, revealed new genes never implicated with <it>Xanthomonas </it>pathogenicity before, including hypothetical ORFs. Among the 8 mutants with no canker symptoms are the <it>hrpB4 </it>and <it>hrpX </it>genes, two genes that belong to type III secretion system (TTSS), two hypothetical ORFS and, surprisingly, the <it>htrA </it>gene, a gene reported as involved with the virulence process in animal-pathogenic bacteria but not described as involved in phytobacteria virulence. Nucleic acid hybridization using labeled cDNA probes showed that some of the mutated genes are differentially expressed when the bacterium is grown in citrus leaves. Finally, comparative genomic analysis revealed that 5 mutated ORFs are in new putative pathogenicity islands.</p> <p>Conclusion</p> <p>The identification of these new genes related with Xcc infection and virulence is a great step towards the understanding of plant-pathogen interactions and could allow the development of strategies to control citrus canker.</p

    Proteomics-based identification of differentially abundant proteins reveals adaptation mechanisms of Xanthomonas citri subsp citri during Citrus sinensis infection

    Background: Xanthomonas citri subsp. citri (Xac) is the causal agent of citrus canker. A proteomic analysis under in planta infectious and non-infectious conditions was conducted in order to increase our knowledge about the adaptive process of Xac during infection. Results: For that, a 2D-based proteomic analysis of Xac at 1, 3 and 5 days after inoculation, in comparison to Xac growth in NB media was carried out and followed by MALDI-TOF-TOF identification of 124 unique differentially abundant proteins. Among them, 79 correspond to up-regulated proteins in at least one of the three stages of infection. Our results indicate an important role of proteins related to biofilm synthesis, lipopolysaccharides biosynthesis, and iron uptake and metabolism as possible modulators of plant innate immunity, and revealed an intricate network of proteins involved in reactive oxygen species adaptation during Plants'Oxidative Burst response. We also identified proteins previously unknown to be involved in Xac-Citrus interaction, including the hypothetical protein XAC3981. A mutant strain for this gene has proved to be non-pathogenic in respect to classical symptoms of citrus canker induced in compatible plants.     O gĂȘnero Cattleya Lindl. (Orchidaceae: Laeliinae) no Rio Grande do Sul, Brasil

    Cattleya presents about 120 species distributed in tropical and subtropical regions of the Americas. In its current delimitation, Cattleya also encompasses the Brazilian species formerly placed within the genera Laelia and Sophronitis, as well as the species of a number of recently segregated genera. This contribution deals with this genus in Rio Grande do Sul State, Southern Brazil, therefore contributing to the knowledge of its orchid flora. This work is based in literature and in the analysis of herbarium vouchers as well as in specimens collected during fieldwork in several localities of Rio Grande do Sul. Five native species were recorded: Cattleya cernua, C. coccinea, C. intermedia, C. purpurata and C. tigrina. All these species dwell within the Atlantic Rain Forest biome. This contribution does present an artificial key for the species, the synonymies, descriptions, illustrations, geographic distribution and comments on habitat and natural history of the species.Cattleya abrange aproximadamente 120 espĂ©cies distribuĂ­das principalmente nas regiĂ”es tropicais e subtropicais das AmĂ©ricas. Em sua delimitação atual, Cattleya inclui tambĂ©m as espĂ©cies brasileiras outrora inseridas nos gĂȘneros Laelia e Sophronitis, bem como as espĂ©cies de gĂȘneros mais recentemente segregados. O presente trabalho trata do estudo taxonĂŽmico desse gĂȘnero no Rio Grande do Sul, contribuindo, portanto, para o conhecimento da flora do estado.     The Complex Puzzle of Interactions Among Functional Food, Gut Microbiota, and Colorectal Cancer

    Colorectal cancer exerts a strong influence on the epidemiological panorama worldwide, and it is directly correlated to etiologic factors that are substantiated by genetic and environmental elements. This complex mixture of factors also has a relationship involving the structural dependence and composition of the gut microbiome, leading to a dysbacteriosis process that may evolve to serious modifications in the intestinal lining, eventually causing the development of a neoplasm. The gastrointestinal tract presents defense strategies and immunological properties that interfere in intestinal permeability, inhibiting the bacterial translocation, thus maintaining the integrity of intestinal homeostasis. The modulation of the intestinal microbiome and the extinction of risk factors associated with intestinal balance losses, especially of environmental factors, make cell and defense alterations impossible. This modulation may be conducted by means of functional foods in the diet, especially soluble fibers, polyunsaturated fatty acids, antioxidants and prebiotics that signal immunomodulatory effects in the intestinal microbiota, with preventive and therapeutic action for colorectal cancer. In summary, this review focuses on the importance of dietary modulation of the intestinal microbiota as an instrument for dysbacteriosis and, consequently, for the prevention of colorectal cancer, suggesting anticarcinogenic, and antiangiogenic properties.     New endoperoxides highly active in vivo and in vitro against artemisinin-resistant Plasmodium falciparum

    Background: The emergence and spread of Plasmodium falciparum resistance to artemisinin-based combination therapy in Southeast Asia prompted the need to develop new endoperoxide-type drugs. Methods: A chemically diverse library of endoperoxides was designed and synthesized. The compounds were screened for in vitro and in vivo anti-malarial activity using, respectively, the SYBR Green I assay and a mouse model. Ring survival and mature stage survival assays were performed against artemisinin-resistant and artemisinin-sensitive P. falciparum strains. Cytotoxicity was evaluated against mammalian cell lines V79 and HepG2, using the MTT assay. Results: The synthesis and anti-malarial activity of 21 new endoperoxide-derived compounds is reported, where the peroxide pharmacophore is part of a trioxolane (ozonide) or a tetraoxane moiety, flanked by adamantane and a substituted cyclohexyl ring. Eight compounds exhibited sub-micromolar anti-malarial activity (IC50 0.3–71.1 nM), no cross-resistance with artemisinin or quinolone derivatives and negligible cytotoxicity towards mammalian cells. From these, six produced ring stage survival < 1% against the resistant strain IPC5202 and three of them totally suppressed Plasmodium berghei parasitaemia in mice after oral administration. Conclusion: The investigated, trioxolane–tetrazole conjugates LC131 and LC136 emerged as potential anti-malarial candidates; they show negligible toxicity towards mammalian cells, ability to kill intra-erythrocytic asexual stages of artemisinin-resistant P. falciparum and capacity to totally suppress P. berghei parasitaemia in mice.info:eu-repo/semantics/publishedVersio
