    Neste artigo procuramos relatar a situação mundial e brasileira com relação aos micronutrientes, em especial sobre os minerais. Os elementos químicos minerais desempenham funções de grande importância no organismo humano, sendo indispensáveis para o desenvolvimento e a saúde dos indivíduos. Ainda não existe uma avaliação global do estado nutricional dos indivíduos em relação a esses micronutrientes no Brasil, mas os estudos existentes apontam para a necessidade do acompanhamento das tendências alimentares que poderiam levar às suas deficiências com conseqüências adversas para a saúde da população e o desenvolvimento do nosso país.In this paper we will try to report the Brazilian micronutrients status, as well as in worldwide, specifically for minerals.     Alkaline phosphatases in the complex chronic kidney disease-mineral and bone disorders

    Alkaline phosphatases (APs) remove the phosphate (dephosphorylation) needed in multiple metabolic processes (from many molecules such as proteins, nucleotides, or pyrophosphate). Therefore, APs are important for bone mineralization but paradoxically they can also be deleterious for other processes, such as vascular calcification and the increasingly known cross-talk between bone and vessels. A proper balance between beneficial and harmful activities is further complicated in the context of chronic kidney disease (CKD). In this narrative review, we will briefly update the complexity of the enzyme, including its different isoforms such as the bone-specific alkaline phosphatase or the most recently discovered B1x. We will also analyze the correlations and potential discrepancies with parathyroid hormone and bone turnover and, most importantly, the valuable recent associations of AP's with cardiovascular disease and/or vascular calcification, and survival. Finally, a basic knowledge of the synthetic and degradation pathways of APs promises to open new therapeutic strategies for the treatment of the CKD-Mineral and Bone Disorder (CKD-MBD) in the near future, as well as for other processes such as sepsis, acute kidney injury, inflammation, endothelial dysfunction, metabolic syndrome or, in diabetes, cardiovascular complications. However, no studies have been done using APs as a primary therapeutic target for clinical outcomes, and therefore, AP's levels cannot yet be used alone as an isolated primary target in the treatment of CKD-MBD. Nonetheless, its diagnostic and prognostic potential should be underlined

    Experimental and computational studies on the formation of cyanate from early metal terminal nitrido ligands and carbon monoxide

    An important challenge in the artificial fixation of N[subscript 2] is to find atom efficient transformations that yield value-added products. Here we explore the coordination complex mediated conversion of ubiquitous species, CO and N[subscript 2], into isocyanate. We have conceptually split the process into three steps: (1) the six-electron splitting of dinitrogen into terminal metal nitrido ligands, (2) the reduction of the complex by two electrons with CO to form an isocyanate linkage, and (3) the one electron reduction of the metal isocyanate complex to regenerate the starting metal complex and release the product. These steps are explored separately in an attempt to understand the limitations of each step and what is required of a coordination complex in order to facilitate a catalytic cycle. The possibility of this cyanate cycle was explored with both Mo and V complexes which have previously been shown to perform select steps in the sequence. Experimental results demonstrate the feasibility of some of the steps and DFT calculations suggest that, although the reduction of the terminal metal nitride complex by carbon monoxide should be thermodynamically favorable, there is a large kinetic barrier associated with the change in spin state which can be avoided in the case of the V complexes by an initial binding of the CO to the metal center followed by rearrangement. This mandates certain minimal design principles for the metal complex: the metal center should be sterically accessible for CO binding and the ligands should not readily succumb to CO insertion reactions.National Science Foundation (U.S.) (CHE-1111357

    Epigenetic control of EMT/MET dynamics: HNF4α impacts DNMT3s through miRs-29

    Background and aims: Epithelial-to-mesenchymal transition (EMT) and the reverse mesenchymal-to-epithelial transition (MET) are manifestations of cellular plasticity that imply a dynamic and profound gene expression reprogramming. While a major epigenetic code controlling the coordinated regulation of a whole transcriptional profile is guaranteed by DNA methylation, DNA methyltransferase (DNMT) activities in EMT/MET dynamics are still largely unexplored. Here, we investigated the molecular mechanisms directly linking HNF4α, the master effector of MET, to the regulation of both de novo of DNMT 3A and 3B. Methods: Correlation among EMT/MET markers, microRNA29 and DNMT3s expression was evaluated by RT-qPCR, Western blotting and immunocytochemical analysis. Functional roles of microRNAs and DNMT3s were tested by anti-miRs, microRNA precursors and chemical inhibitors. ChIP was utilized for investigating HNF4α DNA binding activity. Results: HNF4α silencing was sufficient to induce positive modulation of DNMT3B, in in vitro differentiated hepatocytes as well as in vivo hepatocyte-specific Hnf4α knockout mice, and DNMT3A, in vitro, but not DNMT1. In exploring the molecular mechanisms underlying these observations, evidence have been gathered for (i) the inverse correlation between DNMT3 levels and the expression of their regulators miR-29a and miR- 29b and (ii) the role of HNF4α as a direct regulator of miR-29a-b transcription. Notably, during TGFβ-induced EMT, DNMT3s' pivotal function has been proved, thus suggesting the need for the repression of these DNMTs in the maintenance of a differentiated phenotype. Conclusions: HNF4α maintains hepatocyte identity by regulating miR-29a and -29b expression, which in turn control epigenetic modifications by limiting DNMT3A and DNMT3B levels

    Efeito da suplementação com ferro na biodisponibilidade de zinco em uma dieta regional do nordeste do Brasil

    Foram investigados os efeitos da suplementação com ferro na biodisponibilidade de zinco de uma dieta "regional" do Nordeste (DRNE), em ratos albinos Wistar, consumindo rações à base da referida dieta (DRNE) e rações controle. As rações DRNE, continham 16 mg de Zn/kg e níveis de 35 mg, 70 mg e 140mg Fe/kg. As rações controle foram elaboradas segundo o "Committee on Laboratory Animal Diets", contendo níveis de proteína, ferro e zinco ajustados aos das rações experimentais DRNE. Os parâmetros utilizados para medir a biodisponibilidade do zinco foram: Índice de Absorção Aparente do Zn e nível total de Zn nos fêmures. Os resultados obtidos demonstraram que a suplementação com ferro diminuiu a biodisponibilidade do Zn, e os efeitos dessa interferência foram influenciados pela qualidade da dieta e pelas proporções Fe:Zn. Tal fato deve ser considerado nas práticas que envolvem fortificação de alimentos e/ou suplementos medicamentosos, comuns nas populações com carências nutricionais.The effects of supplementation with iron on the zinc bioavailability of the regional diet of northeastern Brazil (RDN), were investigated. One assay with Wistar rats, feed on RDN and control diets was carried out. The RDN diets contained 16 mg Zn/kg and levels of 35 mg, 70 mg and 140 mg Fe/kg, respectively. The control diets were prepared according to the standards of the Committee on Laboratory Animal Diets, with levels of protein, iron and zinc identical to those of RDN diets. Index of apparent absorption and zinc retained in the femur of the animals were the parameters utilized to measure zinc bioavailability. The results demonstrated that the supplementation with iron decreased the zinc bioavailability, and the effects were seen to affect diet quality and the Fe:Zn ratio. This fact must be taken into consideration in practices such as the fortying of foodstuffs and the administration of vitamin-mineral supplements to populations with nutritional deficiencies

    Serum albumins are efficient delivery systems for the photosensitizer hypericin in photosensitization-based treatments against Staphylococcus aureus

    The naturally occurring photosensitizer hypericin can be effectively delivered to bacterial cells using serum albumins as biocompatible carriers. Using a combination of spectroscopic methods we demonstrate that the photophysics of hypericin is fully preserved when bound to these proteins. Thanks to the excellent transport capabilities of serum albumins, that deliver hypericin to Gram-positive S. aureus, an efficient antibacterial action was observed, with a reduction of up to 8 log in the number of colony-forming units. The photoactive material is fully compatible with the use in food processing environments, and may be exploited as a viable method for decontamination from S. aureus and other Gram-positive bacteria

    Chemical and radiological characterization of meals served by the COSEAS (USP-SP)

    O presente estudo apresenta resultados de análise química de refeições (almoço) oferecidas pelo restaurante do COSEAS (USP), por cinco dias não consecutivos. Essas refeições foram coletadas em triplicata, da mesma forma como oferecida para os usuários, sendo, a seguir, liofilizadas para as análises posteriores. No total, 15 amostras foram coletadas. A análise química de composição centesimal foi realizada segundo as técnicas padronizadas pela AOAC. O conteúdo de alguns elementos minerais (Ca, Fe, K, Na, Se e Zn) foi determinado por análise por ativação neutrônica instrumental. A validação da metodologia foi feita por meio da análise dos materiais de referência. A partir dos dados de concentração, calcularam-se os valores de ingestão de cada nutriente correspondente a esta refeição (40% da ingestão diária total), que foram avaliadas segundo as novas recomendações de nutrientes (Dietary Reference Intakes - DRIs) do National Research Council (USA), considerando a população de mulheres no estágio de vida de 19 a 30 anos. Comparando-se os valores médios encontrados com os valores recomendados, concluiu-se que: para os macronutrientes e os micronutrientes, Fe, Se e Zn, as recomendações foram atingidas; para Ca e K foram deficientes; e para Na excedeu o valor recomendado. Os radionuclídeos 40K, 60Co, 137Cs e 131I foram determinados, nas amostras de dieta, por espectrometria gama; 90Sr por cintilação em meio líquido; e 210Po, 234U, 232Th, 238U, 235U, 228Th, 230Th e 232Th por espectrometria alfa. Verificou-se, a partir dos resultados obtidos, que todos os radionuclídeos analisados se encontram muito abaixo dos limites estabelecidos pela FAO.This study presents the results obtained for the chemical and radiological analyses of lunch meals served in the cafeteria at the University of São Paulo-USP (COSEAS restaurant) in Brazil, on 5 non-consecutive days. On each of the five days, the meals were collected in triplicate and then freeze-dried for analysis totalizing 15 samples. The determination of the proximate composition complied with the AOAC standardized methodologies. The content of Ca, Fe, K, Na, Se, and Zn was determined by instrumental neutron activation analysis and the method validation was performed by certified reference materials analyses. Based on the concentration data, the daily intake of each mineral was calculated (corresponding to 40% of the total daily intake) and compared to the new recommendations set by the National Research Council (USA) considering the 19-30 year-old women range group. Comparing the average values obtained in this study with the recommended values, it was possible to verify that for macronutrients and the micronutrients Fe, Se, and Zn the recommendations were followed; for Ca and K they were deficient and exceeded the recommendation value for Na. The radionuclides 40K, 60Co, 137Cs, and 131I were determined by gamma spectrometry; 90Sr by liquid cintilation and 210Po, 234U, 232Th, 238U, 235U, 228Th, 230Th, and 232Th by alfa spectrometry. All radionuclides are present in concentrations below the limits set by the Food and Agriculture Organization (FAO)
