    Innovative Metallic Microfluidic Device for Intensified Biodiesel Production

    We present a strategy for intensified biodiesel production in a novel metallic microdevice. Additive manufacturing using Selective Laser Melting (SLM) was employed to build the metallic device consisting of multiple micro reactors monolithically integrated with multiple micro heat exchangers. This device allows high conversion rate of biodiesel production with concomitant use of the rejected heat from external source to enhance the reaction temperature and, thereby, its output. The biodiesel production was carried out using soybean oil, ethanol and NaOH as the catalyst. The influences of the reaction temperature and the residence time in the biodiesel production was examined. Biodiesel yield increased with the reaction temperature and a rate of conversion of 99.6% was achieved with a reactor residence time of less than 35 seconds. The work opens up a pathway to exploit waste heat to intensify biodiesel production and contribute significantly to global sustainability

    Achieving excellence in lean implementation at construction companies - A case study from Brazil

    Lean Construction has become a popular concept to improve performance and reduce costs in construction projects. Assessment tools and implementation strategies that focus on lean practices such as the Last Planner System, the use of Kanbans, the routine of Kaizens and, recently, the integration with information technology are also common. However, barriers to the wide adoption of lean principles remain and implementation challenges continue to intrigue scholars. Previous studies suggest that barriers to the successful implementation can be either political, economical, social and/or technical, with the focus on lack of knowledge about lean concepts, resistance to the required organisational culture change, and lack of support from top management. Few studies present the lean implementation process from the practitioners’ perspective. Using the narrative enquiry methodology, the authors aim to describe how a construction company from Brazil matured from the implementation of lean operational tools to achieve excellence in the lean culture and mindset. Key themes that emerge from this study for a successful lean construction journey are; effort to stabilise the environment, knowledge creation and management, transparency in the process to enable simplicity and shared understanding, and building trust for further growth.

    Quark Number Susceptibility with Finite Chemical Potential in Holographic QCD

    We study the quark number susceptibility in holographic QCD with a finite chemical potential or under an external magnetic field at finite temperature. We first consider the quark number susceptibility with the chemical potential. We observe that approaching the critical temperature from high temperature regime, the quark number susceptibility divided by temperature square develops a peak as we increase the chemical potential, which confirms recent lattice QCD results. We discuss this behavior in connection with the existence of the critical end point in the QCD phase diagram. We also consider the quark number susceptibility under the external magnetic field. We predict that the quark number susceptibility exhibits a blow-up behavior at low temperature as we raise the value of the magnetic field.     Malacological survey of Biomphalaria snails in municipalities along the Estrada Real in the southeast of the State of Minas Gerais, Brazil

    INTRODUÇÃO: O aumento das práticas de ecoturismo e turismo rural, em Minas Gerais, Brasil, evidencia a importância de se realizarem estudos sobre a ocorrência de hospedeiros intermediários do Schistosoma mansoni, no estado. O presente trabalho objetivou a busca e identificação das espécies de caramujos Biomphalaria encontrados em municípios mineiros pertencentes à Estrada Real, um importante projeto de turismo brasileiro. MÉTODOS: Os moluscos foram coletados em 36 municípios da Estrada Real, no sudeste de Minas Gerais. A pesquisa foi realizada de agosto de 2005 a setembro de 2009 e todos os locais visitados foram georreferenciados com o uso de GPS. RESULTADOS: Dos 36 municípios estudados, 30 apresentaram a ocorrência de pelo menos uma entre as seis espécies de Biomphalaria: glabrata, tenagophila, straminea, peregrina, occidentalis e schrammi. As três primeiras espécies citadas, reconhecidas como hospedeiras intermediárias do S. mansoni, estavam presentes em 33,3%, 47,2% e 8,3% dos municípios estudados, respectivamente. Os moluscos foram encontrados em diferentes tipos de coleções hídricas e em nenhum deles foi detectada infecção pelo S. mansoni. Houve maior ocorrência de Biomphalaria na área referente ao Caminho Novo (Diamantina/MG ao Rio de Janeiro/RJ). CONCLUSÕES: Considerando-se a ocorrência da esquistossomose, no Estado de Minas Gerais, e as repercussões socioeconômicas que envolvem o projeto Estrada Real, este trabalho aponta para a vulnerabilidade das coleções hídricas devido à presença de moluscos Biomphalaria e enfatiza a necessidade de vigilância epidemiológica e medidas educativas e sanitárias integradas com a comunidade local e setores de turismo.INTRODUCTION: The increasing practice of ecotourism and rural tourism in the State of Minas Gerais, Brazil, highlights the importance of studies concerning the occurrence of potential intermediate hosts of Schistosoma mansoni. This study aimed to identify species of Biomphalaria snails in municipalities along the Estrada Real, an important Brazilian tourism project. METHODS: The specimens were collected in different water collections of 36 municipalities along the Estrada Real in the southeast of the State of Minas Gerais. Biomphalaria species were characterized using both morphological and molecular approaches. The research was conducted between August 2005 and September 2009 and all the sites visited were georeferenced using GPS. RESULTS: Six Biomphalaria species were found in 30 of the 36 municipalities studied: glabrata, tenagophila, straminea, peregrina, occidentalis and schrammi. The first three species of Biomphalaria, recognized as intermediate hosts of S. mansoni, were present in 33.3%, 47.2% and 8.3% of the municipalities studied, respectively. The mollusks were found in different types of water collections and no infection by S. mansoni was detected. The highest occurrence of Biomphalaria concentration was verified in the area covered by the Caminho Novo route (Diamantina/MG to Rio de Janeiro/RJ). CONCLUSIONS: Considering the occurrence of schistosomiasis in the State of Minas Gerais and the socioeconomic repercussions involved in the Estrada Real Project, this work focuses on the vulnerability of water collections due to the presence of Biomphalaria mollusks and emphasizes the need for epidemiological surveillance and sanitary and educational measures integrated with the local community and tourism sectors

    Collagen Type IV-Related Nephropathies in Portugal: Pathogenic COL4A5 Mutations and Clinical Characterization of 22 Families

    Alport syndrome (AS) is caused by pathogenic mutations in the genes encoding α3, α4 or α5 chains of collagen IV (COL4A3/COL4A4/COL4A5), resulting in hematuria, chronic renal failure (CRF), sensorineural hearing loss (SNHL) and ocular abnormalities. Mutations in the X-linked COL4A5 gene have been identified in 85% of the families (XLAS). In this study, 22 of 60 probands (37%) of unrelated Portuguese families, with clinical diagnosis of AS and no evidence of autosomal inheritance, had pathogenic COL4A5 mutations detected by Sanger sequencing and/or multiplex-ligation probe amplification, of which 12 (57%) are novel. Males had more severe and earlier renal and extrarenal complications, but microscopic hematuria was a constant finding irrespective of gender. Nonsense and splice site mutations, as well as small and large deletions, were associated with younger age of onset of SNHL in males, and with higher risk of CRF and SNHL in females. Pathogenic COL4A3 or COL4A4 mutations were subsequently identified in more than half of the families without a pathogenic mutation in COL4A5. The lower than expected prevalence of XLAS in Portuguese families warrants the use of next-generation sequencing for simultaneous COL4A3/COL4A4/COL4A5 analysis, as first-tier approach to the genetic diagnosis of collagen type IV-related nephropathies.info:eu-repo/semantics/publishedVersio

    The Second Transmembrane Domain of P2X7 Contributes to Dilated Pore Formation

    Activation of the purinergic receptor P2X7 leads to the cellular permeability of low molecular weight cations. To determine which domains of P2X7 are necessary for this permeability, we exchanged either the C-terminus or portions of the second transmembrane domain (TM2) with those in P2X1 or P2X4. Replacement of the C-terminus of P2X7 with either P2X1 or P2X4 prevented surface expression of the chimeric receptor. Similarly, chimeric P2X7 containing TM2 from P2X1 or P2X4 had reduced surface expression and no permeability to cationic dyes. Exchanging the N-terminal 10 residues or C-terminal 14 residues of the P2X7 TM2 with the corresponding region of P2X1 TM2 partially restored surface expression and limited pore permeability. To further probe TM2 structure, we replaced single residues in P2X7 TM2 with those in P2X1 or P2X4. We identified multiple substitutions that drastically changed pore permeability without altering surface expression. Three substitutions (Q332P, Y336T, and Y343L) individually reduced pore formation as indicated by decreased dye uptake and also reduced membrane blebbing in response to ATP exposure. Three others substitutions, V335T, S342G, and S342A each enhanced dye uptake, membrane blebbing and cell death. Our results demonstrate a critical role for the TM2 domain of P2X7 in receptor function, and provide a structural basis for differences between purinergic receptors. © 2013 Sun et al
