    The diagnosis of cryptorchidism in horses is often a challenge. Based on the history, clinical and rectal examinations and ultrasonography, a definitive diagnosis is not always possible. Various endocrinological diagnostic assays, such as the determination of testosterone, androstenedione, estrogens, urinary steroids and the anti-Mullerian hormone, which demonstrate the presence of testicular tissue, have been described. These tests all have their advantages and disadvantages, which are discussed in this article in order to help practitioners in the field

    The Influence of Surface Roughness on Nucleate Pool Boiling Heat Transfer

    The effect of surface roughness on pool boiling heat transfer is experimentally explored over a wide range of roughness values in water and Fluorinertâ„¢ FC-77, two fluids with different thermal properties and wetting characteristics. The test surfaces ranged from a polished surface (Ra between 0.027 micrometer and 0.038 micrometer) to electrical discharge machined (EDM) surfaces with a roughness Ra ranging from 1.08 micrometer to 10.0 micrometer. Different trends were observed in the heat transfer coefficient with respect to the surface roughness between the two fluids on the same set of surfaces. For FC-77, the heat transfer coefficient was found to continually increase with increasing roughness. For water, on the other hand, EDM surfaces of intermediate roughness displayed similar heat transfer coefficients that were higher than for the polished surface, while the roughest surface showed the highest heat transfer coefficients. The heat transfer coefficients were more strongly influenced by surface roughness with FC-77 than with water. For FC-77, the roughest surface produced 210% higher heat transfer coefficients than the polished surface while for water, a more modest 100% enhancement was measured between the same set of surfaces. Although the results highlight the inadequacy of characterizing nucleate pool boiling data using Ra, the observed effect of roughness was correlated using hRa m as has been done in several prior studies. The experimental results were compared with predictions from several widely used correlations in the literature

    The RhoGEF Trio Functions in Sculpting Class Specific Dendrite Morphogenesis in Drosophila Sensory Neurons

    As the primary sites of synaptic or sensory input in the nervous system, dendrites play an essential role in processing neuronal and sensory information. Moreover, the specification of class specific dendrite arborization is critically important in establishing neural connectivity and the formation of functional networks. Cytoskeletal modulation provides a key mechanism for establishing, as well as reorganizing, dendritic morphology among distinct neuronal subtypes. While previous studies have established differential roles for the small GTPases Rac and Rho in mediating dendrite morphogenesis, little is known regarding the direct regulators of these genes in mediating distinct dendritic architectures.Here we demonstrate that the RhoGEF Trio is required for the specification of class specific dendritic morphology in dendritic arborization (da) sensory neurons of the Drosophila peripheral nervous system (PNS). Trio is expressed in all da neuron subclasses and loss-of-function analyses indicate that Trio functions cell-autonomously in promoting dendritic branching, field coverage, and refining dendritic outgrowth in various da neuron subtypes. Moreover, overexpression studies demonstrate that Trio acts to promote higher order dendritic branching, including the formation of dendritic filopodia, through Trio GEF1-dependent interactions with Rac1, whereas Trio GEF-2-dependent interactions with Rho1 serve to restrict dendritic extension and higher order branching in da neurons. Finally, we show that de novo dendritic branching, induced by the homeodomain transcription factor Cut, requires Trio activity suggesting these molecules may act in a pathway to mediate dendrite morphogenesis.Collectively, our analyses implicate Trio as an important regulator of class specific da neuron dendrite morphogenesis via interactions with Rac1 and Rho1 and indicate that Trio is required as downstream effector in Cut-mediated regulation of dendrite branching and filopodia formation

    Transcriptional Responses of Cultured Rat Sympathetic Neurons during BMP-7-Induced Dendritic Growth

    Dendrites are the primary site of synapse formation in the vertebrate nervous system; however, relatively little is known about the molecular mechanisms that regulate the initial formation of primary dendrites. Embryonic rat sympathetic neurons cultured under defined conditions extend a single functional axon, but fail to form dendrites. Addition of bone morphogenetic proteins (BMPs) triggers these neurons to extend multiple dendrites without altering axonal growth or cell survival. We used this culture system to examine differential gene expression patterns in naïve vs. BMP-treated sympathetic neurons in order to identify candidate genes involved in regulation of primary dendritogenesis.To determine the critical transcriptional window during BMP-induced dendritic growth, morphometric analysis of microtubule-associated protein (MAP-2)-immunopositive processes was used to quantify dendritic growth in cultures exposed to the transcription inhibitor actinomycin-D added at varying times after addition of BMP-7. BMP-7-induced dendritic growth was blocked when transcription was inhibited within the first 24 hr after adding exogenous BMP-7. Thus, total RNA was isolated from sympathetic neurons exposed to three different experimental conditions: (1) no BMP-7 treatment; (2) treatment with BMP-7 for 6 hr; and (3) treatment with BMP-7 for 24 hr. Affymetrix oligonucleotide microarrays were used to identify differential gene expression under these three culture conditions. BMP-7 significantly regulated 56 unique genes at 6 hr and 185 unique genes at 24 hr. Bioinformatic analyses implicate both established and novel genes and signaling pathways in primary dendritogenesis.This study provides a unique dataset that will be useful in generating testable hypotheses regarding transcriptional control of the initial stages of dendritic growth. Since BMPs selectively promote dendritic growth in central neurons as well, these findings may be generally applicable to dendritic growth in other neuronal cell types

    Nucleate Boiling from Smooth and Rough Surfaces--Part 1: Fabrication and Characterization of an Optically Transparent Heater-Sensor Substrate with Controlled Surface Roughness

This first of two companion papers discusses details of the fabrication, construction, and operation of the experimental facility. Test pieces are fabricated from 50.8 mmx50.8 mmx3.18 mm borosilicate glass squares that are roughened by abrading with diamond compound with particles of known size and then annealed in order to control small-scale roughness features. Heater/sensor devices are fabricated by depositing indium tin oxide (ITO), a transparent electrically conductive material, on top of the roughened glass surfaces. A method for calibrating the ITO devices for use as temperature sensors is developed. Experimental boiling curves are reported for seven surfaces of different roughness tested in the perfluorinated fluorocarbon liquid, FC-72. The measured receding contact angle for FC-72 on the ITO-coated surfaces is reported. The wall superheat during saturated pool boiling at atmospheric pressure was found not to vary consistently with surface average roughness (Ra) values. Qualitative differences between smooth and rough surface boiling visualizations obtained simultaneously from the side and from below the heaters are discussed

    Ceftriaxone as effective as long-acting chloramphenicol in short-course treatment of meningococcal meningitis during epidemics: a randomised non-inferiority study.

    BACKGROUND: In sub-Saharan Africa in the 1990s, more than 600,000 people had epidemic meningococcal meningitis, of whom 10% died. The current recommended treatment by WHO is short-course long-acting oily chloramphenicol. Continuation of the production of this drug is uncertain, so simple alternatives need to be found. We assessed whether the efficacy of single-dose treatment of ceftriaxone was non-inferior to that of oily chloramphenicol for epidemic meningococcal meningitis. METHODS: In 2003, we undertook a randomised, open-label, non-inferiority trial in nine health-care facilities in Niger. Participants with suspected disease who were older than 2 months were randomly assigned to receive either chloramphenicol or ceftriaxone. Primary outcome was treatment failure (defined as death or clinical failure) at 72 h, measured with intention-to-treat and per-protocol analyses. FINDINGS: Of 510 individuals with suspected disease, 247 received ceftriaxone, 256 received chloramphenicol, and seven were lost to follow-up. The treatment failure rate at 72 h for the intention-to-treat analysis was 9% (22 patients) for both drug groups (risk difference 0.3%, 90% CI -3.8 to 4.5). Case fatality rates and clinical failure rates were equivalent in both treatment groups (14 [6%] ceftriaxone vs 12 [5%] chloramphenicol). Results were also similar for both treatment groups in individuals with confirmed meningitis caused by Neisseria meningitidis. No adverse side-effects were reported. INTERPRETATION: Single-dose ceftriaxone provides an alternative treatment for epidemic meningococcal meningitis--its efficacy, ease of use, and low cost favour its use. National and international health partners should consider ceftriaxone as an alternative first-line treatment to chloramphenicol for epidemic meningococcal meningitis
