    Effects of radiation on charge-coupled devices

    The effects of 1 MeV electron irradiation upon the performance of two phase, polysilicon aluminum gate CCDs are reported. Both n- and p-surface channel and n-buried channel devices are investigated using 64- and 128-stage line arrays. Characteristics measured as a function of radiation dose include: Transfer inefficiency, threshold voltage, field effect mobility, interface state density, full well signal level and dark current. Surface channel devices are found to degrade considerably at less than 10 to the 5th power rads (Si) due to the large increase in fast interface state density caused by radiation. Buried channel devices maintain efficient operation to the highest dose levels used

    Absolute Calibration of a Large-diameter Light Source

    A method of absolute calibration for large aperture optical systems is presented, using the example of the Pierre Auger Observatory fluorescence detectors. A 2.5 m diameter light source illuminated by an ultra--violet light emitting diode is calibrated with an overall uncertainty of 2.1 % at a wavelength of 365 nm.Comment: 15 pages, 8 figures.     Growth, Condition, and Trophic Relations of Stocked Trout in Southern Appalachian Mountain Streams

    Stream trout fisheries are among the most popular and valuable in the United States, but many are dependent on hatcheries to sustain fishing and harvest. Thus, understanding the ecology of hatchery‐reared trout stocked in natural environments is fundamental to management. We evaluated the growth, condition, and trophic relations of Brook Trout Salvelinus fontinalis, Brown Trout Salmo trutta, and Rainbow Trout Oncorhynchus mykiss that were stocked in southern Appalachian Mountain streams in western North Carolina. Stocked and wild (naturalized) trout were sampled over time (monthly; September 2012–June 2013) to compare condition and diet composition and to evaluate temporal dynamics of trophic position with stable isotope analysis. Relative weights (Wr) of stocked trout were inversely associated with their stream residence time but were consistently higher than those of wild trout. Weight loss of harvested stocked trout was similar among species and sizes, but fish stocked earlier lost more weight. Overall, 40% of 141 stomachs from stocked trout were empty compared to 15% of wild trout stomachs (N = 26). We identified a much higher rate of piscivory in wild trout (18 times that of stocked trout), and wild trout were 4.3 times more likely to consume gastropods relative to stocked trout. Hatchery‐reared trout were isotopically similar to co‐occurring wild fish for both δ13C and δ15N values but were less variable than wild trout. Differences in sulfur isotope ratios (δ34S) between wild and hatchery‐reared trout indicated that the diets of wild fish were enriched in δ34S relative to the diets of hatchery‐reared fish. Although hatcheryreared trout consumed prey items similar to those of wild fish, differences in consumption or behavior (e.g., reduced feeding) may have resulted in lower condition and negative growth. These findings provide critical insight on the trophic dynamics of stocked trout and may assist in developing and enhancing stream trout fisheries

    Charting the protein complexome in yeast by mass spectrometry

    It has become evident over the past few years that many complex cellular processes, including control of the cell cycle and ubiquitin-dependent proteolysis, are carried out by sophisticated multisubunit protein machines that are dynamic in abundance, post-translational modification state, and composition. To understand better the nature of the macromolecular assemblages that carry out the cell cycle and ubiquitin-dependent proteolysis, we have used mass spectrometry extensively over the past few years to characterize both the composition of various protein complexes and the modification states of their subunits. In this article we review some of our recent efforts, and describe a promising new approach for using mass spectrometry to dissect protein interaction networks

    The prevalence of pseudoscientific ideas and neuromyths among sports coaches

    There has been an exponential growth in research examining the neurological basis of human cognition and learning. Little is known, however, about the extent to which sports coaches are aware of these advances. Consequently, the aim of the present study was to examine the prevalence of pseudoscientific ideas among British and Irish sports coaches. In total, 545 coaches from the United Kingdom and Ireland completed a measure that included questions about how evidence-based theories of the brain might enhance coaching and learning, how they were exposed to these different theories, and their awareness of neuromyths. Results revealed that the coaches believed that an enhanced understanding of the brain helped with their planning and delivery of sports sessions. Goal-setting was the most frequently used strategy. Interestingly, 41.6% of the coaches agreed with statements that promoted neuromyths. The most prevalent neuromyth was “individuals learn better when they receive information in their preferred learning style (e.g., auditory, visual, or kinesthetic)”, which 62% of coaches believed. It is apparent that a relatively large percentage of coaches base aspects of their coaching practice on neuromyths and other pseudoscientific ideas.     Possibilities for pedagogy in Further Education: Harnessing the abundance of literacy

    In this report, it is argued that the most salient factor in the contemporary communicative landscape is the sheer abundance and diversity of possibilities for literacy, and that the extent and nature of students' communicative resources is a central issue in education. The text outlines the conceptual underpinnings of the Literacies for Learning in Further Education project in a social view of literacy, and the associated research design, methodology and analytical framework. It elaborates on the notion of the abundance of literacies in students' everyday lives, and on the potential for harnessing these as resources for the enhancement of learning. It provides case studies of changes in practice that have been undertaken by further education staff in order to draw upon students' everyday literacy practices on Travel and Tourism and Multimedia courses. It ends with some of the broad implications for conceptualising learning that arise from researching through the lens of literacy practices

    Multi-color Molecular Visualization of Signaling Proteins Reveals How C-Terminal Src Kinase Nanoclusters Regulate T Cell Receptor Activation

    Elucidating the mechanisms that controlled T cell activation requires visualization of the spatial organization of multiple proteins on the submicron scale. Here, we use stoichiometrically accurate, multiplexed, singlemolecule super-resolution microscopy (DNA-PAINT) to image the nanoscale spatial architecture of the primary inhibitor of the T cell signaling pathway, Csk, and two binding partners implicated in its membrane association, PAG and TRAF3. Combined with a newly developed co-clustering analysis framework, we find that Csk forms nanoscale clusters proximal to the plasma membrane that are lost post-stimulation and are re-recruited at later time points. Unexpectedly, these clusters do not co-localize with PAG at the membrane but instead provide a ready pool of monomers to downregulate signaling. By generating CRISPR-Cas9 knockout T cells, our data also identify that a major risk factor for autoimmune diseases, the protein tyrosine phosphatase non-receptor type 22 (PTPN22) locus, is essential for Csk nanocluster re-recruitment and for maintenance of the synaptic PAG population
