Synthesis and conformational analysis of leptocarpin derivatives. Influence of modification of the oxirane ringon leptocarpin's cytotoxic activity.

Indexación: ScieloABSTRACT The reaction in acidconditions of Leptocarpin 1, a compound with antitumor activity, formed two new isomeric products, 8b-angeloyl-1b,3b-dihydroxy-4,10-dimethyl,-D11(13) methylen-4Z,9Z-dieneheliangol-6,12-olide 2 and 8b-angeloyl-1b,3b-dihydroxy-4-methyl-D11(13),D11(14)-dimethylen-4Z-eneheliangol-6,12-olide 3, whose structures reported in this study were established by spectroscopy (1H-NMR, 13C-NMR, MS and IR) and confirmedthrough ROESY experiments and theoretical studies by molecular mechanics. The in vitro cytotoxicity of these isomeric compounds was less active than leptocarpin, showing the importance of the oxirane ring in the biological activity. Dopamine receptor D5 signaling plays a dual role in experimental autoimmune encephalomyelitis potentiating Th17-mediated immunity and favoring suppressive activity of regulatory T-cells

Nevertheless, the lack of drugs that might discriminate between DRD1 and DRD5 has made the pharmacological distinction between the two receptors difficult. We have previously shown genetic evidence demonstrating a relevant role of DRD5-signaling in dendritic cells (DCs) favoring the CD4+ T-cell-driven inflammation in EAE. However, the role of DRD5-signaling confined to CD4+ T-cells in the development of EAE is still unknown. Here, we analyzed the functional role of DRD5-signaling in CD4+ T-cell-mediated responses and its relevance in EAE by using a genetic approach. Our results show that DRD5-signaling confined to naive CD4+ T-cells exerts a pro-inflammatory effect promoting the development of EAE with a stronger disease severity. This pro-inflammatory effect observed for DRD5-signaling in naive CD4+ T-cells was related with an exacerbated proliferation in response to T-cell activation and to an increased ability to differentiate toward the Th17 inflammatory phenotype. On the other hand, quite unexpected, our results show that DRD5-signaling confined to Tregs strengthens their suppressive activity, thereby dampening the development of EAE manifestation. This anti-inflammatory effect of DRD5-signaling in Tregs was associated with a selective increase in the expression of glucocorticoid-induced tumor necrosis factor receptor-related protein (GITR), which has been described to play a critical role in the expansion of Tregs. Our findings here indicate a complex role for DRD5-signaling in CD4+ T-cells-driven responses potentiating early inflammation mediated by effector T-cells in EAE, but exacerbating suppressive activity in Tregs and thereby dampening disease manifestation in late EAE stages. © 2018 Osorio-Barrios, Prado, Contreras and Pacheco.https://www.frontiersin.org/articles/10.3389/fncel.2018.00192/ful

The Past in the Present . Looking Inside Collective Reincorporation of FARC-EP Ex-Members

In 2016, the Colombian government signed the peace agreement with the Revolutionary Armed Forces of Colombia-People’s Army (FARC-EP) and aimed to reintegrate more than 13,000 ex-combatants into society based on a collective reincorporation approach. This approach promotes alternatives for social, economic and political reincorporation beyond Disarmament, Demobilisation and Reintegration (DDR) standards by valuing the ex-combatants’ legacy in conflict times. This research aims to analyse under a sociological micro-perspective the meanings, transitions, dimensions and relationships around collective reincorporation. Instead of focusing on a single dimension on reintegration, I analyse how these dimensions are present and interact in FARC-EP ex-members everyday practices. I discuss the influence that former comrades, families, surrounding communities and institutions have on the configuration of ex-combatants’ legacy, identity, social capital, concept of citizenship and agency. Based on a trans-local perspective, I use a multi-sited ethnography approach to make an in-depth analysis of the everyday life of FARC-EP ex-members and their commonalities beyond geographical boundaries in three territories in which they are collectively settled. I find that the building of relationships among FARC-EP ex-members and the use of their agency contribute in not just their reincorporation process, but also in the transformation of the social, political and economic dynamics of their surroundings. I conclude by saying that the social visibility of FARC-EP ex-members becomes an alternative to increase local agency, and a valuable strategy to break the divide between past and present trajectories. Therefore, collective reincorporation is an approach to social transformation in which FARC-EP ex-members through their collective initiatives, become visible agents of change in peacebuilding

Aerobic power, huddling and the efficiency of torpor in the South American marsupial, Dromiciops gliroides.

During periods of cold, small endotherms depend on a continuous supply of food and energy to maintain euthermic body temperature (T(b)), which can be challenging if food is limited. In these conditions, energy-saving strategies are critical to reduce the energetic requirements for survival. Mammals from temperate regions show a wide arrange of such strategies, including torpor and huddling. Here we provide a quantitative description of thermoregulatory capacities and energy-saving strategies in Dromiciops gliroides, a Microbiotherid marsupial inhabiting temperate rain forests. Unlike many mammals from temperate regions, preliminary studies have suggested that this species has low capacity for control and regulation of body temperature, but there is still an incomplete picture of its bioenergetics. In order to more fully understand the physiological capacities of this "living fossil", we measured its scope of aerobic power and the interaction between huddling and torpor. Specifically, we evaluated: (1) the relation between basal (BMR) and maximum metabolic rate (MMR), and (2) the role of huddling on the characteristics of torpor at different temperatures. We found that BMR and MMR were above the expected values for marsupials and the factorial aerobic scope (from [Formula: see text]CO(2)) was 6.0±0.45 (using [Formula: see text]CO(2)) and 6.2±0.23 (using [Formula: see text]O(2)), an unusually low value for mammals. Also, repeatability of physiological variables was non-significant, as in previous studies, suggesting poor time-consistency of energy metabolism. Comparisons of energy expenditure and body temperature (using attached data-loggers) between grouped and isolated individuals showed that at 20°C both average resting metabolic rate and body temperature were higher in groups, essentially because animals remained non-torpid. At 10°C, however, all individuals became torpid and no differences were observed between grouped and isolated individuals. In summary, our study suggests that the main response of Dromiciops gliroides to low ambient temperature is reduced body temperature and torpor, irrespective of huddling. Low aerobic power and low time-consistency of most thermoregulatory traits of Dromiciops gliroides support the idea of poor thermoregulatory abilities in this species

Curso de Especial Interés: Victimología y CriminologíaEl objetivo de este trabajo fue la creación del manual Legal Psychologist in Psychosocial Contexts and University Psychological Services (LEPSYCOUS) el cual permite establecer el perfil disciplinar del psicólogo jurídico en contextos psicosociales y servicios psicológicos universitarios; en primer lugar la definición del perfil disciplinar del psicólogo jurídico permite delimitar las funciones del mismo en los subcampos de aplicación como lo son: la psicología Victimal, Criminológica y la Psicología de la Mediación y conciliación; en segundo lugar sirve como una guía de apoyo de tipo administrativo y legal para que el psicólogo jurídico, en su ejercicio pueda cumplir de manera correcta con las funciones tanto en los contextos psicosociales como en los servicios psicológicos universitarios. Proyecto : Escuela de Repostería : La Pâtisserie

La Pâtisserie tiene como fin establecerse dentro del mercado de los postres en Bogotá, donde se venderán dos tipos de productos y un servicio: el primer tipo de producto es: vender diferentes alimentos como cupcakes, tortas, muffins, galletas y bebidas como tinto o cappuccino entre otros, el segundo es vender utensilios de repostería importados desde Estados Unidos, el servicio que se venderá es una clase personalizada donde se enseñen a hacer las recetas tradicionales y además algunas nuevas que se desarrollen en la tienda. El diferencial del proyecto es el programa de responsabilidad social, en el cual se busca incluir a personas con discapacidad cognitiva, esto se llevará a cabo gracias a la Fundación Best Buddies, la cual se encarga de capacitar y asegurar todas las condiciones para que personas con Síndrome de Down puedan ser empleada. Para ello, se iniciarán con programas de capacitación del personal quien demuestre tener gusto y habilidad para la repostería. Después de llevar a cabo la encuesta se concretó que el segmento de La Pâtisserie son mujeres entre los 25 - 34 años, de estratos altos que residen por el nororiente de Bogotá, a las cuales les gustan los postres y quieren aprender un poco más sobre las recetas. Son mujeres trabajadoras que buscan salir de la rutina y dedicar tiempo a actividades que sean de su interés. Este proyecto se iniciará con una inversión de $70.000.000, en donde se encuentran incluidos el arriendo del local, las adaptaciones necesarias para que el personal con discapacidad pueda estar segura y trabaje tranquilamente, todos los implementos de cocina incluidos muebles y materiales para iniciar con la producción de los productos que se ofrecerán., Gracias a esto según proyecciones se espera tener un crecimiento del 14,9 This project’s differential is the social program, which includes people with cognitive impairment into the business model. ¿El camino hacia la paz o palos de ciego? Impacto del Programa de Desmovilización

El desarme, la desmovilización y la reintegración (DDR) es la iniciativa de consolidación de la paz más utilizada en todo el mundo. A pesar de su popularidad, las evaluaciones empíricas de la contribución que el DDR ha hecho a la seguridad y la paz han sido desatendidas en gran medida. Utilizando la experiencia de Colombia, se calculó el impacto que la desmovilización paramilitar ha tenido sobre las tasas de homicidios a través de Colombia. Utilizando un modelo econométrico de efectos fijos con datos a nivel municipal durante un período de 12 años, se encontró que la desmovilización ha desatado dos fuerzas: por un lado disminuye los homicidios como resultado de una menor actividad paramilitar y por el otro, los aumenta en las áreas de desmovilización en proporción a la densidad de personas desmovilizadas. Se encuentra que la tasa de desmovilización de las AUC incrementa la tasa de homicidios en 2.8/100.000 y que la reducción en las acciones ofensivas ha implicado una disminución en la tasa de asesinatos de 9.9/100.000. Así, el efecto combinado es una reducción de 7.1/100.000 en la tasa de homicidios en los municipios en los que operaron y se desmovilizaron los paramilitares. Se estimó un modelo probit para explorar la heterogeneidad en el cambio de la tasa de homicidio en las áreas de desmovilización. Se encontró que los municipios con presencia de grupos armados y más densos en población, tienen mayorprobabilidad de tener tasas de homicidio más elevadas. Las áreas con mayores tasas de arrestos tienen una probabilidad mayor de tener menos homicidios en el periodo posterior a ladesmovilización.desmovilización de paramilitares, desarme y reintegración, construcción de paz, conflicto en Colombia, efectos fijos, probit.

¿En qué condiciones entran los jóvenes en el mercado laboral en El Salvador? Este cuaderno, el tercero publicado por la FES, se propone indagar sobre la pertinencia (o la falta de pertinencia) de la educación en el mundo del trabajo. Quienes logran terminar la escuela —muchas veces, con sacrificios personales y familiares de por medio— no tienen garantía de que obtendrán un trabajo que les asegure su bienestar.
