    Development of specific PCR assays for the detection of Cryptocaryon irritans

    Cryptocaryon irritans is one of the most important protozoan pathogens of marine fish, causing the “white spot” disease and posing a significant problem to marine aquaculture. In the present study, a C. irritans-specific reverse primer (S15) was designed based on the published sequence of the second internal transcribed spacer (ITS-2) of ribosomal DNA (rDNA) of C. irritans and used together with the conserved forward primer P1 to develop a specific polymerase chain reaction (PCR) assay for direct, rapid, and specific detection of C. irritans. The specificity of these primers was tested with both closely and distantly related ciliates (Pseudokeroronpsis rubra, Pseudokeroronpsis carnae, Euplotes sp. 1, Ichthyophthirius multifiliis, Pseudourostyla cristata, and Paramecium caudaium), and only C. irritans was detected and no product was amplified from any other ciliates examined in this study using the specific primer set P1-S15. The specific PCR assay was able to detect as low as 45 pg of C. irritans DNA and a nested PCR assay using two primer sets (P1/NC2, P1/S15) increased the sensitivity, allowing the detection of a single C. irritans. The species-specific PCR assays should provide useful tools for the diagnosis, prevention, and molecular epidemiological investigations of C. irritans infection in marine fish

    An overview of population-based algorithms for multi-objective optimisation

    In this work we present an overview of the most prominent population-based algorithms and the methodologies used to extend them to multiple objective problems. Although not exact in the mathematical sense, it has long been recognised that population-based multi-objective optimisation techniques for real-world applications are immensely valuable and versatile. These techniques are usually employed when exact optimisation methods are not easily applicable or simply when, due to sheer complexity, such techniques could potentially be very costly. Another advantage is that since a population of decision vectors is considered in each generation these algorithms are implicitly parallelisable and can generate an approximation of the entire Pareto front at each iteration. A critique of their capabilities is also provided

    Otimização multiobjetivo das dimensões dos ambientes de uma residência unifamiliar baseada em simulação energética e estrutural

    Resumo O desenvolvimento da tecnologia computacional nas últimas décadas levou a otimização associada à parametrização a possibilitar o desenvolvimento de projetos de melhor desempenho, com ou sem integração com a simulação. O intuito deste trabalho foi aplicar a otimização baseada em simulação na forma de uma edificação residencial unifamiliar com o objetivo de encontrar a composição que apresentasse o melhor desempenho térmico com o menor custo dos materiais construtivos, considerando-se restrições estruturais. Foram utilizados os programas Rhinoceros e Energy Plus, além de plug-ins para modelagem paramétrica, o intercâmbio de informações entre os programas e a condução da otimização. Foram realizadas duas simulações, a primeira considerando os custos dos componentes construtivos e os graus-hora de aquecimento e resfriamento combinados e a segunda considerando os custos e graus-hora de aquecimento, que produziu resultados mais relevantes. Dois casos de cada solução de Pareto foram selecionados para análise, dentre 19,8 mil e 27,4 mil casos obtidos com uso de algoritmos evolucionários. Apesar das limitações de algumas das ferramentas, foi verificado um grande potencial de aplicação deste método no desenvolvimento de projetos arquitetônicos

    The starting point for the present contribution is that professional translators as well as editors tend to consider the theoretical debate on translation with mistrust. By looking at the translation process in its various phases, this paper aims to demonstrate how theory does, as a matter of fact, provide a 'meta-language' that can be of great help in the training of prospective translators, while at the same time offering useful tools for an evaluation of the translated text that goes beyond a merely impressionistic critique, based on individual taste. Furthermore, this paper discusses a number of theoretical concepts developed in the field of translation theory that can be of use to the translator's analytical work before, during, and after his/her activity of transcodification
