    Adaptive Management for Impacts to Eelgrass Habitat in Gloucester Harbor

    The Massachusetts Office of Coastal Zone Management and the Massachusetts Institute of Technology Sea Grant College Program, along with partners, led an effort to create an eelgrass bank, raise awareness of the value of eelgrass habitat, and facilitate transplanting efforts to Boston Harbor in the summer and fall of 2006. A planned impact to eelgrass habitat in Gloucester Harbor warranted efforts to try to save this valuable and declining resource. This unfortunate circumstance was used to educate interested citizens, students and teachers from regional schools, and government employees. Methods to transplant and store eelgrass were researched and tested in attempt to facilitate restoration of the impact area. Two community events were organized at Pavilion Beach to harvest eelgrass from the impact area. These events were attended by a variety of government (city, state, and federal) and non-government employees, along with students and teachers, and attracted much attention of the citizens of Gloucester. Eelgrass was successfully transplanted to Boston Harbor by the Massachusetts Division of Marine Fisheries. Harvested eelgrass was also maintained in a hydroponic raft system for three months (October-December) and used to set-up an interpretative display in a flow-through tank at the Gloucester Maritime Heritage Center. While the harvested eelgrass was ultimately not transplanted back to the impact corridor, experience in storing eelgrass within hydroponic and tank systems could assist future restoration efforts. By teaming up to save the eelgrass at Pavilion Beach in Gloucester Harbor, project partners demonstrated the advantage of creative, adaptive, and cooperative efforts to manage coastal resources.     Stabilities of nanohydrated thymine radical cations: insights from multiphoton ionization experiments and ab initio calculations

    Multi-photon ionization experiments have been carried out on thymine-water clusters in the gas phase. Metastable H2O loss from T+(H2O)n was observed at n ≥ 3 only. Ab initio quantum-chemical calculations of a large range of optimized T+(H2O)n conformers have been performed up to n = 4, enabling binding energies of water to be derived. These decrease smoothly with n, consistent with the general trend of increasing metastable H2O loss in the experimental data. The lowest-energy conformers of T+(H2O)3 and T+(H2O)4 feature intermolecular bonding via charge-dipole interactions, in contrast with the purely hydrogen-bonded neutrals. We found no evidence for a closed hydration shell at n = 4, also contrasting with studies of neutral clusters

    Eelgrass (Zostera marina L) is the dominant seagrass occurring in eastern Canada and the northeastern United States, where it often forms extensive meadows in coastal and estuarine areas. Eelgrass beds are extremely productive and provide many valuable ecological functions and ecosystem services. They serve as critical feeding and nursery habitat for a wide variety of commercially and recreationally important fish and shellfish and as feeding areas for waterfowl and other waterbirds. Eelgrass detritus is also transported considerable distances to fuel offshore food webs. In addition, eelgrass beds stabilize bottom sediments, dampen wave energy, absorb nutrients from surrounding waters, and retain carbon through burial

    The extreme r-element rich, iron-poor halo giant CS31082-001: Implications for the r-process site(s) and radioactive cosmochronology

    We present a high-resolution spectroscopic analysis of the bright (V=11.7), extreme halo giant CS31082-001 ([Fe/H] = -2.9), obtained in an ESO-VLT Large Programme dedicated to very metal-poor stars. We find CS31082-001 to be extremely rich in r-process elements, comparable in this respect only to the similarly metal-poor, but carbon-enriched, giant CS22892-052. As a result of the extreme overabundance of the heaviest r-process elements, and negligible blending from CH and CN molecular lines, a reliable measurement is obtained of the U II line at 386 nm, for the first time in a halo star, along with numerous lines of Th II, as well as lines of 25 other r-process elements. Abundance estimates for a total of 43 elements are reported in CS31082-001, almost half of the entire periodic table. All elements with 56 \leq Z \leq 72 follow the Solar r-element pattern, reduced by about 1.25 dex ([r/Fe]=+1.7 dex, a factor 50). Pb, in contrast, seems to be below the shifted Solar r-process distribution, possibly indicating an error in the latter, while thorium is more enhanced than the lighter nuclides. Thus, while a universal production ratio for the r-process elements seems to hold in the interval 56 \leq Z \leq 72, it breaks down in the actinide region. When available, the U/Th is thus preferable to Th/Eu for radioactive dating: (i) because of its faster decay rate and smaller sensitivity to observational errors, and (ii) because the inital production ratio of the neighboring nuclides 238U and 232Th is more robustly predicted than the 151Eu/232Th ratio. Our current best estimate for the age of CS31082-001 is 14.0+/-2.4 Gyr

    Clinical features that identify children with primary immunodeficiency diseases

    BACKGROUND: The 10 warning signs of primary immunodeficiency diseases (PID) have been promoted by various organizations in Europe and the United States to predict PID. However, the ability of these warning signs to identify children with PID has not been rigorously tested. OBJECTIVE: The main goal of this study was to determine the effectiveness of these 10 warning signs in predicting defined PID among children who presented to 2 tertiary pediatric immunodeficiency centers in the north of England. METHODS: A retrospective survey of 563 children who presented to 2 pediatric immunodeficiency centers was undertaken. The clinical records of 430 patients with a defined PID and 133 patients for whom detailed investigations failed to establish a specific PID were reviewed. RESULTS: Overall, 96% of the children with PID were referred by hospital clinicians. The strongest identifiers of PID were a family history of immunodeficiency disease in addition to use of intravenous antibiotics for sepsis in children with neutrophil PID and failure to thrive in children with T-lymphocyte PID. With these 3 signs, 96% of patients with neutrophil and complement deficiencies and 89% of children with T-lymphocyte immunodeficiencies could be identified correctly. Family history was the only warning sign that identified children with B-lymphocyte PID. CONCLUSIONS: PID awareness initiatives should be targeted at hospital pediatricians and families with a history of PID rather than the general public. Our results provide the general pediatrician with a simple refinement of 10 warning signs for identifying children with underlying immunodeficiency diseases. </jats:sec

    Absent in melanoma 2 (AIM2) positive profile identifies a poor prognosis of lung adenocarcinoma patients.

    The absent in melanoma 2 (AIM2) inflammasome has been demonstrated as involved in tumor growth. In this study we used human samples of lung adenocarcinoma (LUAD) patients, taking advantage of a mouse model of smoking cessation. Human samples were stratified according to the smoking status, high-risk factor for this type of tumor. Both public transcriptomic and human samples obtained by a clinical trial proved that AIM2 was upregulated either in terms of mRNA or protein, respectively, in the tumor mass according to the TNM stage, but it did not relate to the smoking status, age and sex. The upregulation of AIM2 was correlated to an immunosuppressive environment according to resting/non-active dendritic cells (DCs) and T regulatory cells, as demonstrated in both human samples and by means of an experimental model of smoking mice. Computational analysis showed that AIM2 upregulation was correlated to both an inflammasome profile, responsible for the poor prognosis of non-smoker and smoker LUAD patients, and to a non-inflammasome profile for former smoker. In conclusion, our study demonstrated that AIM2 is involved in lung carcinogenesis either in a canonical and non-canonical manner due to an immunosuppressive microenvironment associated to a dismal prognosis of LUAD patients

    Activation of the AIM2 Receptor in Circulating Cells of Post-COVID-19 Patients With Signs of Lung Fibrosis Is Associated With the Release of IL-1α, IFN-α and TGF-β.

    Severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2), responsible for COVID-19, has caused a global pandemic. Observational studies revealed a condition, herein called as Long-COVID syndrome (PC), that affects both moderately and severely infected patients, reducing quality-of-life. The mechanism/s underlying the onset of fibrotic-like changes in PC are still not well defined. The goal of this study was to understand the involvement of the Absent in melanoma-2 (AIM2) inflammasome in PC-associated lung fibrosis-like changes revealed by chest CT scans. Peripheral blood mononuclear cells (PBMCs) obtained from PC patients who did not develop signs of lung fibrosis were not responsive to AIM2 activation by Poly dA:dT. In sharp contrast, PBMCs from PC patients with signs of lung fibrosis were highly responsive to AIM2 activation, which induced the release of IL-1α, IFN-α and TGF-β. The recognition of Poly dA:dT was not due to the activation of cyclic GMP-AMP (cGAMP) synthase, a stimulator of interferon response (cGAS-STING) pathways, implying a role for AIM2 in PC conditions. The release of IFN-α was caspase-1- and caspase-4-dependent when AIM2 was triggered. Instead, the release of pro-inflammatory IL-1α and pro-fibrogenic TGF-β were inflammasome independent because the inhibition of caspase-1 and caspase-4 did not alter the levels of the two cytokines. Moreover, the responsiveness of AIM2 correlated with higher expression of the receptor in circulating CD14+ cells in PBMCs from patients with signs of lung fibrosis
