    ECOSYSTEM APPROACH TO FISHERIES MANAGEMENT IN THE SOUTHERN BENGUELA: A WORKSHOP OVERVIEW

    A workshop was held in Cape Town in December 2002 to introduce the concept of an ecosystem approach to fisheries (EAF) management in the southern Benguela, and to examine the options for implementing an EAF in South Africa. The workshop considered alternative modelling approaches that may have potential for an ecosystem approach to fisheries. Consensus was that an EAF should be implemented in South Africa through an incremental process, starting immediately. Ecosystem models can be used to provide guidance on reference points and broader management objectives still currently set on the basis of single-species assessments. Such additional information would be incorporated into the decision-making process, and comments received at a management level would also feed back to the modelling process. It was suggested that, at the scientific level, an ecosystem modelling perspective could be incorporated into existing single-species management recommendations by testing them with ecosystem models. Compilation of an “ecosystem considerations” document was recommended to initiate the process. It was proposed that a dedicated EAF working group be established in South Africa to advise on the process of implementing an EAF in the various fisheries, and to provide overarching guidance and to ensure consistency in integrating existing data and information for informing the management process

    Wolf Population Dynamics

    A LARGE, DARK WOLF poked his nose out of the pines in Yellowstone National Park as he thrust a broad foot deep into the snow and plowed ahead. Soon a second animal appeared, then another, and a fourth. A few minutes later, a pack of thirteen lanky wolves had filed out of the pines and onto the open hillside. Wolf packs are the main social units of a wolf population. As numbers of wolves in packs change, so too, then, does the wolf population (Rausch 1967). Trying to understand the factors and mechanisms that affect these changes is what the field of wolf population dynamics is all about. In this chapter, we will explore this topic using two main approaches: (1) meta-analysis using data from studies from many areas and periods, and (2) case histories of key long-term studies. The combination presents a good picture-a picture, however, that is still incomplete. We also caution that the data sets summarized in the analyses represent snapshots of wolf population dynamics under widely varying conditions and population trends, and that the figures used are usually composites or averages. Nevertheless, they should allow generalizations that provide important insight into wolf population dynamics

    A multicentre integration of a computer-led follow-up of prostate cancer is valid and safe

    Background Prostate cancer (CaP) has a rising number of patients requiring routine follow up. In this study, we aimed to test a computer led follow up service for prostate cancer in two UK hospitals. The testing aimed to validate the computer Expert system in making clinical decisions according to the individual patient’s clinical need. The valid model should accurately identify patients with disease recurrence or treatment failure based on their blood test and clinical picture. Methods A clinical decision support system (CDSS) was developed from European (EAU) and national (NICE) guidelines along with knowledge acquired from Urologists. This model was then applied in two UK hospitals to review patients post CaP treatment. These patients’ data (n= 200) were then reviewed by two independent Urology consultants (blinded from the CDSS and other consultant’s rating) and the agreement was calculated by kappa statistics for validation. The second objective aimed to verify the system by estimating the system reliability. Results The two individual urology consultants identified 12 % & 15% of the patients to have potential disease progression and recommended their referral to the Urology care. The kappa coefficient for the agreement between the CDSS and the 2 consultants was 0.81 (p < 0.001) and 0.84 (p < 0.001). The agreement among both specialist was also high with k = 0.83 (p < 0.001). The system reliability was estimated on all cases and this demonstrated 100% repeatability of the decisions. Conclusion The computer led follow up is a valid model for providing safe follow up for prostate cancer

    Polyvalent diazonium polymers provide efficient protection of oncolytic adenovirus Enadenotucirev from neutralising antibodies while maintaining biological activity in vitro and in vivo

    Oncolytic viruses offer many advantages for cancer therapy when administered directly to confined solid tumors. However, the systemic delivery of these viruses is problematic due to host immune response, undesired interactions with blood components and inherent targeting to the liver. Efficacy of systemically administered viruses has been improved by masking viral surface proteins with polymeric materials, through modulation of viral pharmacokinetic profile and accumulation in tumors in vivo. Here we describe a new class of polyvalent reactive based upon poly(N-(2-hydroxypropyl)methacrylamide) (polyHPMA) with diazonium reactive groups and their application in the modification of the chimeric oncolytic virus Enadenotucirev (EnAd). A series of six reactive copolymers with different chain lengths and density of reactive groups was synthesised and used to coat EnAd. Polymer coating was found to be extremely efficient with concentrations as low as 1 mg/mL resulting in complete (>99%) ablation of neutralising antibody binding. Coating efficiency was found to be dependent on both chain length and reactive group density. Coated viruses were found to have reduced transfection activity both in vitro and in vivo with greater protection against neutralising antibodies resulting in lower transgene production. However, in the presence of neutralising antibodies some in vivo transgene expression was maintained for coated virus compared to the uncoated control. Reduction in transgene expression was found to not be solely due to reduced cellular uptake but due to reducing unpackaging of the virus within the cells and reducing replication indicating that polymer coating does not cause permanent inactivation of the virus. These data suggest that virus activity may be modulated by appropriate design of coating polymers while retaining protection against neutralising antibodies

The preparations of the Einstein-Podolsky-Rosen resources as well as the number sum measurement resulting in projection to certain Bell state may be done conditionally with linear optical elements, i.e., beam splitters, phase shifters and zero-one-photon detectors.     Kondo Temperature for the Two-Channel Kondo Models of Tunneling Centers

    The possibility for a two-channel Kondo (2CK2CK) non Fermi liquid state to appear in a metal as a result of the interaction between electrons and movable structural defects is revisited. As usual, the defect is modeled by a heavy particle moving in an almost symmetric double-well potential (DWP). Taking into account only the two lowest states in DWP is known to lead to a Kondo-like Hamiltonian with rather low Kondo temperature, TKT_K. We prove that, in contrast to previous believes, the contribution of higher excited states in DWP does not enhance TKT_K. On the contrary, TKT_K is reduced by three orders of magnitude as compared with the two-level model: the prefactor in TKT_K is determined by the spacing between the second and the third levels in DWP rather than by the electron Fermi energy. Moreover, TKT_K, turns out to be parametrically smaller than the splitting between the two lowest levels. Therefore, there is no microscopic model of movable defects which may justify non-Fermi liquid 2CK2CK phenomenology.Comment: 5 pages, 4 .eps figure

    Effects of commercial formaldehyde inclusion and lysine level on nursery pig performance

Journal of Animal Science, 94, 191-192. doi:10.2527/msasas2016-410Previous research has demonstrated that commercial formaldehyde products may reduce the risk of microbiological hazards in feed, but there is concern about the product binding Lys and reducing growth performance. Therefore, the objective of this experiment was to evaluate the role of 2 commercially-available formaldehyde sources on growth performance of pigs fed a Lys-sufficient or Lys-deficient diet. A total of 299 pigs (PIC 327 × 1050; initial BW 15.2 kg) were used in a 14-d study. Dietary treatments were arranged in a 3 × 2 factorial design with 3 formaldehyde inclusions: none vs. 3.25 kg per tonne SalCURB (Kemin Industries, Inc., Des Moines, IA) vs. 3.0 kg per tonne Termin-8 (Anitox Corp, Lawrenceville, GA) and 2 Lys levels: Adequate (1.25% SID Lys) vs. Low (1.10% SID Lys). Product concentrations were established from supplier recommendations, and diets were treated with in commercial mills utilizing supplier-specific equipment. Pens of pigs were balanced by initial BW and randomly allotted to treatments with 5 pigs per pen and 10 pens per treatment. Data were analyzed in a factorial design using the GLIMMIX procedure of SAS. Overall, there was a tendency (P 0.10). In summary, SalCURB inclusion did not alter nursery pig growth performance compared to the untreated basal diet, regardless of Lys level. However, the inclusion of Termin-8 tended to result in poorer G:F in adequate Lys diets and poorer ADG and G:F in low Lys diets compared to an untreated control

    Correlation of baseline biomarkers with clinical outcomes and response to fulvestrant with vandetanib or placebo in patients with bone predominant metastatic breast cancer: An OCOG ZAMBONEY sub-study

    AbstractBackgroundBone metastases are common in women with breast cancer and often result in skeletal related events (SREs). As the angiogenic factor vascular endothelial growth factor (VEGF) regulates osteoclast activity and is associated with more extensive bone metastases and SRE risk in metastatic breast cancer, we hypothesized that blockade of VEGF signaling could be a therapeutic strategy for inhibiting bone metastases progression and possibly prolonging overall (OS) or progression-free survival (PFS). The Zamboney trial was a randomized placebo-controlled study designed to assess whether patients with bone predominant metastatic breast cancer benefited from addition of the VEGF receptor (VEGFR) targeting agent, vandetanib, to endocrine therapy with fulvestrant. As a companion study, evaluation of biomarkers and their potential association with response to vandetanib or SRE risk was performed.MethodsBaseline overnight fasted serum from enrolled patients was analyzed for levels of various putative biomarkers including; VEGF-A, soluble (s)VEGFR2, sVEGFR3, transforming growth factor (TGF)-β1 and activinA by ELISA. Spearman correlation coefficients and Wilcoxon rank sum tests were used to investigate potential relationships between biomarker values and baseline clinical parameters. Prognostic and predictive ability of each marker was investigated using Cox proportional hazards regression with adjustments for treatment and baseline strata of serum CTx (<400 versus ≥400ng/L).ResultsOf 129 enrolled patients, serum was available for analysis in 101; 51 in vandetanib and 50 in placebo arm. Mean age amongst consenting patients was 59.8 years. Clinical characteristics were not significantly different between patients with or without serum biomarker data and serum markers were similar for patients by treatment arm. Baseline sVEGFR2 was prognostic for OS (HR=0.77, 95% CI=0.61–0.96, p=0.020), and although a modest association was observed, it was not significant for PFS (HR=0.90, 95% CI=0.80–1.01, p=0.085) nor time to first SRE (HR=0.82, 95% CI=0.66–1.02, p=0.079). When interaction terms were evaluated, sVEGFR2 was not found to be predictive of response to vandetanib, although a modest association remained with respect to PFS (interaction p=0.085). No other marker showed any significant prognostic or predictive ability with any measured outcome.ConclusionsIn this clinical trial, sVEGFR2 appeared prognostic for OS, hence validation of sVEGFR2 should be conducted. Moreover, the role of sVEGFR2 in breast cancer bone metastasis progression should be elucidated
