    HgCl2 addition to pore water samples from cold seeps can affect the geochemistry of dissolved inorganic carbon ([DIC], δ13CDIC)

    The conventional use of mercuric chloride (HgCl2) as an antimicrobial agent in water samples for dissolved inorganic carbon (DIC) analysis poses health and environmental risks related to its handling and disposal. Even though there is an increasing interest in quantifying pore water DIC contribution to the ocean carbon cycle and deep-sea acidification, the paucity of comparative studies on marine pore waters prevents the modification of sampling protocols and HgCl2 still remains widely used. Here, we compared DIC concentrations and δ13CDIC composition in pore water samples from methane seepage areas in the Barents Sea and offshore N. Svalbard. Samples were extracted using 0.15 μm rhizon filters and split into two aliquots with 2–3 replicates each. Only one aliquot was treated with 10 μL saturated HgCl2(aq) and all samples were stored in the dark at 4 ◦C, prior to measurements ~30 days later. The samples yielded extremely wide ranges of DIC concentrations, from 1.8 mM to 20.1 mM, and δ13CDIC values, from − 36.0‰ to − 1.6‰ (VPDB), due to variable contributions of methane-derived DIC to the pore water system. Overall, we obtained lower [DIC] (77% samples; N = 26) and heavier δ13C values (79% samples; N = 42) in Hg-treated samples. Isotopic and concentration differences larger than the uncertainties on the mean of replicated measurements (±0.2–0.5‰; ± 0.5 mM) and analytical precision (0.15‰; 0.71 mM) represent the 38% and 19% of the samples, with offsets of up to 7.4‰ and 1.9 mM, respectively. The largest offsets are in agreement with our CO2-degassing model, suggesting an interaction between mercuric chloride and dissolved hydrogen sulfide released by sulfate-driven methane oxidation. We therefore caution against further use of HgCl2 for DIC studies of marine pore waters from cold seeps

    Biogeochemical investigations of methane seepage at the ultraslow-spreading Arctic mid-ocean ridge: Svyatogor ridge, Fram Strait

    The Arctic continental shelves are important reservoirs of methane stored in gas hydrates and deeper geological formations. However, little is known about methane dynamics in deeper oceanic settings such as mid-ocean ridges, mainly due to operational challenges related to their remoteness. This study investigates a recently discovered methane seepage environment at Svyatogor Ridge, a sediment-covered transform fault on the western flank of the Arctic mid-ocean ridge west of Svalbard. Svyatogor Ridge was previously hypothesized to host deep gas hydrates and active fluid flow systems, which is a unique combination for this setting and requires geochemical evidence. Based on sediment and foraminiferal geochemistry, we demonstrate that Svyatogor Ridge hosts a shallow methane cycle with anaerobic oxidation of microbial methane, sustaining chemosynthetic communities at the seafloor. Our geochemical datasets also suggest that methane fluxes were higher in the past than today and long-lasting, with episodes of intense methane oxidation recorded in pre-Holocene sediments. Methane seepage is still ongoing at this location, although no evidence exists that methane is reaching the sea surface. The results provide the first evidence of a methane seepage environment ever reported from the ultra-slow spreading Arctic mid-ocean ridge, thus calling for a reevaluation of the role of this type of ridge in the ocean methane cycle

    STRONTIUM ISOTOPE STRATIGRAPHY AS A CONTRIBUTION FOR DATING MIOCENE SHELF CARBONATES (S. MARINO FM., NORTHERN APENNINES)

    This paper provides new data on strontium isotope stratigraphy applied to the Miocene heterozoan shelfal carbonates of the S. Marino Fm. (Marecchia Valley, northern Apennines). Sr isotopic analyses were carried out on oyster shells, bryozoans and bulk-rocks from the lower-middle carbonate portion of the section. In the upper part of the succession that shows evidence of detrital influx, 87Sr/86Sr analyses were performed on foraminifera tests, separating planktonic and benthic forms. Results were compared with calcareous nannofossil biostratigraphic data from the same levels, in order to test the reliability of Sr dating in mixed carbonate-siliciclastic sediments. Mean ages obtained from oysters range between 16.9 Ma and 16.3 Ma. Very similar results are obtained using bryozoans (16.5 Ma to 16.1 Ma) and bulk-rocks (16.8 Ma to 16.2 Ma). These results allow to better constrain the age of the massive carbonate shelf, referable to the upper Burdigalian. In the upper carbonate-siliciclastic portion of the shelf, numerical ages obtained from planktonic and benthic foraminifera are in good agreement with nannofossil biozones (mean ages respectively around 15.3 Ma and 14.5 Ma) although they display wide confidence intervals. These wide age uncertainties depend on the slow rate of change of marine 87Sr/86Sr through time that characterizes the interval between ~15 and ~13.5 Ma

    Hand-written letters and photo albums linking geoscientists with school classes

    Do we miss something about &laquo;traditional&rdquo; media such as handwritten letters and photography before the digital age? Some of the authors remember this age fondly, and we wanted to see if this fondness could be translated into a science dialogue project with school classes. We designed and carried out a communication process with 4 classes at different schools across Europe. During this process, each class would interact with a single scientist primarily via hand-written questions &amp; letters, and a Polaroid photo album. The scientists would make this unique, one-of-a-kind album whilst on board a research expedition in the Barents Sea. We asked the question whether this process might show any benefits to the school students involved. To answer this, we asked the students to write up their thoughts on communicating with a scientist in this way. We analysed the texts and found that most students thought the letters and polaroid albums were a &ldquo;beautiful experience&rdquo;. Others commented on how important it is to actually put pen to paper and write, since they use (almost) only digital media these days. Most importantly, the students learnt different elements of the science connected to the research expedition, but also about the scientific process in general. And, equally important, some of the students were surprised and thankful that the scientists took the time to communicate with them in such a personal way. These results could possibly have been achieved using other media, however the hand-written letters and Polaroids worked very well. They also seemed to conjure up some of the personal memories that we have about communication not so long ago. Maybe there is something to be said for slowing things down with our science communication projects and making them more personal and unique. This is something that snail-mail and making photo albums forces us to do.</p

    Profibrotic Effects of Endothelin-1 on Fibroblasts Are Mediated by Aldosterone in Vitro: Relevance to the Pathogenesis and Therapy of Systemic Sclerosis and Pulmonary Arterial Hypertension

    Endothelin-1 (ET-1) is a vasoactive and profibrotic peptide that plays a pivotal role in diseases such as systemic sclerosis (SSc) and pulmonary arterial hypertension (PAH), by inducing fibrosis and vascular remodeling. Such effects may be sustained by the induction of aldosterone production and reactive oxygen species (ROS). We have used fibroblasts obtained from skin of healthy donors and SSc patients and commercial fibroblasts from lung to evaluate whether ET-1 is able to stimulate ROS production directly or indirectly through aldosterone induction. We found that ET-1 receptors are present in all types of fibroblasts analyzed, whereas the expression of mineralocorticoid receptor (MCR) is lower in dermal fibroblasts from healthy donors (HDFs) compared to fibroblasts derived from lung (HPFs) or from skin of SSc patients (SScHDFs). ET-1 induces ROS production in HDFs and SScHDFs after 24 h of incubation involving its receptor B (ETB), whereas aldosterone exerts its effects after 40 min of incubation. Moreover, ROS production was inhibited by the pre-incubation of cells with MCR inhibitor. Our results indicate that ET-1 induces ROS indirectly through aldosterone production suggesting that aldosterone may play a pivotal role in the pathogenesis of SSc and PAH

    Endothelin Receptors Expressed by Immune Cells Are Involved in Modulation of Inflammation and in Fibrosis: Relevance to the Pathogenesis of Systemic Sclerosis

    open13noEndothelin-1 (ET-1) plays a pivotal role in vasoconstriction, fibrosis, and inflammation, the key features of systemic sclerosis (SSc). ET-1 receptors (ETA and ETB) are expressed on endothelial cells, smooth muscle cells, and fibroblasts, but their presence on immune cells has not been deeply investigated so far. Endothelin receptors antagonists such as bosentan have beneficial effects on vasoconstriction and fibrosis, but less is known about their potential anti-inflammatory effects. We studied the expression of ET-1 receptors on immune cells (T and B lymphocytes, monocytes, and neutrophils) and the link between ET-1 and inflammation in patients with SSc. We show here that ET-1 exerts a proinflammatory effect in CD4+ T cells, since it induces an increased IFN-γ production; preincubation with antagonists of both receptors reduces IFN-γ production. Moreover, following ET-1 stimulation, neutrophils produce proinflammatory mediators, thus amplifying the effects of activated CD4+ T cells. Our data indicate that ET-1 system is involved in the pathogenesis of inflammation and fibrosis typical of SSc, through the activation of T lymphocytes and neutrophils and the consequent release of proinflammatory and profibrotic cytokines.     Hot Vents Beneath an Icy Ocean: The Aurora Vent Field, Gakkel Ridge, Revealed

    Evidence of hydrothermal venting on the ultra-slow spreading Gakkel Ridge in the Central Arctic Ocean has been available since 2001, with first visual evidence of black smokers on the Aurora Vent Field obtained in 2014. But it was not until 2021 that the first ever remotely operated vehicle (ROV) dives to hydrothermal vents under permanent ice cover in the Arctic were conducted, enabling the collection of vent fluids, rocks, microbes, and fauna. In this paper, we present the methods employed for deep-sea ROV operations under drifting ice. We also provide the first description of the Aurora Vent Field, which includes three actively venting black smokers and diffuse flow on the Aurora mound at ~3,888 m depth on the southern part of the Gakkel Ridge (82.5°N). The biological communities are dominated by a new species of cocculinid limpet, two small gastropods, and a melitid amphipod. The ongoing analyses of Aurora Vent Field samples will contribute to positioning the Gakkel Ridge hydrothermal vents in the global biogeographic puzzle of hydrothermal vents
