    Polycyclic Aromatic Hydrocarbons (PAHs) are pollutants that can cause mutagenic and carcinogenic to humans and cause environmental harm. The present study evaluated the degradation of phenanthrene and anthracene contaminants present in sandy soils using sodium persulfate activated with Fe2+ in acid medium. It was evaluated three sandy soils named A, B and C with different organic carbon tenors of 27, 16 and 31 g kg-1 respectively. After 24 hours of reaction, it was achieved a treatment efficiency of 60, 45 and 11% for the phenanthrene and 97, 96 and 81% for anthracene. There was no change in the composition of organic carbon in soil after the use of the oxidant. However there was a change in the constitution of the soil humic fractions. The use of the persulfate oxidant may be considered promising for use in soil because it does not compete with the organic carbon in the soil

    Infected site-restricted Foxp3+ natural regulatory T cells are specific for microbial antigens

    Natural regulatory T (T reg) cells are involved in control of the immune response, including response to pathogens. Previous work has demonstrated that the repertoire of natural T reg cells may be biased toward self-antigen recognition. Whether they also recognize foreign antigens and how this recognition contributes to their function remain unknown. Our studies addressed the antigenic specificity of natural T reg cells that accumulate at sites of chronic infection with Leishmania major in mice. Our results support the idea that natural T reg cells are able to respond specifically to foreign antigens in that they strongly proliferate in response to Leishmania-infected dendritic cells, they maintain Foxp3 expression, and Leishmania-specific T reg cell lines can be generated from infected mice. Surprisingly, the majority of natural T reg cells at the infected site are Leishmania specific. Further, we showed that parasite-specific natural T reg cells are restricted to sites of infection and that their survival is strictly dependent on parasite persistence

    Nel corso del 2003, nell’ambito del piano di sorveglianza passiva sulle Encefalopatie Spongiformi Trasmissibili (TSEs), per la prima volta in Sardegna, è stata diagnosticata istologicamente ed immunoistochimicamente la scrapie in una capra. Il soggetto proveniva da un gregge nel quale da 2 anni si manifestava una patologia polimorfa caratterizzata talvolta da sintomi neurologici e da una mortalità intorno al 20%. Sulla base della legge attualmente in vigore in Italia si è proceduto all’abbattimento di tutte le capre presenti in allevamento (n = 93). Durante l’abbattimento venivano prelevati da ciascun capo obex, linfonodi retrofaringei e tonsille. L’esame istologico dell’obex non evidenziava quadri lesivi di tipo spongiforme mentre l’esame immunoistochimico del tessuto nervoso e linfatico evidenziava la presenza di cinque soggetti nello stato pre-clinico della malattia

    CD4+CD25+ Regulatory T Cells Can Mediate Suppressor Function in the Absence of Transforming Growth Factor β1 Production and Responsiveness

    CD4+CD25+ regulatory T cells inhibit organ-specific autoimmune diseases induced by CD4+CD25−T cells and are potent suppressors of T cell activation in vitro. Their mechanism of suppression remains unknown, but most in vitro studies suggest that it is cell contact–dependent and cytokine independent. The role of TGF-β1 in CD4+CD25+ suppressor function remains unclear. While most studies have failed to reverse suppression with anti–transforming growth factor (TGF)-β1 in vitro, one recent study has reported that CD4+CD25+ T cells express cell surface TGF-β1 and that suppression can be completely abrogated by high concentrations of anti–TGF-β suggesting that cell-associated TGF-β1 was the primary effector of CD4+CD25+-mediated suppression. Here, we have reevaluated the role of TGF-β1 in CD4+CD25+-mediated suppression. Neutralization of TGF-β1 with either monoclonal antibody (mAb) or soluble TGF-βRII-Fc did not reverse in vitro suppression mediated by resting or activated CD4+CD25+ T cells. Responder T cells from Smad3−/− or dominant-negative TGF-β type RII transgenic (DNRIITg) mice, that are both unresponsive to TGF-β1–induced growth arrest, were as susceptible to CD4+CD25+-mediated suppression as T cells from wild-type mice. Furthermore, CD4+CD25+ T cells from neonatal TGF-β1−/− mice were as suppressive as CD4+CD25+ from TGF-β1+/+ mice. Collectively, these results demonstrate that CD4+CD25+ suppressor function can occur independently of TGF-β1

    Trimethoxylated halogenated chalcones as dual inhibitors of mao-b and bace-1 for the treatment of neurodegenerative disorders

    Six halogenated trimethoxy chalcone derivatives (CH1–CH6) were synthesized and spec-trally characterized. The compounds were further evaluated for their inhibitory potential against monoamine oxidases (MAOs) and β-secretase (BACE-1). Six compounds inhibited MAO-B more effectively than MAO-A, and the 2′,3′,4′-methoxy moiety in CH4–CH6 was more effective for MAO-B inhibition than the 2′,4′,6′-methoxy moiety in CH1–CH3. Compound CH5 most potently inhibited MAO-B, with an IC50 value of 0.46 µM, followed by CH4 (IC50 = 0.84 µM). In 2′,3′,4′-methoxy derivatives (CH4-CH6), the order of inhibition was –Br in CH5 >-Cl in CH4 >-F in CH6 at the para-position in ring B of chalcone. CH4 and CH5 were selective for MAO-B, with selectivity index (SI) values of 15.1 and 31.3, respectively, over MAO-A. CH4 and CH5 moderately inhibited BACE-1 with IC50 values of 13.6 and 19.8 µM, respectively. When CH4 and CH5 were assessed for their cell viability studies on the normal African Green Monkey kidney cell line (VERO) using MTT assays, it was noted that both compounds were found to be safe, and only a slightly toxic effect was observed in concentrations above 200 µg/mL. CH4 and CH5 decreased reactive oxygen species (ROS) levels of VERO cells treated with H2 O2, indicating both compounds retained protective effects on the cells by antioxidant activities. All compounds showed high blood brain barrier permeabilities analyzed by a parallel artificial membrane permeability assay (PAMPA). Molecular docking and ADME prediction of the lead compounds provided more insights into the rationale behind the binding and the CNS drug likeness. From non-test mutagenicity and cardiotoxicity studies, CH4 and CH5 were non-mutagenic and non-/weak-cardiotoxic. These results suggest that CH4 and CH5 could be considered candidates for the cure of neurological dysfunctions

    Serum methylarginines and spirometry-measured lung function in older adults

    Rationale: Methylarginines are endogenous nitric oxide synthase inhibitors that have been implicated in animal models of lung disease but have not previously been examined for their association with spirometric measures of lung function in humans. Objectives: This study measured serum concentrations of asymmetric and symmetric dimethylarginine in a representative sample of older community-dwelling adults and determined their association with spirometric lung function measures. Methods: Data on clinical, lifestyle, and demographic characteristics, methylated arginines, and L-arginine (measured using LC-MS/MS) were collected from a population-based sample of older Australian adults from the Hunter Community Study. The five key lung function measures included as outcomes were Forced Expiratory Volume in 1 second, Forced Vital Capacity, Forced Expiratory Volume in 1 second to Forced Vital Capacity ratio, Percent Predicted Forced Expiratory Volume in 1 second, and Percent Predicted Forced Vital Capacity. Measurements and Main Results: In adjusted analyses there were statistically significant independent associations between a) higher asymmetric dimethylarginine, lower Forced Expiratory Volume in 1 second and lower Forced Vital Capacity; and b) lower L-arginine/asymmetric dimethylarginine ratio, lower Forced Expiratory Volume in 1 second, lower Percent Predicted Forced Expiratory Volume in 1 second and lower Percent Predicted Forced Vital Capacity. By contrast, no significant associations were observed between symmetric dimethylarginine and lung function. Conclusions: After adjusting for clinical, demographic, biochemical, and pharmacological confounders, higher serum asymmetric dimethylarginine was independently associated with a reduction in key measures of lung function. Further research is needed to determine if methylarginines predict the decline in lung function

    ENVIRONMENTAL-FRIENDLY SYNTHESIS OF FLAVONE AND ITS EFFECT ON LETTUCE-APHID INTERACTION

    La búsqueda de compuestos químicos que sean eficaces para el control de plagas y a su vez inocuos para la salud del hombre y el ambiente se presenta como una necesidad. En el presente trabajo se informa un procedimiento simple y compatible con el ambiente para la preparación de un compuesto perteneciente a la familia de los fenilpropanoides (flavona), a través de ciclodeshidratación de 1-(2-hidroxifenil)-3-fenil-1,3-propanodiona en condiciones de impacto ambiental reducido (síntesis en ausencia de solvente, catalizada con ácido sólido recuperable y utilizando calefacción mediante microondas y con tiempos reducidos), y la evaluación del crecimiento de una especie hortícola (lechuga) por la acción de este compuesto. El porcentaje de geminación de las semillas de lechuga disminuyó con 400 y 500 ppm de flavona respecto del control, mientras que a partir de 200 ppm de flavona se observó un menor crecimiento de las plántulas debido a la disminución de la longitud del hipocótilo. Aunque no se observaron efectos por contacto directo de la flavona sobre el cuerpo de los áfidos, pudo identificarse repelencia del insecto en discos de lechuga embebidos en 500 ppm de flavona.The search for chemical compounds that are effective for pest control and in turn harmless to human health and the environment is needed. In this work, a simple and environmentally compatible procedure is reported for the preparation of a compound belonging to the phenylpropanoid (flavone) family, through cyclodehydration of 1- (2-hydroxyphenyl) -3-phenyl-1, 3-propanedione under conditions of reduced environmental impact (synthesis in the absence of solvent, catalyzed with recoverable solid acid and using microwave heating and with reduced times), and the evaluation of the growth of a horticultural species (lettuce) by the action of this compound. The percentage of germination of lettuce seeds decreased with 400 and 500 ppm of flavone compared to the control, while from 200 ppm of flavone a lower growth of the seedlings was observed due to the decrease in the length of the hypocotyl. Although no effects were observed by direct contact of flavone on the aphid body, insect repellency could be identified in lettuce discs embedded in 500 ppm flavone

    The Collective Article “New Mediterranean Biodiversity Records” of the Mediterranean Marine Science journal offers the means to publish biodiversity records in the Mediterranean Sea. The current article has adopted a country-based classification and the countries are listed according to their geographic position, from west to east. New biodiversity data are reported for 7 different countries, although one species reported from Malta is new for the entire Mediterranean basin, and is presumably also present in Israel and Lebanon (see below, under Malta). Italy: the rare native fish Gobius kolombatovici is first reported from the Ionian Sea, whilst the alien jellyfish Rhopilema nomadica and the alien fish Oplegnathus fasciatus are first reported from the entire country. The presence of O. fasciatus from Trieste is concomitantly the first for the entire Adriatic Sea. Finally, the alien bivalve Arcuatula senhousia is reported for the first time from Campania (Tyrrhenian Sea). Tunisia: a bloom of the alien crab ortunus segnis is first reported from the Gulf of Gabes, where it was considered as casual. Malta: the alien flatworm Maritigrella fuscopunctata is recorded in the Mediterranean Sea for the first time, on the basis of 25 specimens. At the same time, web searches include possible unpublished records from Israel and Lebanon. The alien crab P. segnis, already mentioned above, is first formally reported from Malta based on specimens collected in 1972. Concomitantly, the presence of Callinectes sapidus in Maltese waters is excluded since based on misidentifications. Greece: the Atlantic northern brown shrimp Penaeus atzecus, previously known from the Ionian Sea from sporadic records only, is now well established in Greek and international Ionian waters. The alien sea urchin Diadema setosum is reported for the second time from Greece, and its first record from the country is backdated to 2010 in Rhodes Island. The alien lionfish Pterois miles is first reported from Greece and concomitantly from the entire Aegean Sea. Turkey: the alien rhodophyte Antithamnion hubbsii is first reported from Turkey and the entire eastern Mediterranean. New distribution data are also provided for the native fishes Alectis alexandrina and Heptranchias perlo. In particular, the former record consists of a juvenile measuring 21.38 mm total length, whilst the latter by a mature male. Cyprus: the rare native cephalopod Macrotritopus defilippi, and the alien crab Atergatis roseus, sea slug Plocamopherus ocellatus and fish Cheilodipterus novemstriatus are first recorded from the entire country. Lebanon: the alien crabs Actaea savignii and Matuta victor, as well as the alien fish Synanceia verrucosa, are first recorded from the entire country. In addition, the first Mediterranean record of A. savignii is backdated to 2006, whilst the high number of M. victor specimens observed in Lebanon suggest its establishment in the Basin. The Atlantic fishes Paranthias furcifer and Seriola fasciata, and the circumtropical Rachycentron canadum, are also first reported from the country. The P. furcifer record backdates its presence in the Mediterranean to 2007, whilst S. fasciata records backdate its presence in the eastern Mediterranean to 2005. Finally, two of these latter species have been recently ascribed to alien species, but all three species may fit the cryptogenic category, if not a new one, better.peer-reviewe

    Mesenchymal Stromal Cells Improve Salivary Function and Reduce Lymphocytic Infiltrates in Mice with Sjögren's-Like Disease

    Non-obese diabetic (NOD) mice develop Sjögren's-like disease (SS-like) with loss of saliva flow and increased lymphocytic infiltrates in salivary glands (SGs). There are recent reports using multipotent mesenchymal stromal cells (MSCs) as a therapeutic strategy for autoimmune diseases due to their anti-inflammatory and immunomodulatory capabilities. This paper proposed a combined immuno- and cell-based therapy consisting of: A) an injection of complete Freund's adjuvant (CFA) to eradicate autoreactive T lymphocytes, and B) transplantations of MSCs to reselect lymphocytes. The objective of this was to test the effectiveness of CD45(-)/TER119(-) cells (MSCs) in re-establishing salivary function and in reducing the number of lymphocytic infiltrates (foci) in SGs. The second objective was to study if the mechanisms underlying a decrease in inflammation (focus score) was due to CFA, MSCs, or CFA+MSCs combined.Donor MSCs were isolated from bones of male transgenic eGFP mice. Eight week-old female NOD mice received one of the following treatments: insulin, CFA, MSC, or CFA+MSC (combined therapy). Mice were followed for 14 weeks post-therapy. CD45(-)/TER119(-) cells demonstrated characteristics of MSCs as they were positive for Sca-1, CD106, CD105, CD73, CD29, CD44, negative for CD45, TER119, CD11b, had high number of CFU-F, and differentiated into osteocytes, chondrocytes and adipocytes. Both MSC and MSC+CFA groups prevented loss of saliva flow and reduced lymphocytic infiltrations in SGs. Moreover, the influx of T and B cells decreased in all foci in MSC and MSC+CFA groups, while the frequency of Foxp3(+) (T(reg)) cell was increased. MSC-therapy alone reduced inflammation (TNF-α, TGF-β), but the combination of MSC+CFA reduced inflammation and increased the regenerative potential of SGs (FGF-2, EGF).The combined use of MSC+CFA was effective in both preventing saliva secretion loss and reducing lymphocytic influx in salivary glands

    The Genomic and Immune Landscapes of Lethal Metastatic Breast Cancer.

    The detailed molecular characterization of lethal cancers is a prerequisite to understanding resistance to therapy and escape from cancer immunoediting. We performed extensive multi-platform profiling of multi-regional metastases in autopsies from 10 patients with therapy-resistant breast cancer. The integrated genomic and immune landscapes show that metastases propagate and evolve as communities of clones, reveal their predicted neo-antigen landscapes, and show that they can accumulate HLA loss of heterozygosity (LOH). The data further identify variable tumor microenvironments and reveal, through analyses of T cell receptor repertoires, that adaptive immune responses appear to co-evolve with the metastatic genomes. These findings reveal in fine detail the landscapes of lethal metastatic breast cancer.CRUK
