    Nature of behaviours that challenge in residents living in aged care homes: implications for psychosocial interventions and service development

    Background/Aims: Competing theories exist regarding the causes and nature of behaviours that challenge (BC). The present study attempted to provide better insight into these behaviours and determine whether there were any underlying relationships for people living in care homes with and without dementia. Methods: Cross sectional data for 2185 residents living in 63 UK care homes using the Challenging Behaviour Scale were collected. The incidence (i.e. presence/absence) of 25 commonly reported behaviours that challenge staff were determined using dichotomous scoring. Cluster analysis and Mokken scaling were used to examine underlying relationships. Mokken is a method of analysing data to determine whether there is a ‘latent’ structure within a data set. Results: The prevalence of reported BC was 87.5%. Cluster analysis revealed three main clusters: apathy, agitation with internal focus, agitation with active external focus. For seven of the 25 items a hierarchical model emerged, where behaviours at the bottom of the hierarchy tended to occur in the presence of those higher up. Behaviours at the bottom of the hierarchy (dangerous behaviour) were less frequently observed than the items at the top (lack of self-care, verbal aggression). Conclusion: Some common BC may occur in groups and themed clusters. This study has shown that a hierarchical structure of BC in residents may be present. The findings testify to the complexity in the aetiology and treatment of BC and hence the need for focussed high intensity bio-psychosocial interventions to be targeted towards those with high levels of ‘unmet need’. Implications for future research and practice are discussed

    Imaging of the Stellar Population of IC10 with Laser Guide Star Adaptive Optics and the Hubble Space Telescope

    We present adaptive optics (AO) images of the central starburst region of the dwarf irregular galaxy IC10. The Keck 2 telescope laser guide star was used to achieve near diffraction-limited performance at H and K' (Strehls of 18% and 32%, respectively). The images are centered on the putative Wolf-Rayet (W-R) object [MAC92]24. We combine our AO images with F814W data from HST. By comparing the K' vs. [F814W]-K' color-magnitude diagram (CMD) with theoretical isochrones, we find that the stellar population is best represented by at least two bursts of star formation, one ~ 10 Myr ago and one much older (150-500 Myr). Young, blue stars are concentrated in the vicinity of [MAC92]24. This population represents an OB association with a half-light radius of about 3 pc. We resolve the W-R object [MAC92]24 into at least six blue stars. Four of these components have near-IR colors and luminosities that make them robust WN star candidates. By matching the location of C-stars in the CMD with those in the SMC we derive a distance modulus for IC10 of about 24.5 mag. and a foreground reddening of E(B-V) = 0.95. We find a more precise distance by locating the tip of the giant branch in the F814W, H, and K' luminosity functions. We find a weighted mean distance modulus of 24.48 +/- 0.08. The systematic error in this measurement, due to a possible difference in the properties of the RGB populations in IC10 and the SMC, is +/- 0.16 mag.Comment: 13 pages, 13 figures, ApJ in pres

    Mycobacterium tuberculosis RNA Expression Patterns in Sputum Bacteria Indicate Secreted Esx Factors Contributing to Growth are Highly Expressed in Active Disease

    To identify factors contributing to the ability of tubercle bacilli to grow in the lung during active infection, we analyzed RNA expression patterns in bacteria present in patient sputum. Prominent among bacterial transcripts identified were those encoding secreted peptides of the Esat-6 subfamily that includes EsxK and EsxL (Rv1197 and Rv1198). H37Rv esxKL and esxJI transcripts were differentially expressed under different growth conditions, and disruption of these genes altered growth phase kinetics in typical laboratory batch broth cultures. These growth defects, including the reduced intracellular growth of an ΔesxKL mutant in primary human macrophages, were reversed by either low multiplicity co-infection or co-culture with wild-type bacteria, demonstrating the ability of the secreted factors to rescue isogenic mutants. Complementing either only esxL or esxI alone (Rv1198 or Rv1037c) also reduced observed growth defects, indicating these genes encode factors capable of contributing to growth. Our studies indicate that the Mycobacterium tuberculosis Mtb9.9 family secreted factors EsxL and EsxI can act in trans to modulate growth of intracellular bacteria, and are highly expressed during active human lung infection

Host range: Compatible hosts vary in their susceptibility to citrus canker (CC), with grapefruit, lime and lemon being the most susceptible, sweet orange being moderately susceptible, and kumquat and calamondin being amongst the least susceptible. Microbiological properties: Xcc is a rod-shaped (1.5–2.0 X 0.5–0.75 mm), Gram-negative, aerobic bacterium with a single polar flagellum. The bacterium forms yellow colonies on culture media as a result of the production of xanthomonadin. Distribution: Present in South America, the British Virgin Islands, Africa, the Middle East, India, Asia and the South Pacific islands. Localized incidence in the USA, Argentina, Brazil, Bolivia, Uruguay, Senegal, Mali, Burkina Faso, Tanzania, Iran, Saudi Arabia, Yemen and Bangladesh.     Akt regulates centrosome migration and spindle orientation in the early Drosophila melanogaster embryo

    Correct positioning and morphology of the mitotic spindle is achieved through regulating the interaction between microtubules (MTs) and cortical actin. Here we find that, in the Drosophila melanogaster early embryo, reduced levels of the protein kinase Akt result in incomplete centrosome migration around cortical nuclei, bent mitotic spindles, and loss of nuclei into the interior of the embryo. We show that Akt is enriched at the embryonic cortex and is required for phosphorylation of the glycogen synthase kinase-3β homologue Zeste-white 3 kinase (Zw3) and for the cortical localizations of the adenomatosis polyposis coli (APC)–related protein APC2/E-APC and the MT + Tip protein EB1. We also show that reduced levels of Akt result in mislocalization of APC2 in postcellularized embryonic mitoses and misorientation of epithelial mitotic spindles. Together, our results suggest that Akt regulates a complex containing Zw3, Armadillo, APC2, and EB1 and that this complex has a role in stabilizing MT–cortex interactions, facilitating both centrosome separation and mitotic spindle orientation

K.Background: A region on Sus scrofa chromosome 4 (SSC4) surrounding single nucleotide polymorphism (SNP) marker WUR10000125 (WUR) has been reported to be strongly associated with both weight gain and serum viremia in pigs after infection with PRRS virus (PRRSV). A proposed causal mutation in the guanylate binding protein 5 gene (GBP5) is predicted to truncate the encoded protein. To investigate transcriptional differences between WUR genotypes in early host response to PRRSV infection, an RNA-seq experiment was performed on globin depleted whole blood RNA collected on 0, 4, 7, 10 and 14 days post-infection (dpi) from eight littermate pairs with one AB (favorable) and one AA (unfavorable) WUR genotype animal per litter. Results: Gene Ontology (GO) enrichment analysis of transcripts that were differentially expressed (DE) between dpi across both genotypes revealed an inflammatory response for all dpi when compared to day 0. However, at the early time points of 4 and 7dpi, several GO terms had higher enrichment scores compared to later dpi, including inflammatory response (p < 10(-7)), specifically regulation of NFkappaB (p < 0.01), cytokine, and chemokine activity (p < 0.01). At 10 and 14dpi, GO term enrichment indicated a switch to DNA damage response, cell cycle checkpoints, and DNA replication. Few transcripts were DE between WUR genotypes on individual dpi or averaged over all dpi, and little enrichment of any GO term was found. However, there were differences in expression patterns over time between AA and AB animals, which was confirmed by genotype-specific expression patterns of several modules that were identified in weighted gene co-expression network analyses (WGCNA). Minor differences between AA and AB animals were observed in immune response and DNA damage response (p = 0.64 and p = 0.11, respectively), but a significant effect between genotypes pointed to a difference in ion transport/homeostasis and the participation of G-coupled protein receptors (p = 8e-4), which was reinforced by results from regulatory and phenotypic impact factor analyses between genotypes. Conclusion: We propose these pathway differences between WUR genotypes are the result of the inability of the truncated GBP5 of the AA genotyped pigs to inhibit viral entry and replication as quickly as the intact GBP5 protein of the AB genotyped pigs

    Superconductivity in AuNiGe Ohmic contacts to a GaAs-based high mobility two-dimensional electron gas

    To cool a high mobility two-dimensional electron gas (2DEG) at a GaAs-AlGaAs heterojunction to milliKelvin temperatures, we have fabricated low resistance Ohmic contacts based on alloys of Au, Ni, and Ge. The Ohmic contacts have a typical contact resistance of R C ≈ 0.8 ω at 4.2 K, which drops to 0.2 ω below 0.9 K. Scanning electron microscope images establish that the contacts have the same inhomogeneous microstructure that has been observed in previous studies. Measurements of the contact resistance R C, the four-terminal resistance along the top of a single contact, and the vertical resistance RV all show that there is a superconductor in the Ohmic contact, which can be turned completely normal with a magnetic field of 0.15 T. We briefly discuss how this superconductivity may be affecting the electrical transport measurements of 2DEGs, especially how it may hinder the cooling of electrons in a 2DEG below 0.1 K

This sulfonyl transfer proceeds without erosion of either diastereo- or enantiocontrol, and is general across a range of N-sulfonyl substituents (SO2R; R = Ph, 4-MeC6H4, 4-MeOC6H4, 4-NO2C6H4, Me, Et) as well as C(3)-(aryl, heteroaryl, alkyl and alkenyl) and C(4)-(aryl and ester) substitution. Crossover reactions indicate an intermolecular step is operative within the formal migration process, although no crossover from C-sulfonyl products was observed.     High Resolution mid-Infrared Imaging of SN 1987A

    Using the Thermal-Region Camera and Spectrograph (T-ReCS) attached to the Gemini South 8m telescope, we have detected and resolved 10 micron emission at the position of the inner equatorial ring (ER) of supernova SN 1987A at day 6067. ``Hot spots'' similar to those found in the optical and near-IR are clearly present. The morphology of the 10 micron emission is globally similar to the morphology at other wavelengths from X-rays to radio. The observed mid-IR flux in the region of SN1987A is probably dominated by emission from dust in the ER. We have also detected the ER at 20 micron at a 4 sigma level. Assuming that thermal dust radiation is the origin of the mid-IR emission, we derive a dust temperature of 180^{+20}_{-10} K, and a dust mass of 1.- 8. 10^{-5} Mo for the ER. Our observations also show a weak detection of the central ejecta at 10 micron. We show that previous bolometric flux estimates (through day 2100) were not significantly contaminated by this newly discovered emission from the ER. If we assume that the energy input comes from radioactive decays only, our measurements together with the current theoretical models set a temperature of 90 leq T leq 100 K and a mass range of 10^{-4} - 2. 10^{-3} Mo for the dust in the ejecta. With such dust temperatures the estimated thermal emission is 9(+/-3) 10^{35} erg s^{-1} from the inner ring, and 1.5 (+/-0.5) 10^{36} erg s^{-1} from the ejecta.
