    Clastic Sediments in the Butler Cave – Sinking Creek System, Virginia, USA

    The Butler Cave - Sinking Creek System in Bath County, Virginia, consists of a master trunk passage along the axis of a syncline with a trellis arrangement of dip-oriented side caves. The western set of dip passages contain a sequence of massively and chaotically bedded sand and cobble sediments. Massive cobble fills also occur in the strike-oriented trunk passage. Cave passages on the eastern side of the syncline contain mostly sand and silt. The light fraction of the sediments consists predominantly of quartz and rock fragments. The sediments contain several percent heavy minerals composed of iron oxides, zircon, rutile, tourmaline and other minerals. Measurement of the visible and near infrared diffuse reflectance spectra shows at least three populations of sediments to be present: an iron-rich, clay-poor group; a clay-rich group; and a gypsiferous sediment. The iron minerals provided a paleomagnetic signal. Sediments from the trunk passage, deposited by recent underground drainage, contained a normal pole direction. Sediments from the dip passages were paleomagnetically reversed, showing the deposition dates from prior to 780,000 years. In one instance reversed polarity deposits overlie normal polarity, implying a minimum age of 990,000 years for the reversed sediments.   

    Adaptive and Behavioral Changes in Kynurenine 3-Monooxygenase Knockout Mice:Relevance to Psychotic Disorders

    BACKGROUND: Kynurenine 3-monooxygenase converts kynurenine to 3-hydroxykynurenine, and its inhibition shunts the kynurenine pathway-which is implicated as dysfunctional in various psychiatric disorders-toward enhanced synthesis of kynurenic acid, an antagonist of both α7 nicotinic acetylcholine and N-methyl-D-aspartate receptors. Possibly as a result of reduced kynurenine 3-monooxygenase activity, elevated central nervous system levels of kynurenic acid have been found in patients with psychotic disorders, including schizophrenia. METHODS: In the present study, we investigated adaptive-and possibly regulatory-changes in mice with a targeted deletion of Kmo (Kmo-/-) and characterized the kynurenine 3-monooxygenase-deficient mice using six behavioral assays relevant for the study of schizophrenia. RESULTS: Genome-wide differential gene expression analyses in the cerebral cortex and cerebellum of these mice identified a network of schizophrenia- and psychosis-related genes, with more pronounced alterations in cerebellar tissue. Kynurenic acid levels were also increased in these brain regions in Kmo-/- mice, with significantly higher levels in the cerebellum than in the cerebrum. Kmo-/- mice exhibited impairments in contextual memory and spent less time than did controls interacting with an unfamiliar mouse in a social interaction paradigm. The mutant animals displayed increased anxiety-like behavior in the elevated plus maze and in a light/dark box. After a D-amphetamine challenge (5 mg/kg, intraperitoneal), Kmo-/- mice showed potentiated horizontal activity in the open field paradigm. CONCLUSIONS: Taken together, these results demonstrate that the elimination of Kmo in mice is associated with multiple gene and functional alterations that appear to duplicate aspects of the psychopathology of several neuropsychiatric disorders

    Maternal Behavior is Impaired in Female Mice Lacking Type 3 Adenylyl Cyclase

    Although chemosensory signals generated by mouse pups may trigger maternal behavior of females, the mechanism for detection of these signals has not been fully defined. As some odorant receptors are coupled to the type 3 adenylyl cyclase (AC3), we evaluated the role of AC3 for maternal behavior using AC3−/− female mice. Here, we report that maternal behavior is impaired in virgin and postpartum AC3−/− mice. Female AC3−/− mice failed the pup retrieval assay, did not construct well-defined nests, and did not exhibit maternal aggression. Furthermore, AC3−/− females could not detect odorants or pup urine in the odorant habituation test and were unable to detect pups by chemoreception. In contrast to wild-type mice, AC activity in main olfactory epithelium (MOE) preparations from AC3−/− female mice was not stimulated by odorants or pheromones. Moreover, odorants and pheromones did not evoke electro-olfactogram (EOG) responses in the MOE of AC3−/− female mice. We hypothesize that the detection of chemical signals that trigger maternal behavior in female mice depends upon AC3 in the MOE

    Schizophrenia-associated somatic copy-number variants from 12,834 cases reveal recurrent NRXN1 and ABCB11 disruptions

    While germline copy-number variants (CNVs) contribute to schizophrenia (SCZ) risk, the contribution of somatic CNVs (sCNVs)—present in some but not all cells—remains unknown. We identified sCNVs using blood-derived genotype arrays from 12,834 SCZ cases and 11,648 controls, filtering sCNVs at loci recurrently mutated in clonal blood disorders. Likely early-developmental sCNVs were more common in cases (0.91%) than controls (0.51%, p = 2.68e−4), with recurrent somatic deletions of exons 1–5 of the NRXN1 gene in five SCZ cases. Hi-C maps revealed ectopic, allele-specific loops forming between a potential cryptic promoter and non-coding cis-regulatory elements upon 5′ deletions in NRXN1. We also observed recurrent intragenic deletions of ABCB11, encoding a transporter implicated in anti-psychotic response, in five treatment-resistant SCZ cases and showed that ABCB11 is specifically enriched in neurons forming mesocortical and mesolimbic dopaminergic projections. Our results indicate potential roles of sCNVs in SCZ risk

    Attack of the 50-foot social justice warrior: the discursive construction of SJW memes as the monstrous feminine

    This essay considers the origin and meaning of “social justice warrior” (SJW) memes. Despite each term within the phrase suggesting potentially positive connotations, we argue that as deployed within “alt-right” communities, it implies a kind of monstrous feminine: a woman who is unwieldy and out of control. We catalogue and analyze this meme using a visual discourse analysis of texts gathered through Google Images and Reddit. Our findings suggest that the SJW meme is deployed to emphasize opponents as having non-normative, problematic bodies, different brains (ones ruled by emotion rather than logic), and monstrous characteristics. We argue that such discourse is potentially dangerous, but that feminists may have the tools to recreate the SJW as an image of power
