    Case-control study to detect protective factors on pig farms with low Salmonella prevalence

    The prevalence of Salmonella in UK pigs is amongst the highest in Europe, highlighting the risk to public health and the need to investigate on-farm controls. The objective of this study was to identify factors currently in operation on pig farms that had maintained a low Salmonella seroprevalence. For this purpose a case-control study was designed and pig farms with a low (\u3c10%) seroprevalence were compared against two randomly selected control farms, sharing the same geographical region and production type. A total of 11,452 samples, including pooled and individual floor faeces and environmental samples from pigs and their vicinity were tested and prevalence examined. In addition, detailed questionnaires were completed during the farm visits to collect descriptive data for risk factor analysis. It was shown that control farms had significantly higher prevalence compared to the case farms (19.4% and 4.3% for pooled and 6.7% and 0.1% for individual samples, respectively). The two risk factor analyses identified multiple variables associated with Salmonella prevalence including variables related to feed, effectiveness of cleaning and disinfection, biosecurity and batch production

    Consistency of Age Reporting on Death Certificates and Social Security Administration Records Among Elderly African-American Decedents

    This paper investigates the quality of age reporting in vital statistics and Social Security/Medicare data among elderly African-Americans. The authors examine whether the death certificate or Social Security age is more likely to reflect accurately the decedents\u27 true age at death by matching their sample to the US Censuses of 1900, 1910 and 1920, and identify factors associated with consistency of age reporting on death certificates and social security records. The results reveal significant discrepancies in age at death data. Birth record availability and literacy were identified as key predictors of age agreement. The match to an early-life census record showed greater agreement with Social Security age than with death certificate age at death. The results have implications for the collection of age information in surveys of elderly African-Americans

    Myosin-X functions in polarized epithelial cells

    Myosin-X, an unconventional myosin that has been studied primarily in fibroblast-like cells, has been shown to have important functions in polarized epithelial cell junction formation, regulation of paracellular permeability, and epithelial morphogenesis.Myosin-X (Myo10) is an unconventional myosin that localizes to the tips of filopodia and has critical functions in filopodia. Although Myo10 has been studied primarily in nonpolarized, fibroblast-like cells, Myo10 is expressed in vivo in many epithelia-rich tissues, such as kidney. In this study, we investigate the localization and functions of Myo10 in polarized epithelial cells, using Madin-Darby canine kidney II cells as a model system. Calcium-switch experiments demonstrate that, during junction assembly, green fluorescent protein–Myo10 localizes to lateral membrane cell–cell contacts and to filopodia-like structures imaged by total internal reflection fluorescence on the basal surface. Knockdown of Myo10 leads to delayed recruitment of E-cadherin and ZO-1 to junctions, as well as a delay in tight junction barrier formation, as indicated by a delay in the development of peak transepithelial electrical resistance (TER). Although Myo10 knockdown cells eventually mature into monolayers with normal TER, these monolayers do exhibit increased paracellular permeability to fluorescent dextrans. Importantly, knockdown of Myo10 leads to mitotic spindle misorientation, and in three-dimensional culture, Myo10 knockdown cysts exhibit defects in lumen formation. Together these results reveal that Myo10 functions in polarized epithelial cells in junction formation, regulation of paracellular permeability, and epithelial morphogenesis

    A variational approach to waveform design for synthetic aperture imaging

    Abstract. We derive an optimal transmit waveform for filtered backprojectionbased synthetic-aperture imaging. The waveform is optimal in terms of minimising the mean square error (MSE) in the resulting image. Our optimization is performed in two steps: First, we consider the minimum-mean-square-error (MMSE) for an arbitrary but fixed waveform, and derive the corresponding filter for the filtered backprojection reconstruction. Second, the MMSE is further reduced by finding an optimal transmit waveform. The transmit waveform is derived for stochastic models of the scattering objects of interest (targets), other scattering objects (clutter), and additive noise. We express the waveform in terms of spatial spectra for the random fields associated with target and clutter, and the spectrum for the noise process. This approach results in a constraint that involves only the amplitude of the Fourier transform of the transmit waveform. Therefore, considerable flexibility is left for incorporating additional requirements, such as minimal variation of transmit amplitude and phase-coding

    DPP6 regulation of dendritic morphogenesis impacts hippocampal synaptic development

    Dipeptidyl-peptidase 6 is an auxiliary subunit of Kv4-mediated A-type K+ channels that, in addition to enhancing channel surface expression, potently accelerates their kinetics. The dipeptidyl-peptidase 6 gene has been associated with a number of human central nervous system disorders including autism spectrum disorders and schizophrenia. Here we employ knockdown and genetic deletion of dipeptidyl-peptidase 6 to reveal its importance for the formation and stability of dendritic filopodia during early neuronal development. We find that the hippocampal neurons lacking dipeptidyl-peptidase 6 show a sparser dendritic branching pattern along with fewer spines throughout development and into adulthood. In electrophysiological and imaging experiments, we show that these deficits lead to fewer functional synapses and occur independently of the potassium channel subunit Kv4.2. We report that dipeptidyl-peptidase 6 interacts with a filopodia-associated myosin as well as with fibronectin in the extracellular matrix. dipeptidyl-peptidase 6 therefore has an unexpected but important role in cell adhesion and motility, impacting the hippocampal synaptic development and function

    Myosin X (Myo10) is an unconventional myosin with two known isoforms: full-length (FL)-Myo10 that has motor activity, and a recently identified brain-expressed isoform, headless (Hdl)-Myo10, which lacks most of the motor domain. FL-Myo10 is involved in the regulation of filopodia formation in non-neuronal cells; however, the biological function of Hdl-Myo10 remains largely unknown. Here, we show that FL- and Hdl-Myo10 have important, but distinct, roles in the development of dendritic spines and synapses in hippocampal neurons. FL-Myo10 induces formation of dendritic filopodia and modulates filopodia dynamics by trafficking the actin-binding protein vasodilator-stimulated phosphoprotein (VASP) to the tips of filopodia. By contrast, Hdl-Myo10 acts on dendritic spines to enhance spine and synaptic density as well as spine head expansion by increasing the retention of VASP in spines. Thus, this study demonstrates a novel biological function for Hdl-Myo10 and an important new role for both Myo10 isoforms in the development of dendritic spines and synapses

    A Practical Theory of Language-integrated Query

    Language-integrated query is receiving renewed attention, in part because of its support through Microsoft’s LINQ framework. We present a practical theory of language-integrated query based on quotation and normalisation of quoted terms. Our technique supports join queries, abstraction over values and predicates, composition of queries, dynamic generation of queries, and queries with nested intermediate data. Higher-order features prove useful even for constructing first-order queries. We prove a theorem characterising when a host query is guaranteed to generate a single SQL query. We present experimental results confirming our technique works, even in situations where Microsoft’s LINQ framework either fails to produce an SQL query or, in one case, produces an avalanche of SQL queries

    A literature review as an aid to identify strategies for mitigating ostreid herpesvirus 1 in Crassostrea gigas hatchery and nursery systems

    An understanding of husbandry strategies and any associated risk factors is important for designing management control measures that can reduce mortality in Pacific oysters, Crassostrea gigas, caused by ostreid herpesvirus 1 (OsHV‐1). The type of culture facility can be considered in relation to the potential pathways that could lead to the entry of a pathogen and its survival. In addition, the animal host (e.g. age, physiological state, selective breeding programmes), husbandry procedures (e.g. stocking density), the pathogen itself (e.g. pathogenicity, virulence) and environmental effects (e.g. temperature) represent other relevant interconnected factors. However, all these factors provide valuable background information for outlining the mitigation strategies needed by the industry, as well as in the context of surveillance and biosecurity programmes. These control mechanisms for hatchery or nursery areas are related to movement restrictions, water treatment, virus inactivation, the production calendar and practical farm management decisions. This comprehensive literature review compiles information related to such approaches and also includes the different existing guidelines suggested for control of OsHV‐1. Therefore, the review represents a solid foundation for a more critical appraisal currently being developed to support recommendations for disease management strategies.info:eu-repo/semantics/publishedVersio

    Preventive medical care in remote Aboriginal communities in the Northern Territory: a follow-up study of the impact of clinical guidelines, computerised recall and reminder systems, and audit and feedback

    Background Interventions to improve delivery of preventive medical services have been shown to be effective in North America and the UK. However, there are few studies of the extent to which the impact of such interventions has been sustained, or of the impact of such interventions in disadvantaged populations or remote settings. This paper describes the trends in delivery of preventive medical services following a multifaceted intervention in remote community health centres in the Northern Territory of Australia. Methods The intervention comprised the development and dissemination of best practice guidelines supported by an electronic client register, recall and reminder systems and associated staff training, and audit and feedback. Clinical records in seven community health centres were audited at regular intervals against best practice guidelines over a period of three years, with feedback of audit findings to health centre staff and management. Results Levels of service delivery varied between services and between communities. There was an initial improvement in service levels for most services following the intervention, but improvements were in general not fully sustained over the three year period. Conclusions Improvements in service delivery are consistent with the international experience, although baseline and follow-up levels are in many cases higher than reported for comparable studies in North America and the UK. Sustainability of improvements may be achieved by institutionalisation of relevant work practices and enhanced health centre capacity
