    The influence of new 1,2,3-triazolo-1,4-benzodiazepine derivatives on the muscle tone of rodents

    Anxiety disorders represent one of the most prevalent categories of psychiatric illnesses, affecting individuals regardless of gender, age, or social standing. They result in substantial personal and societal costs. The pursuit of novel pharmacological approaches for treating these disorders is driven by the increasing medical necessity to enhance the effectiveness and safety profiles of anxiolytic medications. Due to the fact that benzodiazepines and their derivatives have anti-anxiety, hypnotic-sedative, antidepressant, anticonvulsant, and muscle relaxant properties, they occupy a leading place in the treatment of anxiety disorders. An essential aspect of investigating the pharmacological activity of new triazolobenziazepine derivatives is assessing their impact on rodent muscle tone and coordination of movements. The aim of the work is to find out the influence of new 1,2,3-triazolo-1,4-benzodiazepine derivatives on the muscle tone of rodents in the “vertical grid” test and coordination of movements using the rotarod test. Materials and methods. The objects of the study were 5 new derivatives of 1,2,3-triazolo-1,4-benzodiazepines. Before conducting in vivo experiments, these derivatives were mixed with lactose at a ratio of 1:1000. The “vertical grid” test and rotarod test (rotating rod test) were used to reproduce the model of motor behavior of rodents. Results. The presence of a tendency to the manifestation of a myorelaxant effect in the “vertical grid” test was established. The indicator of the total duration of detention at the facilities was similar and did not differ significantly in the control and experimental groups at doses of 0.50 mg/kg and 0.75 mg/kg. Derivatives MA-252, MA-253 and MA-254 at a dose of 1 mg/kg reduced the total duration of retention on the vertical grid, which indicates their mild muscle relaxant effect. In the rotating rod test, MA-253 derivative at a dose of 1 mg/kg increased the retention time on the rotarod, which demonstrates greater physical endurance of the animals of these experimental groups. Conclusions. The study’s findings indicated that the 1,2,3-triazolo-1,4-benzodiazepine derivatives did not exhibit an adverse impact on movement coordination. Some of these derivatives demonstrated a mild muscle relaxant effect. These results support the need for further research into their influence on spontaneous motor activity. Additionally, there’s a necessity to determine the dosage regimen, establish an effective dose, and adapt it for human use

    Substituted 1H-pyrazolo[3,4-b]pyridine-4- and 1H-pyrazolo[3,4-b]pyridine-6-carboxamides have been synthetized through a Doebner–Ugi multicomponent reaction sequence in a convergent and versatile manner using diversity generation strategies: combination of two multicomponent reactions and conditions-based divergence strategy. The target products contain as pharmacophores pyrazolopyridine and peptidomimetic moieties with four points of diversity introduced from readily available starting materials including scaffold diversity. A small focused compound library of 23 Ugi products was created and screened for antibacterial activity

    Effective microwave-assisted approach to 1,2,3-triazolobenzodiazepinones via tandem Ugi reaction/catalyst-free intramolecular azide–alkyne cycloaddition

    A novel catalyst-free synthetic approach to 1,2,3-triazolobenzodiazepinones has been developed and optimized. The Ugi reaction of 2-azidobenzaldehyde, various amines, isocyanides, and acids followed by microwave-assisted intramolecular azide–alkyne cycloaddition (IAAC) gave a series of target heterocyclic compounds in moderate to excellent yields. Surprisingly, the normally required ruthenium-based catalysts were found to not affect the IAAC, only making isolation of the target compounds harder while the microwave-assisted catalyst-free conditions were effective for both terminal and non-terminal alkyne

    Ensuring power balance in a hybrid power system with a backup generator

    The combination of several non-guaranteed random energy sources (RES), conventional sources, and non-constant consumer loads in a local system leads to stochastic power imbalances. This study objective consists in determining the possibilities of ensuring the power balance in a hybrid power generation system with a standby generator and a search for the methods of calculating the optimal parameters to achieve energy balance. This objective is achieved by simulating the processes inherent in wind and solar power engineering and the regimes of energy consumption through a combination of random functions with a standard probability distribution. Aggregated data on weather factors for several years in a region with a high renewable energy potential which can be used to describe the behavior of wind and solar energy over time were used as experimental data. The use of multiple simulations of random processes with calculated parameters has made it possible to draw conclusions about the presence of certain ratios of power and the generator control modes. These ratios can determine minimum energy and consumption losses, reduce the likelihood of energy imbalance, more efficiently use the reserved power. Specific features of the stochastic nature of RES related to the presence of trends and random fluctuations at short hourly intervals were additionally taken into account. Possibilities of varying the conditions of and switching on and off of the standby generator were provided. The existence of some ranges was established for the installed power of the generator outside which its use becomes inefficient. The proposed approach makes it possible to find the probability of various system states, assess the reliability of energy supply, and minimize unproductive losses

    Стаття присвячена систематизації факторів впливу на вибір гірничовидобувного обладнання при розробці техногенних родовищ. Подано статистичні данні про площі і обсяги потенційних техногенних утворень, придатних для залучення у розробку у якості техногенних родовищ. Наведено найбільш поширене гірничовидобувне обладнання циклічної та поточної дії, придатне для розробки корисних компонентів техногенних родовищ. Запропоновано виділити чотири групи факторів, які впливають на вибір гірничотранспортного обладнання при розробці техногенних родовищ. Розроблено рекомендації щодо доцільності використання певних видів гірничовидобувного устаткування в залежності від походження матеріалів, способу формування, геометрії та фізико-механічних властивостей порід техногенного родовища.Статья посвящена систематизации факторов влияния на выбор горнодобывающего оборудования при разработке техногенных месторождений. Приведены статистические данные о площадях и объемах потенциальных техногенных образований, пригодных для вовлечения в разработку в качестве техногенных месторождений. Выполнен анализ наиболее распростра-ненного горнодобывающего оборудования циклического и непрерывного действия, которое применяется для разработки полезных компонентов техногенных месторождений. Предложено выделить четыре группы факторов, влияющих на выбор горнотранспортного оборудования при разработке техногенных месторождений.
