    Organic Cation Transporter 3 (OCT3) Is Localized to Intracellular and Surface Membranes in Select Glial and Neuronal Cells Within the Basolateral Amygdaloid Complex of Both Rats and Mice

    Organic cation transporter 3 (OCT3) is a high-capacity, low-affinity transporter that mediates corticosterone-sensitive uptake of monoamines including norepinephrine, epinephrine, dopamine, histamine and serotonin. OCT3 is expressed widely throughout the amygdaloid complex and other brain regions where monoamines are key regulators of emotional behaviors affected by stress. However, assessing the contribution of OCT3 to the regulation of monoaminergic neurotransmission and monoamine-dependent regulation of behavior requires fundamental information about the subcellular distribution of OCT3 expression. We used immunofluorescence and immuno-electron microscopy to examine the cellular and subcellular distribution of the transporter in the basolateral amygdaloid complex of the rat and mouse brain. OCT3-immunoreactivity was observed in both glial and neuronal perikarya in both rat and mouse amygdala. Electron microscopic immunolabeling revealed plasma membrane-associated OCT3 immunoreactivity on axonal, dendritic, and astrocytic processes adjacent to a variety of synapses, as well as on neuronal somata. In addition to plasma membrane sites, OCT3 immunolabeling was also observed associated with neuronal and glial endomembranes, including Golgi, mitochondrial and nuclear membranes. Particularly prominent labeling of the outer nuclear membrane was observed in neuronal, astrocytic, microglial and endothelial perikarya. The localization of OCT3 to neuronal and glial plasma membranes adjacent to synaptic sites is consistent with an important role for this transporter in regulating the amplitude, duration, and physical spread of released monoamines, while its localization to mitochondrial and outer nuclear membranes suggests previously undescribed roles for the transporter in the intracellular disposition of monoamines

    THE PROBIOTIC POTENTIAL OF NON-TOXIGENIC BACTEROIDES FRAGILIS AND THE ROLE OF IL-22 IN COLITIS AND TUMORIGENESIS

    Polysaccharide A (PSA), a highly immunogenic capsular component of non-toxigenic Bacteroides fragilis NCTC 9343 (NTBF), has been shown to promote host mucosal immune system development and to suppress inflammation in the gut. In stark contrast, enterotoxigenic Bacteroides fragilis (ETBF) induces IL-17 and is highly associated with inflammatory bowel disease and colorectal cancer, rapidly inducing acute colitis and tumorigenesis in murine hosts. The interactions of these distinct B. fragilis molecular subtypes were studied to determine the impact of NTBF on ETBF disease. In specific pathogen free (SPF) C57BL/6 WT and MinApc716+/- mice, we show that sequential treatment (NTBF then ETBF three days later) blunted colitis and decreased tumor development. Mice receiving simultaneous NTBF and ETBF treatment exhibited severe colitis and pronounced tumor formation. Abrogated disease severity in sequentially treated mice was attributed to NTBF strain dominance and decreased IL-17, but ETBF colonization prior to or simultaneous with NTBF mitigates the anti-inflammatory effect of NTBF. Surprisingly, NTBF-mediated protection was independent of polysaccharide A (PSA), as sequentially infected mice receiving ΔPSA NTBF exhibited similar levels of protection. Further, SPF WT and Min mice mono-associated with PSA-competent or PSA-deficient NTBF exhibit similar T cell-derived regulation, IL-17, and IFNγ responses. Daily NTBF probiotic treatment of mice stably colonized with ETBF failed to disrupt both ETBF strain dominance and niche occupation. Our findings demonstrate that NTBF may offer protection against ETBF disease but only under certain conditions. Disease outcomes are independent of PSA

    Enhanced magnetic and thermoelectric properties in epitaxial polycrystalline SrRuO3 thin film

    Transition metal oxide thin films show versatile electrical, magnetic, and thermal properties which can be tailored by deliberately introducing macroscopic grain boundaries via polycrystalline solids. In this study, we focus on the modification of the magnetic and thermal transport properties by fabricating single- and polycrystalline epitaxial SrRuO3 thin films using pulsed laser epitaxy. Using epitaxial stabilization technique with atomically flat polycrystalline SrTiO3 substrate, epitaxial polycrystalline SrRuO3 thin film with crystalline quality of each grain comparable to that of single-crystalline counterpart is realized. In particular, alleviated compressive strain near the grain boundaries due to coalescence is evidenced structurally, which induced enhancement of ferromagnetic ordering of the polycrystalline epitaxial thin film. The structural variations associated with the grain boundaries further reduce the thermal conductivity without deteriorating the electronic transport, and lead to enhanced thermoelectric efficiency in the epitaxial polycrystalline thin films, compared with their single-crystalline counterpart.Comment: 24 pages, 5 figure

    A Price Worth Paying: The Case for Controlling Marine Emissions in the Pearl River Delta

    The Pearl River Delta (PRD) is a region with a single airshed, but different administrative and legal practices for controlling air quality. Under the Regional Cooperation Plan on Building a Quality Living Area (QLA Plan) released in June 2012 the Governments of Hong Kong, Guangdong and Macau have outlined a strategy to collaborate in reducing emissions from vessels throughout the PRD. This report provides evidence designed to assist policymakers in the region with this objective. It focuses on regulating toxic exhaust emissions from ocean-going vessels (OGVs) -- the most significant contributors of marine emissions. The findings show that marine sources of sulphur dioxide (SO2) emissions currently account for 519 premature deaths per annum in the PRD. These deaths could be reduced by 91% should an Emission Control Area (ECA) mandating the use of fuels with lower sulphur content be introduced. The report also demonstrates that three less comprehensive control measures would also reduce OGV emissions and associated public health impacts by 41-62%. Policymakers are encouraged to introduce these measures as stepping-stones on the way to establishment of an ECA for the PRD

    A randomized controlled trial on the comparative effectiveness of mindfulness-based cognitive therapy and health qigong-based cognitive therapy among Chinese people with depression and anxiety disorders

    Background: The goal of this study was to investigate treatment outcome and related intervention processes of mindfulness-based cognitive therapy versus health qigong-based cognitive therapy versus waitlist control among individuals with mood disorders. Methods: A total of 187 individuals with mood disorders were randomized and allocated into mindfulness-based cognitive therapy, health qigong-based cognitive therapy, or waitlist control groups. All participants were assessed at three time points with regard to depressive and anxiety symptoms, physical and mental health status, perceived stress, sleep quality, and self-efficacy. Linear mixed models analysis was used to test the individual growth model by studying the longitudinal data. Results: Mindfulness-based cognitive therapy and health qigong-based cognitive therapy both produced greater improvements on all outcome measures as compared with waitlist control. Relatively, more reductions of mood symptoms were observed in the health qigong-based cognitive therapy group as compared with the mindfulness-based cognitive therapy group. Health qigong-based cognitive therapy is more conducive to physical health status whereas mindfulness-based cognitive therapy has more favorable mental health outcomes. Individual growth curve models indicated that alterations in perceived stress was the common predictor of mood changes in both intervention groups. Conclusions: The predominant emphasis on physical health in health qigong-based cognitive therapy makes it more acceptable and effective than mindfulness-based cognitive therapy as applied in Chinese individuals with mood disorders.     Development and pilot evaluation of a personalized decision support intervention for low risk prostate cancer patients.

    ObjectivesDevelopment and pilot evaluation of a personalized decision support intervention to help men with early-stage prostate cancer choose among active surveillance, surgery, and radiation.MethodsWe developed a decision aid featuring long-term survival and side effects data, based on focus group input and stakeholder endorsement. We trained premedical students to administer the intervention to newly diagnosed men with low-risk prostate cancer seen at the University of California, San Francisco. Before the intervention, and after the consultation with a urologist, we administered the Decision Quality Instrument for Prostate Cancer (DQI-PC). We hypothesized increases in two knowledge items from the DQI-PC: How many men diagnosed with early-stage prostate cancer will eventually die of prostate cancer? How much would waiting 3 months to make a treatment decision affect chances of survival? Correct answers were: "Most will die of something else" and "A little or not at all."ResultsThe development phase involved 6 patients, 1 family member, 2 physicians, and 5 other health care providers. In our pilot test, 57 men consented, and 44 received the decision support intervention and completed knowledge surveys at both timepoints. Regarding the two knowledge items of interest, before the intervention, 35/56 (63%) answered both correctly, compared to 36/44 (82%) after the medical consultation (P = .04 by chi-square test).ConclusionsThe intervention was associated with increased patient knowledge. Data from this pilot have guided the development of a larger scale randomized clinical trial to improve decision quality in men with prostate cancer being treated in community settings

    Dermoid cysts: Epidemiology and diagnostic approach based on clinical experiences

    Background Dermoid cysts are congenital tumors that are benign. Dermoid cysts with intracranial extension can cause serious neurological complications. It is important, therefore, to determine whether a patient has a dermoid cyst when their chief concern at a doctor’s visit is a mass in the head or neck area. In this study, we present a literature review of dermoid cysts and an analysis of the authors’ experiences, with the goal of providing guidance useful for the diagnosis and treatment of dermoid cysts. Methods This study retrospectively analyzed the medical records of 62 patients who visited the two medical clinics with which the authors are affiliated. The patients were enrolled between October 2003 and January 2017. Results Of the 62 patients analyzed in this study, 32 were 0 to 5 years of age (52%) and 23 were 17 years of age or older (37%). Forty-seven patients underwent 1 or more imaging study during the process of diagnosis. Thirty-two patients were suspected to have a dermoid cyst. Forty-nine patients were analyzed to determine the depth of the cyst. Bone was seen in 43 patients through imaging tests or during actual surgery, and nine of the 43 had bony problems (21%). Conclusions This study found that dermoid cysts were present in many adults, and that a high rate of deep lesions was observed, as well as many cases in which even the bone was affected. These results suggest, therefore, that dermoid cysts should be considered, and medical professionals should actively conduct imaging studies

    Edge Breakdown Suppression of 10 Gbps Avalanche Photodiode

    We have demonstrated a high-speed avalanche photodiode (APD) for a 10 Gbps optical communication system. To achieve a high gain-bandwidth product and reliable operation, the reduction of the multiplication layer thickness and an optimum design of the internal electric eld distribution are essential. One- and two-dimensional analysis were done for this purpose. The suppression of edge breakdown can be achieved by precise control of the thickness of the multiplication layer and charge densities of the eld control layer. Furthermore we suggest a junction curvature shape having negative curvature of the equi-potential line at the device edge. This new design successfully suppressed edge breakdown. The fabricated APD shows high current gain without premature edge breakdown, and a gain-bandwidth of above 80 GHz has been obtained
