    This work tries to reflect on the religious experience, or of the sacred, in the current post modernity based of the individual and collective beliefs of the experiences and common discourses in everyday life. It is approached under an individual perspective since in post modernity is common for each person to affirm their faith in god, which requires the necessary tolerance in a multicultural society that demands respect and acceptance of those who see things from their own perspective and different from most.El presente ensayo trata de reflexionar sobre la experiencia religiosa, o de lo sagrado, que se vive en la posmodernidad y parte de las creencias que se manifiestan en lo individual y colectivo de las vivencias y discursos comunes en la cotidianidad. Se habla desde lo individual puesto que en la posmodernidad es frecuente que cada quien afirme su propia fe en Dios, frente a lo cual hay que guardar la tolerancia necesaria en una sociedad multicultural, que pide el respeto y la aceptación por quienes ven las cosas de modo diverso al propio y de las mayorías

    Sustainability and quality of work at risk: knowledge work intensification

    Desde que en 1991 se realizan las Encuestas Europeas de Condiciones de Trabajo existe consenso sobre la mejora en varias dimensiones de las condiciones/calidad del trabajo (e.g. participación, cualificación, seguridad...). Sin embargo, en el mismo periodo, estas tendencias se han acompañado de una intensificación del trabajo, una dinámica que se considera un riesgo para la sostenibilidad y la calidad del trabajo y por ende, para el bienestar y la salud de los trabajadores. Aunque la literatura internacional apunta a los cambios en la organización del trabajo para explicar esta intensificación del trabajo, los análisis empíricos y estadísticamente representativos son escasos, particularmente en España. En este artículo analizamos el caso español en el periodo 1999-2012, explotando los microdatos oficiales de las últimas cuatro oleadas de las Encuestas Nacionales de Condiciones de Trabajo (ENCTs) que son representativas de la población trabajadora española. Distinguimos dos periodos para considerar los cambios metodológicos entre 2003 y 2006. Tras diferenciar una intensidad de trabajo más mental (cognitiva y emocional) y otra más física, 1) proponemos una operacionalización de la intensidad de trabajo; 2) analizamos su evolución y diferencias en base a ocupación, género y edad; 3) profundizamos en los factores organizativos que condicionan la intensidad del trabajo y 4) discutimos en qué medida estos factores pueden explicar las diferencias de intensidad de las distintas ocupaciones, géneros y grupos de edad.Since the first European Working Conditions Survey back in 1991 trends show that several working conditions/quality of work dimensions are improving (e.g. participation, skills, safety...). However, a worrying work intensification process has emerged during the same period. Work intensity is considered an important risk for the sustainability, quality of work and worker’s health and wellbeing. International literature suggests that changes in work organization are a major driver, but representative empirical analysis are still scarce, particularly in Spain. This article analyzes the Spanish case in the 1999-2012 period. We use official microdata from the National Working Conditions Surveys (representative of the working population in Spain). We consider two distinct periods to account for the methodological changes between 2003 and 2006.     The impact of non-additive genetic associations on age-related complex diseases

    Genome-wide association studies (GWAS) are not fully comprehensive, as current strategies typically test only the additive model, exclude the X chromosome, and use only one reference panel for genotype imputation. We implement an extensive GWAS strategy, GUIDANCE, which improves genotype imputation by using multiple reference panels and includes the analysis of the X chromosome and non-additive models to test for association. We apply this methodology to 62,281 subjects across 22 age-related diseases and identify 94 genome-wide associated loci, including 26 previously unreported. Moreover, we observe that 27.7% of the 94 loci are missed if we use standard imputation strategies with a single reference panel, such as HRC, and only test the additive model. Among the new findings, we identify three novel low-frequency recessive variants with odds ratios larger than 4, which need at least a three-fold larger sample size to be detected under the additive model. This study highlights the benefits of applying innovative strategies to better uncover the genetic architecture of complex diseases. Most genome-wide association studies assume an additive model, exclude the X chromosome, and use one reference panel. Here, the authors implement a strategy including non-additive models and find that the number of loci for age-related traits increases as compared to the additive model alone.Peer reviewe

    Single nucleotide polymorphism discovery in rainbow trout by deep sequencing of a reduced representation library

    <p>Abstract</p> <p>Background</p> <p>To enhance capabilities for genomic analyses in rainbow trout, such as genomic selection, a large suite of polymorphic markers that are amenable to high-throughput genotyping protocols must be identified. Expressed Sequence Tags (ESTs) have been used for single nucleotide polymorphism (SNP) discovery in salmonids. In those strategies, the salmonid semi-tetraploid genomes often led to assemblies of paralogous sequences and therefore resulted in a high rate of false positive SNP identification. Sequencing genomic DNA using primers identified from ESTs proved to be an effective but time consuming methodology of SNP identification in rainbow trout, therefore not suitable for high throughput SNP discovery. In this study, we employed a high-throughput strategy that used pyrosequencing technology to generate data from a reduced representation library constructed with genomic DNA pooled from 96 unrelated rainbow trout that represent the National Center for Cool and Cold Water Aquaculture (NCCCWA) broodstock population.</p> <p>Results</p> <p>The reduced representation library consisted of 440 bp fragments resulting from complete digestion with the restriction enzyme <it>Hae</it>III; sequencing produced 2,000,000 reads providing an average 6 fold coverage of the estimated 150,000 unique genomic restriction fragments (300,000 fragment ends). Three independent data analyses identified 22,022 to 47,128 putative SNPs on 13,140 to 24,627 independent contigs. A set of 384 putative SNPs, randomly selected from the sets produced by the three analyses were genotyped on individual fish to determine the validation rate of putative SNPs among analyses, distinguish apparent SNPs that actually represent paralogous loci in the tetraploid genome, examine Mendelian segregation, and place the validated SNPs on the rainbow trout linkage map. Approximately 48% (183) of the putative SNPs were validated; 167 markers were successfully incorporated into the rainbow trout linkage map. In addition, 2% of the sequences from the validated markers were associated with rainbow trout transcripts.</p> <p>Conclusion</p> <p>The use of reduced representation libraries and pyrosequencing technology proved to be an effective strategy for the discovery of a high number of putative SNPs in rainbow trout; however, modifications to the technique to decrease the false discovery rate resulting from the evolutionary recent genome duplication would be desirable.</p

    Generation of a reference transcriptome for evaluating rainbow trout responses to various stressors

    <p>Abstract</p> <p>Background</p> <p>Fish under intensive culture conditions are exposed to a variety of acute and chronic stressors, including high rearing densities, sub-optimal water quality, and severe thermal fluctuations. Such stressors are inherent in aquaculture production and can induce physiological responses with adverse effects on traits important to producers and consumers, including those associated with growth, nutrition, reproduction, immune response, and fillet quality. Understanding and monitoring the biological mechanisms underlying stress responses will facilitate alleviating their negative effects through selective breeding and changes in management practices, resulting in improved animal welfare and production efficiency.</p> <p>Results</p> <p>Physiological responses to five treatments associated with stress were characterized by measuring plasma lysozyme activity, glucose, lactate, chloride, and cortisol concentrations, in addition to stress-associated transcripts by quantitative PCR. Results indicate that the fish had significant stressor-specific changes in their physiological conditions. Sequencing of a pooled normalized transcriptome library created from gill, brain, liver, spleen, kidney and muscle RNA of control and stressed fish produced 3,160,306 expressed sequence tags which were assembled and annotated. SNP discovery resulted in identification of ~58,000 putative single nucleotide polymorphisms including 24,479 which were predicted to fall within exons. Of these, 4907 were predicted to occupy the first position of a codon and 4110 the second, increasing the probability to impact amino acid sequence variation and potentially gene function.</p> <p>Conclusion</p> <p>We have generated and characterized a reference transcriptome for rainbow trout that represents multiple tissues responding to multiple stressors common to aquaculture production environments. This resource compliments existing public transcriptome data and will facilitate approaches aiming to evaluate gene expression associated with stress in this species.</p
