    Lipid metabolism in development and progression of hepatocellular carcinoma

    Metabolic reprogramming is critically involved in the development and progression of cancer. In particular, lipid metabolism has been investigated as a source of energy, micro-environmental adaptation, and cell signalling in neoplastic cells. However, the specific role of lipid metabolism dysregulation in hepatocellular carcinoma (HCC) has not been widely described yet. Alterations in fatty acid synthesis, β-oxidation, and cellular lipidic composition contribute to initiation and progression of HCC. The aim of this review is to elucidate the mechanisms by which lipid metabolism is involved in hepatocarcinogenesis and tumour adaptation to different conditions, focusing on the transcriptional aberrations with new insights in lipidomics and lipid zonation. This will help detect new putative therapeutic approaches in the second most frequent cause of cancer-related death

    Framing higher education quality from a business perspective: setting the conditions for enhanced value co-creation

    Higher education institutions (HEIs) are complex entities interacting with a variety of stakeholders. Scholars have widely discussed the approaches and models of value co-creation in the higher education context. However, the attention has been primarily focussed on a single category of stakeholders, namely students. This article aims at eliciting the value expectations of private and public sector organizations, emphasizing their potential role in value co-creation dynamics. A case study approach was undertaken in order to shed light on these issues. It involved a medium-sized university established in Northern Italy. Business stakeholders were especially interested in the university’s ability to provide students with practical and soft skills. Moreover, they appreciated the interdisciplinary nature of educational curricula. Sporadic exchanges with the local environment were considered to be a major shortcoming; this was thought to undermine the university’s ability to establish a value co-creation partnership with its business stakeholders, engendering lower quality of educational services

    Physiologic variations of gastrin in serum of German shepherd dog

    Gastrin is a pool of peptide hormones at different molecular weight secreted by gastric and duodenal mucosa during feeding and involved in the intake of proteins. Aim of present study was to establish the gastrin concentration in serum samples of 40 healthy German shepherd dogs belonging to 5 different age groups. A RIA commercial kit has been used, which has been validated for the animal species under study with preliminary dose-response experiments. Results obtained showed specificity of the procedure used for canine gastrin. The obtained reference range (14.6-61.8 pg/ml) resulted to be superimposable to the range reported in literature for italian mongrel dogs (15- 78.6 pg/ml). No differences related to sex have been observed (females 38.2 + 10.4 pg/ml males 38.2 + 12.7 pg/ml). The gastrin concentration resulted to be elevated in puppies aged 3-3.5 months (58 pg/ml) and stabilized in dogs from 7 months to 10 years on values ranging 27-33 pg/ml. Physiological and pathological role of this hormone is discussed. This study supports the critical evaluation of pathological variations of gastrin in serum of the dogs

    BeppoSAX observations of PKS 0528+134

    We report on the BeppoSAX observations of the gamma-ray blazar PKS 0528+134 performed in Feb and Mar 1997, during a multiwavelength campaign involving EGRET and ground based telescopes. The source was in a faint and hard state, with energy spectral index alpha=0.48+-0.04 between 0.1 and 10 keV, and [2-10] keV flux of 2.7E-12 erg/cm2/s. No significant variability was observed. The source was detected in the 20-120 keV band by the PDS, with a flux lying slightly above the extrapolation from lower X-ray energies. Comparing this low state with previous higher states of the source, there is an indication that the X-ray spectrum hardens and the gamma-ray spectrum steepens when the source is fainter.Comment: 4 pages, LateX, 5 figures (included).     Efficacy of a diet containing caseinate hydrolysate on signs of stress in dogs

    The purpose of this randomized, double blind, placebo-controlled trial was to evaluate 3 the efficacy of a diet containing caseinate hydrolysate (CH) on signs of stress in 2 4 groups of dogs (defined as Anxious and Non-anxious) using physiological (serum 5 cortisol and lysozyme, N:L ratios and heart rate) and behavioral parameters. 6 From an initial group of 40 female Beagle dogs, ranging in age from 10 months to 4 7 years (mean = 1.47 years; SD = 0.53) belonging to a dog colony, 32 were selected for 8 this study according to their level of anxiety. A group of 16 Anxious dogs and a group 9 of 16 Non-anxious dogs were identified. 10 A baseline period, aimed to obtain reference values of investigated parameters, 11 preceded the experimental phase. Both groups (Anxious and Non-anxious) were divided 12 into a treatment group, which received the diet containing CH, and a control group 13 which received a placebo diet (PD). Anxious CH and PD groups were balanced for 14 anxiety level. Each dog was evaluated 3 times a day at 4 weeks intervals (T1-T2-T3). 15 Each evaluation lasted 2 days and involved a Reactivity Evaluation Form (REF), a 16 blood sampling, heart rate recording and a 10 min behavioral video recording. Results 17 from REF scores showed that while at T1 Anxious dogs had significantly higher scores 18 (Mann-Whitney test: P<0.001) compared to Non-anxious dogs, no difference was found 19 between Anxious dogs fed with CH diet and Non-anxious fed with PD or CH diet at T3. 20 Behavioral observations evidenced some signs of improvement in Anxious dog fed with 21 CH diet. Cortisol level significantly decreased in Anxious dogs fed with CH diet 22 (Friedman test: P<0.05). Individual differences in physiological measures of stress 23 responses may have contributed to the large variability, making interpretation of these 24 measures difficult. These results suggest that CH may be used as a functional ingredient 25 alleviating stress in dogs

    Postoperative Delirium after elective and emergency surgery: analysis and checking of risk factors. A study protocol

    BACKGROUND: Delirum is common in hospitalized elderly patients and may be associated with increased morbidity, length of stay and patient care costs. Delirium (acute confusional state) is defined as an acute disorder of attention and cognition. In elderly patients, delirium is often an early indicator of patho-physiological disturbances. Despite landmark studies dating back to the 1940s, the pathogenesis of Delirium remains poorly understood. Early investigators noted that Delirium was characterized by global cortical dysfunction that was associated predominantly with specific electroencephalographic changes. It's important to understand the risk factors and incidence of Delirium. Some of the risk factors are already identified in literature and can be summarized in the word "VINDICATE" which stands for: Vascular, Infections, Nutrition, Drugs, Injury, Cardiac, Autoimmune, Tumors, Endocrine. Aims of this study are: to re-evaluate the above mentioned clinical risk factors, adding some others selected from literature, and to test, as risk factors, a pattern of some genes associated to cognitive dysfunction and inflammation possibly related to postoperative Delirium. DESIGN: All patients admitted to our Emergency Unit who are meet our inclusion/exclusion criteria will be recruited. The arising of postoperative Delirium will select incidentally two groups (Delirium/non Delirium) and the forward analysis of correlate risk factors will be performed. As in a typical observational case/control study we will consider all the exposure factors to which our population are submitted towards the outcome (presence of Delirium). Our exposures are the following: ASA, Pain (SVS; VAS), Blood gas analysis (pH; Hb; pO2; pCO2), Residence pharmacological therapy (BDZ; hypnotics; narcotic drugs; alcohol; nitrous derivates), Body temperature, Arterial pressure, Heart frequency, Breath frequency, Na, K, Creatinin, Glicemia, Albumin, Hct, White blood cells, Glasgow Coma Scale (GCS), Cognitive state (SPMSQ), Functional state (ADL and IADL), Psychological Distress (HADS), Cumulative Illness Rating Scale (CIRS), Hypotension (classified in: light; moderate and severe and duration), Blood loss (classified in: < 2 lt and > 2 lt), Blood transfusions (< 2 lt and > 2 lt), Quantity of red cells and plasma transfusions, Visual VAS / SVS (timing: I-II-III post-operative day), Red cells and Plasma transfusions, Blood count evaluation and Saturation (O(2)%), Postoperative analgesia (Emilia-Romagna protocol), Presence of malignant tumoral disease, APACHE Score II.     Circulating Hepatitis B Surface Antigen Particles Carry Hepatocellular microRNAs

    Hepatitis B virus (HBV) produces high quantities of subviral surface antigen particles (HBsAg) which circulate in the blood outnumbering virions of about 1\103–6 times. In individuals coinfected with the defective hepatitis Delta virus (HDV) the small HDV-RNA-genome and Delta antigen circulate as ribonucleoprotein complexes within HBsAg subviral particles. We addressed the question whether subviral HBsAg particles may carry in the same way cellular microRNAs (miRNAs) which are released into the bloodstream within different subcellular forms such as exosomes and microvescicles. Circulating HBsAg particles were isolated from sera of 11 HBsAg carriers by selective immunoprecipitation with monoclonal anti-HBs-IgG, total RNA was extracted and human miRNAs were screened by TaqMan real-time quantitative PCR Arrays. Thirty-nine human miRNAs were found to be significantly associated with the immunoprecipitated HBsAg, as determined by both comparative DDCT analysis and non-parametric tests (Mann-Whitney, p<0.05) with respect to controls. Moreover immunoprecipitated HBsAg particles contained Ago2 protein that could be revealed in ELISA only after 0.5% NP40. HBsAg associated miRNAs were liver-specific (most frequent = miR-27a, miR-30b, miR-122, miR-126 and miR-145) as well as immune regulatory (most frequent = miR-106b and miR-223).     SDS-PAGE-Based Quantitative Assay for Screening of Kidney Stone Disease

    Kidney stone disease is a common health problem in industrialised nations. We developed a SDS-PAGE-based method to quantify Tamm Horsfall glycoprotein (THP) for screening of kidney stone disease. Urinary proteins were extracted by using ammonium sulphate precipitation at 0.27 g salt/mL urine. The resulted pellet was dissolved in TSE buffer. Ten microliters of the urinary proteins extract was loaded and separated on 10% SDS-PAGE under reducing condition. THP migrated as single band in SDS-PAGE. The assay reproducibility and repeatability were 4.8% CV and 2.6% CV, respectively. A total of 117 healthy subjects and 58 stone patients were tested using this assay, and a distinct cut-off (P < 0.05) at 5.6 ÎĽg/mL THP concentration was used to distinguish stone patients from healthy subjects. The sensitivity and specificity of the method were 92.3% and 83.3%, respectively

    Homologous recombination DNA repair defects in PALB2-associated breast cancers

Mono-allelic germline pathogenic variants in the Partner And Localizer of BRCA2 (PALB2) gene predispose to a high-risk of breast cancer development, consistent with the role of PALB2 in homologous recombination (HR) DNA repair. Here, we sought to define the repertoire of somatic genetic alterations in PALB2-associated breast cancers (BCs), and whether PALB2-associated BCs display bi-allelic inactivation of PALB2 and/or genomic features of HR-deficiency (HRD). Twenty-four breast cancer patients with pathogenic PALB2 germline mutations were analyzed by whole-exome sequencing (WES, n = 16) or targeted capture massively parallel sequencing (410 cancer genes, n = 8). Somatic genetic alterations, loss of heterozygosity (LOH) of the PALB2 wild-type allele, large-scale state transitions (LSTs) and mutational signatures were defined. PALB2-associated BCs were found to be heterogeneous at the genetic level, with PIK3CA (29%), PALB2 (21%), TP53 (21%), and NOTCH3 (17%) being the genes most frequently affected by somatic mutations. Bi-allelic PALB2 inactivation was found in 16 of the 24 cases (67%), either through LOH (n = 11) or second somatic mutations (n = 5) of the wild-type allele. High LST scores were found in all 12 PALB2-associated BCs with bi-allelic PALB2 inactivation sequenced by WES, of which eight displayed the HRD-related mutational signature 3. In addition, bi-allelic inactivation of PALB2 was significantly associated with high LST scores. Our findings suggest that the identification of bi-allelic PALB2 inactivation in PALB2-associated BCs is required for the personalization of HR-directed therapies, such as platinum salts and/or PARP inhibitors, as the vast majority of PALB2-associated BCs without PALB2 bi-allelic inactivation lack genomic features of HRD
