    The effect of antimicrobials on verocytotoxin bacteriophage transduction under bovine rumen fluid and broth conditions

    peer-reviewedThe verocytotoxin genes in verocytotoxigenic Escherichia coli (VTEC) are carried by bacteriophages, incorporated into the bacterial genome (prophage). Antibiotics may promote phage replication and release to infect other cells (transduction), thus leading to the emergence of new VTEC strains. This study investigated transduction of a verocytotoxin2-encoding bacteriophage (3538(vtx2::cat)) under laboratory conditions, including the effect of antibiotic treatments. Luria-Bertani Miller broth and rumen fluid (raw and sterilised by irradiation) were inoculated with the donor (C600φ3538(Δvtx2::cat)) and recipient (E. coli C600::kanamycinR) strains (4 log10 cfu/mL) and incubated at 38°C. Antibiotic treatments (minimal inhibitory and sub-inhibitory concentrations of ampicillin, cefquinome, oxytetracycline and sodium sulfamethazine) were applied after 3 h. Samples were tested for donor, recipient, cell-free phage and transductants at times t = 0, 3, 4, 6, 27 (24 h post-antibiotic treatment) and 51 h. Free phage was detected in the untreated broth and rumen samples, as were the transductants confirmed by polymerase chain reaction. The antibiotic treatments did not significantly (P > 0.01) increase the concentrations of free phage or transductants detected. It was therefore concluded that, under laboratory conditions, the antibiotics tested did not induce bacteriophage lysis, release and infection of new bacterial cells beyond that constitutively found in the phage population

    A classification test for junior high school girls in physical education

    There is a need in physical education for a performance test which classifies according to ability at the junior high school age and which is efficient in time and equipment. The development of such a test was the objective of this study. A test was desired which could be administered to a class of thirty-five girls in thirty minutes of activity, uncomplicated in form and the equipment needed, and which could be administered by one teacher and yield results easily interpreted and acceptably valid and reliable. A review of literature supported homogeneous classification as desirable in physical education. The factors of ability in performance were studied. Methods of classifying which have been used were analyzed, and the more applicable performance tests and items were examined to shape a new test

    Listening and learning: giving voice to trans experiences of disasters

    This article gives voice to trans experiences of disasters, investigating their specific vulnerabilities and resilient capacities. We draw on findings from a project on lesbian, gay, bisexual and trans (LGBT) experiences of recent Australian and New Zealand disasters. We present and analyse trans voices from a survey conducted across multiple case study sites and insights from interview data with a trans person who experienced the 2011 Brisbane floods. Conceptually, to provide a robust understanding of trans experiences of disasters, we bring socially sensitive disaster studies into conversation with trans geographies. Disaster studies have begun to examine LGBT experiences, with some suggestion that trans people are most vulnerable. We advance this work by focusing on trans lives. Trans geographies, in turn, underline the importance of space, place and the body in understanding trans lives, and the need to examine the lived reality of trans people’s everyday geographies rather than embodiment as an abstract concept. Applying these insights to the trans voices in our project, we examine four themes that highlight impediments to and possibilities for trans-inclusive disaster planning: apprehension with emergency services and support; concerns about home and displacement; anxiety about compromising the trans body; and the potential of trans and queer interpersonal networks for capacity building. We offer suggestions for trans-inclusive disaster planning and preparedness, and indicate how the insights from trans experience can enrich disaster planning and preparedness for wider social groups.Australian Research Council-DP13010265

    Supply chains and energy security in a low carbon transition

    This special edition to be published in Applied Energy brings together a range of papers that explore the complex, multi-dimensional and inter-related issues associated with the supply or value chains that make up energy systems and how a focus on them can bring new insights for energy security in a low carbon transition. Dealing with the trilemma of maintaining energy security, reducing greenhouse gas emissions and maintaining affordability for economies and end users are key issues for all countries, but there are synergies and trade-offs in simultaneously dealing with these different objectives. Currently, industrialised energy systems are dominated by supply chains based on fossil fuels and these, for the most part, have been effective in enabling energy security and affordability. However, they are increasingly struggling to do this, particularly in respect to efforts to tackle climate change, given that the energy sector is responsible for around two-thirds of the global greenhouse gas emissions [1]. A key challenge is therefore how to decarbonise energy systems, whilst also ensuring energy security and affordability. This special issue, through a focus on supply chains, particularly considers the interactions and relationships between energy security and decarbonisation. Energy security is a property of energy systems and their ability to withstand short-term shocks and longer-term stresses depends on other important system properties including resilience, robustness, flexibility and stability [2]. Energy systems are essentially a supply chain comprising of multiple and interrelated sub-chains based around different fuels, technologies, infrastructures, and actors, operating at different scales and locations – from extraction/imports and conversion through to end use [3]. These supply chains have become increasingly globalised and are influenced by the on-going shifts in global supply and demand. Thus the aim of this special issue is to explore and discuss how to enable the development of a secure and sustainable energy system through a better understanding of both existing and emerging low carbon energy supply chains as well as of new approaches to the design and management of energy systems. In part, because moving from a system dominated by fossil fuels to one based on low carbon creates a new set of risks and uncertainties for energy security as well as new opportunities. A large number of submissions from over 18 countries were received for this special edition and 16 papers were accepted after peer review. These address a variety of issues and we have chosen to discuss the findings under two key themes, although many of the papers cut across these: (1) Insights from, and for, supply chain analysis. (2) Insights for energy security and its management. We then provide in (3) a summary of insights and research gaps.     Real-time PCR methods for the detection of blown pack spoilage causing Clostridium species; C. estertheticum, C. gasigenes and C. ruminantium

    A set of real-time PCR methods for the detection of C. estertheticum, C. gasigenes and C. ruminantium, the causative agents of blown pack spoilage (BPS) in vacuum packaged beef, was developed. Robust validation of the sensitivity and specificity was carried out in the three matrices (beef meat drip, wet environmental swabs and dry environmental swabs) as encountered in our testing laboratory and against Clostridium strains (n = 76) and non-Clostridium strains (n = 36). It was possible to detect 4–5 spores per ml for C. estertheticum, 2 spores per ml for C. gasigenes and 8 spores per ml for C. ruminantium, without the need for enrichment of the samples. This high sensitivity is particularly important for the beef sector, not just for testing spoiled product but also in the early detection of contaminated beef and in validation of sporicidal cleaning procedures for critical pieces of equipment such as the vacuum packaging machine, which have the potential to contaminate large volumes of product

    Genetic characterisation of a subset of Campylobacter jejuni isolates from clinical and poultry sources in Ireland

    peer reviewedCampylobacter spp. is a significant and prevalent public health hazard globally. Campylobacter jejuni is the most frequently recovered species from human cases and poultry are considered the most important reservoir for its transmission to humans. In this study, 30 Campylobacter jejuni isolates were selected from clinical (n = 15) and broiler (n = 15) sources from a larger cohort, based on source, virulence, and antimicrobial resistance profiles. The objective of this study was to further characterise the genomes of these isolates including MLST types, population structure, pan-genome, as well as virulence and antimicrobial resistance determinants. A total of 18 sequence types and 12 clonal complexes were identified. The most common clonal complex was ST-45, which was found in both clinical and broiler samples. We characterised the biological functions that were associated with the core and accessory genomes of the isolates in this study. No significant difference in the prevalence of virulence or antimicrobial resistance determinants was observed between clinical and broiler isolates, although genes associated with severe illness such as neuABC, wlaN and cstIII were only detected in clinical isolates. The ubiquity of virulence factors associated with motility, invasion and cytolethal distending toxin (CDT) synthesis in both clinical and broiler C. jejuni genomes and genetic similarities between groups of broiler and clinical C. jejuni reaffirm that C. jejuni from poultry remains a significant threat to public health.Teagas

    Time reclaimed: temporality and the experience of meaningful work

    The importance of meaningful work has been identified in scholarly writings across a range of disciplines. However, empirical studies remain sparse and the potential relevance of the concept of temporality, hitherto somewhat neglected even in wider sociological studies of organizations, has not been considered in terms of the light that it can shed on the experience of work as meaningful. These two disparate bodies of thought are brought together to generate new accounts of work meaningfulness through the lens of temporality. Findings from a qualitative study of workers in three occupations with ostensibly distinct temporal landscapes are reported. All jobs had the potential to be both meaningful and meaningless; meaningfulness arose episodically through work experiences that were shared, autonomous and temporally complex. Schutz’s notion of the ‘vivid present’ emerged as relevant to understanding how work is rendered meaningful within an individual’s personal and social system of relevances
