    The Headlines This unique research project revealed fascinating insights into the spiritual flourishing of very young children and how this is nourished within church toddler groups in England. The findings have uncovered evidence of very young children exploring self-understanding and the nurturing of relationships with others. We found that church toddler groups contribute to children’s spiritual flourishing through: • Providing an environment that fosters a non-threatening connection with spirituality and faith. • Nurturing and sustaining relationships with families. • Offering a safe place for young families to come together, play together, and feel supported in their spiritual well-being. • Enabling a sense of belonging to the church community through engagement in the church toddler group. However, we also found that: • There is a need for adults to have a deeper awareness of the significant role they play in enabling the spiritual flourishing of very young children. • A common misunderstanding of spiritual nurture in terms of faith development hampers focused attention on the spiritual nurture of very young children

    Severe pneumonia due to Parachlamydia acanthamoebae following intranasal inoculation: a mice model.

    Parachlamydia acanthamoebae is an obligate intracellular bacterium naturally infecting free-living amoebae. The role of this bacterium as an agent of pneumonia is suggested by sero-epidemiological studies and molecular surveys. Furthermore, P. acanthamoebae may escape macrophages microbicidal effectors. Recently, we demonstrated that intratracheal inoculation of P. acanthamoebae induced pneumonia in 100% of infected mice. However, the intratracheal route of infection is not the natural way of infection and we therefore developed an intranasal murine model. Mice inoculated with P. acanthamoebae by intranasal inoculation lost 18% of their weight up to 8 days post-inoculation. All mice presented histological signs of pneumonia at day 2, 4, 7, and 10 post-inoculation, whereas no control mice harboured signs of pneumonia. A 5-fold increase in bacterial load was observed from day 0 to day 4 post-inoculation. Lungs of inoculated mice were positive by Parachlamydia-specific immunohistochemistry 4 days post-inoculation, and P. acanthamoebae were localized within macrophages. Thus, we demonstrated that P. acanthamoebae induce a severe pneumonia in mice. This animal model (i) further supports the role of P. acanthamoebae as an agent of pneumonia, confirming the third Koch postulate, and (ii) identified alveolar macrophages as one of the initial cells where P. acanthamoebae is localized following infection

    Linking dominant rainfall-runoff event hydrologic response dynamics with nitrate and chloride load estimates of three boreal Shield catchments

    Understanding hydrological dynamics in boreal Shield catchments is critical for projecting changes in stream runoff and chemistry in a region that is sensitive to climate change. Previous work has mostly focused on a limited number of events over one or a few seasons because of the relative scarcity of high-frequency datasets and automated tools for rainfall-runoff event delineation. For the boreal region, a greater understanding of seasonality in hydrologic response and solute export related to rainfall-initiated events is needed, as significant shifts in hydrologic regime from climate change are expected. This study aimed to help resolve these knowledge gaps by assessing event-scale hydrologic response dynamics and stream loads of nitrate and chloride using long-term data from three boreal Shield catchments. Hydrometric and stream chemistry data from 2001-2018 were analyzed to delineate rainfall-runoff events and estimate event nitrate and chloride loads. Event hydrologic response and loads were highly variable, especially with respect to catchment runoff initiation. Only subtle differences in hydrologic response dynamics were observed between summer and fall events, while seasonal differences in event nitrate and chloride loads were mostly statistically significant. Interestingly, a wide range of rainfall-runoff events classified by response magnitude and timing were associated with differences in nitrate and chloride export.     Inter-sectoral and multilevel coordination alone do not reduce deforestation and advance environmental justice:Why bold contestation works when collaboration fails

    Policy makers, academics, and conservationists often posit that poor coordination between different land use sectors, and between levels of governance, as an underlying challenge for reducing deforestation and forest degradation. This paper analyzes this argument using data from interviews with over 500 respondents from government, nongovernmental organizations, private companies, local and indigenous communities, activists, and individuals involved in 35 diverse land use initiatives in three countries: Peru, Indonesia, and Mexico. We find that while there is strong evidence of widespread coordination failures between sectors and levels, more fundamental political issues preclude effective coordination. We argue that political coalitions act to oppose environmental objectives and to impede their opponents from participating in land use governance. Moreover, we find that where coordination between actors does occur, it does not necessarily produce environmentally sustainable and socially just land use outcomes. Where we do find successful initiatives to reduce deforestation and benefit local people, effective coordination between well-informed actors is often present, but it does not occur spontaneously, and is instead driven by political organizing over time by activists, local people, nongovernmental organizations, and international donors. We suggest that the global environmental community must recognize explicitly these political dimensions of land use governance in order to successfully collaborate with local people to reduce deforestation

    Integrated modelling and multiscale gyrokinetic validation study of ETG turbulence in a JET hybrid H-mode scenario

    Previous studies with first-principle-based integrated modelling suggested that ETG turbulence may lead to an anti-GyroBohm isotope scaling in JET high-performance hybrid H-mode scenarios. A dedicated comparison study against higher-fidelity turbulence modelling invalidates this claim. Ion-scale turbulence with magnetic field perturbations included, can match the power balance fluxes within temperature gradient error margins. Multiscale gyrokinetic simulations from two distinct codes produce no significant ETG heat flux, demonstrating that simple rules-of-thumb are insufficient criteria for its onset

    Caspase-11 Activation in Response to Bacterial Secretion Systems That Access the Host Cytosol

    Inflammasome activation is important for antimicrobial defense because it induces cell death and regulates the secretion of IL-1 family cytokines, which play a critical role in inflammatory responses. The inflammasome activates caspase-1 to process and secrete IL-1β. However, the mechanisms governing IL-1α release are less clear. Recently, a non-canonical inflammasome was described that activates caspase-11 and mediates pyroptosis and release of IL-1α and IL-1β. Caspase-11 activation in response to Gram-negative bacteria requires Toll-like receptor 4 (TLR4) and TIR-domain-containing adaptor-inducing interferon-β (TRIF)-dependent interferon production. Whether additional bacterial signals trigger caspase-11 activation is unknown. Many bacterial pathogens use specialized secretion systems to translocate effector proteins into the cytosol of host cells. These secretion systems can also deliver flagellin into the cytosol, which triggers caspase-1 activation and pyroptosis. However, even in the absence of flagellin, these secretion systems induce inflammasome activation and the release of IL-1α and IL-1β, but the inflammasome pathways that mediate this response are unclear. We observe rapid IL-1α and IL-1β release and cell death in response to the type IV or type III secretion systems of Legionella pneumophila and Yersinia pseudotuberculosis. Unlike IL-1β, IL-1α secretion does not require caspase-1. Instead, caspase-11 activation is required for both IL-1α secretion and cell death in response to the activity of these secretion systems. Interestingly, whereas caspase-11 promotes IL-1β release in response to the type IV secretion system through the NLRP3/ASC inflammasome, caspase-11-dependent release of IL-1α is independent of both the NAIP5/NLRC4 and NLRP3/ASC inflammasomes as well as TRIF and type I interferon signaling. Furthermore, we find both overlapping and non-redundant roles for IL-1α and IL-1β in mediating neutrophil recruitment and bacterial clearance in response to pulmonary infection by L. pneumophila. Our findings demonstrate that virulent, but not avirulent, bacteria trigger a rapid caspase-11-dependent innate immune response important for host defense

    Safety and Immunogenicity of the Live Attenuated Varicella Vaccine Following T Replete or T Cell-Depleted Related and Unrelated Allogeneic Hematopoietic Cell Transplantation (alloHCT)

    There are limited studies assessing the live attenuated varicella vaccine following allogeneic hematopoietic cell transplantation (alloHCT). Because of the morbidity of varicella acquired after childhood, we immunized and retrospectively analyzed the safety and immunogenicity of this vaccine in 46 varicella zoster virus (VZV) seronegative patients <20 years old at HCT who achieved a CD4 cell count ≥200/μL, were off immunosuppression, and responded to ≥1 post-HCT vaccines. Two vaccinated patients lacking follow-up titers were excluded from analysis. Stem cells were derived from an HLA-matched sibling (n = 18) or an alternative (HLA mismatched related or unrelated) donor (n = 26). Median time to vaccination was 4 years. Sixty-four percent of patients seroconverted following 1 immunization. There was no significant difference in response between recipients of a matched related or alternative donor graft (P = .2) or between those given a T cell-depleted or T-replete alternative donor graft (P = .27). Three of 44 patients developed a self-limited varicella-like rash within 2.5 weeks of immunization. With a median follow-up of 29.1 (range: 6.9-167.1) months, there were no subsequent cases of varicella-like rashes. No patient developed shingles. This study suggests that this vaccine is safe and immunogenic when given according to preset clinical and immunologic milestones, warranting larger prospective studies in patients ≥24 months following HCT as outlined in current post-HCT vaccine guidelines

    Hospitality research continues to broaden through an ever-increasing dialogue and alignment with a greater number of academic disciplines. This paper demonstrates how an enhanced understanding of hospitality can be achieved through synergy between archaeology, the classics and sociology. It focuses on classical Roman life, in particular Pompeii, to illustrate the potential for research synergy and collaboration, to advance the debate on hospitality research and to encourage divergence in research approaches. It demonstrates evidence of commercial hospitality activities through the excavation hotels, bars and taverns, restaurants and fast food sites. The paper also provides an example of the benefits to be gained from multidisciplinary analysis of hospitality and tourism

    Sociological and Communication-Theoretical Perspectives on the Commercialization of the Sciences

    Both self-organization and organization are important for the further development of the sciences: the two dynamics condition and enable each other. Commercial and public considerations can interact and "interpenetrate" in historical organization; different codes of communication are then "recombined." However, self-organization in the symbolically generalized codes of communication can be expected to operate at the global level. The Triple Helix model allows for both a neo-institutional appreciation in terms of historical networks of university-industry-government relations and a neo-evolutionary interpretation in terms of three functions: (i) novelty production, (i) wealth generation, and (iii) political control. Using this model, one can appreciate both subdynamics. The mutual information in three dimensions enables us to measure the trade-off between organization and self-organization as a possible synergy. The question of optimization between commercial and public interests in the different sciences can thus be made empirical.Comment: Science & Education (forthcoming
