    Observational evidence for a broken Li Spite plateau and mass-dependent Li depletion

    We present NLTE Li abundances for 88 stars in the metallicity range -3.5 < [Fe/H] < -1.0. The effective temperatures are based on the infrared flux method with improved E(B-V) values obtained mostly from interstellar NaI D lines. The Li abundances were derived through MARCS models and high-quality UVES+VLT, HIRES+Keck and FIES+NOT spectra, and complemented with reliable equivalent widths from the literature. The less-depleted stars with [Fe/H] < -2.5 and [Fe/H] > -2.5 fall into two well-defined plateaus of A_{Li} = 2.18 (sigma = 0.04) and A_{Li} = 2.27 (sigma = 0.05), respectively. We show that the two plateaus are flat, unlike previous claims for a steep monotonic decrease in Li abundances with decreasing metallicities. At all metallicities we uncover a fine-structure in the Li abundances of Spite plateau stars, which we trace to Li depletion that depends on both metallicity and mass. Models including atomic diffusion and turbulent mixing seem to reproduce the observed Li depletion assuming a primordial Li abundance A_{Li} = 2.64, which agrees well with current predictions (A_{Li} = 2.72) from standard Big Bang nucleosynthesis. Adopting the Kurucz overshooting model atmospheres increases the Li abundance by +0.08 dex to A_{Li} = 2.72, which perfectly agrees with BBN+WMAP.Comment: A&A Letters, in pres

    Occurrence of phytoplasma associated with sugarcane yellow leaf syndrome in three regions of São Paulo State, Brazil

    Uma doença conhecida como síndrome do amarelecimento foliar da cana-de-açúcar ou amarelinho causou danos expressivos à cultura no estado de São Paulo, na década de noventa. A síndrome também foi registrada em outros países e investigações sobre a etiologia evidenciaram a associação da doença com vírus e fitoplasma, de forma isolada ou simultânea. A doença é potencialmente importante e ocorre endemicamente no estado paulista, por esta razão o presente estudo buscou demonstrar a ocorrência do fitoplasma em três tradicionais regiões produtoras de cana, localizadas no estado de São Paulo, através da detecção molecular deste agente em plantas naturalmente infectadas. Para isto, plantas sintomáticas das variedades SP71-6163, SP71-6180 e SP89-1115 foram amostradas em Piracicaba, Jaú e Ribeirão Preto e a partir do tecido foliar foi extraído o DNA total. PCR duplo foi processado com os "primers" P1/Tint-16F2n/R2 e os produtos foram analisados por eletroforese em gel de agarose. Amplificações de fragmentos de DNA de 1,2 kb evidenciaram a presença de fitoplasma em 36% das plantas sintomáticas e revelaram a sua ocorrência nas três regiões amostradas. Os resultados demonstraram a associação do fitoplasma com a doença e a importância de se manter uma atenta vigilância nos campos e viveiros, bem como buscar genótipos com bom nível de resistência nos programas de melhoramento.Sugarcane yellow leaf syndrome caused serious damage to crops in the São Paulo State, Brazil, in the 1990's. The syndrome was also reported in other countries and investigations into the etiology revealed an association between the disease and virus and/or phytoplasma. The disease is potentially important and occurs endemically in that State, and for this reason the present study was conducted in order to demonstrate the occurrence of phytoplasma in three traditional sugarcane-producing areas, in São Paulo State, through molecular detection from naturally infected plants. Symptomatic plants belonging to varieties SP71-6163, SP71-6180 and SP89-1115 were sampled from Piracicaba, Jaú and Ribeirão Preto, and total DNA was extracted from foliar tissues. Nested PCR was conducted with primer pairs P1/Tint-16F2n/R2, and the amplified products were analyzed by electrophoresis on agarose gels. Amplified DNA fragments of 1.2 kb evidenced the presence of phytoplasma in 36% of symptomatic plants and revealed its occurrence in all sampled regions. The results demonstrated that phytoplasma is associated with the disease and that it is important to keep a safe inspection of nurseries and monitoring plants in the field, as well as to select sugarcane genotypes with a good level of resistance in breeding programs.Coordenação de Aperfeiçoamento de Pessoal Nível Superior - CAPE

    Comparative experimental study between L-Hydro treated pulmonary homograft and fresh pulmonary homograft

    OBJETIVO: Buscando novas formas de preservação de tecidos utilizamos o polietileno-glicol, método L-Hydro (LH), que consiste na extração controlada de substâncias antigênicas e incorporação de agente antinflamatório e antitrombótico. MÉTODOS: Em dez carneiros jovens, substituímos o tronco pulmonar, em sete, por homoenxertos pulmonares (HP) tratados pelo processo L-H e, em três, por HP a fresco, implantados ortotopicamente, seguidos por 320 dias. Os carneiros foram avaliados por exames laboratoriais e ecocardiográficos. Ao cabo dos 320 dias foram sacrificados, procedendo-se à avaliação hemodinâmica, radiológica, macro/microscópica, óptica e eletrônica, varredura e transmissão. Resultados foram analisados pelo teste t de Student de amostras independentes para dados contínuos, análise de variância para medidas repetidas, pelo teste exato de Fisher para dados categóricos. RESULTADOS: Evolução clínica e exames laboratoriais não conseguiram estabelecer diferenças significativas entre os grupos. Ecocardiograma revelou diferença quanto ao gradiente médio pulmonar, significativa aos 10 meses, maior no grupo controle. Avaliação radiológica e macroscópica não estabeleceu diferenças. Na avaliação microscópica, óptica/eletrônica, células de revestimento e intersticiais foram encontradas nos dois grupos igualmente. O porcentual de revestimento celular calculado nos dois grupos foi semelhante. Nódulos de celularidade foram observados somente no grupo de homoenxertos a fresco. CONCLUSÕES: Estes dados indicam que os dois grupos apresentaram desempenho clínico e hemodinâmico semelhante. Ao ecocardiograma o grupo LH apresentou melhor desempenho, e evidências histológicas de repopulação celular intersticial e endotelial. Na análise macro/microscópica, óptica/eletrônica, o grupo L-Hydro apresentou macroscopia, estrutura histológica e ultraestrutural semelhante ao homoenxerto fresco, à exceção de nódulos de maior celularidade intersticial, presentes apenas no homoenxerto a fresco.OBJECTIVE: In an effort to make available homografts preserved in a simpler and less costly way, we evaluated the polyethyleneglycol, L-Hydro (LH) method, that consists in the controlled extraction of antigenic substances and the incorporation of anti-inflammatory and anti-thrombotic agent. METHODS: We substituted the pulmonary trunk in ten ovines, seven received LH treated pulmonary homografts and three, fresh pulmonary homografts, orthotopically implanted and followed-up for 320 days. Ovines where evaluated by means of laboratory tests, echocardiographic exams. At the 320 days, were euthanized, hemodynamic, radiology, macroscopic, optic/electronic microscopic, scanning/transmission evaluations were performed. Results were analyzed by Student t test of independent samples for continuous data, by variance analysis of repeated measures, and by Fisher exact test for categorical data. RESULTS: We couldn't establish relevant differences in clinical evolution and laboratory tests between groups. Echocardiogram revealed difference in pulmonary medium gradient, which was significant 10 months follow-up, higher in the control group. Radiologic and macroscopic evaluations didn't established differences. In the optic/electronic microscopic evaluation, liner and interstitial cells were equally found in both groups. The cell liner percent calculated in both groups was similar. Cellularity nodules were observed only infresh homograft group. CONCLUSIONS: These data indicate that both groups presented similar clinical/hemodynamic performances. The LH group's echocardiogram presented a better performance. It also presented histological evidences of interstitial and endothelial cell repopulation. In the macro/optic and electronic microscopic analysis, group L-H presented macroscopy/histological structure and ultra-structural similar to the fresh group, with the exception of nodules with higher interstitial cellularity, present only in the fresh homograft group.Fapes

    The Chemical Homogeneity of Sun-like Stars in the Solar Neighborhood

    The compositions of stars are a critical diagnostic tool for many topics in astronomy such as the evolution of our Galaxy, the formation of planets, and the uniqueness of the Sun. Previous spectroscopic measurements indicate a large intrinsic variation in the elemental abundance patterns of stars with similar overall metal content. However, systematic errors arising from inaccuracies in stellar models are known to be a limiting factor in such studies, and thus it is uncertain to what extent the observed diversity of stellar abundance patterns is real. Here we report the abundances of 30 elements with precisions of 2% for 79 Sun-like stars within 100 parsecs. Systematic errors are minimized in this study by focusing on solar twin stars and performing a line-by-line differential analysis using high-resolution, high-signal-to-noise spectra. We resolve [X/Fe] abundance trends in galactic chemical evolution at precisions of 10310^{-3} dex Gyr1^{-1} and reveal that stars with similar ages and metallicities have nearly identical abundance patterns. Contrary to previous results, we find that the ratios of carbon-to-oxygen and magnesium-to-silicon in solar metallicity stars are homogeneous to within 10% throughout the solar neighborhood, implying that exoplanets may exhibit much less compositional diversity than previously thought. Finally, we demonstrate that the Sun has a subtle deficiency in refractory material relative to >80% of solar twins (at 2σ\sigma confidence), suggesting a possible signpost for planetary systems like our own.Comment: ApJ accepted versio

    Replacement of pulmonary artery trunk in sheep using tubular valved heterograft in non-aldehydic preservation

    INTRODUCTION: The cardiac bioprostheses are related to thromboembolic events, infectious and degenerative diseases. Wear is mainly attributed to the denaturation of collagen. Glutaraldehyde, the predominant method of preservation of bioprostheses, favors the calcification process and limits their durability. Several techniques try to contain the degenerative process of bioprostheses. OBJECTIVES: To evaluate the process of calcification in vivo pulmonary valve heterografts preserved in non-aldehydic (L-Hydro®). METHODS: Seventeen sheep underwent replacement of the pulmonary artery valved tubular grafts of bovine pericardium. The animals were divided into two groups: Group L-Hydro® (test / n = 14) and Group Glutaraldehyde (control / n = 3). About 150 days after implantation the animals were sacrificed, necropsied and implants subjected to a pathological study, radiological evaluation and measurement of calcium by atomic absorption spectrophotometry. Statistical analysis was obtained through the Fisher's exact test, Student's t or Mann-Whitney test (significance: 5%). RESULTS: The radiological evaluation, the macroscopic and microscopic measurement of serum calcium by atomic absorption spectrophotometry showed increased calcification of the prosthetic group Glutaraldehyde, when compared to denture-HydroR Group L (P = 0.001). Seven animals in Group L-Hydro® (50%) had adherence of the leaflets to the wall of the tube (P = 0.228). CONCLUSIONS: Prostheses preserved in L-Hydro® were more resistant to calcification when compared with glutaraldehyde preserved.INTRODUÇÃO: As biopróteses valvares cardíacas estão relacionadas a eventos tromboembólicos, infecciosos e degenerativos. Seu desgaste é atribuído principalmente à desnaturação do colágeno. O glutaraldeído, método predominante de preservação de biopróteses, favorece o processo de calcificação e limita sua durabilidade. Diversas técnicas tentam conter o processo degenerativo das biopróteses. OBJETIVO: Avaliar o processo de calcificação, in vivo, de heteroenxertos pulmonares valvados, preservados em meio não-aldeídico (L-Hydro®). MÉTODOS: Dezessete carneiros foram submetidos à substituição do tronco da artéria pulmonar por enxerto tubular valvado de pericárdio bovino. Os animais foram distribuídos em dois grupos: Grupo L-Hydro® (teste / n=14) e Grupo Glutaraldeído (controle /n=3). Cerca de 150 dias pós-implante os animais foram sacrificados, necropsiados e as próteses submetidas a estudo anatomopatológico, avaliação radiológica e dosagem do cálcio por espectrofotometria de absorção atômica. A análise estatística foi obtida por meio dos testes exato de Fisher, T de Student ou Mann-Whitney (significância: 5%). RESULTADOS: A avaliação radiológica, macroscopia, microscopia e dosagem de cálcio por espectrofotometria de absorção atômica demonstraram maior calcificação nas próteses do Grupo Glutaraldeído, quando comparadas às próteses do Grupo L-Hydro® (P=0,001). Sete animais do Grupo L-Hydro® (50%) apresentaram aderência das cúspides à parede do tubo (P=0,228).     Pyruvate dehydrogenase kinase regulates vascular inflammation in atherosclerosis and increases cardiovascular risk

    Aims Recent studies have revealed a close connection between cellular metabolism and the chronic inflammatory process of atherosclerosis. While the link between systemic metabolism and atherosclerosis is well established, the implications of altered metabolism in the artery wall are less understood. Pyruvate dehydrogenase kinase (PDK)-dependent inhibition of pyruvate dehydrogenase (PDH) has been identified as a major metabolic step regulating inflammation. Whether the PDK/PDH axis plays a role in vascular inflammation and atherosclerotic cardiovascular disease remains unclear. Methods and results Gene profiling of human atherosclerotic plaques revealed a strong correlation between PDK1 and PDK4 transcript levels and the expression of pro-inflammatory and destabilizing genes. Remarkably, the PDK1 and PDK4 expression correlated with a more vulnerable plaque phenotype, and PDK1 expression was found to predict future major adverse cardiovascular events. Using the small-molecule PDK inhibitor dichloroacetate (DCA) that restores arterial PDH activity, we demonstrated that the PDK/PDH axis is a major immunometabolic pathway, regulating immune cell polarization, plaque development, and fibrous cap formation in Apoe−/− mice. Surprisingly, we discovered that DCA regulates succinate release and mitigates its GPR91-dependent signals promoting NLRP3 inflammasome activation and IL-1β secretion by macrophages in the plaque. Conclusions We have demonstrated for the first time that the PDK/PDH axis is associated with vascular inflammation in humans and particularly that the PDK1 isozyme is associated with more severe disease and could predict secondary cardiovascular events. Moreover, we demonstrate that targeting the PDK/PDH axis with DCA skews the immune system, inhibits vascular inflammation and atherogenesis, and promotes plaque stability features in Apoe−/− mice. These results point toward a promising treatment to combat atherosclerosis

    ITCH E3 ubiquitin ligase downregulation compromises hepatic degradation of branched-chain amino acids

    Objective: Metabolic syndrome, obesity, and steatosis are characterized by a range of dysregulations including defects in ubiquitin ligase tagging proteins for degradation. The identification of novel hepatic genes associated with fatty liver disease and metabolic dysregulation may be relevant to unravelling new mechanisms involved in liver disease progression METHODS: Through integrative analysis of liver transcriptomic and metabolomic obtained from obese subjects with steatosis, we identified itchy E ubiquitin protein ligase (ITCH) as a gene downregulated in human hepatic tissue in relation to steatosis grade. Wild-type or ITCH knockout mouse models of non-alcoholic fatty liver disease (NAFLD) and obesity-related hepatocellular carcinoma were analyzed to dissect the causal role of ITCH in steatosis RESULTS: We show that ITCH regulation of branched-chain amino acids (BCAAs) degradation enzymes is impaired in obese women with grade 3 compared with grade 0 steatosis, and that ITCH acts as a gatekeeper whose loss results in elevation of circulating BCAAs associated with hepatic steatosis. When ITCH expression was specifically restored in the liver of ITCH knockout mice, ACADSB mRNA and protein are restored, and BCAA levels are normalized both in liver and plasma CONCLUSIONS: Our data support a novel functional role for ITCH in the hepatic regulation of BCAA metabolism and suggest that targeting ITCH in a liver-specific manner might help delay the progression of metabolic hepatic diseases and insulin resistance

    The Solar Twin Planet Search: V. Close-in, low-mass planet candidates and evidence of planet accretion in the solar twin HIP 68468

    Context. More than two thousand exoplanets have been discovered to date. Of these, only a small fraction have been detected around solar twins, which are key stars because we can obtain accurate elemental abundances especially for them, which is crucial for studying the planet-star chemical connection with the highest precision. Aims. We aim to use solar twins to characterise the relationship between planet architecture and stellar chemical composition. Methods. We obtained high-precision (1 m s-1) radial velocities with the HARPS spectrograph on the ESO 3.6 m telescope at La Silla Observatory and determined precise stellar elemental abundances (~0.01 dex) using spectra obtained with the MIKE spectrograph on the Magellan 6.5 m telescope. Results. Our data indicate the presence of a planet with a minimum mass of 26 ± 4 Earth masses around the solar twin HIP 68468. The planet is more massive than Neptune (17 Earth masses), but unlike the distant Neptune in our solar system (30 AU), HIP 68468c is close-in, with a semi-major axis of 0.66 AU, similar to that of Venus. The data also suggest the presence of a super-Earth with a minimum mass of 2.9 ± 0.8 Earth masses at 0.03 AU; if the planet is confirmed, it will be the fifth least massive radial velocity planet candidate discovery to date and the first super-Earth around a solar twin. Both isochrones (5.9 ± 0.4 Gyr) and the abundance ratio [Y/Mg] (6.4 ± 0.8 Gyr) indicate an age of about 6 billion years. The star is enhanced in refractory elements when compared to the Sun, and the refractory enrichment is even stronger after corrections for Galactic chemical evolution. We determined a nonlocal thermodynamic equilibrium Li abundance of 1.52 ± 0.03 dex, which is four times higher than what would be expected for the age of HIP 68468. The older age is also supported by the low log (R'HK) (–5.05) and low jitter (<1 m s-1). Engulfment of a rocky planet of 6 Earth masses can explain the enhancement in both lithium and the refractory elements. Conclusions. The super-Neptune planet candidate is too massive for in situ formation, and therefore its current location is most likely the result of planet migration that could also have driven other planets towards its host star, enhancing thus the abundance of lithium and refractory elements in HIP 68468. The intriguing evidence of planet accretion warrants further observations to verify the existence of the planets that are indicated by our data and to better constrain the nature of the planetary system around this unique starJ.M. acknowledges support from FAPESP (2012/24392-2) and CNPq (Bolsa de Produtividade).
