    Bone allografts in the osteosynthesis of traumatic fractures

    Nos últimos anos, assistimos a importantes avanços científicos na área da segurança microbiológica, na imunologia e no conhecimento do comportamento biológico dos aloenxertos do aparelho locomotor, assim como a alterações da legislação que regulamenta as transplantações de orgãos e tecidos de origem humana, o que conduziu a alterações importantes na organização dos Bancos de Tecidos em todo o mundo. Os aloenxertos ósseos podem estar indicados na osteossíntese de fracturas ósseas traumáticas e no tratamento das complicações desse tipo de lesões, área que não tem sido suficientemente divulgada na literatura internacional. No período compreendido entre 1982 e 2007, o BancoTecidos dos HUC disponibilizou 5231 aloenxertos do aparelho locomotor para aplicação clínica. De entre estes e no período compreendido entre os anos 1994 e 2006, foram usados 1078 aloenxertos ósseos na osteossíntese de fracturas ósseas traumáticas e no tratamento de complicações de fracturas, na condição de medida terapêutica complementar: 500 esponjosos granulados, 98 maciços e 480 desmineralizados. O número das fracturas ósseas traumáticas e dos casos com complicações de fracturas foi de 420, distribuídos da seguinte forma: 290 localizados no fémur, 84 na tíbia, 32 no úmero e 14 no rádio. As lesões da extremidade distal do fémur representaram a causa mais frequente da aplicação dos aloenxertos. Nesta série, não foram confirmados casos de infecção associada ao aloenxerto, nem qualquer caso de transmissão de doenças virais aos receptores. A consolidação das fracturas foi conseguida entre os 3 e os 6 meses. Uma reabsorção parcial do enxerto foi verificada em 21 casos (5%), em correlação directa com as deficientes condições vasculares do leito receptor e com a técnica cirúrgica. A aplicação de aloenxerto ósseo no tratamento de fracturas ósseas e das suas complicações é um procedimento seguro e com resultados satisfatórios

    Relationship between soil fungal diversity and temperature in the maritime Antarctic

    Soil fungi have pivotal ecological roles as decomposers, pathogens and symbionts1, 2. Alterations to their diversity arising from climate change could have substantial effects on ecosystems, particularly those undergoing rapid warming that contain few species3, 4. Here, we report a study using pyrosequencing to assess fungal diversity in 29 soils sampled from a 1,650 km climatic gradient through the maritime Antarctic, the most rapidly warming region in the Southern Hemisphere5, 6. Using a ‘space-for-time’ substitution approach, we show that soil fungal diversity is higher in warmer habitats, with increases of 4.7 (observed) and 11.3 (predicted) fungal taxa per degree Celsius rise in surface temperature along the transect. Among 22 predictor variables, air temperature was the strongest and most consistent predictor of diversity. We propose that the current rapid warming in the maritime Antarctic (0.34 °C per decade6) will facilitate the colonization of soil by a wider diversity of fungi than at present, with data from regression models suggesting 20–27% increases in fungal species richness in the southernmost soils by 2100. Such increases in diversity, which provide a sentinel for changes at lower latitudes, are likely to have substantial effects on nutrient cycling and, ultimately, productivity in the species-poor soils of maritime Antarctica

    Rhizobium strains competitiveness on bean nodulation in Cerrado soils.

    ABSTRACT: The objective of this work was to identify the most competitive and effective Rhizobium strains in order to increase common bean yield by nitrogen fixation as alternative or complementation to the nitrogen fertilization. Competitiveness tests were lead in axenic conditions, in Cerrado soil pots and in three field experiments, with native Rhizobium strains that were previously identified, according to their effectiveness and genetic variability. The identification of strains in nodules was performed using serological tests (axenic conditions) ? agglutination and enzyme linked immunosorbent (Elisa) assays ? and random amplified polymorfic DNA (RAPD) (Cerrado soil). Plant yield was determined using the dry weight (greenhouse conditions), total N and grain yield (field experiments). Among the analyzed Rhizobium strains, native strain SLA 2.2 and commercial strain CIAT 899 were the dominant nodules in plants of the most productive plots, presenting yield productivity similar or higher to those obtained in treatments where 20 kg ha-1 of N were applied. RESUMO: O objetivo deste trabalho foi identificar as estirpes de Rhizobium mais efetivas e competitivas, a fim de maximizar a produtividade do feijoeiro por meio da fixação de nitrogênio, como alternativa à adubação nitrogenada. Foram conduzidos testes de competitividade em condições axênicas, em vasos com solo do Cerrado e em três experimentos de campo, com estirpes de Rhizobium nativas, previamente selecionadas quanto à efetividade e à variabilidade genética. A identificação das estirpes nos nódulos foi efetuada por meio das técnicas de aglutinação e ensaio imunoabsorvente de ligação de enzimas (Elisa), em condições de casa de vegetação, e pela técnica de DNA polimórfico amplificado ao acaso (RAPD), em solo de Cerrado. A produtividade das plantas foi determinada pela produção de matéria seca, teor de N e produção de grãos (condições de campo). A estirpe nativa SLA 2.2 e a estirpe comercial CIAT 899 foram dominantes nos nódulos das plantas das parcelas mais produtivas, com índices de produtividade iguais ou superiores aos obtidos nos tratamentos em que foram aplicados 20 kg ha-1 de N

    High potential for weathering and climate effects of non-vascular vegetation in the Late Ordovician

    It has been hypothesized that predecessors of today’s bryophytes significantly increased global chemical weathering in the Late Ordovician, thus reducing atmospheric CO2 concentration and contributing to climate cooling and an interval of glaciations. Studies that try to quantify the enhancement of weathering by non-vascular vegetation, however, are usually limited to small areas and low numbers of species, which hampers extrapolating to the global scale and to past climatic conditions. Here we present a spatially explicit modelling approach to simulate global weathering by non-vascular vegetation in the Late Ordovician. We estimate a potential global weathering flux of 2.8 (km3 rock) yr−1, defined here as volume of primary minerals affected by chemical transformation. This is around three times larger than today’s global chemical weathering flux. Moreover, we find that simulated weathering is highly sensitive to atmospheric CO2 concentration. This implies a strong negative feedback between weathering by non-vascular vegetation and Ordovician climate

    Na recuperacao e manutencao da fertilidade dos solos degradados, a decomposicao apresenta-se particularmente importante sendo a razao celulose/N um dos principais fatores que influenciam a efetividade desses processo. Neste trabalho avaliou-se a ocorrencia e a dinamica da populacao de microorganismos celuloliticos nos solos de diferentes modelos de reflorestamento de uma area degradada do Projeto Jaiba/MG. Esses modelos incluiram especies arboreas de eucalipto, leguminosas e nao leguminosas nativas. Observaram-se diferencas significativas no numero de celuloliticos entre as estacoes e os locais estudados. Nas areas impactadas ocorreu elevacao menos intensa dos microrganismos evidenciando o efeito limitante do impacto sobre essa populacao microbiana. Nos modelos de plantio avaliados o numero de celuloliticos diferiu significativamente, sendo o mais elevado no modelo representado pelo plantio do maior numero de especies. em todos os experimentos, a maior populacao de celuloliticos ocorreu nos consorcios entre leguminosas e outras especies vegetais nativas e a menor nas areas de plantio de eucalipto, sugerindo uma acao limitante dessa planta sobre o crescimento desses microrganismos. Esses resultados evidenciaram o papel dos celuloliticos como bioindicadores da qualidade do solo e da resposta as diferentes praticas de manejo

    Diverse soil carbon dynamics expressed at the molecular level

    The stability and potential vulnerability of soil organic matter (SOM) to global change remains incompletely understood due to the complex processes involved in its formation and turnover. Here we combine compound-specific radiocarbon analysis with fraction-specific and bulk-level radiocarbon measurements in order to further elucidate controls on SOM dynamics in a temperate and sub-alpine forested ecosystem. Radiocarbon contents of individual organic compounds isolated from the same soil interval generally exhibit greater variation than those among corresponding operationally-defined fractions. Notably, markedly older ages of long-chain plant leaf wax lipids (n-alkanoic acids) imply that they reflect a highly stable carbon pool. Furthermore, marked 14C variations among shorter- and longer-chain n-alkanoic acid homologues suggest that they track different SOM pools. Extremes in SOM dynamics thus manifest themselves within a single compound class. This exploratory study highlights the potential of compound-specific radiocarbon analysis for understanding SOM dynamics in ecosystems potentially vulnerable to global change

    Improving land surface models with FLUXNET data

    There is a growing consensus that land surface models (LSMs) that simulate terrestrial biosphere exchanges of matter and energy must be better constrained with data to quantify and address their uncertainties. FLUXNET, an international network of sites that measure the land surface exchanges of carbon, water and energy using the eddy covariance technique, is a prime source of data for model improvement. Here we outline a multi-stage process for "fusing" (i.e. linking) LSMs with FLUXNET data to generate better models with quantifiable uncertainty. First, we describe FLUXNET data availability, and its random and systematic biases. We then introduce methods for assessing LSM model runs against FLUXNET observations in temporal and spatial domains. These assessments are a prelude to more formal model-data fusion (MDF). MDF links model to data, based on error weightings. In theory, MDF produces optimal analyses of the modelled system, but there are practical problems. We first discuss how to set model errors and initial conditions. In both cases incorrect assumptions will affect the outcome of the MDF. We then review the problem of equifinality, whereby multiple combinations of parameters can produce similar model output. Fusing multiple independent and orthogonal data provides a means to limit equifinality. We then show how parameter probability density functions (PDFs) from MDF can be used to interpret model validity, and to propagate errors into model outputs. Posterior parameter distributions are a useful way to assess the success of MDF, combined with a determination of whether model residuals are Gaussian. If the MDF scheme provides evidence for temporal variation in parameters, then that is indicative of a critical missing dynamic process. A comparison of parameter PDFs generated with the same model from multiple FLUXNET sites can provide insights into the concept and validity of plant functional types (PFT) – we would expect similar parameter estimates among sites sharing a single PFT. We conclude by identifying five major model-data fusion challenges for the FLUXNET and LSM communities: (1) to determine appropriate use of current data and to explore the information gained in using longer time series; (2) to avoid confounding effects of missing process representation on parameter estimation; (3) to assimilate more data types, including those from earth observation; (4) to fully quantify uncertainties arising from data bias, model structure, and initial conditions problems; and (5) to carefully test current model concepts (e.g. PFTs) and guide development of new concepts

    Using an in-vitro biofilm model to assess the virulence potential of Bacterial Vaginosis or non-Bacterial Vaginosis Gardnerella vaginalis isolates

    Gardnerella vaginalis is the most common species found in bacterial vaginosis (BV). However, it is also present in a significant proportion of healthy women and G. vaginalis vaginal colonization does not always lead to BV. In an effort to better understand the differences between G. vaginalis isolated from women with a positive (BV) versus a negative (non-BV) diagnosis of BV, we compared the virulence potential of 7 BV and 7 non-BV G. vaginalis isolates and assessed the virulence factors related to biofilm formation, namely: initial adhesion and cytotoxic effect, biofilm accumulation, susceptibility to antibiotics, and transcript levels of the known vaginolysin, and sialidase genes. Furthermore, we also determined the ability of G. vaginalis to displace lactobacilli previously adhered to HeLa cells. Our results showed that non-BV strains were less virulent than BV strains, as suggested by the lower cytotoxicity and initial adhesion to Hela cells. Significant differences in expression of known virulence genes were also detected, further suggesting a higher virulence potential of the BV associated G. vaginalis. Importantly, we demonstrated that BV associated G. vaginalis were able to displace pre-coated vaginal protective lactobacilli and we hypothesize this to be a trigger for BV development.European Union funds (FEDER/COMPETE) and by national funds (FCT) under the project with reference FCOMP-01-0124-FEDER-008991 (PTDC/BIA-MIC/098228/2008).
