    Évaluation de la performance de pratiques de gestion optimales installĂ©es en sĂ©rie : le cas du marchĂ© public de Longueuil

    Cette Ă©tude de cas menĂ©e au MarchĂ© public de Longueuil avait pour objectif d’évaluer la performance hydraulique et l’efficacitĂ© Ă©puratoire de PGOs installĂ©s en sĂ©rie dans un climat nordique comme au QuĂ©bec. Pour ce faire, un protocole de suivi quantitatif et qualitatif a Ă©tĂ© dĂ©veloppĂ© pour Ă©tudier deux chaĂźnes de traitement: une chaĂźne constituĂ©e d’une tranchĂ©e drainante, d’un bassin sec et d’un bassin avec retenue permanente et une seconde constituĂ©e de biorĂ©tentions et d’un bassin avec retenue permanente. Le suivi quantitatif des eaux pluviales a dĂ©montrĂ© l’efficacitĂ© des PGOs Ă  retenir les eaux de ruissellement. Les biorĂ©tentions permettent de rĂ©duire, en moyenne, les volumes et les dĂ©bits de pointe de 70 % et 94 % respectivement. Les rĂ©sultats ont Ă©galement dĂ©montrĂ© que la tranchĂ©e drainante, le bassin sec et le bassin avec retenue permanente permettent, en moyenne, un dĂ©calage des dĂ©bits respectifs de 30 minutes, 10 minutes et d’un peu plus d’une heure. CombinĂ©es, les PGOs permettent le dĂ©calage des dĂ©bits de prĂšs de deux heures, ce qui est souhaitĂ©. Les campagnes d’échantillonnage ont montrĂ© des rĂ©sultats favorables pour l’enlĂšvement des matiĂšres en suspension et des matiĂšres organiques au niveau de la tranchĂ©e drainante et des cellules de biorĂ©tention. En effet, des taux moyens d’enlĂšvement de matiĂšres en suspension de 79 % et 84 % ont respectivement Ă©tĂ© obtenus pour ces deux PGOs. Toutefois, pour les cellules de biorĂ©tention un relargage de phosphore a, Ă  l’occasion, Ă©tĂ© observĂ©. Il s’est avĂ©rĂ© que le bassin sec relargue Ă©galement le phosphore, mais aussi des matiĂšres en suspension. Par contre, l’ajout du bassin Ă  retenue permanente Ă  l’aval permet de contrĂŽler ce relargage en effectuant un traitement supplĂ©mentaire. L’amĂ©nagement d’une PGO ayant pour fonction de traiter les eaux est donc un choix judicieux pour pallier le relĂąchement des contaminants de celle Ă  l’amont. À la lumiĂšre des de ces rĂ©sultats, l’emploi de chaĂźnes de traitement est donc avantageux comparativement Ă  l’utilisation de PGOs seules

    Association between Abdominal Fat (DXA) and Its Subcomponents (CT Scan) before and after Weight Loss in Obese Postmenopausal Women: A MONET Study

    Introduction. Subcutaneous fat (ScF) and visceral fat (VF) measurements using CT scan are expensive and may imply significant radiation doses. Cross-sectional studies using CT scan showed that ScF and VF are significantly correlated with abdominal fat measured by DXA (AF-DXA). The association has not been studied after a weight loss. Objective. To determine (1) the associations between AF-DXA and ScF and VF before and after weight loss and (2) the associations between their changes. Methods. 137 overweight/obese postmenopausal women were divided in two groups (1-caloric restriction or 2-caloric restriction + resistance training). AF was assessed using DXA and CT scan. Results. Correlations between AF-DXA and ScF (before: r = 0.87, after; r = 0.87; P < .01) and, AF-DXA and VF (before: r = 0.61, after; r = 0.69; P < .01) are not different before and after the weight loss. Correlations between delta AF-DXA and delta ScF (r = 0.72; P < .01) or delta VF (r = 0.51; P < .01) were found. Conclusion. The use of AF-DXA as a surrogate for VF after weight loss is questionable, but may be interesting for ScF

    Cell wall biochemical alterations during Agrobacterium -mediated expression of haemagglutinin-based influenza virus-like vaccine particles in tobacco

    International audienceInfluenza virus‐like particles (VLPs) have been shown to induce a safe and potent immune response through both humoral and cellular responses. They represent promising novel influenza vaccines. Plant‐based biotechnology allows for the large‐scale production of VLPs of biopharmaceutical interest using different model organisms, including Nicotiana benthamiana plants. Through this platform, influenza VLPs bud from the plasma membrane and accumulate between the membrane and the plant cell wall. To design and optimize efficient production processes, a better understanding of the plant cell wall composition of infiltrated tobacco leaves is a major interest for the plant biotechnology industry. In this study, we have investigated the alteration of the biochemical composition of the cell walls of N. benthamiana leaves subjected to abiotic and biotic stresses induced by the Agrobacterium‐mediated transient transformation and the resulting high expression levels of influenza VLPs. Results show that abiotic stress due to vacuum infiltration without Agrobacterium did not induce any detectable modification of the leaf cell wall when compared to non infiltrated leaves. In contrast, various chemical changes of the leaf cell wall were observed post‐Agrobacterium infiltration. Indeed, Agrobacterium infection induced deposition of callose and lignin, modified the pectin methylesterification and increased both arabinosylation of RG‐I side chains and the expression of arabinogalactan proteins. Moreover, these modifications were slightly greater in plants expressing haemagglutinin‐based VLP than in plants infiltrated with the Agrobacterium strain containing only the p19 suppressor of silencing

    Chronic disease patients’ experience with telehealth interventions and self-care strategies during the first wave of the COVID-19 pandemic

    Purpose. During COVID-19 pandemic, Québec (Canada) Public Health Agency asked Family Medicine Group (FMG) to change their practice such as recommending the use of telehealth interventions. However, it is unknown if telehealth meets patients' needs in terms of self-care, especially those suffered from physical and mental chronic condition (PCDs). This study aimed to explore, from a PCD's perspective, the response of FMG about their self-care needs during COVID-19 pandemic. Methods. We conducted an exploratory qualitative descriptive study using semi-structured interviews to reach 40 PCDs from three regions of Québec. We explored FMGs' responses, PCDs needs, coping strategies regarding the COVID-19 and the changes in primary care services. We used the transactional theory of stress and coping to inform the data collection and analysis and the COREQ appraisal grid to report this study. Results. PCDs appreciated telehealth and perceived that the accessibility was increased. Moreover, family physicians from FMG conducted the majority of the follow-ups although interdisciplinary teams were available. The coping strategies raised by the PCDs were primarily aimed at maintaining their health status, while some of them aimed to reduce stress levels. Conclusions. PCDs seem to appreciate telehealth but believe it requires improvements, and it is not adequate for all types of follow-ups. PCDs adopt coping strategies to face the pandemic that were not always optimal for their health.     Field Longevity of a Fluorescent Protein Marker in an Engineered Strain of the Pink Bollworm, Pectinophora gossypiella (Saunders)

    The cotton pest, pink bollworm (Pectinophora gossypiella (Saunders)), is a significant pest in most cotton-growing areas around the world. In southwestern USA and northern Mexico, pink bollworm is the target of the sterile insect technique (SIT), which relies on the mass-release of sterile pink bollworm adults to over-flood the wild population and thereby reduce it over time. Sterile moths reared for release are currently marked with a dye provided in their larval diet. There are concerns, however, that this marker fails from time to time, leading to sterile moths being misidentified in monitoring traps as wild moths. This can lead to expensive reactionary releases of sterile moths. We have developed a genetically marked strain that is engineered to express a fluorescent protein, DsRed2, which is easily screened under a specialised microscope. In order to test this marker under field conditions, we placed wild-type and genetically marked moths on traps and placed them in field cages. The moths were then screened, in a double-blind fashion, for DsRed2 fluorescence at regular intervals to determine marker reliability over time. The marker was shown to be robust in very high temperatures and generally proved reliable for a week or longer. More importantly, genotyping of moths on traps by PCR screening of the moths was 100% correct. Our findings indicate that this strain - and fluorescent protein markers in general - could make a valuable contribution to SIT

    Changes in Culture Expanded Human Amniotic Epithelial Cells: Implications for Potential Therapeutic Applications

    Human amniotic epithelial cells (hAEC) isolated from term placenta have stem cell-like properties, differentiate into tissue specific cells and reduce lung and liver inflammation and fibrosis following transplantation into disease models established in mice. These features together with their low immunogenicity and immunosuppressive properties make hAEC an attractive source of cells for potential therapeutic applications. However, generation of large cell numbers required for therapies through serial expansion in xenobiotic-free media may be a limiting factor. We investigated if hAEC could be expanded in xenobiotic-free media and if expansion altered their differentiation capacity, immunophenotype, immunosuppressive properties and production of immunomodulatory factors. Serial expansion in xenobiotic-free media was limited with cumulative cell numbers and population doubling times significantly lower than controls maintained in fetal calf serum. The epithelial morphology of primary hAEC changed into mesenchymal-stromal like cells by passage 4–5 (P4–P5) with down regulation of epithelial markers CK7, CD49f, EpCAM and E-cadherin and elevation of mesenchymal-stromal markers CD44, CD105, CD146 and vimentin. The P5 hAEC expanded in xenobiotic-free medium differentiated into osteocyte and alveolar epithelium-like cells, but not chondrocyte, hepatocyte, α- and ÎČ-pancreatic-like cells. Expression of HLA Class IA, Class II and co-stimulatory molecules CD80, CD86 and CD40 remained unaltered. The P5 hAEC suppressed mitogen stimulated T cell proliferation, but were less suppressive compared with primary hAEC at higher splenocyte ratios. Primary and P5 hAEC did not secrete the immunosuppressive factors IL-10 and HGF, whereas TGF-ÎČ1 and HLA-G were reduced and IL-6 elevated in P5 hAEC. These findings suggest that primary and expanded hAEC may be suitable for different cellular therapeutic applications
