Sp8 exhibits reciprocal induction with Fgf8 but has an opposing effect on anterior-posterior cortical area patterning
Telencephalic patterning centers, defined by the discrete expression domains of distinct morphogens, Fgfs in the commissural plate (CoP), Wnts and Bmps in the cortical hem, and a ventral domain of Sonic hedgehog (Shh), are postulated to establish during development the initial patterning of the telencepahlon, including the neocortex. We show that the expression patterns of Sp5, Sp8, and Sp9, members of the Sp8-like family that are homologues of Drosophila buttonhead, correlate during early embryonic development with these three telencephalic patterning centers. To study potential functional relationships, we focused on Sp8, because it is transiently expressed in the CoP coincident with the expression of Fgf8, a morphogen implicated in area patterning of the neocortex. We also show that Sp8 is expressed in cortical progenitors in a high to low anterior-medial to posterior-lateral gradient across the ventricular zone. We used in utero electroporation of full-length and chimeric expression constructs to perform gain-of-function and loss-of-function studies of interactions between Sp8 and Fgf8 and their roles in cortical area patterning. We show that Fgf8 and Sp8 exhibit reciprocal induction in vivo in the embryonic telencephalon. Sp8 also induces downstream targets of Fgf8, including ETS transcription factors. In vitro assays show that Sp8 binds Fgf8 regulatory elements and is a direct transcriptional activator of Fgf8. We also show that Sp8 induction of Fgf8 is repressed by Emx2 in vitro, suggesting a mechanism to limit Fgf8 expression to the CoP. In vivo expression of a dominant negative Sp8 in the CoP indicates that Sp8 maintains expression of Fgf8 and also its effect on area patterning. Ectopic expression of Sp8 in anterior or posterior cortical poles induces significant anterior or posterior shifts in area patterning, respectively, paralleled by changes in expression of gene markers of positional identity. These effects of Sp8 on area patterning oppose those induced by ectopic expression of Fgf8, suggesting that in parallel to regulating Fgf8 expression, Sp8 also activates a distinct signaling pathway for cortical area patterning. In summary, Sp8 and Fgf8 robustly induce one another, and may act to balance the anterior-posterior area patterning of the cortex

Neuregulin repellent signaling via ErbB4 restricts GABAergic interneurons to migratory paths from ganglionic eminence to cortical destinations
<p>Abstract</p> <p>Background</p> <p>Cortical GABAergic interneurons (INs) are generated in the medial ganglionic eminence (MGE) and migrate tangentially into cortex. Because most, if not all, migrating MGE-derived INs express the neuregulin (NRG) receptor, ErbB4, we investigated influences of Nrg1 isoforms and Nrg3 on IN migration through ventral telencephalon (vTel) and within cortex.</p> <p>Results</p> <p>During IN migration, NRG expression domains and distributions of ErbB4-expressing, MGE-derived INs are complementary with minimal overlap, both in vTel and cortex. In wild-type mice, within fields of NRG expression, these INs are focused at positions of low or absent NRG expression. However, in ErbB4-/- HER4<sup>heart </sup>mutant mice in which INs lack ErbB4, these complementary patterns are degraded with considerable overlap evident between IN distribution and NRG expression domains. These findings suggest that NRGs are repellents for migrating ErbB4-expressing INs, a function supported by <it>in vitro </it>and <it>in vivo </it>experiments. First, in collagen co-cultures, MGE-derived cells preferentially migrate away from a source of secreted NRGs. Second, cells migrating from wild-type MGE explants on living forebrain slices from wild-type embryonic mice tend to avoid endogenous NRG expression domains, whereas this avoidance behavior is not exhibited by ErbB4-deficient cells migrating from MGE explants and instead they have a radial pattern with a more uniform distribution. Third, ectopic NRG expression in the IN migration pathway produced by <it>in utero </it>electroporation blocks IN migration and results in cortex distal to the blockade being largely devoid of INs. Finally, fewer INs reach cortex in ErbB4 mutants, indicating that NRG-ErbB4 signaling is required for directing IN migration from the MGE to cortex.</p> <p>Conclusions</p> <p>Our results show that NRGs act as repellents for migrating ErbB4-expressing, MGE-derived GABAergic INs and that the patterned expression of NRGs funnels INs as they migrate from the MGE to their cortical destinations.</p
Sticking under wet conditions: the remarkable attachment abilities of the torrent frog, staurois guttatus
Tree frogs climb smooth surfaces utilising capillary forces arising from an air-fluid interface around their toe pads, whereas torrent frogs are able to climb in wet environments near waterfalls where the integrity of the meniscus is at risk. This study compares the adhesive capabilities of a torrent frog to a tree frog, investigating possible adaptations for adhesion under wet conditions. We challenged both frog species to cling to a platform which could be tilted from the horizontal to an upside-down orientation, testing the frogs on different levels of roughness and water flow. On dry, smooth surfaces, both frog species stayed attached to overhanging slopes equally well. In contrast, under both low and high flow rate conditions, the torrent frogs performed significantly better, even adhering under conditions where their toe pads were submerged in water, abolishing the meniscus that underlies capillarity. Using a transparent platform where areas of contact are illuminated, we measured the contact area of frogs during platform rotation under dry conditions. Both frog species not only used the contact area of their pads to adhere, but also large parts of their belly and thigh skin. In the tree frogs, the belly and thighs often detached on steeper slopes, whereas the torrent frogs increased the use of these areas as the slope angle increased. Probing small areas of the different skin parts with a force transducer revealed that forces declined significantly in wet conditions, with only minor differences between the frog species. The superior abilities of the torrent frogs were thus due to the large contact area they used on steep, overhanging surfaces. Decoupling heavy sparticles in Effective SUSY scenarios: Unification, Higgs masses and tachyon bounds
Using two-loop renormalization group equations implementing the decoupling of
heavy scalars, Effective SUSY scenarios are studied in the limit in which there
is a single low energy Higgs field. Gauge coupling unification is shown to hold
with similar or better precision than in standard MSSM scenarios. b-tau
unification is examined, and Higgs masses are computed using the effective
potential, including two-loop contributions from scalars. A 125 GeV Higgs is
compatible with stops/sbottoms at around 300 GeV with non-universal boundary
conditions at the scale of the heavy sparticles if some of the trilinear
couplings at this scale take values of the order of 1-2 TeV; if more
constrained boundary conditions inspired by msugra or gauge mediation are set
at a higher scale, heavier colored sparticles are required in general. Finally,
since the decoupled RG flow for third-generation scalar masses departs very
significantly from the MSSM DR-bar one, tachyon bounds for light scalars are
revisited and shown to be relaxed by up to a TeV or more.Comment: 35 pages, 17 figures. v2: Updated some scans, allowing for changes in

Further evidence for the involvement of EFL1 in a Shwachman-Diamond-like syndrome and expansion of the phenotypic features.
Recent evidence has implicated EFL1 in a phenotype overlapping Shwachman-Diamond syndrome (SDS), with the functional interplay between EFL1 and the previously known causative gene SBDS accounting for the similarity in clinical features. Relatively little is known about the phenotypes associated with pathogenic variants in the EFL1 gene, but the initial indication was that phenotypes may be more severe, when compared with SDS. We report a pediatric patient who presented with a metaphyseal dysplasia and was found to have biallelic variants in EFL1 on reanalysis of trio whole-exome sequencing data. The variant had not been initially reported because of the research laboratory's focus on de novo variants. Subsequent phenotyping revealed variability in her manifestations. Although her metaphyseal abnormalities were more severe than in the original reported cohort with EFL1 variants, the bone marrow abnormalities were generally mild, and there was equivocal evidence for pancreatic insufficiency. Despite the limited number of reported patients, variants in EFL1 appear to cause a broader spectrum of symptoms that overlap with those seen in SDS. Our report adds to the evidence of EFL1 being associated with an SDS-like phenotype and provides information adding to our understanding of the phenotypic variability of this disorder. Our report also highlights the value of exome data reanalysis when a diagnosis is not initially apparent
The Overall Coefficient of the Two-loop Superstring Amplitude Using Pure Spinors
Using the results recently obtained for computing integrals over
(non-minimal) pure spinor superspace, we compute the coefficient of the
massless two-loop four-point amplitude from first principles. Contrasting with
the mathematical difficulties in the RNS formalism where unknown normalizations
of chiral determinant formulae force the two-loop coefficient to be determined
only indirectly through factorization, the computation in the pure spinor

LHC Discovery Potential for Non-Standard Higgs Bosons in the 3b Channel
In a variety of well motivated models, such as two Higgs Doublet Models
(2HDMs) and the Minimal Supersymmetric Standard Model (MSSM), there are neutral
Higgs bosons that have significantly enhanced couplings to b-quarks and tau
leptons in comparison to those of the SM Higgs. These so called non-standard
Higgs bosons could be copiously produced at the LHC in association with b
quarks, and subsequently decay into b-quark pairs. However, this production
channel suffers from large irreducible QCD backgrounds. We propose a new search
strategy for non-standard neutral Higgs bosons at the 7 TeV LHC in the 3b's
final state topology. We perform a simulation of the signal and backgrounds,
using state of the art tools and methods for different sets of selection cuts,
and conclude that neutral Higgs bosons with couplings to b-quarks of about 0.3
or larger, and masses up to 400 GeV, could be seen with a luminosity of 30
fb^{-1}. In the case of the MSSM we also discuss the complementarity between
the 3b channel and the inclusive tau pair channel in exploring the

A 125 GeV SM-like Higgs in the MSSM and the rate
We consider the possibility of a Standard Model (SM)-like Higgs in the
context of the Minimal Supersymmetric Standard Model (MSSM), with a mass of
about 125 GeV and with a production times decay rate into two photons which is
similar or somewhat larger than the SM one. The relatively large value of the
SM-like Higgs mass demands stops in the several hundred GeV mass range with
somewhat large mixing, or a large hierarchy between the two stop masses in the
case that one of the two stops is light. We find that, in general, if the
heaviest stop mass is smaller than a few TeV, the rate of gluon fusion
production of Higgs bosons decaying into two photons tends to be somewhat
suppressed with respect to the SM one in this region of parameters. However, we
show that an enhancement of the photon decay rate may be obtained for light
third generation sleptons with large mixing, which can be naturally obtained
for large values of and sizable values of the Higgsino mass

Comparison of Short-Term Estrogenicity Tests for Identification of Hormone-Disrupting Chemicals
The aim of this study was to compare results obtained by eight different short-term assays of estrogenlike actions of chemicals conducted in 10 different laboratories in five countries. Twenty chemicals were selected to represent direct-acting estrogens, compounds with estrogenic metabolites, estrogenic antagonists, and a known cytotoxic agent. Also included in the test panel were 17β-estradiol as a positive control and ethanol as solvent control. The test compounds were coded before distribution. Test methods included direct binding to the estrogen receptor (ER), proliferation of MCF-7 cells, transient reporter gene expression in MCF-7 cells, reporter gene expression in yeast strains stably transfected with the human ER and an estrogen-responsive reporter gene, and vitellogenin production in juvenile rainbow trout. 17β-Estradiol, 17α-ethynyl estradiol, and diethylstilbestrol induced a strong estrogenic response in all test systems. Colchicine caused cytotoxicity only. Bisphenol A induced an estrogenic response in all assays. The results obtained for the remaining test compounds—tamoxifen, ICI 182.780, testosterone, bisphenol A dimethacrylate, 4-n-octylphenol, 4-n-nonylphenol, nonylphenol dodecylethoxylate, butylbenzylphthalate, dibutylphthalate, methoxychlor, o,p′-DDT, p,p′-DDE, endosulfan, chlomequat chloride, and ethanol—varied among the assays. The results demonstrate that careful standardization is necessary to obtain a reasonable degree of reproducibility. Also, similar methods vary in their sensitivity to estrogenic compounds. Thus, short-term tests are useful for screening purposes, but the methods must be further validated by additional interlaboratory and interassay comparisons to document the reliability of the methods
