    Arbitrary order 2D virtual elements for polygonal meshes: Part II, inelastic problem

    The present paper is the second part of a twofold work, whose first part is reported in [3], concerning a newly developed Virtual Element Method (VEM) for 2D continuum problems. The first part of the work proposed a study for linear elastic problem. The aim of this part is to explore the features of the VEM formulation when material nonlinearity is considered, showing that the accuracy and easiness of implementation discovered in the analysis inherent to the first part of the work are still retained. Three different nonlinear constitutive laws are considered in the VEM formulation. In particular, the generalized viscoplastic model, the classical Mises plasticity with isotropic/kinematic hardening and a shape memory alloy (SMA) constitutive law are implemented. The versatility with respect to all the considered nonlinear material constitutive laws is demonstrated through several numerical examples, also remarking that the proposed 2D VEM formulation can be straightforwardly implemented as in a standard nonlinear structural finite element method (FEM) framework

    Il ruolo della costruzione di scenari nella spiegazione dei disturbi macrolinguistici della schizofrenia

    Riassunto: Al centro di questo articolo è l’analisi dei fondamenti neurocognitivi dei deficit macrolinguistici, discorsivo-narrativi nello specifico, osservabili nei pazienti affetti da schizofrenia. Tale analisi è condotta in riferimento alle ricerche svolte negli ultimi decenni nell’ambito della psicolinguistica e delle neuroscienze. I risultati di queste ricerche hanno mostrato che alcuni aspetti dei disturbi macroelaborativi che caratterizzano il profilo linguistico dei pazienti schizofrenici possono essere spiegati chiamando in causa i deficit di tre sistemi cognitivi: teoria della mente, funzioni esecutive, memoria semantica. In questo articolo si sostiene che un tale quadro esplicativo vada integrato attraverso il riferimento a un ulteriore processo cognitivo cruciale per l’elaborazione macrolinguistica: la costruzione di scenari. Tale processo è fondamentale per la comprensione e produzione discorsiva in quanto responsabile della costruzione della rappresentazione globale degli eventi del discorso narrativo. Alla luce di queste considerazioni, in questo lavoro si propone che un ruolo cruciale nella spiegazione di alcuni aspetti dei disturbi macrolinguistici nella schizofrenia sia svolto da una compromissione nel processo di costruzione di scenari mentali e si avanza l’ipotesi che tale compromissione sia determinata, a sua volta, da problematiche che agiscono sul piano del funzionamento delle aree cerebrali preposte alla percezione e all’assemblaggio multimodale delle rappresentazioni sensomotorie.Parole chiave: Abilità macrolinguistiche; Costruzione di scenari; Elaborazione discorsiva: Embodied Cognition; Narrazione; Schizofrenia; Percezione; Modelli mentaliThe role scenario building in the explanation of macrolinguistic disorders in schizophreniaAbstract: This article analyzes the neurocognitive foundations of macrolinguistic deficits, specifically narrative, observable in patients with schizophrenia. This analysis is conducted with reference to the investigations carried out in recent decades in the field of psycholinguistics and neuroscience. The results of these investigations showed that some aspects of macrolinguistic deficits of schizophrenics can be explained in terms of impairments of three cognitive systems: theory of mind, executive functions, and semantic memory. In this article, it is suggested that this explanatory framework has to be integrated with the reference to a further cognitive process: scenarios construction. Such process turns out to be critical for discourse comprehension and production as it is responsible for the building of the global representation of events of a narrative. In the light of these considerations, it is claimed that a crucial role in the explanation of some aspects of macrolinguistic disorders in schizophrenia is played by an impairment in the process of constructing mental scenarios.     222Rn daughters influence on scaler mode of the ARGO-YBJ detector

    The ARGO-YBJ experiment is a full coverage air shower array; its lowest energy threshold is reached using the "scaler mode technique". Working in this mode, the signals generated by any particle hitting each cluster are put in coincidence every 150 ns and read by four independent scaler channels, giving the counting rates of multiplicity \geq1, \geq2, \geq3 and \geq4 (C1, C2, C3 and C4, respectively). The study of these counting rates pointed out a different behaviour of C1 respect to C2, C3 and C4, suggesting that C1 is detecting not only cosmic rays. This work shows that the radon (222Rn) gamma emitter daughters present in the ARGO-YBJ building air are contributing to C1 counts at the level of 1 Hz each Bq/m3 of radon. The uncertainty about this contribution is great, because of the high variability of 222Rn concentration and the building ventilation. The radon monitoring will allow the C1 correction improving the sensitivity of the ARGO-YBJ experiment at its lowest energy threshold.Comment: 4 pages, 3 figures, Proceedings of the 32nd International Cosmic Ray Conference (ICRC

    Road traffic crashes are a main cause of physical harm and mortality. That means a heavy burden in terms of short, medium and long-term disability and public costs for society. Among the most relevant cases of severe accidents is driving under the influence of alcohol and psychoactive drugs. That\u2019s the reason of the international strong effort to combating the phenomenon. In Italy the draft law concerning the criminal offence of vehicular homicide and road traffic personal injuries is currently being discussed. Previously the law n. 120/2010 has already changed the regulation in matter of drink-driving and driving under the influence of controlled substances with stiffer penalties for transgressors and (at least some) guidance as to assess the infringement. The draft law suggests that clinical and toxicological evidences of actual driving impairment has to be proved to reach significance in court, nevertheless no standard and effective procedure has being established for detection, at least for what concerns controlled substances. The purpose of this article is to put in evidence, in accordance with a medico-legal perspective, some of the issues that emerge from this complex matter (i.e. the need of a list of the impairing substance under detection, of well-defined legal cut-off values, of a better definition of the problems following the therapeutic use of drugs of abuse and other concerns), issues, those, that are now more troubling whereas the introduction of new crimes and punishments is becoming more concrete

    Non-Alcoholic Fatty Liver Disease: From Pathogenesis to Clinical Impact

    Non-Alcoholic Fatty Liver Disease (NAFLD) is caused by the accumulation of fat in over 5% of hepatocytes in the absence of alcohol consumption. NAFLD is considered the hepatic manifestation of metabolic syndrome (MS). Recently, an expert consensus suggested as more appropriate the term MAFLD (metabolic-associated fatty liver disease). Insulin resistance (IR) plays a key role in the development of NAFLD, as it causes an increase in hepatic lipogenesis and an inhibition of adipose tissue lipolysis. Beyond the imbalance of adipokine levels, the increase in the mass of visceral adipose tissue also determines an increase in free fatty acid (FFA) levels. In turn, an excess of FFA is able to determine IR through the inhibition of the post-receptor insulin signal. Adipocytes secrete chemokines, which are able to enroll macrophages inside the adipose tissue, responsible, in turn, for the increased levels of TNF-. The latter, as well as resistin and other pro-inflammatory cytokines such as IL-6, enhances insulin resistance and correlates with endothelial dysfunction and an increased cardiovascular (CV) risk. In this review, the role of diet, intestinal microbiota, genetic and epigenetic factors, low-degree chronic systemic inflammation, mitochondrial dysfunction, and endoplasmic reticulum stress on NAFLD have been addressed. Finally, the clinical impact of NAFLD on cardiovascular and renal outcomes, and its direct link with type 2 diabetes have been discussed

    The Candoglia Marble and the “Veneranda Fabbrica del Duomo di Milano”: A Renowned Georesource to Be Potentially Designed as Global Heritage Stone

    Marbles from Alpine area have been widely employed to build and decorate masterpieces and buildings which often represent the cultural heritage of an area (statuary, historic buildings and sculptures). Candoglia marble, object of the present research, is one of the most famous and appreciated marbles from Alpine area; it has been quarried since Roman times in the Verbano-Cusio-Ossola (VCO; Piemonte—NW Italy) extractive area. Candoglia Marble outcrops are present as lenses within the high-grade paragneisses of the Ivrea Zone, a visible section of deep continental crust characterised by amphibolite- to granulite-facies metamorphism (Palaeozoic period). Candoglia calcitic marble (80–85% CaCO3 and the 15–20% other minerals) shows a characteristic pink to gray colour and a coarse-grained texture (>3 mm): frequent centimetre-thick dark-greenish silicate layers (mainly represented by diopside and tremolite) characterize the texture of the marble. It has been largely used in local rural constructions and historical buildings, but its most famous application has been (and still is) for the “Duomo di Milano” construction (fourteenth century). The Veneranda Fabbrica del Duomo di Milano carried out the anthropogenic activities dealing with the Candoglia marble exploitation; it has to be highlighted that the company have managed the Marble exploitation during the last seven centuries and that the quarry itself is a tangible sign of the development of extraction and heritage in the VCO area. Candoglia marble can be recognized as a significant example of a “Global Heritage Stone Resource”: its exploitation from quarry to building (the Duomo di Milano) well represents the close correlation between stone and cultural heritage, between georesources and humankind development

    Long-Term Safety of Antifibrotic Drugs in IPF: A Real-World Experience

    Pirfenidone and nintedanib are the only two drugs approved for the treatment of idiopathic pulmonary fibrosis (IPF). Both proved to be safe and well-tolerated in clinical trials, but real-world data and direct comparisons are scarce. This real-life study explored the safety profile of pirfenidone and nintedanib with a prolonged follow-up. We retrospectively collected clinical status, adverse events (AEs), and treatment changes from IPF patients who had started an antifibrotic treatment at our centre from December 2011 to December 2020, including 192 patients treated with pirfenidone and 89 with nintedanib. The majority of patients in both groups experienced one or more AEs during the follow-up. A higher proportion of AEs in the nintedanib group were effectively treated with behavioural modifications or additional medications compared with the pirfenidone group (52.5% vs. 40.6%, p = 0.04). Overall, a difference in the impact of AEs due to nintedanib versus pirfenidone resulted in a lower permanent discontinuation of therapy (8.3% vs. 18.3%, p = 0.02), with the latter being associated with a higher risk of drug discontinuation at 48 months after initiation (OR = 2.52, p = 0.03). Our study confirms the safety profile of antifibrotic drugs in IPF but highlights that AEs due to nintedanib are usually easier to manage and lead to fewer cases of permanent discontinuation of therapy

    The adherent/invasive escherichia coli (AIEC) strain LF82 invades and persists in human prostate cell lineRWPE-1 activating a strong inflammatory response

    Adherent/invasive Escherichia coli (AIEC) strains have recently been receiving increased attention because they are more prevalent and persistent in the intestine of Crohn's disease (CD) patients than in healthy subjects. Since AIEC strains show a high percentage of similarity to extraintestinal pathogenic E. coli (ExPEC), neonatal meningitis-associated E. coli (NMEC), and uropathogenic E. coli (UPEC) strains, here we compared AIEC strain LF82 with a UPEC isolate (strain EC73) to assess whether LF82 would be able to infect prostate cells as an extraintestinal target. The virulence phenotypes of both strains were determined by using the RWPE-1 prostate cell line. The results obtained indicated that LF82 and EC73 are able to adhere to, invade, and survive within prostate epithelial cells. Invasion was confirmed by immunofluorescence and electron microscopy. Moreover, cytochalasin D and colchicine strongly inhibited bacterial uptake of both strains, indicating the involvement of actin microfilaments and microtubules in host cell invasion. Moreover, both strains belong to phylogenetic group B2 and are strong biofilm producers. In silico analysis reveals that LF82 shares with UPEC strains several virulence factors: namely, type 1 pili, the group II capsule, the vacuolating autotransporter toxin, four iron uptake systems, and the pathogenic island (PAI). Furthermore, compared to EC73, LF82 induces in RWPE-1 cells a marked increase of phosphorylation of mitogen-activated protein kinases (MAPKs) and of NF-κB already by 5 min postinfection, thus inducing a strong inflammatory response.
