Exome-wide association study of pancreatic cancer risk
We conducted a case-control exome-wide association study to discover germline variants in coding regions that affect risk for pancreatic cancer, combining data from 5 studies. We analyzed exome and genome sequencing data from 437 patients with pancreatic cancer (cases) and 1922 individuals not known to have cancer (controls). In the primary analysis, BRCA2 had the strongest enrichment for rare inactivating variants (17/437 cases vs 3/1922 controls) (P=3.27x10(-6); exome-wide statistical significance threshold P<2.5x10(-6)). Cases had more rare inactivating variants in DNA repair genes than controls, even after excluding 13 genes known to predispose to pancreatic cancer (adjusted odds ratio, 1.35, P=.045). At the suggestive threshold (P<.001), 6 genes were enriched for rare damaging variants (UHMK1, AP1G2, DNTA, CHST6, FGFR3, and EPHA1) and 7 genes had associations with pancreatic cancer risk, based on the sequence-kernel association test. We confirmed variants in BRCA2 as the most common high-penetrant genetic factor associated with pancreatic cancer and we also identified candidate pancreatic cancer genes. Large collaborations and novel approaches are needed to overcome the genetic heterogeneity of pancreatic cancer predisposition
Institutional, social and individual behavioural effects of energy feedback in public buildings across eleven European cities

The Publisher's final version can be found by following the DOI link.Better understanding of the factors influencing how people use energy in public buildings can help deliver more effective CO2 reduction strategies. This paper describes the institutional, social and individual behavioural effects of communication campaigns in over 500 public buildings in 11 European cities. These campaigns involved engaging with staff to reduce energy use through feedback services based on information from sub hourly meter readings.
A summative evaluation was conducted to understand impacts of different information provision in these cities. Qualitative data were gathered through a set of interviews with 40 building professionals at the central or building level. These interviews identified differences in how the energy efficiency communication-based campaigns were implemented at each site and elicited factors to explain how users’ perceptions and understanding changed as a result of the interventions. The evaluation framework helped to identify not only improvements in the delivery of communication-based campaigns, but also the communication factors that impacted on individual behaviour change. The research highlighted the influence of institutional and social effects on individual beliefs and norms. To achieve more effective change in attitudes to reduce use, energy feedback needs to be supported with engagement activities, such as energy coaches, campaigns, and interactive online fora
From AKNS to derivative NLS hierarchies via deformations of associative products
Using deformations of associative products, derivative nonlinear Schrodinger
(DNLS) hierarchies are recovered as AKNS-type hierarchies. Since the latter can
also be formulated as Gelfand-Dickey-type Lax hierarchies, a recently developed
method to obtain 'functional representations' can be applied. We actually
consider hierarchies with dependent variables in any (possibly noncommutative)
associative algebra, e.g., an algebra of matrices of functions. This also

Weak-anchoring effects in a thin pinned ridge of nematic liquid crystal
A theoretical investigation of weak-anchoring effects in a thin
two-dimensional pinned static ridge of nematic liquid crystal resting on a flat
solid substrate in an atmosphere of passive gas is performed. Specifically, we
solve a reduced version of the general system of governing equations recently
derived by Cousins et al. [Proc. Roy. Soc. A}, 478(2259):20210849, 2022] valid
for a symmetric thin ridge under the one-constant approximation of the
Frank--Oseen bulk elastic energy with pinned contact lines to determine the
shape of the ridge and the behaviour of the director within it. Numerical
investigations covering a wide range of parameter values indicate that the
energetically-preferred solutions can be classified in terms of the
Jenkins--Barratt--Barbero--Barberi critical thickness into five qualitatively
different types of solution. In particular, the theoretical results suggest
that anchoring breaking occurs close to the contact lines. The theoretical
predictions are supported by the results of physical experiments for a ridge of
the nematic 4'-pentyl-4-biphenylcarbonitrile (5CB). In particular, these
experiments show that the homeotropic anchoring at the gas--nematic interface
is broken close to the contact lines by the stronger rubbed planar anchoring at
the nematic--substrate interface. A comparison between the experimental values
of and the theoretical predictions for the effective refractive index of the
ridge gives a first estimate of the anchoring strength of an interface between
air and 5CB to be at a temperature

Practitioner-informed improvements to early childhood intervention performance checklists and practice guides
Results from four early childhood practitioner field tests of performance checklists and early intervention practice guides are reported. Findings from the first field test were used to make changes and improvements in the checklists and practice guides evaluated in the second and third field tests, and findings from the latter two field tests were used to improve the checklist and practice guide evaluated in the fourth field test. Results indicated that changes made in response to practitioners’ suggestions and feedback were associated with (1) progressive increases in the practitioners’ social validity judgments of the checklists, practice guides, and checklist-practice guide correspondence, and (2) progressive decreases in the number of practitioner suggestions and feedback for improving the early intervention materials. The field-test research demonstrates the importance of practitioner input, suggestions, and feedback for improving the usefulness of early childhood intervention practices

The fibroblast mitogen platelet-derived growth factor -BB (PDGF-BB) induces a transient expression of the orphan nuclear receptor NR4A1 (also named Nur77, TR3 or NGFIB). The aim of the present study was to investigate the pathways through which NR4A1 is induced by PDGF-BB and its functional role. We demonstrate that in PDGF-BB stimulated NIH3T3 cells, the MEK1/2 inhibitor CI-1040 strongly represses NR4A1 expression, whereas Erk5 downregulation delays the expression, but does not block it. Moreover, we report that treatment with the NF-κB inhibitor BAY11-7082 suppresses NR4A1 mRNA and protein expression. The majority of NR4A1 in NIH3T3 was found to be localized in the cytoplasm and only a fraction was translocated to the nucleus after continued PDGF-BB treatment. Silencing NR4A1 slightly increased the proliferation rate of NIH3T3 cells; however, it did not affect the chemotactic or survival abilities conferred by PDGF-BB. Moreover, overexpression of NR4A1 promoted anchorage-independent growth of NIH3T3 cells and the glioblastoma cell lines U-105MG and U-251MG. Thus, whereas NR4A1, induced by PDGF-BB, suppresses cell growth on a solid surface, it increases anchorage-independent growth
