Ab initio circular dichroism with the Yambo code: applications to dipeptides

    Get PDF
Circular dichroism (CD) spectroscopy is a useful technique for characterizing chiral molecules. It is more sensitive than total absorption to molecule conformation, and it is routinely used to identify enantiomers. We present here a first principles implementation of CD with application to three cyclo-dipeptides. Our CD approach for molecules has been integrated in the 5.0 release of the Yambo code, distributed under GPL

Tra profondità e superficie. Anatomia, visione e scrittura in Valerio Magrelli e Michel Tournier

At the outer limit of the body, the skin is a threshold between inside and outside, depth and surface, visible and invisible. In his project of integrating psychology and body growth, Didier Anzieu considers the epidermis to be a double-envelope surface, impermeable and porous at the same time, provided with an inner side which encloses the content of the body and records any slight change which occurs in it, and an outer side, a boundary between the body and the world which accumulates the traces left by experience, by the passing of time and by forces acting outside the body. Anatomy becomes a metaphor for writing both in Valerio Magrelli and Michel Tournier. In Magrelli’s Nel condominio di carne, both the image of an eye looking inwards and the X-ray vision become synonyms for introspection and starting point for the development of writing. Tournier’s artistic path, on the contrary, seems to develop along two parallel lines: the refusal of an epistemology of the deep associating interior, secret, truth, and the celebration of surface. The sense of horror aroused by the dissection of interior anatomy, as well as the image of an open window which widens the view onto the external world, overturn the self-perceptive point of view by reversing the direction of sight along which writing unfolds. As Tournier’s sensibility for photography might therefore correspond with the exaltation of externality as well as with his choice of a centrifugal writing, so Magrelli’s radiological interest corresponds with the celebration of depth and with the autobiographical inclination of his poetics. At the outer limit of the body, the skin is a threshold between inside and outside, depth and surface, visible and invisible. Ab Initio Spectroscopic Investigation of Pharmacologically Relevant Chiral Molecules: The Cases of Avibactam, Cephems, and Idelalisib as Benchmarks for Antibiotics and Anticancer Drugs

Abstract: The ability to accurately measure or predict several physicochemical properties of molecules which play a role as active substances in drugs can be of strategic importance for pharmacological applications, in addition to its possible interest in fundamental research. Chirality is a relevant feature in the characterization of drug molecules: enantiomers can show different pharmacological activity and adverse effects. The ability to separate stereoisomers and to assign their absolute configuration can thus be crucial. Circular dichroism (CD) spectra are a useful tool to distinguish between enantiomers. In this work we apply an in-house developed code, based on an efficient DFT approach for circular dichroism, to fully characterize the molecular optical properties in the case of few selected fundamental molecules for current medical and pharmaceutical research, namely avibactam, as representative of non b-lactam inhibitors, two cephems (cefepime and cefoxitin), as examples of b-lactam antibiotics, and idelalisib, as a recent relevant anticancer active substance to treat major leukemias. For the above molecules, in addition to their optical absorption spectra, we calculate their CD spectra within state-of-the-art computational techniques. We then investigate both the conformational and chemical sensitivity of absorption and CD spectra for the chosen molecules. The outcomes of the present research could be of fundamental importance to gain additional information on molecules involved in therapeutic protocols for severe diseases or in drug design

Given the difficulties in diagnosis of type 1 GD in adults because of disease heterogeneity and lack of awareness, appropriate diagnostic algorithms or flow-charts starting from non-specific findings may help. Case reports help to establish the usefulness of our proposed flowchart in patients presenting with \u201cunexplained hyperferritinemia\u201d

REGULUS Iodine Electric Propulsion System Integration in CubeSats’ Platforms and Testing

    Get PDF
REGULUS is an electric propulsion (EP) system for CubeSats at TRL8 and now waiting for the IoD flight in late 2020. REGULUS system is provided for integration with all electronics, fluidic line, iodine tank and structures for total mass below 3 kg. Thanks in particular to the Magnetically Enhanced RF Plasma Thruster (MEPT) technology and the use of iodine propellant, the system can provide 3000Ns of total impulse in a 93.8 x 95.0 x 150.0 mm volume performance, fitting in a 1.5U Cubesat. REGULUS includes the whole propulsion package for integration in CubeSats and MicroSats as well as small CubeSat carriers. The system is composed by the thruster, the electronics (PPUs and PCU) the fluidic line and the tank. The main features of REGULUS are the presence of a simple architecture, a thruster with no neutralizer and grids, no high DC-voltage PPU and the use of solid iodine as propellant, that can be substituted with Xenon fluidic line and tank when required. Its first mission will be onboard of Unisat-7 by GAUSS. The flight will take place in late 2020 in a Soyuz flight. During the mission, REGULUS will allow Unisat-7 to perform an orbit descending maneuver, drag compensation in VLEO and decommissioning

Clinical and molecular epidemiology of erythropoietic protoporphyria in Italy

Background: Erythropoietic protoporphyria (EPP) is a rare inherited disease associated with heme metabolism, characterized by severe life-long photosensitivity and liver involvement. Objectives: To provide epidemiological data of EPP in Italy. Materials and Methods: Prospective/retrospective data of EPP patients were collected by an Italian network of porphyria specialist centres (Gruppo Italiano Porfiria, GrIP) over a 20-year period (1996-2017). Results: In total, 179 patients (79 females) with a clinical and biochemical diagnosis of EPP were assessed, revealing a prevalence of 3.15 cases per million persons and an incidence of 0.13 cases per million persons/year. Incidence significantly increased after 2009 (due to the availability of alfa-melanotide, which effectively limits skin photosensitivity). Mean age at diagnosis was 28 years, with only 22 patients (12.2%) diagnosed 6410 years old. Gene mutations were assessed in 173 (96.6%) patients; most (164; 91.3%) were FECH mutations on one allele in association with the hypomorphic variant, c.315-48C, on the other (classic EPP), and nine (5.2%) were ALAS2 mutations (X-linked EPP). Only one case of autosomal recessive EPP was observed. Of the 42 different FECH mutations, 15 are novel, three mutations collectively accounted for 45.9% (75/164) of the mutations (c.215dupT [27.2%], c.901_902delTG [11.5%] and c.67\u2009+\u20095G\u2009>\u2009A [7.2%]), and frameshift mutations were prevalent (33.3%). A form of light protection was used by 109/179 (60.8%) patients, and 100 (56%) had at least one \u3b1-melanotide implant. Three cases of severe acute liver involvement, requiring OLT, were observed. Conclusions: These data define, for the first time, the clinical and molecular epidemiology of EPP in Italy

Prescription appropriateness of anti-diabetes drugs in elderly patients hospitalized in a clinical setting: evidence from the REPOSI Register

Diabetes is an increasing global health burden with the highest prevalence (24.0%) observed in elderly people. Older diabetic adults have a greater risk of hospitalization and several geriatric syndromes than older nondiabetic adults. For these conditions, special care is required in prescribing therapies including anti- diabetes drugs. Aim of this study was to evaluate the appropriateness and the adherence to safety recommendations in the prescriptions of glucose-lowering drugs in hospitalized elderly patients with diabetes. Data for this cross-sectional study were obtained from the REgistro POliterapie-Società Italiana Medicina Interna (REPOSI) that collected clinical information on patients aged ≥ 65 years acutely admitted to Italian internal medicine and geriatric non-intensive care units (ICU) from 2010 up to 2019. Prescription appropriateness was assessed according to the 2019 AGS Beers Criteria and anti-diabetes drug data sheets.Among 5349 patients, 1624 (30.3%) had diagnosis of type 2 diabetes. At admission, 37.7% of diabetic patients received treatment with metformin, 37.3% insulin therapy, 16.4% sulfonylureas, and 11.4% glinides. Surprisingly, only 3.1% of diabetic patients were treated with new classes of anti- diabetes drugs. According to prescription criteria, at admission 15.4% of patients treated with metformin and 2.6% with sulfonylureas received inappropriately these treatments. At discharge, the inappropriateness of metformin therapy decreased (10.2%, P < 0.0001). According to Beers criteria, the inappropriate prescriptions of sulfonylureas raised to 29% both at admission and at discharge. This study shows a poor adherence to current guidelines on diabetes management in hospitalized elderly people with a high prevalence of inappropriate use of sulfonylureas according to the Beers criteria
