    Immediate Biomechanical Effects of Providing Adaptive Assistance With an Ankle Exoskeleton in Individuals After Stroke

    Recent studies on ankle exoskeletons have shown the feasibility of this technology for post-stroke gait rehabilitation. The main contribution of the present work is a comprehensive experimental analysis and protocol that focused on evaluating a wide range of biomechanical, usability and users’ perception metrics under three different walking conditions: without exoskeleton, with an ankle exoskeleton unpowered, and with an ankle exoskeleton powered. To carry out this study, we developed the ABLE-S exoskeleton that can provide time-adapted ankle plantarflexion and dorsiflexion assistance. Tests with five participants with chronic stroke showed that walking with the ABLE-S exoskeleton significantly corrected foot drop by 25 % while reducing hip compensatory movements by 21 %. Furthermore, asymmetrical spatial gait patterns were significantly reduced by 51 % together with a significant increase in the average foot tilting angle at heel strike by 349 %. The total time to don, doff and set-up the device was of 7.86 ± 2.90 minutes. Finally, 80 % of the participants indicated that they were satisfied with their walking performance while wearing the exoskeleton, and 60 % would use the device for community ambulation.     Blood DNA methylation signature of diet quality, and association with cardiometabolic traits

    BACKGROUND: Diet quality might influence cardiometabolic health through epigenetic changes, but this has been little investigated in adults. Our aim was to identify Cytosine-phosphate-Guanine (CpG) dinucleotides associated with diet quality by conducting an epigenome-wide association study (EWAS) based on blood DNA methylation (DNAm), and to assess how diet-related CpGs associate with inherited susceptibility to cardiometabolic traits: body mass index (BMI), systolic blood pressure (SBP), triglycerides, type 2 diabetes (T2D) and coronary heart disease (CHD). METHODS: Meta-EWAS including 5,274 participants in four cohorts from Spain, the US and the UK. We derived three dietary scores (exposures) to measure adherence to a Mediterranean diet (MMDS), to a healthy plant-based diet (HPDI) and to the Dietary Approaches to Stop Hypertension (DASH). Blood DNAm (outcome) was assessed with the Infinium arrays Human Methylation 450 K BeadChip and MethylationEPIC BeadChip. For each diet score, we performed linear EWAS adjusted for age, sex, blood cells, smoking and technical variables, and BMI in a second set of models. We also conducted Mendelian randomization analyses to assess the potential causal relationship between diet-related CpGs and cardiometabolic traits. RESULTS: We found 18 differentially methylated CpGs associated with dietary scores (p-value < 1.08 × 10-7; Bonferroni correction), of which 12 were previously associated with cardiometabolic traits. Enrichment analysis revealed overrepresentation of diet-associated genes in pathways involved in inflammation and cardiovascular disease. Mendelian randomization analyses suggested that genetically determined methylation levels corresponding to lower diet quality at cg02079413 (SNORA54), cg02107842 (MAST4), and cg23761815 (SLC29A3) were causally associated with higher BMI, and at cg05399785 (WDR8) with greater SBP; and methylation levels associated with higher diet quality at cg00711496 (PRMT1) with lower BMI, T2D risk and CHD risk, and at cg0557921 (AHRR) with lower CHD risk. CONCLUSIONS: Diet quality in adults was related to differential methylation in blood at 18 CpGs, some of which related to cardiometabolic health.Availability: The R code for the analysis is available in the following Github repository: https://github.com/jorgedb98/B64_DIAMETR.git

    Cell type-specific changes in Wnt signaling and neuronal differentiation in the developing mouse cortex after prenatal alcohol exposure during neurogenesis

    Fetal Alcohol Spectrum Disorder (FASD) encompasses an array of effects of prenatal alcohol exposure (PAE), including physical abnormalities and cognitive and behavioral deficits. Disruptions of cortical development have been implicated in multiple PAE studies, with deficits including decreased progenitor proliferation, disrupted neuronal differentiation, aberrant radial migration of pyramidal neurons, and decreased cortical thickness. While several mechanisms of alcohol teratogenicity have been explored, how specific cell types in the brain at different developmental time points may be differentially affected by PAE is still poorly understood. In this study, we used single nucleus RNA sequencing (snRNAseq) to investigate whether moderate PAE from neurulation through peak cortical neurogenesis induces cell type-specific transcriptomic changes in the developing murine brain. Cluster analysis identified 25 neuronal cell types, including subtypes of radial glial cells (RGCs), intermediate progenitor cells (IPCs), projection neurons, and interneurons. Only Wnt-expressing cortical hem RGCs showed a significant decrease in the percentage of cells after PAE, with no cell types showing PAE-induced apoptosis as measured by caspase expression. Cell cycle analysis revealed only a subtype of RGCs expressing the downstream Wnt signaling transcription factor Tcf7l2 had a decreased percentage of cells in the G2/M phase of the cell cycle, suggesting decreased proliferation in this RGC subtype and further implicating disrupted Wnt signaling after PAE at this early developmental timepoint. An increased pseudotime score in IPC and projection neuron cell types indicated that PAE led to increased or premature differentiation of these cells. Biological processes affected by PAE included the upregulation of pathways related to synaptic activity and neuronal differentiation and downregulation of pathways related to chromosome structure and the cell cycle. Several cell types showed a decrease in Wnt signaling pathways, with several genes related to Wnt signaling altered by PAE in multiple cell types. As Wnt has been shown to promote proliferation and inhibit differentiation at earlier stages in development, the downregulation of Wnt signaling may have resulted in premature neuronal maturation of projection neurons and their intermediate progenitors. Overall, these findings provide further insight into the cell type-specific effects of PAE during early corticogenesis

    Physical characterization of long-lasting hybrid eruptions: the 2021 tajogaite eruption of Cumbre Vieja (La Palma, Canary Islands)

    Long-lasting, hybrid eruptions can be of complex description and classification, especially when associated with multiple eruptive styles and multiple products. The 2021 Tajogaite eruption of La Palma, Canary Islands, was associated with a magma-gas decoupled system that resulted in the simultaneous emission of lava flows and tephra plumes from various vents. Even though the tephra blanket (∼2 × 107 m3) represents only 7%–16% of the total erupted volume, it provides fundamental insights into the overall eruptive dynamics. Tephra was mostly dispersed NE-SW due to a complex regional and local wind patterns and was subdivided into 3 units and 11 layers that well correlate at different distances from the vent and with both tremor data and lava emission rate. While plume height varied at the temporal scale of a few hours, the average mass eruption rate associated with the tephra blanket of the different units remained relatively constant (∼3–4 × 103 kg s−1). In contrast, the emission rate of lava largely increased after the first week and remained higher than the overall emission of tephra throughout the whole eruption (average value of ∼6 × 104 kg s−1).     Dietary diversity and depression: cross-sectional and longitudinal analyses in Spanish adult population with metabolic syndrome. Findings from PREDIMED-Plus trial

    Objective: To examine the cross-sectional and longitudinal (2-year follow-up) associations between dietary diversity (DD) and depressive symptoms. Design: An energy-adjusted dietary diversity score (DDS) was assessed using a validated FFQ and was categorised into quartiles (Q). The variety in each food group was classified into four categories of diversity (C). Depressive symptoms were assessed with Beck Depression Inventory-II (Beck II) questionnaire and depression cases defined as physician-diagnosed or Beck II >= 18. Linear and logistic regression models were used. Setting: Spanish older adults with metabolic syndrome (MetS). Participants: A total of 6625 adults aged 55–75 years from the PREDIMED-Plus study with overweight or obesity and MetS. Results: Total DDS was inversely and statistically significantly associated with depression in the cross-sectional analysis conducted; OR Q4 v. Q1 = 0·76 (95 % CI (0·64, 0·90)). This was driven by high diversity compared to low diversity (C3 v. C1) of vegetables (OR = 0·75, 95 % CI (0·57, 0·93)), cereals (OR = 0·72 (95 % CI (0·56, 0·94)) and proteins (OR = 0·27, 95 % CI (0·11, 0·62)). In the longitudinal analysis, there was no significant association between the baseline DDS and changes in depressive symptoms after 2 years of follow-up, except for DD in vegetables C4 v. C1 = (β = 0·70, 95 % CI (0·05, 1·35)). Conclusions: According to our results, DD is inversely associated with depressive symptoms, but eating more diverse does not seem to reduce the risk of future depression.     The Evolution of Primate Short-Term Memory.

    Short-term memory is implicated in a range of cognitive abilities and is critical for understanding primate cognitive evolution. To investigate the effects of phylogeny, ecology and sociality on short-term memory, we tested the largest and most diverse primate sample to date (421 non-human primates across 41 species) in an experimental delayed-response task. Our results confirm previous findings that longer delays decrease memory performance across species and taxa. Our analyses demonstrate a considerable contribution of phylogeny over ecological and social factors on the distribution of short-term memory performance in primates; closely related species had more similar short-term memory abilities. Overall, individuals in the branch of Hominoidea performed better compared to Cercopithecoidea, who in turn performed above Platyrrhini and Strepsirrhini. Interdependencies between phylogeny and socioecology of a given species presented an obstacle to disentangling the effects of each of these factors on the evolution of short-term memory capacity. However, this study offers an important step forward in understanding the interspecies and individual variation in short-term memory ability by providing the first phylogenetic reconstruction of this trait’s evolutionary history.     En un país -Francia- donde el campo del teatro está estructurado culturalmente durante décadas, el Teatro Aplicado es una noción que a menudo aparece como ancillar, frente a un arte institucionali-zado, incluso mirificado. Por un lado, estaría el Teatro, puro, noble, auténtico y por otro, estarían sus avatares: el teatro de empresa, el teatro para el desarrollo personal, el teatro para patologías, etc. Si tienen la misma fuente, su consanguinidad no deja de asustar. ¿cómo pueden unos artistas que crean alejados de cualquier coacción exterior pertenecer a la misma familia del teatro que unos actores o directores que "obedecen" a un encargo, en un contexto específico, con un público muchas veces participantes de talleres ... y que son por tanto prisioneros, en cierto modo, de un arte instru-mental izado? A este problema ético, este libro intenta responder, a través de ejemplos concretos, para una mayor comprensión inrerculrural Francia/ Colombia.In a country -France- where the field of theater has been culturally structured for decades, Applied Theater is a notion that often appears as an ancillary, in the face of an institutionalized, even mirified, art. On the one hand, there would be the Theater, pure, noble, authentic and on the other, there would be its ups and downs: company theater, theater for personal development, theater for pathologies, etc. If they have the same source, their consanguinity does not stop frightening. How can some artists who create far from any external coercion belong to the same theater family as some actors or directors who "obey" a commission, in a specific context, with an audience that is often workshop participants... are they therefore prisoners, in a certain way, of an instrumented art?
