Pobreza econômica e sociocultural no abuso de drogas: da responsabilidade individual à sociopolítica
Research on drug abuse has often ignored users’
own opinions and perceptions about their addiction.
In this study, we wanted to hear their voice on the
reasons and motives why they engaged in drug
abuse, and on the consequences this behavior had
on their lives. Data were collected in Portugal from
interviews with fifteen people under treatment for
addictive behavior relative to alcohol and illegal
drugs. The interviews were analyzed through
Thematic Analysis and revealed the existence of
several structural factors impacting on the lives of
the participants, namely, gender discrimination,
poor schooling, socioeconomic marginalization
and exclusion associated to insufficient and
inadequate public policies. The action of more
symbolic structural factors – for instance, their
widespread beliefs on drug addiction as a result of
free will – through its internalization by families,
friends and by addicts themselves, became visible
in interviewees’ narratives, in which the rejection
by close ones, as well as their own feelings of
guilt, sadness and self-disapproval, are prominent
features. As a conclusion, we call attention to
the need for an integrated public policy in the
educational, health and justice areas, and the
implementation of awareness-raising actions aimed
at the general public, in order to attenuate the
impact of structural factors on the lives of current
and potential drug addicts.A pesquisa sobre o abuso de drogas tem
frequentemente ignorado as opiniões dos
próprios usuários e as suas perceções sobre a
sua dependência. Neste estudo, quisemos ouvir
a sua voz sobre as razões e os motivos por que
se engajaram no abuso de drogas e sobre as
consequências que este comportamento teve
nas suas vidas. Os dados foram coletados em
Portugal a partir de entrevistas com quinze
pessoas sob tratamento para comportamento
aditivo em relação a álcool e drogas ilegais.
As entrevistas foram analisadas através de
análise temática e revelaram a existência de
diversos fatores estruturais impactando na
vida dos participantes, ou seja, discriminação
de gênero, escolaridade baixa, marginalização
socioeconômica e exclusão associados a políticas
públicas insuficientes e inadequadas. A ação de
fatores estruturais mais simbólicos – por exemplo,
suas crenças generalizadas na toxicodependência
como resultado do livre-arbítrio – através de sua
interiorização pelas famílias, amigos e pelos
viciados em si, tornou-se visível nas narrativas dos
entrevistados, em que a rejeição pelos próximos,
além de seus próprios sentimentos de culpa,
tristeza e autodesaprovação, são características
proeminentes. Como conclusão, chamamos
atenção para a necessidade de uma política pública
integrada nas áreas de educação, saúde e justiça,
e a implementação de ações de sensibilização
visando o público em geral, a fim de atenuar o
impacto dos fatores estruturais sobre a vida dos
atuais e potenciais viciados em drogas
Potencial terapêutico de miR-9 e miR-29 na doença de Alzheimer
A demência é uma síndrome que está relacionada com a idade, apresenta várias causas e são raras as que se podem tratar ou prevenir eficazmente. A forma de demência mais frequente é a doença de Alzheimer, uma doença cerebral progressiva e degenerativa, irreversível e sem cura. A doença de Alzheimer é caracterizada por várias alterações morfológicas/patológicas, sendo as principais a acumulação extracelular de péptidos ß-amilóides, denominados como placas Aß, assim como a acumulação intracelular da proteína tau, a qual está associada a lesões intraneuronais e fibrilares que consistem em agregações de filamentos helicoidais emparelhados, compostos pela proteína tau hiperfosforilada, associada a microtúbulos, também conhecidos como emaranhados neurofibrilares. Diversas evidências mostram ainda que a acumulação de péptidos Aß leva também a um aumento da severidade da patologia tau. Como referido, atualmente a doença de Alzheimer, bem como outras doenças neurodegenerativas, não apresentam cura e as terapias existentes apenas diminuem temporariamente os sintomas. Desta forma uma abordagem promissora poderá ser a utilização de certos miARNs para o silenciamento da expressão de várias proteínas envolvidas no surgimento e progressão da doença de Alzheimer. Os miARNs representam a classe de ARNs não codificantes que atuam como potentes reguladores da pós-transcrição da expressão génica. Desta forma através dos microARNs consegue-se a inibição pós-transcricional da expressão das respetivas proteínas, causadoras do surgimento e progressão de várias doenças. Este trabalho teve como principal objetivo avaliar o efeito do pre-miR-9 e pre-miR-29, como agentes terapêuticos na DA, procurando a diminuição da expressão de proteínas alvo envolvidas na patologia, nomeadamente a APP, BACE1 e presenilina, na linha celular N2a695. Com a realização do projeto verificou-se que tanto o pre-miR-9, como o pre-miR-29b têm efeito nas proteínas da via amilóide em estudo. De forma geral foi observado um maior efeito pelo pre-miR-9 na APP e na presenilina e um maior efeito do pre-miR-29b na BACE1. No entanto foi ainda obtido um maior silenciamento da expressão da presenilina por parte do pre-miR-29b em estudos de western blot mas, por RT-qPCR e imunocitoquímica não se verificou uma diferença significativa entre a sinergia do pre-miR-9 e pre-miR-29b e o seu uso individual. Relativamente à BACE1 verificou-se um silenciamento superior após transfeção das células com o pre-miR-29b relativamente ao pre-miR-9 ou à co-transfeção com ambos os pre-miARNs. Em relação à APP verificou-se a inibição da sua expressão, induzida quer pelo pre-miR-9 quer pelo pre-miR-29b, isoladamente ou transferidos em simultâneo. Em suma, com o trabalho realizado foi possível verificar uma relação quer do pre-miR-9, quer do pre-miR-29b na regulação da expressão das proteínas estudadas (presenilina, BACE1 e APP). Num futuro próximo, pretende-se confirmar e aprofundar esta análise, especialmente nos casos que não foram previamente reportados. De uma forma mais específica, será interessante confirmar a ação regulatória do pre-miR-29b sobre a expressão da presenilina, assim como confirmar a possível relação destes pre-miRs com o silenciamento da expressão da APP, que não foram ainda evidenciados na literatura. O conhecimento e melhor compreensão destas vias associadas ao silenciamento génico é de extrema importância, visto que estes pre-miRs poderão ser agentes terapêuticos com interesse de aplicação no tratamento da doença de Alzheimer.Dementia is a syndrome which is related to the age, it has several causes and it is rare how can be effectively treated or prevented. The most common form of dementia is Alzheimer's disease, a progressive and degenerative brain disease, irreversible and without cure. Alzheimer's disease is characterized by several morphological/pathological changes, being the most relevant the extracellular accumulation of ß-amyloid peptides, called Aß plaques, as well as the intracellular accumulation of tau protein, which is associated with intraneuronal and fibrillary lesions consisting of paired helical filaments composed of the hyperphosphorylated protein, associated with microtubules, also known as neurofibrillary. Several evidences also show that the ß-amyloid accumulation can also lead to an increased tau pathology severity. As noted, currently Alzheimer's disease as well as the other neurodegenerative diseases do not present an effective treatment and the therapies may only decrease the symptoms. In this way a promising approach can be the use of certain miRNAs for the silencing of several proteins involved in the onset and progression of Alzheimer's disease. The miRNAs represent a class of non-coding RNAs that act as post-transcriptional regulators of gene expression. Thus, through the use of microRNAs, it is possible to inhibit the expression of target proteins, involved on the onset and progression of various diseases. This work had as main objective to evaluate the effect of pre-miR-9 and pre-mi-29 as therapeutic agents in AD, looking for the inhibition of particular proteins currently associated to the disease, namely the APP, BACE1 and presenilin, in the N2a695 cell line. With this project it was concluded that both pre-miR-9 and pre-miR-29b have an effect on the proteins of the amyloid pathway. In general, there was a greater effect of pre-miR-9 on APP and presenilin expression and a greater effect of pre-miR-29b on BACE1. However, it was also observed the inhibition of presenilin by pre-miR-29b in western blot studies, but by RT-qPCR and immunocytochemistry there was no significant difference between the synergy of pre-miR-9 and pre-miR- 29b and its individual use. Relative to BACE1 there was superior silencing after cell´s transfection with pre-miR-29b relative to pre-miR-9 or co-transfection with both pre-miRNAs. Regarding APP, the inhibition of its expression was induced either by pre-miR-9 or pre-miR-29b, when isolated, or transferred simultaneously. In conclusion, the pre-miR-9 and pre-miR-29b in the regulation of the expression of the proteins studied (presenilin, BACE1 and APP) could be verified. In the near future, it is intended to confirm and deepen this analysis, especially in the cases which have not previously been reported. More specifically, it will be interesting to confirm the regulatory action of pre-miR-29b on the expression of presenilin, as well as confirm the possible relationship of these pre-miRs with the silencing of APP, which have not yet been evidenced in the literature. The knowledge and better understanding of these pathways associated with gene silencing assumes an extreme importance, since these pre-miRs may be therapeutic agents of interest for the treatment of Alzheimer's disease
A assimetria dos ciclos económicos: evidência internacional
São testadas duas formas distintas de assimetria cíclica, a deepness e a steepness, através da aplicação do teste do coeficiente de enviesamento e do teste triples. A assimetria é analisada em vinte e três países, segundo uma perspectiva secular do PIB real per capita (1820/70 a 2008). Os resultados dos testes sugerem que a deepness não é relevante a nível internacional. Para a steepness, os testes revelam resultados contraditórios, sendo que o coeficiente de enviesamento evidencia a presença de assimetria em onze dos vinte e três países e o teste triples em seis países. A aplicação do teste do coeficiente de enviesamento e
do teste triples permite cotejar os resultados obtidos e aferir da robustez dos resultados. Ao contrário do sugerido na literatura, baseada em dados trimestrais, a nossa investigação não é conclusiva acerca da presença, ao nível internacional, de assimetria nos ciclos económicos.We test, using the coefficient of skewness and the triples test, two types of cyclical
asymmetry – the deepness and the steepness. The asymmetry, for twenty-three countries, is
examined, from over a century data of real GDP per capita (1820/70 to 2008). There is no
international evidence of deepness. For the steepness, the tests show mixed results. Indeed,
the coefficient of skewness reveal presence of asymmetry for eleven countries, and the
triples test for six ones. The assessment of these two tests allows us to judge the results and
the robustness of the analyses. On contrary of the most of literature, based on quarterly
data, our research is mixed about international evidence of business cycle asymmetries
Beliefs on Marital Violence and Self-Reported Dating Violence: A Comparative Study of Cape Verdean and Portuguese Adolescents
In this study, we compare Portuguese and
Cape Verdean youths’ upholding of traditional beliefs about
intimate partner violence (IPV) and the frequency of selfreported
violent behavior in dating relationships. The sample
(n =404) consisted of 183 Cape Verdean and 221 Portuguese
secondary school students of both sexes (56 % female; mean
age = 16). We used two questionnaires that had previously
been validated in the Portuguese population. The results revealed
that young Cape Verdean adolescents uphold stronger
traditional beliefs than Portuguese adolescents do, but there
were no differences in overall prevalence of abuse between
the two samples. The relationship between traditional beliefs
and self-reported violence was significant only in the
Cape Verdean sample, suggesting that campaigns against
IPV have not, so far, been as effective in Cape Verde as in
