    Automated processing of series of micro-CT scans

    For some applications of high-resolution X-ray Tomography (micro-CT) scanning, a large set of similar samples is to be analyzed in order to obtain statistically significant results. The complete process, including the micro-CT scan itself, the reconstruction and the analysis is almost identical for every sample. However, in a typical workflow every step is manually performed for every individual sample. This could be optimised by automation of this process, which results in less human intervention and thus a smaller cost and a lower risk to human error. We developed a reliable method to semi-automatically scan several stacked samples and automatically reconstruct the resulting series of data sets. The reconstruction step includes the manual reconstruction of one data set in order to optimize the reconstruction parameters, which can then be used for the rest of the batch. In future work, the automatic handling of the next step in the micro-CT workflow, 3D analysis, will also be improved

    Job satisfaction in relation to communication in health care among nurses : a narrative review and pratical recommendations

    Worldwide, nurse shortage and high turnover rates are observed. Job satisfaction is a major determinant of retention and is influenced by intraorganizational communication and perceived communication satisfaction. This article presents a narrative review on communication satisfaction, job satisfaction, and their mutual relationship as well as their impact on turnover intention and burnout risk in the nursing profession. A literature search was conducted in the databases PubMed, Web of Science, and The Cochrane Library, and 47 articles were included. Descriptive analysis identified different types of social networks in the health care workplace. There is a positive association between communication and job satisfaction among nurses, translating into decreased turnover intention and burnout risk. Job satisfaction is required both for organizational stability as for coguaranteeing patient safety. This will be best achieved through an organization-wide multimodal prevention and intervention program, aimed at optimizing different modalities of interprofessional communication, workload, and job satisfaction

    Electronic band gaps of confined linear carbon chains ranging from polyyne to carbyne

    Ultra long linear carbon chains of more than 6000 carbon atoms have recently been synthesized within double-walled carbon nanotubes, and they show a promising new route to one--atom--wide semiconductors with a direct band gap. Theoretical studies predicted that this band gap can be tuned by the length of the chains, the end groups, and their interactions with the environment. However, different density functionals lead to very different values of the band gap of infinitely long carbyne. In this work, we applied resonant Raman excitation spectroscopy with more than 50 laser wavelengths to determine for the first time the band gap of long carbon chains encapsulated inside DWCNTs. The experimentally determined band gaps ranging from 2.253 to 1.848 eV follow a linear relation with Raman frequency. This lower bound is the smallest band gap of linear carbon chains observed so far. The comparison with experimental data obtained for short chains in gas phase or in solution demonstrates the effect of the DWCNT encapsulation, leading to an essential downshift of the band gap. This is explained by the interaction between the carbon chain and the host tube, which greatly modifies the chain's bond length alternation.Comment: 7 pages, 5 figure

    RpoS-independent evolution reveals the importance of attenuated cAMPCRP regulation in high hydrostatic pressure resistance acquisition in E. coli

    High hydrostatic pressure (HHP) processing is an attractive non-thermal alternative to food pasteurization. Nevertheless, the large inter- and intra-species variations in HHP resistance among foodborne pathogens and the ease by which they can acquire extreme resistance are an issue of increasing concern. Since RpoS activity has been considered as a central determinant in the HHP resistance of E. coli and its pathovars, this study probed for the potential of an E. coli MG1655 ΔrpoS mutant to acquire HHP resistance by directed evolution. Despite the higher initial HHP sensitivity of the ΔrpoS mutant compared to the wild-type strain, evolved lineages of the former readily managed to restore or even succeed wild-type levels of resistance. A number of these ΔrpoS derivatives were affected in cAMP/CRP regulation, and this could be causally related to their HHP resistance. Subsequent inspection revealed that some of previously isolated HHP-resistant mutants derived from the wild-type strain also incurred a causal decrease in cAMP/CRP regulation. cAMP/CRP attenuated HHP-resistant mutants also exhibited higher resistance to fosfomycin, a preferred treatment for STEC infections. As such, this study reveals attenuation of cAMP/CRP regulation as a relevant and RpoS-independent evolutionary route towards HHP resistance in E. coli that coincides with fosfomycin resistance

    Protein aggregates act as a deterministic disruptor during bacterial cell size homeostasis

    Mechanisms underlying deviant cell size fluctuations among clonal bacterial siblings are generally considered to be cryptic and stochastic in nature. However, by scrutinizing heat-stressed populations of the model bacterium Escherichia coli, we uncovered the existence of a deterministic asymmetry in cell division that is caused by the presence of intracellular protein aggregates (PAs). While these structures typically locate at the cell pole and segregate asymmetrically among daughter cells, we now show that the presence of a polar PA consistently causes a more distal off-center positioning of the FtsZ division septum. The resulting increased length of PA-inheriting siblings persists over multiple generations and could be observed in both E. coli and Bacillus subtilis populations. Closer investigation suggests that a PA can physically perturb the nucleoid structure, which subsequently leads to asymmetric septation

    The expression of virulence increases outer-membrane permeability and sensitivity to envelope stress in Salmonella Typhimurium

    Environmental cues modulate the expression of virulence in bacterial pathogens. However, while cues that upregulate virulence are often intuitive and mechanistically well understood, this is less so for cues that downregulate virulence. In this study, we noticed that upregulation of the HilD virulence regulon in Salmonella Typhimurium ( S .Tm) sensitized cells to membrane stress mediated by cholate, Tris/EDTA or heat. Further monitoring of membrane status and stress resistance of S .Tm cells in relation to virulence expression, revealed that co-expressed virulence factors embedded in the envelope (including the Type Three Secretion System 1 and the flagella) increased permeability, and stress sensitivity of the membrane. Importantly, pretreating the bacteria by sublethal stress inhibited virulence expression and restored stress resistance. As such, these results demonstrate a trade-off between virulence and stress resistance, which explains the downregulation of virulence expression in response to harsh environments in S .Tm

    Conservative solutions for progress: on solution types when combining QCA with in-depth Process-Tracing

    What is the most appropriate QCA solution type when engaging in a multimethod design that includes QCA and in-depth process-tracing (PT)? While either the intermediate or the parsimonious solution are generally favored in QCA-only studies, we identify important challenges that can emerge when selecting those solutions in a QCA-PT multimethod study. We particularly highlight the risk of mechanistic heterogeneity, omitted conditions, and draw the attention on the issue of generalization. We discuss each of these intertwined challenges in depth, and explain why the conservative solution is useful to consider in addressing them. We substantiate our arguments by drawing on a recently completed evaluation study that was commissioned by the Flemish ESF Agency in Belgium. In the study, we combined QCA and theory-guided in-depth process-tracing to uncover under what combinations of conditions (QCA) a training programme would lead to successful training transfer and how (PT) this happened in the successful cases.
