    Design of a Microgravity Hybrid Inflatable Airlock

    Spacewalks, or extra-vehicular activities (EVAs), are a critical component of human space exploration for science activities and habitat construction and maintenance. For NASA's proposed lunar Gateway system, an airlock module is required for vehicle maintenance, repair, and exploration. Traditional airlock structures are fully metallic, with two chambers, known as an equipment lock and a crew lock. The larger volume, called the equipment lock, serves as the storage, logistics and electronics area, while the smaller volume, called the crew lock, serves as the volume to transition from the vacuum of space to the pressurized cabin. A traditional metallic structure design offers mass efficiency for these elements, but cannot offer volume efficiency. The potential to use an inflatable fabric pressure shell supplemented by a metallic support structure allows for efficiency in both mass and volume. Inflatable structures are being used for human habitable space modules, starting with the Bigelow Expandable Activities Module on the International Space Station. They are high-strength fabric-based structures that are compactly stowed for launch and then, once in space, they are expanded and rigidized with internal pressure. They provide significant launch volume savings over metallic structures. For Gateway, a hybrid airlock design is proposed with both metallic and inflatable structural elements, taking advantage of each material's capabilities. A metallic equipment lock serves as both a docking node and provides pressurized volume for pre-EVA activities including pre-breathe and suit donning/doffing. A rigid equipment lock offers stowage space during launch for integrated hardware and suits. Adding an integrated inflatable crew lock provides the volume required for EVAs with minimal use of launch volume. Using dual inflatable crew locks provides redundancy and the capability to move large pieces of equipment into and out of the vehicle for repair and maintenance. The inflatable crew lock is deflated and packaged in the launch shroud and expanded after installation on the Gateway. This packing capability allows additional volume to be added to the equipment lock and fully utilize the capability of the launch vehicle. This report outlines the work completed to design, analyze, and test the systems of a microgravity airlock with inflatable crew locks. In detail, it includes launch vehicles, structural sizing of the metallic equipment lock, the fabric layers of the inflatable crew lock, the internal structure of the crew lock, the space suit interface elements, the crew restraint system, the hatches and pass-throughs, the material and thermal elements, and the crew operations for the usage of the system.     Human Mars Entry, Descent, and Landing Architecture Study: Phase 3 Summary

    Over the past four years, NASA has directed the Entry, Descent and Landing Architecture Study (EDLAS) to evaluate candidate technologies to deliver human-scale vehicles (carrying 20t payloads) to the surface of Mars. The study focused on two vehicles in Phase 3 (during 2018 and 2019), one low and one mid lift-to-drag (L/D) vehicle. This paper summarizes updates to the structural and aeroshell outer mold line design for the mid-L/D Mars entry lander vehicle that occurred during EDLAS Phase 3. The design efforts used parametric optimization to arrive at a solution for total vehicle primary and secondary structure mass, and vehicle center-of-gravity location. Additionally, the end of Phase 2 provided an updated surface payload manifest that required three 22 t landers. This paper also summarizes the results of the packaging feasibility study of the updated payload configurations for those three reference missions.     Tumor De Células De La Granulosa: Diagnóstico, Tratamiento Hormonal E Intervención Quirúrgica En Yegua Criolla Colombiana

    Severe reproductive problems in mares are related to ovarian tumors. Among these, the Granulosa Cell Tumor (GCT) is the most commonly diagnosed, although it represents only 2.5% of all the tumors in horses. The following investigation is a case study of GCT in one creole adult mare, which presented a similar behavior as the male, virilism and anestrus. An ultrasonographic examination was used to identify the tumor and GnRH was administered as hormonal treatment; in the following weeks an ovariectomy was done as a surgical treatment. The histopathologic examination of the neoplasic tissue evidenced cellular proliferation with similar morphologic characteristics as the granulosa cells, which is the definitive diagnosis for GCT. The hormonal and surgical treatment was satisfactory, verified by the cessation of abnormal behaviors and by the return of the cyclicity of the remaining ovary. It is expected that in the following months the mare can be used again to reproduction.Graves problemas reproductivos se han asociado a tumores ováricos en yeguas. Dentro de estos el Tumor de Células de la Granulosa (TCG) sigue siendo el más diagnosticado, aunque sólo representa el 2.5% de todos los tumores del equino. El siguiente es un estudio de caso de TCG en una yegua criolla adulta que manifestó comportamiento similar al macho, virilismo y anestro. Se utilizó el examen ultrasonográfico para identificar el tumor y se administró GnRH como tratamiento hormonal; en semanas posteriores se procedió a la ovariectomia como tratamiento quirúrgico. El examen histopatológico del tejido neoplásico evidenció proliferación celular con características morfológicamente semejantes a las células de la granulosa, siendo el diagnóstico definitivo TCG. Se concluye que el tratamiento hormonal y quirúrgico fue satisfactorio, lo cual se pudo comprobar por el cese de comportamientos anormales y el retorno de la ciclicidad del ovario restante, se espera que en meses posteriores la yegua pueda ser utilizada nuevamente para reproducción

    Wildlife Reservoirs of Canine Distemper Virus Resulted in a Major Outbreak in Danish Farmed Mink (<em>Neovison vison</em>)

    A major outbreak of canine distemper virus (CDV) in Danish farmed mink (Neovison vison) started in the late summer period of 2012. At the same time, a high number of diseased and dead wildlife species such as foxes, raccoon dogs, and ferrets were observed. To track the origin of the outbreak virus full-length sequencing of the receptor binding surface protein hemagglutinin (H) was performed on 26 CDV's collected from mink and 10 CDV's collected from wildlife species. Subsequent phylogenetic analyses showed that the virus circulating in the mink farms and wildlife were highly identical with an identity at the nucleotide level of 99.45% to 100%. The sequences could be grouped by single nucleotide polymorphisms according to geographical distribution of mink farms and wildlife. The signaling lymphocytic activation molecule (SLAM) receptor binding region in most viruses from both mink and wildlife contained G at position 530 and Y at position 549; however, three mink viruses had an Y549H substitution. The outbreak viruses clustered phylogenetically in the European lineage and were highly identical to wildlife viruses from Germany and Hungary (99.29% – 99.62%). The study furthermore revealed that fleas (Ceratophyllus sciurorum) contained CDV and that vertical transmission of CDV occurred in a wild ferret. The study provides evidence that wildlife species, such as foxes, play an important role in the transmission of CDV to farmed mink and that the virus may be maintained in the wild animal reservoir between outbreaks

    Assessing mechanical integrity of spinal fusion by in situ endochondral osteoinduction in the murine model

    <p>Abstract</p> <p>Background</p> <p>Historically, radiographs, micro-computed tomography (micro-CT) exams, palpation and histology have been used to assess fusions in a mouse spine. The objective of this study was to develop a faster, cheaper, reproducible test to directly quantify the mechanical integrity of spinal fusions in mice.</p> <p>Methods</p> <p>Fusions were induced in ten mice spine using a previously described technique of in situ endochondral ossification, harvested with soft tissue, and cast in radiolucent alginate material for handling. Using a validated software package and a customized mechanical apparatus that flexed and extended the spinal column, the amount of intervertebral motion between adjacent vertebral discs was determined with static flexed and extended lateral spine radiographs. Micro-CT images of the same were also blindly reviewed for fusion.</p> <p>Results</p> <p>Mean intervertebral motion between control, non-fused, spinal vertebral discs was 6.1 ± 0.2° during spine flexion/extension. In fusion samples, adjacent vertebrae with less than 3.5° intervertebral motion had fusions documented by micro-CT inspection.</p> <p>Conclusions</p> <p>Measuring the amount of intervertebral rotation between vertebrae during spine flexion/extension is a relatively simple, cheap (<$100), clinically relevant, and fast test for assessing the mechanical success of spinal fusion in mice that compared favorably to the standard, micro-CT.</p

    The SIB Swiss Institute of Bioinformatics (www.isb-sib.ch) provides world-class bioinformatics databases, software tools, services and training to the international life science community in academia and industry. These solutions allow life scientists to turn the exponentially growing amount of data into knowledge. Here, we provide an overview of SIB's resources and competence areas, with a strong focus on curated databases and SIB's most popular and widely used resources. In particular, SIB's Bioinformatics resource portal ExPASy features over 150 resources, including UniProtKB/Swiss-Prot, ENZYME, PROSITE, neXtProt, STRING, UniCarbKB, SugarBindDB, SwissRegulon, EPD, arrayMap, Bgee, SWISS-MODEL Repository, OMA, OrthoDB and other databases, which are briefly described in this article

    Discutindo a educação ambiental no cotidiano escolar: desenvolvimento de projetos na escola formação inicial e continuada de professores

    A presente pesquisa buscou discutir como a Educação Ambiental (EA) vem sendo trabalhada, no Ensino Fundamental e como os docentes desta escola compreendem e vem inserindo a EA no cotidiano escolar., em uma escola estadual do município de Tangará da Serra/MT, Brasil. Para tanto, realizou-se entrevistas com os professores que fazem parte de um projeto interdisciplinar de EA na escola pesquisada. Verificou-se que o projeto da escola não vem conseguindo alcançar os objetivos propostos por: desconhecimento do mesmo, pelos professores; formação deficiente dos professores, não entendimento da EA como processo de ensino-aprendizagem, falta de recursos didáticos, planejamento inadequado das atividades. A partir dessa constatação, procurou-se debater a impossibilidade de tratar do tema fora do trabalho interdisciplinar, bem como, e principalmente, a importância de um estudo mais aprofundado de EA, vinculando teoria e prática, tanto na formação docente, como em projetos escolares, a fim de fugir do tradicional vínculo “EA e ecologia, lixo e horta”.Facultad de Humanidades y Ciencias de la Educació

    Background The immunological escape of tumors represents one of the main ob- stacles to the treatment of malignancies. The blockade of PD-1 or CTLA-4 receptors represented a milestone in the history of immunotherapy. However, immune checkpoint inhibitors seem to be effective in specific cohorts of patients. It has been proposed that their efficacy relies on the presence of an immunological response. Thus, we hypothesized that disruption of the PD-L1/PD-1 axis would synergize with our oncolytic vaccine platform PeptiCRAd. Methods We used murine B16OVA in vivo tumor models and flow cytometry analysis to investigate the immunological background. Results First, we found that high-burden B16OVA tumors were refractory to combination immunotherapy. However, with a more aggressive schedule, tumors with a lower burden were more susceptible to the combination of PeptiCRAd and PD-L1 blockade. The therapy signifi- cantly increased the median survival of mice (Fig. 7). Interestingly, the reduced growth of contralaterally injected B16F10 cells sug- gested the presence of a long lasting immunological memory also against non-targeted antigens. Concerning the functional state of tumor infiltrating lymphocytes (TILs), we found that all the immune therapies would enhance the percentage of activated (PD-1pos TIM- 3neg) T lymphocytes and reduce the amount of exhausted (PD-1pos TIM-3pos) cells compared to placebo. As expected, we found that PeptiCRAd monotherapy could increase the number of antigen spe- cific CD8+ T cells compared to other treatments. However, only the combination with PD-L1 blockade could significantly increase the ra- tio between activated and exhausted pentamer positive cells (p= 0.0058), suggesting that by disrupting the PD-1/PD-L1 axis we could decrease the amount of dysfunctional antigen specific T cells. We ob- served that the anatomical location deeply influenced the state of CD4+ and CD8+ T lymphocytes. In fact, TIM-3 expression was in- creased by 2 fold on TILs compared to splenic and lymphoid T cells. In the CD8+ compartment, the expression of PD-1 on the surface seemed to be restricted to the tumor micro-environment, while CD4 + T cells had a high expression of PD-1 also in lymphoid organs. Interestingly, we found that the levels of PD-1 were significantly higher on CD8+ T cells than on CD4+ T cells into the tumor micro- environment (p < 0.0001). Conclusions In conclusion, we demonstrated that the efficacy of immune check- point inhibitors might be strongly enhanced by their combination with cancer vaccines. PeptiCRAd was able to increase the number of antigen-specific T cells and PD-L1 blockade prevented their exhaus- tion, resulting in long-lasting immunological memory and increased median survival
