UIDB/04209/2020 UIDP/04209/2020 UIDB/05021/2020 UIDP/05021/2020In the parliamentary election held on 30 January 2022 in Portugal, Chega, a populist and anti-estab-lishment party of the radical-right wing won 7.18% of the votes and 12 seats in Parliament, becoming the third major political force of the 15th Legislature. Its 12 Members of Parliament (MPs) include only one woman, Rita Matias, who is also the youngest MP in the Chamber. During the election campaign, Rita Matias presented her-self as an anti-feminist, a statement that gained public and media attention after her election into public office. In this chapter, our purpose is to understand the relevance and significance of this position within the broader framework of the connection between gender and populism and to understand the extent to which this orien-tation contributes to establishing the political-ideological project of Chega. To try to establish this relationship between gender and right-wing populism, we propose to explore two aspects that we assume are interconnec-ted: gender performance structuring the populist style and ideology. Our investigation has a twofold focus: Rita Matias Parliamentary interventions, between March 29th and September 30th; and journalistic texts, concerning her declarations, published between February 1st and September 30th, 2022. As a conclusion, we point out that the declaration of anti-feminism made during the campaign brought, both to the media and to Parliament, a new angle that had not yet been explored and embodies the populist style and radical right-wing ideology of Chega for gender’s issues.

Illiberalism and the Deinstitutionalization of Public Diplomacy: The Rise of Hungary and Viktor Orbán in American Conservative Media

The promotion of Hungary and Viktor Orbán among American conservatives is often presented as a warning of conservative embrace of illiberal politics. While acknowledging the draw of Hungary’s illiberal policies as the motivating factor for American conservative interest in Hungary, our focus seeks to answer to what extent this embrace of Hungary can be considered a form of public diplomacy. We examined the frequency and substance of mentions of Hungary and Viktor Orbán in 1643 articles within 13 American conservative media outlets to track how the narrative around the country and the prime minister has evolved over the past four years, bearing in mind the impact of Tucker Carlson’s interview with Viktor Orbán in late 2021. We found both an increase in the quantity of articles focused on Hungary and Viktor Orbán as well as a largely positive trend defending and praising the policies of Hungary and the prime minister. We also observed a strong focus on Orbán as the primary actor of Hungarian public diplomacy and argue that this hyper-presidentialized focus exemplifies the deinstitutionalization of public diplomacy, along with other elements that contribute to the enhancement of Orbán as an individual public diplomacy actor

The guanine nucleotide exchange factor RIC8 regulates conidial germination through Gα proteins in Neurospora crassa.

Heterotrimeric G protein signaling is essential for normal hyphal growth in the filamentous fungus Neurospora crassa. We have previously demonstrated that the non-receptor guanine nucleotide exchange factor RIC8 acts upstream of the Gα proteins GNA-1 and GNA-3 to regulate hyphal extension. Here we demonstrate that regulation of hyphal extension results at least in part, from an important role in control of asexual spore (conidia) germination. Loss of GNA-3 leads to a drastic reduction in conidial germination, which is exacerbated in the absence of GNA-1. Mutation of RIC8 leads to a reduction in germination similar to that in the Δgna-1, Δgna-3 double mutant, suggesting that RIC8 regulates conidial germination through both GNA-1 and GNA-3. Support for a more significant role for GNA-3 is indicated by the observation that expression of a GTPase-deficient, constitutively active gna-3 allele in the Δric8 mutant leads to a significant increase in conidial germination. Localization of the three Gα proteins during conidial germination was probed through analysis of cells expressing fluorescently tagged proteins. Functional TagRFP fusions of each of the three Gα subunits were constructed through insertion of TagRFP in a conserved loop region of the Gα subunits. The results demonstrated that GNA-1 localizes to the plasma membrane and vacuoles, and also to septa throughout conidial germination. GNA-2 and GNA-3 localize to both the plasma membrane and vacuoles during early germination, but are then found in intracellular vacuoles later during hyphal outgrowth

Molecular Analysis of TTF-1 and TTF-2 Genes in Patients with Early Onset Papillary Thyroid Carcinoma

Two common variants, close from TTF-1 and TTF-2, were shown to predispose to thyroid cancer (TC) in European populations. We aimed to investigate whether TTF-1 and TTF-2 variants might contribute to TC early onset (EO). Tumor samples from eighteen patients with papillary TC (PTC), who underwent total thyroidectomy at an age of ≤21, were screened for TTF-1 and TTF-2 variants. No TTF-1 variants were documented; two novel germinal TTF-2 variants, c.200C>G (p.A67G) and c.510C>A (p.A170A), were identified in two patients. Two already described TTF-2 variants were also documented; the allelic frequency among patients was not different from that observed among controls. Moreover, RET/PTC rearrangements and the BRAFV600E mutation were identified in 5/18 and 2/18 PTCs, respectively. Thyroglobulin (TG) and thyroid peroxidase (TPO) expression was found to be significantly decreased in tumors, and the lowest level of TPO expression occurred in a tumor harboring both the p.A67GTTF-2 variant and a RET/PTC3 rearrangement

Instituto de Investigación Médica Mercedes y Martín Ferreyra; Argentina.Although sex hormones are usually considered the main architects of sexual dimorphisms, recent studies have demonstrated that sex chromosomes can also induce sex differences in somatic gene expression in the absence of hormonal differences. Ngn3 is a Notch regulated gene that, in developing neurons, is involved in neurite extension and remodeling. Previous results showed that hypothalamic neurons carrying the XX sex chromosomes present a higher expression of Ngn3 and a faster rate of development than XY neurons, irrespectively of gonadal hormones. Using the Four Core Genotypes (FCG) mouse model, here we analyzed the expression of X-linked genes involved in neuronal growth and differentiation which are probable candidates to regulate Ngn3 expression. By qPCR, we have evaluated the expression of Ddx3x, Eif2s3x, Kdm6a, Syp, Mecp2 and Usp9x in primary hypothalamic cultures from E15 FCG mice. Ddx3x, Eif2s3x and Kdm6a showed higher expression levels in XX neurons than in XY neurons, regardless of the embryo sex. Importantly, Kdm6a is an epigenetic regulator codifying for a histone demethylase, whereas Ddx3x and Eif2s3x codify translation regulators. Thereby, it is possible to hypothesize that some of these genes might be regulating Ngn3 expression and neuronal development. Further experiments blocking these X-linked genes are required to determine the effect of this specific down regulation over Ngn3 and neuronal development.Fil: Cabrera Zapata, Lucas Ezequiel. We have previously observed that intracerebroventricular allopregnanolone (ALLO) injection produced an anxiolytic effect and inhibited sexual receptivity when the test was performed in a separate manner. Also, ALLO reverts learning deficit in female rats in the hippocampi. To study the behavioral effects of an acute treatment with ALLO in the right lateral ventricle we used two approaches: a- A battery test to analyze the anxiety and mating behavior. And b- The avoidance test and novel object recognition test to evaluate its effect on memory and learning. Ovariectomized rats were injected with estrogen and progesterone. After it ALLO or vehicle were administered into the right lateral ventricle. To reach the objective (a) rats were put in a sequential battery test in the next order: 1-Open field. 2- Plus maze task. 3- Mating behavior. For the aim (b) it was performed a Novel Object Recognition Test and Step-down Inhibitory Avoidance Task. ALLO did not affect locomotors-exploratory behavior. Animals treated with ALLO, spent more time and had more entries into the open arm in a plus maze task and lordosis quotient was lower than in the control group. ALLO increased the latency in step down test and had no effects on discrimination index test in NORT. Here we demonstrated that one pharmacological dose of ALLO in ovariectomized primed rats is enough to generate all changes observed in the battery test. Moreover, the acute treatment with ALLO in lateral ventricle enhanced the memory acquisition in an avoidance task.Fil: Pelegrina, Laura Tatiana.
