    Control of assembly of extra-axonemal structures: The paraflagellar rod of trypanosomes

    Eukaryotic flagella are complex microtubule based organelles and in many organisms there are extra axonemal structures present, including the outer dense fibres of mammalian sperm and the paraflagellar rod (PFR) of trypanosomes. Flagellum assembly is a complex process occurring across three main compartments, the cytoplasm, the transition fibre-transition zone, and the flagellum. It begins with translation of protein components, followed by their sorting and trafficking into the flagellum, transport to the assembly site and then incorporation. Flagella are formed from over 500 proteins; the principles governing axonemal component assembly are relatively clear. However, the coordination and sites of extra-axonemal structure assembly processes are less clear. We have discovered two cytoplasmic proteins in T. brucei that are required for PFR formation, PFR assembly factors 1 and 2. Deletion of either PFR-AF1 or PFR-AF2 dramatically disrupted PFR formation and caused a reduction in the amount of major PFR proteins. The presence of cytoplasmic factors required for PFR formation aligns with the concept of processes occurring across multiple compartments to facilitate axoneme assembly and this is likely a common theme for extra-axonemal structure assembly

    Ricci-flat deformation of orbifolds and localized tachyonic modes

    We study Ricci-flat deformations of orbifolds in type II theory. We obtain a simple formula for mass corrections to the twisted modes due to the deformations, and apply it to originally tachyonic and massless states in several examples. In the case of supersymmetric orbifolds, we find that tachyonic states appear when the deformation breaks all the supersymmetries. We also study nonsupersymmetric orbifolds C^2/Z_{2N(2N+1)}, which is T-dual to N type 0 NS5-branes. For N>=2, we compute mass corrections for states, which have string scale tachyonic masses. We find that the corrected masses coincide to ones obtained by solving the wave equation for the tachyon field in the smeared type 0 NS5-brane background geometry. For N=1, we show that the unstable mode representing the bubble creation is the unique tachyonic mode.Comment: 20 pages, minor collection

    Structure-antifouling activity relationship and molecular targets of bio-inspired(Thio)xanthones

    The development of alternative ecological and effective antifouling technologies is still challenging. Synthesis of nature-inspired compounds has been exploited, given the potential to assure commercial supplies of potential ecofriendly antifouling agents. In this direction, the antifouling activity of a series of nineteen synthetic small molecules, with chemical similarities with natural products, were exploited in this work. Six (4, 5, 7, 10, 15 and 17) of the tested xanthones showed in vivo activity toward the settlement of Mytilus galloprovincialis larvae (EC50: 3.53–28.60 µM) and low toxicity to this macrofouling species (LC50 > 500 µM and LC50/EC50: 17.42–141.64), and two of them (7 and 10) showed no general marine ecotoxicity (<10% of Artemia salina mortality) after 48 h of exposure. Regarding the mechanism of action in mussel larvae, the best performance compounds 4 and 5 might be acting by the inhibition of acetylcholinesterase activity (in vitro and in silico studies), while 7 and 10 showed specific targets (proteomic studies) directly related with the mussel adhesive structure (byssal threads), given by the alterations in the expression of Mytilus collagen proteins (PreCols) and proximal thread proteins (TMPs).     A comparative study of the dynamic critical behavior of the four-state Potts like models

    We investigate the short-time critical dynamics of the Baxter-Wu (BW) and n=3n=3 Turban (3TU) models to estimate their global persistence exponent θg\theta _{g}. We conclude that this new dynamical exponent can be useful in detecting differences between the critical behavior of these models which are very difficult to obtain in usual simulations. In addition, we estimate again the dynamical exponents of the four-state Potts (FSP) model in order to compare them with results previously obtained for the BW and 3TU models and to decide between two sets of estimates presented in the current literature. We also revisit the short-time dynamics of the 3TU model in order to check if, as already found for the FSP model, the anomalous dimension of the initial magnetization x0x_{0} could be equal to zero

    Biochemical parameters of silver catfish (Rhamdia quelen) after transport with eugenol or essential oil of Lippia alba added to the water

    The transport of live fish is a routine practice in aquaculture and constitutes a considerable source of stress to the animals. The addition of anesthetic to the water used for fish transport can prevent or mitigate the deleterious effects of transport stress. This study investigated the effects of the addition of eugenol (EUG) (1.5 or 3.0 mu L L-1) and essential oil of Lippia alba (EOL) (10 or 20 mu L L-1) on metabolic parameters (glycogen, lactate and total protein levels) in liver and muscle, acetylcholinesterase activity (AChE) in muscle and brain, and the levels of protein carbonyl (PC), thiobarbituric acid reactive substances (TBARS) and nonprotein thiol groups (NPSH) and activity of glutathione-S-transferase in the liver of silver catfish (Rhamdia quelen; Quoy and Gaimard, 1824) transported for four hours in plastic bags (loading density of 169.2 g L-1). The addition of various concentrations of EUG (1.5 or 3.0 mu L L-1) and EOL (10 or 20 mu L L-1) to the transport water is advisable for the transportation of silver catfish, since both concentrations of these substances increased the levels of NPSH antioxidant and decreased the TBARS levels in the liver.     Closed String Tachyon Condensation at c=1

    The c=1 matrix model, with or without a type 0 hat, has an exact quantum solution corresponding to closed string tachyon condensation along a null surface. The condensation occurs, and spacetime dissolves, at a finite retarded time on I^+. The outgoing quantum state of tachyon fluctuations in this time-dependent background is computed using both the collective field and exact fermion pictures. Perturbative particle production induced by the moving tachyon wall is shown to be similar to that induced by a soft moving mirror. Hence, despite the fact that I^+ for the tachyon is geodesicaly incomplete, quantum correlations in the incoming state are unitarily transmitted to the outgoing state in perturbation theory. It is also shown that, non-perturbatively, information can leak across the tachyon wall, and tachyon scattering is not unitary.     Off-Shell Interactions for Closed-String Tachyons

    Off-shell interactions for localized closed-string tachyons in C/Z_N superstring backgrounds are analyzed and a conjecture for the effective height of the tachyon potential is elaborated. At large N, some of the relevant tachyons are nearly massless and their interactions can be deduced from the S-matrix. The cubic interactions between these tachyons and the massless fields are computed in a closed form using orbifold CFT techniques. The cubic interaction between nearly-massless tachyons with different charges is shown to vanish and thus condensation of one tachyon does not source the others. It is shown that to leading order in N, the quartic contact interaction vanishes and the massless exchanges completely account for the four point scattering amplitude. This indicates that it is necessary to go beyond quartic interactions or to include other fields to test the conjecture for the height of the tachyon potential.Comment: 37 pages, 3 figures, LaTeX, JHEP class.
