Condition of Brazilian sardine, Sardinella brasiliensis (Steindacher, 1879) larvae in the Sao Sebastiao inner and middle continental shelf (Sao Paulo, Brazil)
Growth and mortality estimates of Sardinella brasiliensis in the southeastern Brazilian Bight
Length frequency analisys were applied for sardine data collected from commercial catches throughout the southeastern Brazilian coast in 1977-1987. Age/length keys were obtained by otolith ring countings and utilized to transform length into age composition. Growth parameters were estimated by the von Bertalanffy growth equation using age and length data for each year and for all the period of investigation. The mean growth parameters for the entire period were estimated as L∞ = 271 mm and K= 0.59 year-1. Instantaneous total mortality (Z) coefficients per year were estimated using catch curves and methods based on the mean length of the fish caught. Total mortality rate for the entire period was obtained through the average of the annual values. Natural mortality (M) was estimated using the Paul/s empirical equation (1980a), and fishing mortality (F) by the difference between total and natural mortality values.The results were Z = 3.6 year-1; M = 0.96 year-1; and F= 2.6 year-1.Estudo sobre o crescimento e a mortalidade de Sardinella brasiliensis, da costa sudeste do Brasil, foi realizado para o período 1977 a 1987. As análises foram efetuadas a partir de dados existentes sobre distribuições de freqüência de comprimento de amostras da captura comercial. Chaves idade/comprimento, construidas a partir de leitura de anéis de crescimento em otólitos, dentro do Programa Integrado de Estudos Biológicos sobre a Sardinha - PIEBS, foram utilizadas para transformar as distribuiçes de comprimento em idade. Os parâmetros de crescimento da equação de von Bertalanffy foram estimados, anualmente e para o período como um todo, a partir de métodos que utilizam dados de idade e comprimento. Os parâmetros médios encontrados para o período foram: L∞ = 271 mm e K= 0,59 ano-1. Os coeficientes instantâneos de mortalidade total (Z) anuais foram calculados pelas curvas de captura e por métodos baseados no comprimento médio dos indivíduos nas capturas. A mortalidade total para todo o período foi obtida pela média dos valores anuais. O coeficiente instantâneo de mortalidade natural (M) foi estimado pela equação empírica de Pauly (1980a), usando um fator de correção de 0,8 para clupeoides, e o coeficiente instantâneo de mortalidade por pesca (F), pela diferença entre a mortalidade total e a natural. Os valores encontrados foram: Z= 3,6 ano-1, M = 0,96 ano-1 e F= 2,6 ano-1
Enterohemorrhagic Escherichia coli O157: H7 from healthy dairy cattle in Mid-West Brazil: occurrence and molecular characterization
Enterohemorrhagic Escherichia coli (EHEC) serotype O157:H7 represents the major Shiga toxin-producing E. coli (STEC) strain related to large outbreaks and severe diseases such as hemorrhagic colitis (HC) and the potentially lethal hemolytic uremic syndrome (HUS). The aim of this study was to report the occurrence and molecular characterization of O157:H7 isolates obtained by rectal swab from 52 healthy dairy cattle belonging to 21 farms in Mid-West of Brazil. Detection of 16SrRNA, stx1, stx2, rfbO157, fliCh7, eae, ehxA, saa, cnf1, chuA, yjaA and TSPE4.C2 genes was performed by PCR. The isolates were further characterized by serotyping. Two hundred and sixty E. coli isolates were obtained, of which 126 were characterized as STEC. Two isolates from the same cow were identified as serotype O157:H7. Both isolates presented the stx2, eae, ehxA, saa and cnf1 virulence factor genes and the chuA gene in the phylogenetic classification (virulent group D), suggesting that they were clones. The prevalence of O157:H7 was found to be 1.92% (1/52 animals), demonstrating that healthy dairy cattle from farms in the Mid-West of Brazil are an important reservoir for highly pathogenic E. coli O157:H7
Atlas of marine bony fish otoliths (Sagittae) of Southeastern - Southern Brazil Part I: Gadiformes (Macrouridae, Moridae, Bregmacerotidae, Phycidae and Merlucciidae); Part II: Perciformes (Carangidae, Sciaenidae, Scombridae and Serranidae)
The drawings, detailed pictures, precise descriptions and measurements that characterize otoliths must be made available for studies in various areas, including taxonomy, phylogeny, ecology, fisheries, paleontology, diversity, predator-prey relationships and modeling. The Collection of Teleostei Fish Otoliths of Southeastern-Southern Brazil (COSS-Brasil) of IOUSP contains 45,000 pairs of otoliths from 210 species. This publication is the first in a series that will constitute an atlas of Teleostei otoliths for southeastern-southern Brazil and presents the results of the morphologic and morphometric analyses of 11 Gadiformes and 36 Perciformes species by means of the most commonly used features, measurements and indices. Three otoliths of each species were illustrated and photographed whenever possible. The frequency of occurrence was calculated for each characteristic by total length classes (TL), and the ontogenetic differences were analyzed (multiple χ2 test; significance 0.05). Morphometric analyses were conducted for each characteristic per total length (TL) class and for the whole sample, and the ontogenetic differences were analyzed
Detection of multidrug-resistant tuberculosis from stored DNA samples : a multicenter study
Background: In low-income countries, rapid detection of tuberculosis (TB) drug resistance is often restricted by the difficulties of transporting and storing sputum samples from remote health centers to the reference laboratories where molecular tests are available. The aim of this study was to evaluate the performance of four transport and storage systems for molecular detection of rifampicin (RIF) and isoniazid (INH) resistance. Methods: This was a multicenter study. Molecular detection of RIF and INH resistance was performed directly from smear-positive TB sputa spotted on a slide, FTA card, GenoCard, and ethanol using the Genotype MTBDRplus assay. The performance of the DNA extraction method from each storage support to detect drug resistance was assessed by calculating their sensitivity and specificity compared to the phenotypic method. Results: From all sites, the overall sensitivity and specificity for RIF-resistance detection was 88% and 85%, respectively, for slides, 86% and 92%, respectively, for GenoCard, 87% and 89%, respectively, for FTA card, and 88% and 92%, respectively, for ethanol. For INH-resistance detection, the overall sensitivity and specificity was 82% and 90%, respectively, for slides, 85% and 96%, respectively, for GenoCard, 86% and 92%, respectively, for FTA card, and 86% and 94%, respectively, for ethanol. Conclusion: Smear slides and filter cards showed to be very useful tools to facilitate DNA extraction from sputum samples with the potential to accelerate the detection of drug resistance in remote areas
Distribution of the pilS gene in Escherichia coli pathovars, its transfer ability and influence in the typical enteropathogenic E. coli adherence phenotype
Typical enteropathogenic Escherichia coli strains (tEPEC) cause attaching/effacing lesions in eukaryotic cells and produce the bundle-forming pilus (BFP), which interweaves and aggregates bacteria, resulting in the localized adherence (LA) pattern on eukaryotic cells. Previously, we identified tEPEC strains (serotype O119:H6) that exhibited LA simultaneously with an aggregative adherence (AA)-like pattern (LA/AA-like+). Remarkably, AA is characteristically produced by strains of enteroaggregative E. coli (EAEC), another diarrheagenic E. coli pathovar. In one LA/AA-like + strain (Ec404/03), we identified a conjugative plasmid containing the pil operon, which encodes the Pil fimbriae. Moreover, a pil operon associated with an AA pattern and plasmid transfer had been previously described in the EAEC C1096 strain. In this study, we investigated the occurrence of the two pilS alleles (pilSEc404 and pilSC1096) in tEPEC strains of different serotypes, origins and years of isolation. We also examined the potential relationship of pilS with the AA-like phenotype, its ability to be transferred by conjugation, and occurrence among strains of the other E. coli pathovars. The pilS alleles were found in 90 (55.2%) of 163 tEPEC strains, with pilSEc404 occurring more often (30.7%) than pilSC1096 (25.1%). About 21 tEPEC serotypes carried pilS. The pilS alleles were found in tEPEC strains from Chile, Peru and different Brazilian cities, with the oldest strain being isolated in 1966. No absolute correlation was found between the presence of pilS and the AA-like pattern. Conjugative pilS transfer was detected in 26.2% of pilSEc404+ strains and in 65.1% of pilSC1096+ strains, but only pilSEc404+ transconjugants were AA-like+, thus suggesting that the latter allele might need a different genetic background to express this phenotype. pilS was found in all other E. coli pathovars, where it was most prevalent in enterotoxigenic E. coli. Diarrhoeagenic Escherichia coli and Escherichia albertii in Brazil: pathotypes and serotypes over a 6-year period of surveillance
Diarrhoeagenic Escherichia coli (DEC) is a leading cause of infectious diarrhoea worldwide. In recent years, Escherichia albertii has also been implicated as a cause of human enteric diseases. This study describes the occurrence of E. coli pathotypes and serotypes associated with enteric illness and haemolytic uremic syndrome (HUS) isolated in Brazil from 2011 to 2016. Pathotypes isolated included enteropathogenic E. coli (EPEC), enteroaggregative E. coli (EAEC), enterotoxigenic E. coli (ETEC), enteroinvasive E. coli (EIEC) and Shiga toxin-producing E. coli (STEC). PCR of stool enrichments for DEC pathotypes was employed, and E. albertii was also sought. O:H serotyping was performed on all DEC isolates. A total of 683 DEC and 10 E. albertii strains were isolated from 5047 clinical samples. The frequencies of DEC pathotypes were 52.6% (359/683) for EPEC, 325% for EAEC, 6.3% for ETEC, 4.4% for ETEC and 4.2% for STEC. DEC strains occurred in patients from 3 months to 96 years old, but EPEC, EAEC and STEC were most prevalent among children. Both typical and atypical isolates of EPEC and EAEC were recovered and presented great serotype heterogeneity. HUS cases were only associated with STEC serotype O157:H7. Two E. albertii isolates belonged to serogroup O113 and one had the stx2f gene. The higher prevalence of atypical EPEC in relation to EAEC in community-acquired diarrhoea in Brazil suggests a shift in the trend of DEC pathotypes circulation as previously EAEC predominated. This is the first report of E. albertii isolation from active surveillance.
