    Assessing methods to improve benthic fish sampling in a stony headwater stream

    Electrofishing is a well-established and widely used method for surveying fish populations. Nonetheless, its effectiveness is impacted by numerous factors, including water chemistry, habitat type and fish species. Both physiological and behavioural responses make bottom-dwelling ā€˜benthicā€™ fish which lack swim bladders (e.g. European bullhead Cottus gobio) particularly difficult to survey by electrofishing. We compare the performance and practicalities of electrofishing for benthic fish at a rocky northern English headwater stream with two sampling methods originally designed for crayfish surveys; the triple drawdown method which involves repeated dewatering of a site, and the Pritchard Trap method which involves sunken traps filled with natural substrate that samples a small, fixed (0.25 m2) area of river bed. Both the Pritchard trapping and triple drawdown methods provided similar high-density population density estimates for bullhead which were at least 2.5ā€“5 times higher than predicted from electrofishing derived sweep depletion curves. Electrofishing and the triple drawdown method are both resource-intensive, requiring expensive equipment and a team of trained operatives. These approaches also pose a risk to fish and non-target organisms. In contrast, Pritchard Traps provide a cost-effective passive, low risk survey method requiring minimal training and only one operative. Pritchard traps, therefore, show particular promise for benthic fish surveying and monitoring

    Ketamine coadministration attenuates morphine tolerance and leads to increased brain concentrations of both drugs in the rat

    Background and Purpose The effects of ketamine in attenuating morphine tolerance have been suggested to result from a pharmacodynamic interaction. We studied whether ketamine might increase brain morphine concentrations in acute coadministration, in morphine tolerance and morphine withdrawal. Experimental Approach Morphine minipumps (6mg center dot day(-1)) induced tolerance during 5 days in Sprague-Dawley rats, after which s.c. ketamine (10mg center dot kg(-1)) was administered. Tail flick, hot plate and rotarod tests were used for behavioural testing. Serum levels and whole tissue brain and liver concentrations of morphine, morphine-3-glucuronide, ketamine and norketamine were measured using HPLC-tandem mass spectrometry. Key Results In morphine-naive rats, ketamine caused no antinociception whereas in morphine-tolerant rats there was significant antinociception (57% maximum possible effect in the tail flick test 90min after administration) lasting up to 150min. In the brain of morphine-tolerant ketamine-treated rats, the morphine, ketamine and norketamine concentrations were 2.1-, 1.4- and 3.4-fold, respectively, compared with the rats treated with morphine or ketamine only. In the liver of morphine-tolerant ketamine-treated rats, ketamine concentration was sixfold compared with morphine-naive rats. After a 2 day morphine withdrawal period, smaller but parallel concentration changes were observed. In acute coadministration, ketamine increased the brain morphine concentration by 20%, but no increase in ketamine concentrations or increased antinociception was observed. Conclusions and Implications The ability of ketamine to induce antinociception in rats made tolerant to morphine may also be due to increased brain concentrations of morphine, ketamine and norketamine. The relevance of these findings needs to be assessed in humans.Peer reviewe

    Non-Fermi liquid behavior below the NĆ©el temperature in the frustrated heavy fermion magnet UAuā‚‚

    The term Fermi liquid is almost synonymous with the metallic state. The association is known to break down at quantum critical points (QCPs), but these require precise values of tuning parameters, such as pressure and applied magnetic field, to exactly suppress a continuous phase transition temperature to the absolute zero. Three-dimensional non-Fermi liquid states, apart from superconductivity, that are unshackled from a QCP are much rarer and are not currently well understood. Here, we report that the triangular lattice system uranium diauride (UAu2) forms such a state with a non-Fermi liquid low-temperature heat capacity [Formula: see text] and electrical resistivity [Formula: see text] far below its NĆ©el temperature. The magnetic order itself has a novel structure and is accompanied by weak charge modulation that is not simply due to magnetostriction. The charge modulation continues to grow in amplitude with decreasing temperature, suggesting that charge degrees of freedom play an important role in the non-Fermi liquid behavior. In contrast with QCPs, the heat capacity and resistivity we find are unusually resilient in magnetic field. Our results suggest that a combination of magnetic frustration and Kondo physics may result in the emergence of this novel state

    The Square Kilometre Array (SKA) will offer an unprecedented view onto the early Universe, using interferometric observations of the redshifted 21cm line. The 21cm line probes the thermal and ionization state of the cosmic gas, which is governed by the birth and evolution of the first structures in our Universe. Here we show how the evolution of the 21cm signal will allow us to study when the first generations of galaxies appeared, what were their properties, and what was the structure of the intergalactic medium. We highlight qualitative trends which will offer robust insights into the early Universe

    Preliminary genetic evidence of two different populations of Opisthorchis viverrini in Lao PDR

    Opisthorchis viverrini is a major public health concern in Southeast Asia. Various reports have suggested that this parasite may represent a species complex, with genetic structure in the region perhaps being dictated by geographical factors and different species of intermediate hosts. We used four microsatellite loci to analyze O. viverrini adult worms originating from six species of cyprinid fish in Thailand and Lao PDR. Two distinct O. viverrini populations were observed. In Ban Phai, Thailand, only one subgroup occurred, hosted by two different fish species. Both subgroups occurred in fish from That Luang, Lao PDR, but were represented to very different degrees among the fish hosts there. Our data suggest that, although geographical separation is more important than fish host specificity in influencing genetic structure, it is possible that two species of Opisthorchis, with little interbreeding, are present near Vientiane in Lao PDR

    Feedback driven by enzyme catalyzed reactions occurs widely in biology and has been well characterized in single celled organisms such as yeast. There are still few examples of robust enzyme oscillators in vitro that might be used to study nonlinear dynamical behavior. One of the simplest is the ureaā€“urease reaction that displays autocatalysis driven by the increase in pH accompanying the production of ammonia. A clock reaction was obtained from low to high pH in batch reactor and bistability and oscillations were reported in a continuous flow rector. However, the oscillations were found to be irreproducible and one contributing factor may be the lack of stability of the enzyme in solution at room temperature. Here, we investigated the effect of immobilizing urease in thiol-poly(ethylene glycol) acrylate (PEGDA) hydrogel beads, prepared using emulsion polymerization, on the ureaā€“urease reaction. The resultant mm-sized beads were found to reproduce the pH clock and, under the conditions employed here, the stability of the enzyme was increased from hours to days

    Monitoring of urothelial cancer disease status after treatment by digital droplet PCR liquid biopsy assays

    Objectives Real-time monitoring of disease status would be beneficial for timely decision making in the treatment of urothelial cancer (UC), and may accelerate the evaluation of clinical trials. Use of cell free tumor DNA (cftDNA) as a biomarker in liquid biopsy is minimally invasive and its successful use has been reported in various cancer types, including UC. The objective of this study was to evaluate the use of digital droplet PCR (ddPCR)-based assays to monitor UC after treatment. Method and materials Blood, urine and matching formalin fixed, paraffin embedded diagnostic specimens were collected from 20 patients diagnosed with stage T1 (nā€Æ=ā€Æ2) and T2/T3 (nā€Æ=ā€Æ18) disease. SNaPshot assays, Sanger sequencing and whole exome sequencing were used to identify tumor-specific mutations, and somatic mutation status was confirmed using patient-matched DNAs extracted from buffy coats and peripheral blood mononucleocytes. The ddPCR assays of the tumor-specific mutations were used to detect the fractional abundance of cftDNA in plasma and urine. Results SNaPshot and Sanger sequencing identified point mutations in 70% of the patients that were assayable by ddPCR. Cases of remission and relapse monitored by assays for PIK3CA E542K and TP53 Y163C mutations in plasma and urine concurred with clinical observations up to 48 months from the start of chemotherapy. A new ddPCR assay for the telomerase reverse transcriptase (TERT) promoter (-124) mutation was developed. The TERT assay was able to detect mutations in cases below the limit of detection by SNaPshot. Whole exome sequencing identified a novel mutation, CNTNAP4 G727*. A ddPCR assay designed to detect this mutation was able to distinguish mutant from wild-type alleles. Conclusions The study demonstrated that ddPCR assays could be used to detect cftDNA in liquid biopsy monitoring of the post-therapy disease status in patients with UC. Overall, 70% of the patients in our study harbored mutations that were assayable by ddPCR

    Relationship of anxiety with joint pain and its management: A population survey

    Objectives The aims of the present study was to examine the associations between the severity of pain and anxiety in a community population reporting joint pain, and to investigate the management of joint pain in the presence of comorbid anxiety. Methods A population survey was carried out of people aged ā‰„45 years, registered with eight general practices in Cheshire, Shropshire and Staffordshire, UK. Respondents were asked to report pain intensity in their hands, hips, knees and feet (on a numerical rating scale), anxiety symptoms (Generalized Anxiety Disorder sevenā€item [GADā€7] scale) and guidelineā€recommended treatments used to manage pain. Clinical anxiety was defined by a GADā€7 score of 10 or more. Results A total of 11,222 respondents with joint pain were included in the analysis, with 1,802 (16.1%) reporting clinical anxiety. Respondents reporting more severe pain were more likely to report clinical anxiety (severe versus mild pain, odds ratio [OR] 5.36, 95% confidence interval [CI] 4.56 to 6.31). The number of pain sites was also positively associated with clinical anxiety (four versus one site; OR 3.64, 95% CI 3.09 to 4.30). Those with clinical anxiety were less likely to undertake general fitness exercises (OR 0.60, 95% CI 0.52 to 0.70), but more likely to diet (OR 1.43, 95% CI 1.21 to 1.69), use walking aids (OR 1.53, 95% CI 1.32 to 1.77) and assistive devices (OR 1.24, 95% CI 1.04 to 1.49), and more likely to use opioids (OR 1.34, 95% CI 1.18 to1.52). Conclusions Anxiety is common among patients presenting to primary care with joint pain. Patients with anxiety are likely to manage their joint pain differently to those without. Caseā€finding to identify and treat anxiety would be appropriate in this population, with caution about opioid prescribing and consideration of exercise as an intervention

    The periodic topography of ice stream beds: Insights from the Fourier spectra of mega-scale glacial lineations

    Ice stream bed topography contains key evidence for the ways ice streams interact with, and are potentially controlled by, their beds. Here we present the first application of two-dimensional Fourier analysis to 22 marine and terrestrial topographies from 5 regions in Antarctica and Canada, with and without mega-scale glacial lineations (MSGLs). We find that the topography of MSGL-rich ice stream sedimentary beds is characterized by multiple, periodic wavelengths between 300 and 1200 m and amplitudes from decimeters to a few meters. This periodic topography is consistent with the idea that instability is a key element to the formation of MSGL bedforms. Dominant wavelengths vary among locations and, on one paleo ice stream bed, increase along the direction of ice flow by 1.7 Ā± 0.52% kmāˆ’1. We suggest that these changes are likely to reflect pattern evolution via downstream wavelength coarsening, even under potentially steady ice stream geometry and flow conditions. The amplitude of MSGLs is smaller than that of other fluvial and glacial topographies but within the same order of magnitude. However, MSGLs are a striking component of ice stream beds because the topographic amplitude of features not aligned with ice flow is reduced by an order of magnitude relative to those oriented with the flow direction. This study represents the first attempt to automatically derive the spectral signatures of MSGLs. It highlights the plausibility of identifying these landform assemblages using automated techniques and provides a benchmark for numerical models of ice stream flow and subglacial landscape evolution.The research was funded by the NE/J004766/1 UK NERC New Investigator and a SAGES 728 PECRE grants awarded to MS

    Fortnightly changes in water transport direction across the mouth of a narrow estuary

    This research investigates the dynamics of the axial tidal flow and residual circulation at the lower Guadiana Estuary, south Portugal, a narrow mesotidal estuary with low freshwater inputs. Current data were collected near the deepest part of the channel for 21 months and across the channel during two (spring and neap) tidal cycles. Results indicate that at the deep channel, depth-averaged currents are stronger and longer during the ebb at spring and during the flood at neap, resulting in opposite water transport directions at a fortnightly time scale. The net water transport across the entire channel is up-estuary at spring and down-estuary at neap, i.e., opposite to the one at the deep channel. At spring tide, when the estuary is considered to be well mixed, the observed pattern of circulation (outflow in the deep channel, inflow over the shoals) results from the combination of the Stokes transport and compensating return flow, which varies laterally with the bathymetry. At neap tide (in particular for those of lowest amplitude each month), inflows at the deep channel are consistently associated with the development of gravitational circulation. Comparisons with previous studies suggest that the baroclinic pressure gradient (rather than internal tidal asymmetries) is the main driver of the residual water transport. Our observations also indicate that the flushing out of the water accumulated up-estuary (at spring) may also produce strong unidirectional barotropic outflow across the entire channel around neap tide.info:eu-repo/semantics/publishedVersio
