    Liver-specific deletion of the Plpp3 gene alters plasma lipid composition and worsens atherosclerosis in apoE -/- mice

    The PLPP3 gene encodes for a ubiquitous enzyme that dephosphorylates several lipid substrates. Genome-wide association studies identified PLPP3 as a gene that plays a role in coronary artery disease susceptibility. The aim of the study was to investigate the effect of Plpp3 deletion on atherosclerosis development in mice. Because the constitutive deletion of Plpp3 in mice is lethal, conditional Plpp3 hepatocyte-specific null mice were generated by crossing floxed Plpp3 mice with animals expressing Cre recombinase under control of the albumin promoter. The mice were crossed onto the athero-prone apoE -/- background to obtain Plpp3 f/f apoE -/- Alb-Cre + and Plpp3 f/f apoE -/- Alb-Cre - offspring, the latter of which were used as controls. The mice were fed chow or a Western diet for 32 or 12 weeks, respectively. On the Western diet, Alb-Cre + mice developed more atherosclerosis than Alb-Cre - mice, both at the aortic sinus and aorta. Lipidomic analysis showed that hepatic Plpp3 deletion significantly modified the levels of several plasma lipids involved in atherosclerosis, including lactosylceramides, lysophosphatidic acids, and lysophosphatidylinositols. In conclusion, Plpp3 ablation in mice worsened atherosclerosis development. Lipidomic analysis suggested that the hepatic Plpp3 deletion may promote atherosclerosis by increasing plasma levels of several low-abundant pro-atherogenic lipids, thus providing a molecular basis for the observed results

    Avaliação da carcaça de cabritos Saanen alimentados com dietas com grãos de oleaginosas

    Foram utilizados 16 cabritos Saanen não castrados dos 15,16±1,60kg até 30,61±0,33kg de peso vivo, distribuídos em delineamento inteiramente ao acaso, alimentados com dietas (tratamentos) com grãos de linhaça, girassol ou canola. A dieta-controle foi formulada com feno de aveia, farelo de soja, milho moído e suplemento vitamínico-mineral. Os pesos da carcaça quente e fria e os rendimentos da carcaça e de cortes comerciais não foram modificados pelos tratamentos. A dieta com grãos de canola resultou em maior perda por resfriamento da carcaça e menor proporção de gordura no lombo, e a proporção de osso no lombo foi maior para os animais que receberam linhaça ou canola. Os teores de umidade, proteína, lipídios totais e cinzas do músculo Longissimus dorsi não foram alterados com a adição dos grãos de oleaginosas. A dieta com grãos de linhaça resultou em aumento do teor de ômega-3 no músculo, e o teor de colesterol e a razão ômega-6/ômega-3 foram menores nos tratamentos que continham linhaça, girassol e canola. A inclusão dos grãos de oleaginosas nas dietas não alterou as características quantitativas da carcaça de cabritos Saanen, mas melhorou a qualidade da carne, que apresentou menos colesterol e mais ômega-3, com o uso de linhaça

    DLG4-related synaptopathy: a new rare brain disorder

    PURPOSE: Postsynaptic density protein-95 (PSD-95), encoded by DLG4, regulates excitatory synaptic function in the brain. Here we present the clinical and genetic features of 53 patients (42 previously unpublished) with DLG4 variants.METHODS: The clinical and genetic information were collected through GeneMatcher collaboration. All the individuals were investigated by local clinicians and the gene variants were identified by clinical exome/genome sequencing.RESULTS: The clinical picture was predominated by early onset global developmental delay, intellectual disability, autism spectrum disorder, and attention deficit-hyperactivity disorder, all of which point to a brain disorder. Marfanoid habitus, which was previously suggested to be a characteristic feature of DLG4-related phenotypes, was found in only nine individuals and despite some overlapping features, a distinct facial dysmorphism could not be established. Of the 45 different DLG4 variants, 39 were predicted to lead to loss of protein function and the majority occurred de novo (four with unknown origin). The six missense variants identified were suggested to lead to structural or functional changes by protein modeling studies.CONCLUSION: The present study shows that clinical manifestations associated with DLG4 overlap with those found in other neurodevelopmental disorders of synaptic dysfunction; thus, we designate this group of disorders as DLG4-related synaptopathy.Genetics of disease, diagnosis and treatmen

    Characterising acute and chronic care needs: insights from the Global Burden of Disease Study 2019

    Chronic care manages long-term, progressive conditions, while acute care addresses short-term conditions. Chronic conditions increasingly strain health systems, which are often unprepared for these demands. This study examines the burden of conditions requiring acute versus chronic care, including sequelae. Conditions and sequelae from the Global Burden of Diseases Study 2019 were classified into acute or chronic care categories. Data were analysed by age, sex, and socio-demographic index, presenting total numbers and contributions to burden metrics such as Disability-Adjusted Life Years (DALYs), Years Lived with Disability (YLD), and Years of Life Lost (YLL). Approximately 68% of DALYs were attributed to chronic care, while 27% were due to acute care. Chronic care needs increased with age, representing 86% of YLDs and 71% of YLLs, and accounting for 93% of YLDs from sequelae. These findings highlight that chronic care needs far exceed acute care needs globally, necessitating health systems to adapt accordingly
